Abstract

Plant soursop (Annona muricata L.) is one of the Annonaceae familia. This plant has well-known in the society environment and it has been using as a traditional medicine. One of benefits of the soursop is can used as an algernative of cancer treatment. Until now not yet known scientifically whether soursop leaves have anticancer activity.This study aims to determine the potential toxicity of bioactive substances from the ethanol extract, of ethyl acetate and n-hexane of the soursop leaves as an anticancer agent by using Brine Shrimp Lethality Test (BSLT) method. This experimental As many as 100 larvas were use as research objects which divided into 3 groups. Each group consist of 10 larvas and the group repetition were done by randomly, three times per group. The extract concentration used in medium is 500, 100, 10 and as a control 0 ppm. Data obtained from counting the number of dead larvas (letal concentration 50 ppm) after 24 hours treatment. Based on the data obtained from LC50 of extract ethanol, ethyl acetate and n-hexane soursop leaves then determined by using probit analysis. The water content which contained in simplicia (drifed leaves) is 6,49%. The probit analysis results show that the ethanol extract have LC50 values of 86,45 ppm, 92,81 ppm of ethyl acetate extract and n-hexane extracts 37601,05 ppm. The distribution of ethanol and ethyl acetate extracts of soursop leaves in the experiments shown any potential toxicity against Artemia salina Leach, while the n-hexane extract of soursop leaves show no any potential toxicity to against Artemia salina Leach larvas.

Abstract

Psidium guajava Linn. known as guava leaves is a medicinal plant that has been used to treat diseases, including burns medicine. From the results of the study note that the extract of guava leaves ethanol can heal burns. To facilitate its use, then made ointment preparations of guava ethanol extracts. The purpose of this research is to make an ointment formula ethanol extract of guava leaves which has a healing effectiveness on a second degree burns. Extraction of guava leaf made by the process of maceration, then the liquid extract was concentrated by rotary evaporator to obtain a concentrated extract of guava leaf. An ointment formulations ethanol extract of guava leaf are made with concentrations respectively 30%, 35%, and 40. The effectiveness test of the healing of burns grade II using 5 groups: negative control (ointment base), positive control (Bioplacenton®), ointments ethanol extract of guava leaves 30%, 35%, and 40%. The study was conducted . of male white rats with severe between 200 – 400 grams. Observations up to 21st day. These result by using Tukey Test indicate that the concentration of guava leaf extract 35% , The results of this study indicate that the variation concentration of guava leaf extract gives a healing effect that does not differ significantly to healing second degree burns in male white rats.

Abstract

This study was adescriptiveanalysis ofexperimentaldesign. Analysis of sildenafilcitrate compounds using Thin Layer Chromatography (TLC) and Liquid Chromatography Mass Spectrometry (LC-MS) were sampled in two districts, namely sub-district, Bogor Barat and Tanah Sareal thus obtained 32 samples, of which 18 samples of Bogor Barat coded A and 14 samples from Tanah Sareal coded B. TLC results were eluted at Silica gel 60 F254 with eluent methanol:chloroform (4:1), there were 5 samples showed close Rf value standard sildenafil citrate is 0.83, the sample code A4 value of Rf 0.84, A13 Rf value of 0.84, A17 Rf value of 0.83, B11 Rf value of 0.83, and B14 Rf value of 0.84. Results LC-MS where separation using column Phenomenex Synergi Fusion RP-100A, the pumping system with a mobile phase gradient of 0.2 M ammonium acetate pH 7 in water and 0.2 M ammonium acetate pH 7 in acetonitrile, was obtained the samples for A4, A13, A17, B11 dan B14 amounted to 198.01; 1734.14; 226.00; 156.77; 443.87 μg/kg

Abstract

Diuretics is a substance that can stimulate of urine output. The content of flavonoid compunds contained in water extracts of avocado leaves have a diuretic activity. Testing is done withgiving a water extract of avocado leaves per oral to 30 male rats strain (Sparague-Dawley) with weight 200 grams were divided into 6 groups. Frequency of urination with water extract of avocado leaves 5 % , 10 % , 15 % and 20 % respectively which is 2 , 5 , 9 and 20 times with observation for 3 hours. Furosemide tablets urination frequency was 25 times over 3 hours. The result showed that the water extract of avocado leaves 20% have diuretic activity is equivalent to Furosemide tablets at a dose of 7.2 mg / 200 mg BB in male rats.

Abstract

Acne is skin disease caused due to a blockage of the oil gland channel and infection by the bacteria Propionibacterium acnes. Ethanol extract of 70% basil leaves has been known to be potent as antibacterial to Propionibacterium acnes. This study aims to determine the physical quality of gel preparation and antibacterial activity analysis of gel ethanol extract preparation of 70% basil leaves (Ocimum americanum L.) to Propionibacterium acnes bacteria. In this research, ethanol extract 70% of basil leaf is formulated into gel preparation form with various concentration of extract that is 45%, 55%, and 55% then in physical quality test include: organoleptic test, pH, spreading, and viscosity. The gel preparation was then analysed for its antibacterial activity using the diffusion method for the wells. Based on ethanol extract research, 70% of basil leaves had antibacterial activity with drag zone diameter of 5,20 mm (45% extract concentration), 6,06 mm (concentration of extract 50%) and 7,13 mm (concentration of extract 55%). In this study can be concluded that the higher concentration of ethanol extract 70% basil leaves the higher the inhibit zone diameter.

Abstract

Kapsul adalah bentuk sediaan obat terbungkus cangkang kapsul keras atau lunak. Cangkang kapsul dibuat dari gelatin dengan atau tanpa zat tambahan lain. Gelatin merupakan suatu zat yang diperoleh dari hidrolisa parsial dari kulit atau tulang hewan. Sumber gelatin yang dapat digunakan sebagai bahan dasar pembentuk kapsul cangkang keras diantaranya tulang atau kulit dari sapi, babi, ikan. Ikan lele dapat digunakan sebagai bahan dasar untuk pembuatan gelatin. Penelitian ini bertujuan untuk membuat dan mengevaluasi karakteristik kapsul cangkang keras dari gelatin yang dibuat dari gelatin tulang ikan lele dumbo (Clarias garepinus). Proses pembuatan gelatin dilakukan dengan menggunakan cara asam. Hasil pengujian gelatin menunjukan nilai pH gelatin 4,85, kadar air 13,5 %, viskositas 180-190 Cps, kadar abu 3,2% dan kekuatan gel 150 g/cm2. Gelatin yang dihasilkan kemudian digunakan untuk pembuatan cangkang kapsul keras yang dibedakan menjadi 2 formula dengan memvariasikan jumlah gelatin yang digunakan (Formula I = 25 g dan formula II = 35 g gelatin). Evaluasi terhadap karakteristik cangkang kapsul yang meliputi dimensi cangkang kapsul, pengukuran volume cangkang kapsul, waktu hancur dalam air dan larutan asam, kadar air dan kerapuhan. Hasil pengujian karakteristik yang tidak memenuhi persyaratan pada formula I adalah penetapan kadar air dan kerapuhan, sedangkan pada formula II hanya pada penetapan kadar air.

Abstract

Jumlah penderita diabetes melitus meningkatkan setiap tahunnya sehingga kebutuhan insulin meningkat. Insulin merupakan obat utama untuk pasien diabetes mellitus tipe 1 (DMT1). Dalam beberapa kasus, diabetes mellitus tipe 2 (DMT2) memerlukan insulin untuk pengobatannya. Saat ini, Escherichia coli, Saccharomyces cerevisiae, Pichia pastoris, dan Hansenula polymorpha merupakan sistem ekspresi yang digunakan secara luas untuk memproduksi prekursor insulin. Prekursor insulin diubah menjadi insulin secara enzimatik. Pada penelitian ini, digunakan Pichia pastoris untuk memproduksi prekursor insulin karena mampu memproduksi prekursor insulin dalam jumlah besar dan disekresikan ke luar sel Pichia pastoris sehingga mempermudah proses pemurnian. Penelitian diawali dengan regenerasi dan seleksi Pichia pastoris. Pichia pastoris terpilih ditumbuhkan dalam media fermentasi kemudian prekursor insulin yang dihasilkan dikarakterisasi dengan elektroforesis SDS-PAGE dan kadar insulin diukur dengan Elisa. Hasil karakterisasi menunjukkan prekursor insulin berupa pita berukuran sekitar 7 KDa dengan kadar prekursor insulin 6,88 µUI/mL.
