Abstract

Covid 19 pandemic has caused changes in the socioeconomic conditions, affects parents' ability to sustain their children nutritious food. If nutrients are insufficient for a long term is causing growth to be stunted.

Abstract

Glioma merupakan tumor otak primer yang cukup sering ditemukan di Indonesia di antara keganasan otak lainnya. Penderita glioma mempunyai angka harapan hidup yang rendah dikarenakan seringnya mengalami resistansi terapi. Hipoksia merupakan salah satu faktor yang mempengaruhi respons terapi radiasi. Penelitian ini menganalisis ekpresi mRNA HIF-1?, sebagai petanda jaringan hipoksia, pada glioma derajat rendah dibandingkan dengan glioma derajat tinggi. Sampel terdiri dari 15 jaringan glioma derajat rendah, 6 jaringan glioma derajat tinggi, dan 21 sel lekosit dari penderita glioma sebagai kontrol sel normal. Ekspresi relatif mRNA HIF-1? ditentukan secara kuantitatif dengan menggunakan Real Time RT-PCR. Hasil penelitian menunjukkan bahwa ekspresi mRNA HIF-1? pada glioma derajat tinggi lebih tinggi dibandingkan dengan glioma derajat rendah, namun tidak bermakna secara statistik. Hal yang menarik yaitu ditemukan peningkatan ekspresi mRNA HIF-1? dengan nilai yang bervariasi pada 73 % sampel glioma derajat rendah dibandingkan dengan sel lekosit sebagai kontrol sel normal. Begitu juga pada glioma derajat tinggi ditemukan peningkatan mRNA HIF-1? dengan nilai yang bervariasi pada 100% sampel. Dengan demikian, dapat disimpulkan bahwa terdapat variasi derajat hipoksia pada glioma derajat rendah dan derajat tinggi yang mungkin mempengaruhi keberhasilan terapi radiasi.Kata kunci: Glioma, mRNA HIF-1?, hipoksia

Abstract

Background: HIF-2α is a transcription factor in hypoxic condition, and high expression levels of it correlate with the concepts of metastasis, therapy opposition and reduced quality of prognosis in various forms of cancerous growth. Due to the exceedingly infiltrative ability of brain glioma cells, gliomas cannot be completely deteriorated with surgery and the relapse rate is high. This study goal to identify the relative expression of HIF-2α gene in the direction of glioma malignancy and its classification. Methods: Specimens used in this research comprise of 20 glioma samples obtained from glioma patients in Cipto Mangunkusumo Hospital. Relative expression of HIF-2α was measured by utilizing quantitative Real Time-Polymerase Chain Reaction (RT-PCR). Cycle threshold (CT) values were achieved correlated with the amplified DNA, and then the relative expression was attained by using Livak method of calculation. Results: The results produced a greater average of relative expression of HIF-2α in the grade III and IV types (18.64; n=7) rather than in the lower grades (5.68; n=13). However, the data is statistically inconsequential. Conclusions: High-grade glioma tends to express HIF-2α mRNA higher compared to the lower grade. Therefore, it is possible to use HIF-2α as a prognostic marker for glioma- diagnosed patients, although additional experiments need to be performed to strengthen these facts.

Abstract

Background: Fasting may increase the activity of endogenous antioxidants and protect against oxidative stress. However, the effects of different fasting durations on the liver have not been reported. Objective: The purpose of this study is to determine the effect of intermittent and prolonged fasting on oxidative stress markers in the liver tissue and plasma of New Zealand White rabbits. Methods: New Zealand White rabbits were divided into three groups: control, intermittent fasting (IF), and prolonged fasting (PF), with each group consisting of five rabbits. The control group was provided with food ad libitum; the IF group fasted for 16 hours, while the PF group fasted for 40 hours, followed by an eight-hour non-fasting period for six days. In liver tissue and plasma, oxidative stress indicators (catalase, carbonyl, GSH) were evaluated. Results: In the IF group, liver GSH was significantly higher than in the control group. However, neither liver carbonyl nor catalase levels changed significantly in the IF group. In the IF group, plasma carbonyl was significantly lower than in the PF group. In addition, there was no significant differences between groups in plasma catalase and GSH levels. Conclusion: Intermittent fasting and prolonged fasting could significantly increase liver GSH levels of New Zealand White rabbits. In addition, intermittent fasting is more effective than prolonged fasting at preventing oxidative stress.

Abstract

Calorie restriction (CR) is the most effective method for delaying aging and preventing the onset of age-related diseases. Sirtuins constitute a family of nicotinamide adenine dinucleotide (NAD)-dependent histone deacetylases. Their activity can be regulated by NAD+/NADH levels, which are influenced by nutrient intake, a variable acted upon by CR. This review elaborates on the link between CR and sirtuin1 (SIRT1). It retrieved articles from several sources, such as ClinicalKey, PubMed, and ScienceDirect. It discusses the up-to-date knowledge of how SIRT1 acts as a nutrient sensor and regulator of molecular mechanisms. These mechanisms include the control of the cell cycle, enhancing mitochondrial quality control, activating fatty acid oxidation, and stimulating anti-inflammatory effects. Disruptions in the aforementioned mechanisms are the basis of aging. CR increases the expression of SIRT1, which enhances the biogenesis and dynamics of mitochondria, resulting in an antiaging effect. In CR, SIRT1 is activated and stimulates different pathways, especially those related to mitochondrial activity and effectiveness, leading to an antiaging effect in collaboration with other antiaging biomarkers
