Abstract

ABSTRACTIntroduction: Breast cancer is a multifactorial disease as a result of interaction between ge- netic and environmental factors such as hormonal, infection, chemical material, and radiation. Until now, it took as the most occurred of cancer in women, which have the highest mortality in the world. However, its cause still unknown. According to epidemiologic and clinical - laboratory studies, it was found that there are many risk factors which have important roles in female breast cancer like menarche, menopause, endogenous and exogenous hormones, family his- tory (genetics), parity, lactation, obesity, physical activity, diet, alcohol, smoking, environmental factors and history of biopsy and breast examination. The aims of this study to evaluate and compare the clinicopathological features in the four breast cancer subtypes defined by immu- nohistochemistry expression of ER, PR, and HER-2: ER/PR+, HER-2+ (Luminal B); ER/ PR+,HER-2- (Luminal A); ER/PR-, HER-2+ (HER-2 type) and ER-/PR-, HER-2- (Triple nega- tive) type. Methods: This study was a retrospective study of 89 invasive breast cancer. Clini- cal and pathologic features of the four subtypes were compared. Results: ER/PR+, HER2+; ER/PR+, HER2-; ER/PR-, HER2+; and ER/PR-,HER2- types were 10 (11.2%), 35 (39.3%), 27 (30.3%), and 17 (19.1%) samples. Subjects with ER/PR-, HER2- where are likely to be younger (p<0.001). In tumor subtypes with HER2+, the number of subjects with poorly differentiated was larger than the total number of well and moderate differentiated (p < 0.001). There was no significant difference of metastatic to lymph node status in all subtypes. Conclusion: Subtype luminal A of breast cancer have a high number than other subtypes. There was a correlation between overexpression of HER-2 with poorly differentiated of breast cancer but not with a metastatic capability of tumor cells to lymph nodes. Keywords : Luminal A, Luminal B, Her-2 type, Triple negative, breast cancer

Abstract

Objective: This study aimed to determinelevels of expression ET-1 in serum and renal tissue associated withchanges in kidney structure.Method: The rats were divided into two groups. The Obese group was given a high-fat-induced diet chowand a standard diet chow for the control group. ET-1 protein expression and localization in the glomerulusof the kidney were examined. The serum of blood checked for ET-Iusing ELISA method and assessed theET-1 expression of the glomeruliby Immunohistochemistry (IHC). Mesangial expansion is an indicator toevaluate the degree of glomerular damage by Periodic Acid–Schiff(PAS staining). Associations between theimmunoreactivity for ET-1 in glomeruli and the degree of kidney injury were examined.Results: Lee Index was significantly higher in the obese group than in the control group (p<0.05). ET-1immunoreactivity was considerably higher in the obese group than the control rat. It was a strong correlationwith enhancingmesangial expansion glomeruli using PAS staining of the renal tissue (rs=0.765; p= 0.001).Contrast to ET-1 levels in the serum revealed no significant in obese rat groups (p>0.05).Conclusion: These data confirm the role of ET-1 will provide changes in the levels of glomeruli impairment,but serum levels may well fail to reflect the local expression at the tissue level.

Abstract

Penelitian invitro tentang aktivitas antiproliferatif daun Parang Romang (Boehmeria Virgata (Forst) Guill) telah diteliti pada sel Hela, untuk itu perlu dilakukan uji toksisitas untuk menentukan dosis dan keamanan dari tanaman tersebut pada hewan uji. Salah satu paramater dari uji toksisitas adalah pengamatan terhadap gambaran histopatologi organ. Penelitian ini bertujuan untuk pengaruh setelah pemberian dosis tunggal dan berulang ekstrak etanol daun parang romang terhadap perubahan histopatologi pada organ hati dan ginjal dari tikus putih jantan dan betina (Rattus novergicus). Pada pemberian dosis tunggal tikus dibagi menjadi 3 kelompok yaitu kelompok kontrol, kelompok 2000 mg/kg BB, dan kelompok 5000 mg/kg BB kemudian dilakukan pengamatan selama 14 hari dan dikorbankan pada hari ke 15. Sedangkan pada pemberian dosis berulang selama 28 hari terdapat 4 kelompok, yaitu kelompok kontrol, kelompok 250 mg/kg BB, kelompok 500 mg/kg BB, dan kelompok 1000 mg/kg BB dan dikorbankan pada hari ke 29. Semua kelompok kontrol hanya diberikan akuades. Hasil dari penelitian menunjukkan terdapat pengaruh setelah pemberian dosis tunggal dan berulang ekstrak etanol daun parang romang (Boehmeria virgata Linn (Forst) Guill) terhadap gambaran histopatologi organ hati dan ginjal berupa degenerasi hidrofik dan pembesaran glomerulus dengan derajat kerusakan ringan  sampai sedang, baik pada tikus putih jantan maupun betina (Rattus novergicus).

Abstract

Mutasi pada protein anti-apoptosis BCL-2 merupakan salah satu penyebab terjadinya kanker payudara. Beberapa studi in vitro telah menunjukkan bahwa Eupatorium triplinerve Vahl, yang mengandung flavonoid, timohidrokuinon, terpenoid, karoten, vitamin C, stigmasterol, dan kumarin memiliki efek sitotoksik terhadap sel kanker payudara. Kumarin telah dilaporkan oleh berbagai penelitian berperan dalam berbagai jalur penghambatan terjadinya kanker. Tujuan penelitian ini adalah untuk melakukan dan menilai docking dari turunan senyawa kumarin yang terdapat pada tanaman Eupatorium triplinerve Vahl, yakni ayapanin dan ayapin, terhadap protein BCL-2. Docking dilakukan dengan menggunakan program command Autodock Vina pada personal computer (PC) dengan sistem operasi Windows 10 64-bit. Struktur protein BCL-2 dengan kode PDB 6QGG diunduh dari RCSB Protein Data Bank dan struktur 3D dari ayapanin dan ayapin diambil dari situs PubChem. Proses moleculer docking dilakukan secara bertahap dari penyiapan ligan dan protein, validasi metode docking, proses docking, hingga analisis data. Energi ikatan yang diperoleh dari docking antara senyawa ayapanin dan ayapin terhadap protein BCL-2 adalah -6.3 kcal/mol dan -6.9 kcal/mol dibandingkan dengan energi ikatan ligan bawaan, yakni -10.3 pada kantung aktif yang sama. Dengan demikian, ayapanin dan ayapanin memiliki potensi sebagai senyawa inhibitor protein anti-apoptosis BCL-2.

Abstract

Abstract Objective: This study was aimed to investigate the protective effect of Ajwa date palm extract (Phoenix Dactylifera L.) against liver damage in white rats (Rattus norvegicus) induced by Meloxicam. Methods: The samples used in this study were twenty five male white rats divided into five groups with body weight 200 – 300gr, namely a healthy control group, negative control given Meloxicam 30mg/kgBB, treatment group 1 was given Ajwa date fruit extract 75mg/kgBW and Meloxicam 30mg/kgBW, treatment group 2 was given Ajwa date fruit extract 150mg/kgBW and Meloxicam 30mg/kgBW, and treatment group 3 was given Ajwa date fruit extract 300mg/kgBW and Meloxicam 30mg/BW for 14 days. On the 15th day, the rats were anesthetized for blood tests and euthanized for liver organ observation. Results: The result showed by giving Ajwa date palm extract with higher doses and Meloxicam 30mg/kgBW, resulted a decrease in ALT and AST enzyme levels which were close to normal values ​​in rats. Histological observation showed extensive damage to liver cell tissue, with signs of inflammation, hemorrhage, necrosis, and hydropic degeneration. By giving high doses of Ajwa date palm extract and Meloxicam 30mg/kgBW, gave a view of a decrease degree on liver tissue damage. Conclusion: The extract of Ajwa date palm (Phoenix Dactylifera L.) can provide a protective effect against liver damage in white rats (Rattus Norvegicus) induced by Meloxicam with decreased levels of liver function enzymes which are Alananine Transaminase (ALT) and Aspartate Aminotransferase (AST), along in the changes of liver histopathological view which showed a decrease degree of liver cell tissue damage.
