Abstract

The inheritance pattern of resistant tomato to root-knot nematode was studied. GM2 accession and Gondol Putih (GP) cultivars were used as resistant and susceptible parent, respectively. Parental plants, F1 generation, and the F2 generation were grown individually in the sterile soil. One thousand of larvae stage 2 (L2) nematode of M. incognita was infested in each plant. Data of root gall intensity, the number of egg mass, population of L2 nematodes in root and soil were analyzed with the Kormogorov-Smirnov’s test, Chi-square test, and potence ratio. The F2 selected resistant plants then were grown and self fertilized to identify of homozygote plants (F2) related to nematode resistant and good fruit characters. The result showed that the resistant to root-knot nematode was controlled by a dominant gene. Positive correlation between damage levels of roots, eggs mass number and L2 larvae population in the root was significant. Eleven selected plants, conferred the homozygous resistant gene, was prospective lines to be used pedigree or single seed descent selection in producing variety with resistant to nematode, high yield and quality of fruit.   Keywords: tomato, root-knot nematode, resistant gene, dominant, homozygous, correlation

Abstract

Perennial crop leaves contain polysaccharides, polyphenols, and other secondary metabolites in high concentration. The presence of those compounds inhibit enzymatic activities and amplification of DNA. The existing extraction methods were not able to dissociate the metabolites contaminants of sapodilla (Manilkara zapota (L.) van Royen) leaves and thus resulting in low quality of extracted DNA. The aim of this experiment was to develop an effective method to extract DNA from mature leaf samples of sapodilla (Manilkara zapota (L.) van Royen). Fifth modification of Doyle & Doyle DNA extraction protocol with modified concentration of buffer reagent (consisted of: CTAB 2.8%; NaCl 2.5M; mercaptoethanol 3%, and PVP 2.5%) and repetition of some phase purification (liquid nitrogen; three times CIAA; two times ethanol 70%, RNAse 1µl) generated high quality DNA and clear band of PCR amplification using RAPD primers.   Keywords: buffer modifications, DNA extraction, sapodilla

Abstract

This study was conducted to evaluate the heritability, heterosis, and correlation among firmness, total soluble solid (TSS), and color in the F1 hybrids between two day-neutral (DN) and six short-day (SD) cultivars. The firmness of DN cultivars and the sweetness of SD cultivars were considered as a criterion in choosing parent cultivars. Parents and eleven F1 progenies were grown in completely randomized block design. Heritability of TSS, color (a*: opponent channel between red/magenta and green), and firmness was 0.67, 0.55, and 0.49, respectively. Most of the DN × SD hybrids had higher firmness and TSS than the crossing of SD × SD and DN × DN, respectively. In the DN × SD progenies, all heterosis values of TSS and most of heterosis of firmness were positive and negative, respectively. The genetic advance of TSS was higher than firmness. ‘DNKW002’ was a better donor for firmness and can be used as female parent in producing DN cultivar with high TSS. Correlation between firmness and TSS was negative, hence the simultaneous selection was recommended. Principle component analysis (PCA) based on characteristics indicated three groups: group of DN parents and hybrids of DN × DN, group of SD parents and SD × SD hybrids, and group of DN × SD and SD × DN hybrids.
