Abstract

Penelitian tentang aktivitas antioksidan, sitotoksisitas, dan GC MS ekstrak metanol alga hijau Boergesenia forbesii (Harvey) Feldmann telah dilaksanakan. Uji aktivitas antioksidan dilakukan dengan metode DPPH (1,1-diphenyl-2-picrylhydrazil). Uji sitotoksisitas terhadap Artemia salina Leach dilakukan dengan metode BSLT (Brine Shrimp Lethality Test). Identifikasi komponen kimia ekstrak dilakukan dengan menggunakan GC MS. Hasil uji menunjukkan bahwa ekstrak metanol alga hijau Boergesenia forbesii memiliki aktivitas antioksidan sangat lemah (IC50= 585,79ppm). Uji sitotoksisitas terhadap Artemia salena Leach menunjukkan bahwa ekstrak berkolerasi positif sebagai anti kanker (LC50 = 508,74 ppm). Berdasarkan analisis GC MS dari ekstrak metanol alga hijau Boergesenia forbesii menunjukkan 28 senyawa. Komponen utama dari ekstrak metanol alga hijau Boergesenia forbesii adalah Methyl Stearate, Phytol, Methyl Arachidonate, dan 9-Octadecenamide,(z)-.

Abstract

This study aims to determine the effect of using a solution of Trichloroacetic Acid (TCA) on the levels of Malondialdehyde (MDA) in Blood Plasma before and after being stored at low temperatures. Malondialdehyde (MDA) level test was carried out using the thiobarbituric acid-reactive substance (TBARS) test method using a spectrophotometer. Data analysis was carried out statistically using the t test. The test results showed that TCA stored at cold temperatures showed more color changes compared to the use of TCA stored at room temperature which did not change color after being given a solution of Thiobarbituric acid (TBA) and heated. The value of MDA levels in samples using TCA solutions stored at cold temperatures was greater than samples using TCA solutions stored at room temperature. The results of the statistical test on the t test showed that t count > t table and p < 0.05, it was concluded that there was a significant effect and difference between samples using TCA solution stored at cold temperatures and samples using TCA solution stored at room temperature. room. From these results, further research is needed to validate the method used so that the data obtained has been validated and can be used as a reference for practicum methods and research on MDA levels at the Research Laboratory, Faculty of Medicine and Health Sciences (FKIK), Bengkulu University.

Abstract

Extraction of a plant in traditional medicine is a very crucial stage for the success of the treatment. For example, nangka kuning (Vincetoxicum villosum (Blume) Kuntze) which has been scientifically researched, its leaves are extracted with ethanol solvent using the maceration method. Meanwhile, by local people,nangka kuning leaves are processed by boiling using water. This attracted the attention of researchers to re-analyze the content of secondary metabolites in nangka kuning leaf extract made with different solvents and methods. Researchers carried out the extraction using water with heating (infusion), ethanol without heating (maceration) and chloroform without heating (maceration). The results obtained from the three types of extracts contained secondary metabolites of alkaloids in the three extracts. While steroids are only found in extracts with a water solvent. Then the tannins are found in the extract with water and ethanol as solvents. Negative results from the three extracts were shown for secondary metabolites of flavonoids and saponins. So it can be concluded that nangka kuning leaf extract which has the most optimal secondary metabolite is the extract using water as a solvent by heating.
