Abstract

Study on exploration of immunocontraception agents for wildlife fertility regulation has been conducted.The aim of this study was carried to find out the protein which functionable in mice embryos implantation. Theoutcome of this study should be applied to control the fertility of wildlife animals. This research conducted duringten months in BSF Laboratory of Biology Department FMIPA UNDIP and 3rd Unit of UGM LPPT. Adult femaleand male mice with 28 – 30 grams in weight were used as laboratory animals. Mice were divide into two groups, onegroup as positive control group without mated, the other were mated group. Mice handled and breeded in laboratorycondition. The precise day of pregnancy were determined with vaginal plug existences in female mice. Protein wereisolated from uterus on day 1st to 5th of pregnancy. Protein isolation and separation with electrophoresis were basedon Bio-rad manual. This study show that there are no differences in protein band between positive control group andpregnant group. These study suggested that protein determining embryo implantation process is regulatory proteinwhich it has expressed in short time and fast withdrawl.

Abstract

Neem leaf (Azadirachta indica) was known as infertility herb. Neem leaf contain of flavonoid compounds which inhibit the damage of cell. This research aimed to examine the effect of neem leaf on hepotocyte and liver lobules diameter. The total of labolatory animals in this research were 12 male Rattus novergicus. This research was conducted by using the Complete Randomized Design (CRD). The animal was treated by neem leaf extracted with 12, 16, and 20 mg/animal/day (PO, P1, P2, P3) respectively. The treatment was treated during 14 days. The liver was made as histological slide by using HE stain with 3-4 µ in thickness. The observed parameters were hepatocyte diameter, liver lobules diameter, the final body weight and liver weight. Data here analyzed by ANOVA test. All the analysis were performed with 5% significant level. The results showed that the giving of neem leaf extract had no significant effects (p>0.05) on hepatocyte diameter, liver lobules diameter, liver weight and final body weight. It could be concluded that the giving of ethanolic neem leaves extract with 20 mg/animal/day for 14 days did not affect the hepatocyte diameter, liver diameter, liver weight and final body weight

Abstract

Research about animal reproduction can be observed from many aspects, i.e. oestrous cycle. The oestrouscycle is the manifestation of reproduction hormones changes, especially oestradiol 17- B. The oestradiol 17- B hormones causes changes of tissues structures of reproductive tracts. This research have been done toanalyze the correlation between changes of oestradiol 17- B hormones and uterine endometrial thicknessalong the oestrous cycles. The adult female Swiss Webster mice were used as laboratory animal. The phaseof oestrous cycle determined by vaginal smears. Blood and uterine samples collected in each oestrousphases. Hormones concentration counted with RIA, the thickness of endometrial determined on uterinehistological slides with paraffin methods and stained with HE. This search showed that a positivecorrelation between oestradiol 17- B hormones and uterine endometrial thickness along the oestrous cycle.It also showed that oestradiol 17- B hormones have a proliferative effects on reproductive tissues.

Abstract

Soybeans contains phytoestrogen that influence reproductive process. Phytoestrogen administration withchronical dosage along growth periode cause the neuroendocrine system disturbance. Commonly in humanintake of phytoestrogen from soybeans or other soybeans product. So it is important to research beneficialand adverse effects of soybeans consumption with chronical dosage to reproductive process, particulary iscorelation between the sum of ovarium follicles and the estrogen level of mouse (Mus musculus) after dailyconsumption of soybeans along 40 days. The objective of this study was to know the corelation betweenthe sum of ovarium follicle and the estrogen level of mouse, by experimental design. Adult female micewere use as experimental animal, divided into two groups, there were control and treatment. Thegroup of treatment was administrated with 0,975 mg soybeans flour, it was contain ± 0,00156 mgphytoestrogen. The control group was administreted only drinking water. The result of this study showsthat were not significantly difference between control and treatment group. It concluded the corelationbetween the sum of ovarium follicles and the estrogen level were not influenced with daily consumption offitoestrogen.

Abstract

Phytoestrogen has been known have many advantages in human health. It has estrogenic effect because it similarity in structure with natural estrogen. It action done by interact with estrogen receptor and inhibit the interaction of natural estrogen. The aim of this research was to evaluate the effect of soybean starch as phytoestrogen resources on reproduction of M. musculus, by the endometrial thickness and the sum ofendometrial glands aspects. The adult female Swiss Webster mice were used as animal treatment. The mice were treated with 0,975 mg/kg BW/day of soybean starch and 0,148g/kgBW/day. Uterine samples collected from every estrous phase made as histological slides with paraffin methods and stained with H.E.with 6 μ in thickness. Anova with factorial design at 5% significances were used to analyzed the data. This search showed that there was no significances between control and treatment group so it can be concluded that the result were caused by the act of endogen estrogen in animal treatment.

Abstract

Study to detect the expression of Wnt4 protein in the uterus of Swiss Webster mice with immunohistochemistry method has been done. Laboratory animals that were used are adult Swiss Webster mice weighing 25-30 grams. Pregnancy is determined by the presence of vaginal plug in female mice after breeding. Uterus was isolated on seventh days of gestation. The detection of protein expression was done by paraffin method using mouse anti-Wnt4 (Santa Cruz Biotechnology) as primary antibody with 1:250 in dilution. Visualization of Wnt4 protein was done by DAB as chromogen. The positive reaction to the primary antibody showed in perimetrium, myometrium, and endometrium in uterine tissue. The positive reaction to the primary antibody were expressed on blood vessels also. It could be concluded that the Wnt4 protein as signal molecule regulates the embryo development by circulation system. This study offer as source to study Wnt4 protein expression pattern as molecular marker in early development of organism.   Keywords: Wnt4, immunohystochemistry, Mus musculus

Abstract

The aim of this research was to evaluate the effect of semencryopreservation using TES-Tris yolk citrat (TES-TYC) medium onmotility of the sperm. Semen fulfilling inclusion criteria with WHO 1999 criteria. The sperm motility was counted as initial data. The semen was then mixed with TES-TYC medium and cryopreserved in liquid nitrogen. After one mounth the semen was thawed and recount its sperm motility. Data obtained showed that the motility of post freezing sperm cryopreserved was lower compared to the motility of pre freezing sperm. It could be concluded that cryopreservation caused decrease integrity and death of sperm during cryopreservation, that showed the motility of post freezing sperm cryopreserved was lower compared to the motility of prefreezing sperm.

Abstract

Penelitian ini bertujuan untuk mengetahui struktur hepar dan rasio bobot hepar terhadap bobot tubuh mencit (Mus musculus L) jantan setelah pemberian ekstrak etanol daun mimba (Azadirachta indica A.Juss). Penelitian ini menggunakan 2 kelompok perlakuan yaitu Kontrol (akuades) dan Perlakuan (mimba). Perlakuan diberikan secara oral selama 21 hari. Hepar ditimbang dan dibuat preparat menggunakan metode parafin dengan pewarnaan Hematoksilin Eosin. Pengukuran diameter hepatosit dilakukan dalam 4 bidang pandang. Bobot hepar dan bobot tubuh digunakan untuk menghitung nilai HSI. Data dianalisis dengan uji t. Hasil analisis menunjukan adanya pengaruh nyata terhadap diameter hepatosit tetapi menunjukan pengaruh tidak nyata terhadap bobot tubuh, bobot hepar dan nilai Hepatosomatik indek (HSI). Hal ini dapat disimpulkan bahwa pemberian ekstrak etanol daun mimba (Azadirachta indica) sebagai obat tradisional dengan dosis 14 mg/KgBB tidak mempengaruhi struktur hepar dan rasio bobot hepar terhadap bobot tubuh mencit (Mus musculus) jantan. Kata kunci : Daun Mimba, Hepar, Diameter Hepatosit dan HSI.

Abstract

Neem (Azadirachta indica A. Juss) is one of the traditional medicines used by local people as antioxidants, anti-hyperlipidemia, anti-inflammatory, and treatment for other diseases such as heart disease. This study aimed to analyze the effect of neem leaf ethanolic extract on aortic wall thickness and aortic lumen diameter of white rats induced by high-fat feed. The male Wistar rats (Rattus norvegicus L.) with 2 months of age were used. This research was an experimental study with a Randomized Complete Block Design consisted of 6 treatments (P0: commercial feed, P1: high-fat feed and duck egg yolk 2.5 ml/200 g BW per oral, P2: P1 + 8 mg/200 g BW simvastatin, P3-P5: P1+ neem leaf extract of 75, 100, and 125 mg/200 g BW respectively) and 4 replications. Aortic preparations were made by the paraffin method and Hematoxylin-Eosin staining. Data were analyzed using ANOVA followed by Duncan Multiple Range Test with 95% confidence level. The result showed that the decrease in wall thickening and lumen narrowing was getting higher along with the increasing doses of ethanol extract of neem leaves. Based on the result of this research, it was found that the high doses of neem leaf ethanolic extract has the same ability with simvastatin to reduce aortic wall thickning and aortic lumen narrowing of white rats. This study is give a new information that the neem leaves can be used as an alternative medicine for cardiovascular disease.

Abstract

Azadirachta indica A. Juss (Neem) is a medicinal plant that contains antioxidants, antihyperlipidemia, and has antidiabetic activity. This research aimed to examine the effect of neem leaf ethanolic extract on blood glucose levels of white rats given high-fat rations. The male white rats (Rattus norvegicus L.) with two months of age were used. The treatment of test animals consisted of six treatment groups and four replications, namely: the control group (P0); given high fat rations + duck egg yolk 2.5 ml/200gBW (P1); given high-fat rations + 8 mg/200gBW simvastatin (P2); and P1+ the dose of neem leaf ethanolic extract of 75; 100; and 125 mg/200gBW (P3; P4; and P5). Blood-glucose levels were analyzed by colorimetric enzymatic method using Roche Diagnostics Cobas C reagents kits. Blood-glucose level was determined using Roche / Hitachi cobas c systems automatically. Homogeneous and normally distributed data of blood-glucose levels, body weight and feed consumption were analyzed statistically using ANOVA followed by Duncan's test with a significance level of 5% using SPSS 16.0 software. The results indicated that giving the neem leaf ethanolic extract of could reduce the blood glucose levels. In this study, it was concluded that the neem leaf ethanolic extract of could potentially be used as an alternative to cure diabetes mellitus.
