Abstract

A tetrapeptide, an analog of  trypsin-modulating oostatic factor (TMOF), has been synthesised by using a method of solid-phase peptide synthesis on 2-chlorotrityl chloride resin. Fmoc strategy was applied on the synthesis. The formation of peptide bond was facilitated by DIC/oxime as coupling reagent. After all of amino acids were attached on the resin, tetrapeptidyl resin was added by a mixture of TFA:water:EDT(90:5:5) in dichloromethane to cleave the peptide.Crude peptide was purified by reverse-phase column chromatography and the purified peptide was analysed by TOF ES-MS spectroscopy.

Abstract

Crop cabbage worm (Crocidolomia pavonana) is one of the pests that has a bad effect on the cabbage production in Indonesia. One of the efforts that has been applied to control this pest is by using bayrusil. However, the bayrusil can cause negative effects such as poisoning and environmentally polluting effects. To prevent the undesired effects, it has been developed the use of environmentally friendly insecticide and one of them is the use of insecticidal peptide. Tripsin-modulating oostatic factor (TMOF) and ist analogues is one of the peptides that has an insecticidal activity. The aim of this research is to synthesise a linear tetrapeptide (DPAP) an analogues of TMOF and to test insecticidal activities against crop cabbage worm. A DPAP has been synthesised by Solid Phase Peptide Synthesis on 2-chlorotrytil chloride resin. Fmoc strategy was applied on the synthesis and a combination DIC/Oxime was employed in the coupling reaction.The tetrapeptidyl resin was cleaved by TFA:water:EDT (90:5:5). The peptide was purified by reverse-phase column chromatography and characterized by TOF ES-MS spectroscopy. A linear peptide was biologically tested towards the C. pavonana, showing percentage of mortality values at 1000 ppm.

Abstract

Antioxidant pentapeptides are pentapeptide compounds that have antioxidant activity. One of the pentapeptide compounds that have antioxidant activity is FWKVV. FWKVV is a linear pentapeptide with the amino acid sequence phenylalanine-tryptophan-lysine-valine-valine, which was first isolated to hydrolyzate the muscle protein of Miiuy croaker (Miichthysmiiuy). In addition to isolation, FWKVV compounds can be produced by the peptide synthesis method because this method requires a shorter time than the isolation method from natural materials. Synthesis methods commonly used are solution-phase peptide synthesis and solid-phase peptide synthesis (SPPS). However, the SPSS method is more efficient because it does not require purification in every process. The purpose of this study was to synthesize FWKVV compounds using the SPPS method and test their antioxidant activity. FWKVV has been synthesized using the SPPS method with HBTU/HOBt coupling reagent and Fmoc protective group. The FWKVV crud produced was 148.8 mg and had antioxidant activity against DPPH radicals with an IC50 value of 4.2 mg/mL.

Abstract

Pumpkin fruit (Cucurbita moschata. D) belongs the Cucurbitaceae family which is a functional vegetable widely distributed in Indonesia because it has nutritional value and health benefits. This study aims to determine the value of total flavonoid content and antioxidant activity in the ethanol extract of unripe, mature and ripened pumpkin. Simplicia powder was extracted by maceration method using 96% solvent. Testing the total flavonoid content with the addition of AlCl3 at a wavelength of 425 nm and the antioxidant activity test was carried out using DPPH (1-1-diphenyl-2-picrylhydrazyl) as a free radical with a maximum wavelength of 515 nm using microplate reader. The results of the study concluded that the ethanolic extract of pumpkin flesh had a total flavonoid content of 0,146 mgQE/100g in unripe fruit, 0,221 mgQE/100g in mature pumpkin, 0,191 mgQE/100g in ripened pumpkin. The antioxidant activity of the ethanol extract of unripe, mature, and ripened pumpkin fruit obtained was not active or did not completely reduce free radicals.

Abstract

Body lotion merupakan salah satu kosmetik untuk merawat kulit. Cemaran yang terdapat dalam body lotion salah satunya adalah logam berat. Timbal merupakan salah satu jenis logam berat yang memiliki dampak negatif bagi kesehatan manusia. Penelitian ini bertujuan untuk mengetahui ada tidaknya logam timbal (Pb) dalam body lotion dan menentukan kadar logam timbal (Pb) dalam body lotion yang beredar di pasar Jember. Sampel dipreparasi secara destruksi basah. Analisis kandungan logam timbal (Pb) menggunakan metode reaksi warna dan endapan. Analisis kadar logam timbal (Pb) menggunakan metode Spektrofotometri Serapan Atom. Hasil penelitian secara kualitatif menunjukkan bahwa ketiga sampel body lotion tidak menunjukkan perubahan pada reagen NaOH dan HCl encer, sedangkan pada reagen KI ketiga sampel menunjukkan perubahan warna dan endapan. Hasil secara kuantitatif menunjukkan bahwa sampel A, B dan C tidak mengandung timbal. Dari hasil penelitian yang telah dilakukan maka dapat disimpulkan bahwa body lotion yang beredar di pasar Jember tidak mengandung logam timbal (Pb).

Abstract

Tanaman makadamia termasuk famili Protaceae yang berasal dari Australia. Tanaman kacang makadamia memiliki sifat sebagai antioksidan yang dihasilkan oleh tocopherols, tocotrienols dan squalen. Senyawa fitokimia genus makadamia memiliki aktivitas sebagai antioksidan, antiinflamasi, antidislipidemia dan antimikroba. Penelitian ini bertujuan untuk menentukan kadar flavonoid total ekstrak etanol daun makadamia dan uji aktivitas antioksidan dengan menggunakan metode DPPH. Sampel daun makadamia dimaserasi menggunakan pelarut etanol 96%. Analisis kadar flavonoid total dilakukan dengan metode spektrofotometer UV-Vis dan dinyatakan dalam Quercetin Equivalent (QE). Uji aktivitas antioksidan menggunakan metode DPPH dengan menghitung nilai IC50. Hasil analisis menunjukkan bahwa kadar flavonoid total ekstrak etanol 25,39 ± 0,22 mg QE/g ekstrak, sedangkan aktivitas antioksidan ekstrak etanol daun makadamia (Macadamia integrifolia) memiliki nilai IC50 24,99 μg/mL dan termasuk intensitas sangat kuat.

Abstract

Tanaman sirih hijau (Piper betle L.) merupakan salah satu jenis tumbuhan yang banyak dimanfaatkan untuk pengobatan. Tanaman sirih merupakan tanaman yang tumbuh subur di sepanjang Asia tropis hingga Afrika Timur dan menyebar hampir di seluruh wilayah Indonesia. Daun sirih hijau mengandung minyak atsiri yang komponen penyusunnya merupakan senyawa fenol yang mampu menjadi senyawa anti bakterisidal, fungisidal, maupun germisidal. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri daun sirih hijau terhadap bakteri Escherichia coli. Pembuatan ekstrak daun sirih hijau dilakukan menggunakan metode maserasi menggunakan etanol dan n-heksan. Uji aktivitas antibakteri dilakukan dengan metode difusi agar. Penelitian ini menggunakan 3 konsentrasi yaitu 20%, 30%, dan 35%, serta kontrol positif (Chloramphenicol) dan kontrol negatif (DMSO). Hasil menunjukkan ekstrak etanol daun sirih hijau konsentrasi 35% dapat menghambat pertumbuhan bakteri Escherichia coli dengan daya hambat 19,65 ± 7,97 mm, sedangkan ekstrak n-heksan daun sirih hijau konsentrasi 35% dapat menghambat pertumbuhan bakteri Escherichia coli dengan daya hambat 10,17 ± 4,25 mm. Hal ini menunjukkan daun sirih hijau memiliki kemampuan sebagai antibakteri.
