Abstract

Malaria still become one of major health burden in Indonesia especially in remote areas of east Indonesia. Golden standard of malaria parasite detection is still microscopic technique using polychromatic light source whether from halogen or natural light source. A microscopic technique has a lot of benefits but still have weaknesses, such as time-consuming and bias on the reading by microscopist, because of artifact in the image. Aims of this study were to designed malaria parasites detection tool that is robust, fast, convenient and clear by minimizing artifact on the slide. Design of this study was laboratory experimental which modified simple microscope into an automatic microscope with table movement and webcam recording using a microcontroller and monochromatic light source. The wavelength of the light sources was 402nm(blue), 532 nm (green) and 650 nm (red), the intensity of each source differed. The reading of the slide image was conducted by two certified microscopists, who read 60 images of a thick and thin slide with three different live stage of Plasmodium falciparum live, which wearing, trophozoite and schizont. This study showed that modification of microscope was succeeded with automatic movement and webcam recording, process time in one step movement and recording approximately 10 seconds or 17minutes for 100 fields of view as confirmation process. The monochromatic light source has proven to give a clear and contrast field of view when the intensities were higher than 40 mW and the certified microscopist able to identified Plasmodium falciparum parasites. Data analysis of microscopist reading used nonparametric statistic Friedman by SPSS showed that correlation between images using monochromatic and polychromatic lights have meaningless differences in a thick and thin slide. However, hemozoin as a marker of Plasmodium falciparum parasite was less detected by monochromatic light used in this study.

Abstract

Lactic acid bacteria have been reported to be useful as a healt adjunct and are commonly added to food as delivery mechanism. As a probiotic, lactic acid bacteria should have the ability to survive in the digestion process such as resistance towards acidic pH, varieties of bile salt concentrations and enteric pathogan. In this study, lactic acid bacteria were isolated from various sources. Twenty isolated of lactic acid bacteria selected for their resistancy toeards acidic pH (pH 2.5), 0.3% bile salt and their antagonistic activity against enteric pathogan (Esherichia coli, Staphylococcus aureus and Bacillus cereus). The result showed that most of all strauns had good resistance to acidic pH and there were no significant difference among theme (p>0.05). More over, all strains showed tolerance to 0,3% bile salt concentration (varietis among isolates were significant p>0.05). All strains showed inhibition activity against enteric pathogan bacteria, this was estimated from the size of the diameter of the inhibition zones. (The inhibition were significantly different among them (p>0,05)). The best five stains that fulfilled these properties L casei FNCC262, L. acidophilus FNCC116, KL-3 isolate, Da-1 isolate, and Db-2 isolate. These strains could be used as probiotics in further and applications. Key words : acid-tolerant, bile tolerant, antagonistic activity, lactic acid bacteria

Abstract

The road infrastructure has a significant influence on the development facilities of the city as acenter of a new generation. Bandar Lampung as the capital of Lampung province was not im -mune to these problems. Development of a very dynamic with a diverse urban population, trig-gered over the land due to the complexity of the construction of roads that will lead to many nega-tive issues in the future. Problem caused by the increasing development of road infrastructure inand on the outskirts of the city, will increase the rate of population growth to the high impact ofurban land usage that is not in line with the regional spatial plan. To suppress the effect of infra-structure development on land conversion, required policy analysis tool. The making of policysimulation modeling of land usage on the development of road infrastructure using dynamic sys-tem modeling. The models created using causality approach (causal loop), starting from the basicmodeling that is a mapping of the real system of urban land usage which is then converted into 4model. Model of transportation, demography, population and land usage activities, which are as-sociated with a causal relationship. The models will be aided by the GIS as spatial data analysisinput, the models simulate some policy scenarios. The most optimal scenario results to reducingthe rate of population growth, reduce the level of unemployment, increased GDP, and control thepattern of land usage. By performing simulations using these scenarios, all the desired purposecan be met and brought a good impact on the growth of Bandar Lampung city.
