Abstract

This research was conducted during July to November 2020. The MDR bacteria were S. aureus, E. coli, and V. harveyi MDR test microbes. The symbiont microbes were identified through molecular analyses (PCR 16S rDNA). Isolation of symbiont microbes from R. mucronata resulted in 16 isolates, while isolation from A. iilicifolius resulted in 14 isolates. Based on the antimicrobial qualitative test against S. aureus, 8 out of 16 microbial isolates from R. mucronata were found to show antimicrobial properties. The testing of A. ilicifolius symbiont microbes against S. aureus showed 8 out of 14 isolates with antimicrobial properties. The test against E. coli resulted in 2 out of 16 microbial isolates from R. mucronata and 5 out of 14 isolates from A. ilicifolius with antimicrobial properties. The test against V. harveyi resulted in two out of 16 microbial isolates from R.mucronata and 4 out of 14 isolates from A. ilicifolius with antimicrobial properties. The quantitative test found 2 isolates from R. mucronta, namely isolates RM10 and RM12, with antimicrobial properties against MDR strain E. coli, with the best isolate being RM10, which produced 11.22 mm of inhibition zone diameter. Furthermore, the selection of isolates was based on the size of the inhibition zone, the clearness of the inhibition zone and the potential for antibacterial activity. Based on their overall antimicrobial potential against the test microbes, four isolates were selected.  Molecular analyses of RM12 isolate showed 95% homology with Bacillus subtilis, of RM 10 isolate showed 97% homology with Bacillus oceanisediminis, of AC isolate showed 96% homology with Paracoccus caeni, and of AC 5 isolate showed 89% homology with Bacillus circulans. The study found four isolates with antimicrobial potency against MDR pathogenic microbes. The symbiont microbes taken from R. mucronata and A. ilicifolius were determined to be of the genus Bacillus and Paracoccus.

Abstract

The light organ is an electronic device that can emit light. However, there are light organs in animals that can produce light naturally. Loligo duvaucelii is a species whose biolumenesence comes from fluorescent bacteria that live in symbiosis in its ink sacs. This study aims to determine in detail the construction of the squid light organ using the transmission electron microscopy (TEM) method. The results showed that this type of squid has a pair of light organs attached to the dorso-lateral ink sac. The light organ is spherical, some are found on the surface and some are embedded on the wall of the ink sac. It consists of a lens that is located on the outer surface of the ink sac, and a sac of light organs (embedded on the wall of the ink sac) with channels connecting the pocket to the mantle cavity. The wall of the sac of the light organ consists of three layers, namely the innermost layer which is multi-fold with microvilli on the cell surface and between the folds of the sac populated with bacteria, the dense layer that acts as a reflector, and the pigment layer. Cilia are observed on the surface of the duct connecting the sac with the mantle cavity. This study concluded that the construction of the squid light organ has a convex-shaped lens structure and is muscular. In the pockets of light organs, a dense population of bacteria is found. The reflector consists of many layers, and the pigment layer contains many granules.

Abstract

Mangroves prove a habitat for Batillaria zonalis as nursery ground, feeding ground and reproductive ground. This research was conducted from September 2016 - August 2017 and to determine the spasial and temporal pattern  with based on mangrove zone  and environmental characteristics. The gastropods and environmental characteristics were analyzed using Principal Component Analysis (PCA). The results showed that significant changes in gastropod assemblages were primarily due to changes in the water quality and season. Correlation between gastropod and physico-chemical parameters in A. marina  revealed significant relationship with gastropod B. zonalis distribution.
