Abstract

The aim of this research was to study the effect of Binahong extract on the growth of Nile tilapia. A number of 225 fish measuring 10-12 cm in length with an average weight of 15 g was obtained from Fishery Development Center and Training, Tateli. The fish were put  in oxygenated plastic bags and transported to Laboratory of Aquaculture Technology. The fish was kept in aquarium equipped with an aerator with a density of 15 fish per aquaria. The test material used was the stem of Binahong plant stem, macerated with alcohol 70% for 3 days. The extract was then filtered with whatman filter paper and extract was concentrated using a Rotary Evaporator. The extract was further diluted to obtain  concentrations of 12.5, 25, 50, and 100 mg / mL Binahong  extract as treatment was given to the fish after mixed first into the pellet. The fish was fed with treatment feed for 30 days. Control feed was not supplemented with binahong extract. Growth data was measured at day 30 after treatment. The results showed that the extract of binahong had significant effect on fish growth (p <0.01) with the best result achieved at concentration of 100 mg/mL. In conclusion, supplementation of  binahong extract into fish feed could improve growth of  fish in which the best growth was obtained in fish fed diet containing 100 mg/mL of binahong extract.Keywords: Medicinal plants, tilapia, Anredera cordifolia, growth, maceration

Abstract

This study was conducted to determine the effect of yeast as a counterweight of fibrous raw material in Tilapia  feed formulations on growth and feed efficiency value. The research results found that the biggest absolute growth of Tilapia fry occurred in treatment A: 5% yeast (9,2 g),  followed by  C: 15% yeast (8,6 g), D: 20% yeast (7,14 g), and B: 10% yeast (5,03 g). The highest feed efficiency value occurred in treatment A (23,75%), followed by C (22,45%), D (18,69%) and B (13,96%). The parameter of water quality measured during experiment included temperature 26-28o C and pH  7. The range value of water quality in each treatment was considered to be proper for the living condition of fish fry. Keywords: yeast, Oreochromis niloticus, growth, feed efficiency

Abstract

This study aimed to identify the bacterial disease in cultured eel elvers (Anguilla marmorata). The samples used for this study were taken from the Tatelu Freshwater Aquaculture Center. The samples taken were elvers (juvenile life stage of A. marmorata)  which showed clinical symptoms of bacterial infection such as bleeding in the chest, abdomen, and the base of the fin, passive movement, weakness, and loss of balance of the body, loss of appetite, some wounds on back fin, chest, and tail, and the body was not slippery/shiny. Three elvers with the size of 4-6 cm, and weigh about 0.2 g were taken as samples. The organ chosen to be targeted for isolation was head kidney. Each sample was isolated twice as replicates and streaked on Triptic Soy Agar (TSA) medium. First replicates were identified in the Tatelu Health Lab of Freshwater Aquaculture, and the second replicates were identified in North Sulawesi Provincial Health Lab. Bacterial identification was done through a series of observations and morphology of bacterial colonies through gram stain, followed by biochemical tests through oxidase test, catalase test, Triple Sugar Iron Agar (TSIA) test, H2S production test, motility test, O/F test, and Simmons Citrate Agar (SCA) test. Water quality parameters that were measured included temperature, disolve oxygen, pH, and ammonia. Based on identification of bacteria in cultured elvers in Tatelu Freshwater Aquaculture Center, the elver samples had been infected with bacterial  Aeromonas hydrophila. Abstract

The objetive of research was to examine the effect of yeast cell Saccharomyces cereviciae on non-specific immune response of nile tilapia, Oreochromis niloticus. Juveniles weighing 31.83 g in average were cultured in five concrete tanks with a density of 30 fish per tank. The fish were fed pellet supplemented with yeast cell at five different doses (0, 10, 20, 30, 40 g/kg pellet) for three consecutive weeks as much as 4%/bb/day, twice daily. At the end of feeding, three fish from each tank were sampled to measure the immune parameters namely total leucocyte count and phagocytosis activity of phagocyte cells. Research results showed that after three weeks of feeding, total leucocyte and phagocytosis activity of fish fed pellet supplemented with yeast cells were significantly different as compared to those of control fish.  The highest total leucocyte and phagocytosis activity were achieved in fish fed pellet supplemented with 10 g of yeast cells/kg pellet.  It was concluded that the use of yeast cells for three weeks could enhance the nonspecific immune response of nile tilapia. Abstract

The objective of this study was to determine the minimum concentration of extract in inhibiting motile motile aeromonad septicaemia. The material used was the stems and leaves of the plant P. grandiflora as much as 10 g. The crushed simplicia was then macerated with aquades with a ratio of 1:10,  then filtered with filter paper and, put in an Erlenmeyer, and evaporated in an incubator at temperature of 30°C. The sensitivity test was performed by placing a paper disc containing extract on the agar medium that had been scratched with bacterial colonies. Clear zone formed around the paper disc was compared with standard classification table for clarifying the inhibition level.  The results showed that stem and leaf extract had inhibition zone against Aeromonas hydrophila bacteria that caused motile aeromonad septicaemia. The best result was obtain at concentration of extract 3.25 mg / L) with an average diameter of 22.67 mm and for leaf extract had a moderate to strong zone for all treatments. These proved that stem and leaf extract of P. grandiflora plant had antibacterial activity and can be used as control of MAS disease caused by A. hydrophila bacteria. Abstract

The research aimed to study the growth  of seaweed Kappaphycus alvarezii cultured for 42 days  (from May to June 2016) in net pen with different initial weights at Talengen Bay, Island of Sangihe, North Sulawesi Province. The research used Complete Randomized Design with three treatments, each with three replication. The treatments included A: 50 g, B: 100 g, and C: 150 g of initial weight. Data collected were absolute, relative and daily growth.  Data were analyzed using analysis of variance and Least Significance Different Test was used to determine the difference between treatments.  Statistical analysis used JMP (SAS-institute). Research results showed the best absolute growth was achieved in treatment C namely 300.55 g, the best relative growth was observed  in treatment A as much as 284.78% and the best daily growth rate was in treatment A as much as 3.26%. Abstract

The research intends to determine the optimal concentration of honey addition in sperm dilution on sperm motility, fertilization and eggs hatchability of Nile Tolapia (Oreochromis niloticus). Experimental design used was a Complete Randomized Design (CRD) with three times repetition of four treatments. The concentration of honey addition used for treatment are A (0 ml of honey in 100 ml of physiological NaCl), B (0.60 ml of honey in 99.40 ml of physiological NaCl), C (0.65 ml of honey in 99.35 ml of physiological NaCl), D (0.70 ml of honey in 99.30 ml of physiological NaCl). Parent used consisted of five female parent and five male parent with 250 – 500 mg weight. Sperm dilution containers used was 12 plastic cups and the container fertilization and hatching eggs container used was 12 brass plates.  Spermatozoa motility observation performed immediately after dilution process. Fertilization observation was performed after 12 hours of fertilization process, and Egg Hatchability observation was performed after 60 hours of fertilization.  The observation results showed that the addition of honey in sperm dilution contributed significant effect on spermatozoa motility, fertilization and eggs hatchability of Nile Tolapia (Oreochromis niloticus). This observation showed that the addition of 0.70 ml of honey in a sperm dilution was the best treatment seen from the average percentage of spermatozoa motility (73,33 %), fertilization (80,00%) and eggs hatchability (77,33 %.) Abstract

Field practice activities conducted in laboratory incluiding the process of carcass removal from its shells was done in the Laboratory of Nutrition and Fish Feed Technology, and also analysis of Kijing Taiwan carcass chemical composition was done in the Research and Industrial Standardization Institute of Manado. The purpose of field practice are: Analyzing the Proximate composition, Vitamin C, Calcium, and Energy of Kijing Taiwan carcass taiwan in wet and dry form. Providing scientific information about the nutrient content of Kijing Taiwan in wet and dry form. Acquire skills in operating the Proximate instruments of Vitamin C, Calcium, and Energy in the laboratory. Based on the results of the Kijing Taiwan (Anodonta woodiana) carcass proximate analysis  in wet form, the data showed, 11.59% of protein, 80.66% of water, 0.26% of fat, 3.06% of ash, 0.23% of crude fiber, 4.20% of carbohydrate, 65.5 kcal of energy, 0.59% of calcium, 3.86 mg/100 g of Vitamin C. While the dry form of the data showed, 54.84% of protein, 12.18% of water, 4.08% of fat, 14.92% of ash, 0.53% of crude fiber, 13.45% of carbohydrates, 309.88 kcal of energy, 0.55% of calcium, 14.88% of vitamin C mg/100 g. Abstract

This study aims to determine the effect of carrot extract on the phagocyte activity of Vibrio alginolyticus in white snapper. This study used a completely randomized design (CRD) with 4 treatments and 3 replications. The treatments used respectively as follows, treatment A: 0 µl extract/individu, B: 100 µl extract/individu, C: 150 μl extract/individu, and D: 200 μl extract/individu. The average value of phagocytic activity tended to increase, namely treatment A 8.67%; treatment B 9.33%; treatment C 11.00%; and treatment D 14.00%. Carrot extract can increase phagocytic activity of white snapper. An effective dose that can increase the immune response of white snapper was 200 µl extract/individu of fish.

Abstract

ABSTRACT The objective of this research was to analyse the water physical-chemical qualities at aquaculture area in Lake Tondano North Sulawesi. Water quality was measured in situ and in laboratotium. In situ measurement included temperature, pH, and turbidity, while in laboratory, water quality measured included dissolved oxygen, nitrogen, phosphate. It was found that water temperature ranged from 25 - 27 o C, turbidity 0.20 - 4,0 m, and dissolved oxygen 5 – 9 mg/L, Nitrogen 0,0334 - 0,0697 mg/ L, phosphate 0 - 0,0219 mg/ L. It was concluded that water quality around Paleloan Village was appropriate for aquaculture activity. Keywords: water quality, aquaculture, Paleloan Village
