Abstract

Background: Preeclampsia is one of the factors causing the high maternal mortality rate. The risk of morbidity and mortality is higher in Early Onset Preeclampsia (EOPE). Failure of spiral artery remodeling can cause oxidative stress that can inhibit placental development and increase trophoblast apoptosis. Objective: This study aims to analyze the oxidative stress and apoptosis of EOPE placentas. Methods: This study is an observational study with a cross-sectional design. A total of 31 EOPE placentas and 31 normal term placentas were used to measure the concentration of malondialdehyde (MDA) and the relative mRNA expression of FOXO3 and CASP3 using the spectrophotometric and RT-qPCR methods. Results: There was no difference in MDA concentration (p = 0.580) and FOXO3 (p = 0.467) and CASP3 (p = 0.243) mRNA expression in the normal and EOPE groups. There was a strong positive correlation between FOXO3 and CASP3 mRNA expression in the normal (p= 0.0001; r = 0.938) and EOPE groups (p = 0.0001; r = 0.855). There was no correlation between MDA concentration to FOXO3 (p = 0.124; r = 0.282) and CASP3 (p = 0.569; r = 0.106) mRNA expression in normal placenta. There was positive correlation between MDA concentration to FOXO3 (p = 0.016; r = 0.429) and CASP3 mRNA expression in EOPE placenta (p = 0.028; r = 0.395). Conclusion: These results indicate that cell integrity is still maintained through the autophagy process and the level of apoptosis in the EOPE placenta is regulated by ROS through FOXO3.

Abstract

Carbohydrate is the first energy source used in metabolic processes. Failure in carbohydrate metabolism can cause metabolic diseases, and many studies are focused on its therapy. Until now, there is no specific therapy approved. Despite that, one of the main alternatives is regarding the involvement of FOXO1 protein in metabolic disease progress. As a transcription factor for gluconeogenesis genes, FOXO1 can increase blood glucose levels. It also involves various signaling pathways in carbohydrate metabolisms such as PI3K/Akt and PKA in which many proteins act as FOXO1 regulators through posttranslational modification. The vital role of FOXO1 in carbohydrate metabolism provides an opportunity to make FOXO1 the main target of metabolic disease therapy. Various proteins and natural compounds can either directly or indirectly regulate FOXO1 activity. It can be an option to be used to control blood glucose levels by targeting FOXO1

Abstract

Abstract

Clean and healthy living behavior (PHBS) in the household is one of the government's programs to improve people's welfare. PHBS indicators in homes include using clean water, washing hands with soap and clean water, and using healthy latrines. Lack of knowledge and public awareness of the importance of PHBS and inadequate health support facilities and infrastructure can be a factor causing high rates of morbidity and mortality due to a disease. One of the means of supporting public health is the availability of water sources. The decrease in river water discharge in Lembah Sari Village can affect sanitation related to PHBS in the community. We have conducted PHBS counseling activities in Lembah Sari Village using PowerPoint media, posters, and teaching aids. It was an effort to help people know, want, and practice PHBS. After receiving counseling, the community became more aware of PHBS. The community seemed enthusiastic and participated in practicing washing hands with soap as an indicator of PHBS. This activity is expected to be applied by the community daily.

Abstract

Alginate is used as an emulsifier in food production. Its non-toxic, biocompatible, biodegradable, and hydrophilic properties are very promising for industrial use. Alginate can be obtained from brown seaweed because the alginate content in brown seaweed reaches 40% of its dry weight. The alginate extraction process can be carried out in 5 main stages, 1) extraction process using mineral acids, 2) filtering or centrifugation process, 3) re-soaking of the residue using an alkaline solution, 4) precipitation process, and 5) purification. Emulsifiers are molecules that adsorb on the surface of the droplets formed during the homogenization process and form a protective membrane that keeps the droplets from agglomerating. The use of alginate as an emulsifier in food has been carried out by several researchers. The results indicate that alginate substitution in food products has a significant effect on emulsion stability, where the stability of emulsions using alginate is higher than that of emulsions without alginate substitution. In addition, there was a positive interaction between alginate-substituted food products and food products without alginate substitution (control) on storage time. This research aims to provide an overview of the method or process of extracting alginate from brown seaweed (Phaeophyceae) and utilizing alginate as an emulsifier for food.

Abstract

Diabetes mellitus is a metabolic disease characterized by high blood glucose levels. The cause of glucose control failure is decreased insulin production by pancreatic β-cells and insulin resistance. Both lead to the obstacle of glucose uptake into cells. The mechanism of glucose uptake into cells is crucial in carbohydrate metabolism. This mechanism aims to produce energy in the form of ATP. The main signaling pathway after the process of glucose uptake is the PI3K/Akt pathway. This pathway involves many proteins activated by phosphorylation mechanisms. One of the proteins involved in this pathway is PTEN, a PI3K regulator. PTEN activity can dephosphorylate PI3K so that the insulin signaling pathway becomes blocked and glucose cannot be uptaken into cells. It causes blood glucose levels to increase. The role of PTEN in inhibiting the PI3K/Akt pathway seems to be a crucial matter to observe. By inhibiting PTEN activity, the insulin signaling pathway is expected to work properly. We have searched, read, analyzed, and summarized various studies regarding the potential of PTEN to reduce diabetes mellitus cases. In some research articles, the use of active compounds and therapy using stem cells to inhibit PTEN activity has shown good progress in the insulin signaling pathway. Based on these, it can be an option to make PTEN a target for diabetes mellitus therapy.

Abstract

Ras-like proteins from rat brain (Rab) is one of the crucial proteins in the process of intracellular transport. Rab plays a role in delivering vesicles from the donor membrane to the acceptor membrane. Vesicles contain various molecules that are important in cell growth and development. These molecules are generally involved in some signaling pathways. In carrying out its functions, Rab needs to be activated first. The activation process of Rab involves various proteins. Thus, Rab activation and deactivation control are critical to regulating the movement of the transferred molecules. In cancer, there is an uncontrolled activation and deactivation of Rab. Rab's involvement in various signaling pathways by providing molecules needed for growth and development is a basis for increasing the speed of growth and development of cancer cells. Although Rab is not the main factor causing cancer, it can help the development of cancer. So, it can be one of the keys to controlling the transfer of molecules involved in the growth and development of cancer cells. There are several methods related to the activation of Rab for molecular transport. Rab activation inhibition by Rab inhibitors and inhibitors of Rab activating proteins and the use of miRNAs can prevent molecule transport. Thus, control of Rab activation can be an alternative to suppress the growth and development of cancer cells.
