Abstract

African catfish, namely lele dumbo, is an economically important fresh water fish in Nusa Tenggara Barat. There are three strains of African catfish that are well distributed among farmers. Genetic information of three strain catfish stocks is needed in order to sustain their development program. Genetic variations of the three strains of catfish, i.e. Sangkuriang, Masamo, Paiton has been evaluated using RAPD markers. DNA genome was extracted using phenol-chloroform methods. DNA was amplified using 20 primers (OPA 1 – 20). The results showed that three of the 20 primers have good amplification products. There was no significant genetic differences among three strains of catfish analyzed (P>0.05). Genetic variation of three African catfish stocks ranges from low to middle value. The highest genetic variation is found in strain Masamo with heterozygosity of 0.273 (with 70% polymorphism loci), followed by Sangkuriang 0.189 (60%) and Paiton 0.147 (40%). Hybrid candidates of Masamo-Sangkuriang have eterozygosity ranges between 0.256 - 0.306 (with 66.6 7 – 73.77% polymorphism loci). Strain Masamo and Paiton has more close genetic relationship than among the both and Sangkuriang.

Abstract

Ikan kalui merupakan nama lokal dari ikan gurami di Kabupaten Lima Puluh Kota, Sumatera Barat termasuk jenis ikan ekonomis tinggi. Terdapat lima strain ikan kalui yang tersebar di pembudidaya, yaitu Tambago, Palapah, Krista, Jepun, dan Merah. Penelitian ini dilakukan untuk mengamati keragaman genetik strain ikan kalui atau gurami dengan menggunakan penanda RAPD. Sebanyak 50 sampel DNA ikan kalui diekstraksi dari sirip dan diamplifikasi secara random dengan menggunakan empat primer terbaik dari 20 primer OPA yaitu OPA-02, OPA-04, OPA-06, dan OPA-07. Hasil menunjukkan bahwa perbedaan yang nyata secara genetik hanya terdapat antara strain ikan kalui Merah dengan Tambago dan Krista. Variasi genetik tertinggi diamati pada ikan kalui strain Krista dengan nilai heterogenitas 0,1756 kemudian diikuti berturut-turut oleh strain Merah (0,1735); Palapah (0,1480); Jepun (0,0594); dan Tambago (0,0203). Jarak rata-rata Nei genetik adalah 0,407, dengan nilai terendah yang teramati antara strain Tambago dan Palapah.Kalui is a local name of giant gouramy fish in West Sumatera Province that categorized as an high economically fish. Five strains of kalui are distributed to farmers i.e. Tambago, Palapah, Krista, Jepun and Merah. This research was conducted to observe the genetic variation of Kalui strains using RAPD marker. A total of 50 samples of whole DNA was extracted from kalui-giant gouramy finclip and randomly amplified using four of the best 20 primers (OPA i.e. OPA-02, OPA-04, OPA-06 and OPA-07). The results showed that Significant genetic differences were only observed between strain Merah-Tambago and Merah-Krista. The highest variability was observed in Krista with heterogeneity value of 0.1735 followed by Merah (0,1735), Palapah (0,1480), Jepun (0,0594) and Tambago (0,0203). The average Nei genetic distance was 0.407, with the lowest was observed between Tambago and Palapah.

Abstract

Program pemuliaan ikan mas varietas Punten dilakukan dengan seleksi individu terhadap karakter bobot ikan. Pembentukan populasi dasar untuk kegiatan seleksi dilakukan dengan memijahkan secara massal induk ikan mas yang terdiri atas 20 induk betina dan 21 induk jantan yang dikoleksi dari daerah Punten, Kepanjen (delapan betina dan enam jantan), Kediri (tujuh betina dan 12 jantan), Sragen (27 betina dan 10 jantan), dan Blitar (15 betina dan 11 jantan). Larva umur 10 hari dipelihara selama empat bulan. Selanjutnya dilakukan penjarangan sebesar 50% dan benih dipelihara selama 14 bulan untuk dilakukan seleksi dengan panduan hasil sampling 250 ekor individu setiap populasi. Seleksi terhadap calon induk dilakukan saat umur 18 bulan pada populasi jantan dan betina secara terpisah dengan memilih berdasarkan 10% bobot ikan yang terbaik. Calon induk yang terseleksi kemudian dipelihara hingga matang gonad, kemudian dipilih sebanyak 150 pasang dan dipijahkan secara massal. Didapatkan respons positif dari hasil seleksi berdasarkan bobot ikan, yaitu 49,89 g atau 3,66% (populasi ikan jantan) dan 168,47 g atau 11,43% (populasi ikan betina). Nilai heritabilitas untuk bobot ikan adalah 0,238 (jantan) dan 0,505 (betina).Punten carp breeding programs were carried out by individual selection for body weight trait. The base population for selection activities were conducted by mass breeding of parent consisted of 20 female and 21 male collected from area Punten, eight female and six male (Kepanjen), seven female and 12 male (Kediri), 27 female and 10 male (Sragen), 15 female and 11 male (Blitar). Larvae 10 days old reared for four moths. Then after spacing out 50% of total harvest, the offspring reared for 14 months for selection activity based on the sampling of 250 individual each population. Selection of broodstock candidates performed since 18 months age on male and female populations separately by selecting based on 10% of fish with best body weight. Candidates selected broodstocks were then maintained until mature. In oder to produce the next generation 150 pairs were sets and held for mass spawning. The results revealed that selection response were positive, 49.89 g (3.66%) for male and 168.47 (11.43%) for female. Heritability for body weight is 0.238 (male) and 0.505 (female).

Abstract

Penetapan kawasan industri budidaya ikan nila merupakan salah satu sarana untuk mengembangkan industri yang berwawasan lingkungan serta memberikan kemudahan dan daya tarik untuk berinvestasi. Kabupaten Musi Rawas sebagai produsen ikan nila terbesar di Indonesia berpotensi untuk dikembangkan sebagai kawasan sentra industri budidaya ikan nila. Tujuan penelitian ini adalah untuk menentukan kesenjangan kondisi eksisting dan kondisi ideal pada aspek kelayakan umum dan teknis Kabupaten Musi Rawas yang berpotensi untuk dikembangkan sebagai kawasan sentra industri budidaya ikan nila. Diharapkan agar kawasan industri yang dikembangkan sesuai dengan tata ruang, meminimalisasi dampak negatif dan mengembangkan dampak positif terhadap lingkungan hidup, berdaya guna dan berhasil guna, sehingga pada gilirannya mampu menarik peluang investasi bagi pengembangan industri di daerah. Aspek yang dianalisis meliputi aspek kelayakan umum dan kelayakan teknis. Aspek kelayakan umum meliputi: dimensi infrastruktur, masyarakat dan bisnis, sumberdaya, kelembagaan, teknologi, kebijakan, dan pemasaran. Aspek kelayakan teknis meliputi pembenihan, pembesaran di kolam air deras, pembesaran di kolam air tenang, dan pengolahan produk perikanan. Berdasarkan analisis kesenjangan, Kabupaten Musi Rawas termasuk dalam kategori SEDANG, yaitu secara umum dan teknis Kabupaten Musi Rawas memiliki beberapa kriteria sebagai sebuah kawasan sentra industri budidaya ikan nila namun masih diperlukan adanya pengembangan pada beberapa hal seperti: optimalisasi balai benih ikan, penerapan teknologi tepat guna dan peningkatan pemasaran produk hasil perikanan didukung kebijakan yang menfasilitasi kebutuhan akan meningkatkan kelayakan sebagai lokasi sentra industri budidaya ikan nila. Rekomendasi kebijakan yang perlu diambil oleh pemerintah pusat dan daerah untuk pengembangan kawasan sentra industri di Kabupaten Musi Rawas diantaranya melalui optimalisasi peranan balai benih ikan, transfer teknologi tepat guna, diversifikasi produk olahan, penguatan potensi pasar melalui promosi, penguatan kelembagaan produksi, pengolah dan pemasaran hasil perikanan, dan resolusi konflik pemanfaatan dan pengelolaan air irigasi untuk kebutuhan pertanian dan perikanan.Determination of area for Tilapia culture will be meaningful for business development, eco-friendly, sustainability, and attractiveness by the investor. Musi Rawas Regency as the largest tilapia producer in Indonesia is highly potential areas to be developed as a center of tilapia farming industry. The purpose of this study is to determine the general and technical feasibility condition of Musi Rawas Regency which will be developed as a center of tilapia farming industry. It is expected that industrial zones developed in accordance with the spatial plan, will minimize the negative impact to the environment, efficient and effective so that the ability to attract opportunities for industrial development in the region. Aspects analyzed for general eligibility were infrastructure, society and business, resources, institutions, technologies, policies, and market. The aspect of the techniques is the feasibility of hatchery, farmings in flowing water pond, stagnant water ponds, and fish product processing. Based on the gap analysis, Musi Rawas Regency is belonged to the moderate category, in which generally and technically Musi Rawas Regency already has several criteria as a center of tilapia industry but still a need for development on several items such as optimization of the hatchery, application of appropriate technology and enhancement of marketing for fishery products supported by the facilitating policy. Required policy recommendations to be implemented by the central and regional governments for the development of industrial centers in Musi Rawas Regency are optimization of fish seed center, transfer of appropriate technology, diversification of fish products, strengthening of the market potential through promotion, strengthening of production institutions, fish product processing and marketing, and resolution of utilization and management of irrigation water for agricultural and fishery needs.
