Animal Production is a peer-reviewed journal published by the Faculty of Animal Science, Jenderal Soedirman University in association with the Animal Scientist Society of Indonesia. The journal was established in 1999 and available online since May 4, 2011. Animal Production was initially published twice a year, then three times a year starting in 2005. The journal coverage focused on small scale livestock farming and technologies which includes the recent science development in animal production, including all aspects of nutrition, breeding, reproduction, post-harvest processing and socio-economics. All articles are available online. Full text available in pdf format and can be downloaded for free. Abstract

Abstract.  This study was aimed to determine the influences of turmeric extract supplementation on water holding capacity, cooking loss, pH value and tenderness of broiler chicken meat Data analysis was subject to completely randomized design 5 treatments namely T0, T1, T2, T3 and T4 containing non-turmeric extract, 100 mg/kgBW/day, 200 mg/kgBW/day, 300 mg/kgBW/day and 400 mg/kgBW/day, respectively. Each unit of experiment administered 3 heads with four replications. The results indicated no effect from turmeric extract supplementation on water holding capacity, cooking loss, pH value and tenderness of broiler chicken meat. The average treatments of T0, T1, T2, T3, T4 had water holding capacities of  39.86, 37.58, 36.41, 36.94, respectively; cooking losses of 26.00, 27.58, 27.57, 27.11, and 27.49%, respectively; tenderness of 1.97, 1.95, 1.63, 1.77 and 1.99 Nmm, respectively, and final Body weights of 1,618.5, 1,568, 1,692.5, 1,651.75 and 1,462 g/head, respectively. However, a highly significant influence was observed on the pH values of 6.46, 6.04, 6.21, 6.08 and 5.98. The results indicated that none of the turmeric extract supplementation increased water holding capacity, cooking losses, tenderness and body weight. Key words: broiler, cooking loss, pH values, tenderness, water holding capacity, turmeric Abstrak. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak kunyit terhadap daya ikat air, susut masak, nilai pH dan keempukan daging ayam broiler. Manfaat penelitian yaitu tersedianya informasi ilmiah tentang ekstrak kunyit terhadap danging ayam broiler. Perlakuan yang diterapkan adalah T0 tanpa ekstrak kunyit, T1 100, T2 200, T3 300, dan T4 400 mg/kgBB/hari. Data analisis yang digunakan adalah rancangan acak lengkap yang terdiri dari 5 perlakuan dan 4 replikasi. Setiap unit percobaan terdiri dari 3 ekor. Hasil penelitian menunjukkan tidak ada pengaruh pemberian ekstrak kunyit terhadap daya ikat air, susut masak, dan keempukan daging ayam broiler. Rataan untuk perlakuan T0, T1, T2, T3, T4 pada Daya Ikat Air masing–masing 39,86; 37,58; 36,41; 36,94; 34,78%; susut masak 26,00; 27,58; 27,57; 27,11; 27,49%, keempukan 1,97; 1,95; 1,63; 1,77; 1,99 Nmm, dan bobot badan akhir 1.618,5; 1.568; 1.692,5; 1.651,75; 1.462g/ekor. Namun, memberikan pengaruh sangat nyata pada nilai pH 6,46; 6,04; 6,21; 6,08; 5,98. Hasil  menunjukkan bahwa pemberian ekstrak kunyit  tidak meningkatkan daya ikat air, susut masak, keempukan dan bobot badan. Kata Kunci : Broiler, Susut Masak, pH, Keempukan, Daya Ikat Air, Kunyit

Abstract

Abstract. This study aimed to describe the sustainability of forage system in the small holder dairy cattle  in the plateau in East Java, in particular related to the nutrient content. The method used was survey (interviews, questionnaires, field observations and sampling) at the cooperation unit, farmers, livestock and farming location in one of the areas of dairy cattle cooperation in the plateau (Cooperation of SAE Pujon-Malang). The data obtained were analyzed through descriptive and regression statistics. The results showed that forage system dominantly given during dry and rainy seasons are elephant grass and corn stalks. Linear regression equation for the nutrient content of elephant grass is TDN= 40.516 + 1.404 CP, while corn trees is TDN= 56.212 + 0.740 CP. The conclusion showed that the dependent variable is largely influenced by external factors (environment). Improved continuity of availability of forage can be done by increasing the feeding system in the region (plateau) as well as the support from outside the region. Key words: plateau, dairy cattle, forage  Abstrak. Penelitian ini bertujuan untuk mendiskripsikan keberlanjutan sistem pakan hijauan pada  peternakan sapi perah rakyat di wilayah dataran tinggi di Jawa Timur, khususnya tentang kandungan nutrisi. Metode yang digunakan adalah survei (wawancara, pengisian angket/kuesioner, observasi,  dan pengambilan sampel) di koperasi, petani ternak dan lokasi peternakan di salah satu wilayah koperasi persusuan di dataran tinggi (Koperasi SAE Pujon-Malang). Data yang didapat dianalisis dengan regresi dan statistik diskriptif. Hasil penelitian menunjukkan bahwa pakan hijaun yang dominan di  musim kemarau dan hujan adalah rumput gajah dan tebon jagung. Persamaan regresi linier untuk kandungan nutrisi rumput gajah adalah TDN= 40,516  + 1,404 PK,  sedangkan tebon jagung adalah TDN= 56,212 + 0,740 PK. Kesimpulannya adalah variabel dependen sebagian besar dipengaruhi oleh faktor luar (lingkungan). Peningkatan kontinuitas ketersediaan pakan hijauan dapat dilakukan dengan peningkatan sistem pakan  di wilayah (dataran tinggi)  dan dukungan sistem pakan dari luar wilayah. Kata kunci: dataran tinggi, sapi perah, pakan hijauan

Abstract

Abstract. The objective of this research was to evaluate the hydrolyzed chicken feather based on pepsin digestibility and nutrient content, after physico-chemical and biological process. It was carried out by experimental methods at feed and nutrition laboratory. The treatments were hydrolyzed feather meals immersed in 0.5% NaOH and Na2S solution for 0, 2, 4, 6 and 8 hours, each treatment was repeated three times. The results showed that chemical treatment (NaOH-Na2S) in various time of incubation at 60oC followed by fermentation using Bacillus sp. MTS at 37oC for four days decreased the protein of hydrolyzed feather (78.88 to 73.06%), but increased the keratin fiber (1.9 to 3.26%). Pepsin digestibility informed that the increasing incubation time from 0, 2, 4, 6 to 8 hours resulted in higher solubility than that of control (30.2% at 8 hours vs 15.4% at 0 hours). Processing chicken feather  by  0.5% NaOH and Na2S solution at 60oC for 6 hours followed by fermentation increased the value of pepsin digestibility.  Key words: hydrolyzed, Bacillus sp. MTS, feather, solubility Abstrak. Penelitian ini bertujuan mengevaluasi kualitas nutrien tepung bulu ayam hasil proses hidrolisis secara fisiko-kimia dan biologis menggunakan Bacillus sp. MTS. Metode eksperimental digunakan dalam penelitian yang menggunakan dua tahap proses hidrolisis yaitu tahap 1: setelah perebusan bulu dalam larutan NaOH maka bulu direndam dalam larutan  0.5% NaOH dan Na2S pada 600C dan tahap 2: fermentasi bulu selama empat hari pada suhu 370C. Perlakuan berupa waktu inkubasi yaitu 0, 2, 4, 6 dan 8 jam diterapkan pada tahap kedua dengan ulangan sebanyak tiga kali. Perlakuan fisiko-kimia yang dilanjutkan fermentasi menggunakan bakteri spesifik penghasil enzim-enzim pendegradasi keratin bulu menurunkan kadar  protein tepung bulu  (78,88% menjadi 73,06%) dan meningkatkan kadar serat tepung bulu (1,9 menjadi 3,26%). Uji kelarutan protein tepung bulu dalam pepsin menginfromasikan bahwa proses tahap 1 menghasilkan nilai kelarutan protein tepung bulu yang meningkat dua kali dibanding kontrol (30,2% pada 8 jam vs 15,4% pada 0 jam inkubasi) atau enam kali dibanding tepung bulu tanpa hidrolisis (5%). Pengolahan bulu ayam menggunakan cara pemanasan, perendaman dalam larutan NaOH dan Na2S selama 6 jam pada 600C serta fermentasi menghasilkan tepung bulu dengan daya larut dalam pepsin  lebih baik dibanding tanpa pengolahan.  Kata kunci: hidrolisis, tepung-bulu, Bacillus sp. Abstract

Abstract. The aims of this research were to describe the characteristics of Kaligesing goat farmers; to analyze the farmers’ perceptions on the role of extension workers as conduit of information, as mentors, organizers and dynamic factor, technicians and liaisons; to discover the relationship between the farmers’ characteristics with farmers’ perceptions; and to investigate the relationship between farmers’ perceptions to goat maintenance management. The respondents were goat farmers in Kaligesing, Purworejo, Central Java Province. Data were obtained from questionnaire survey method. Determining location, the research applied combination of stratified sampling method with purposive random sampling. Total respondents were 159 farmers with nine farmer groups as samples. Analysis was subject to Spearman Rank, resulting that age, education level and farming experience were not significant to the farmers’ perceptions to the role of extension workers, but the ownership of livestock had a very significant relationship with a correlation coefficient of 0.240, group classes had also very significant relationship with a correlation coefficient of 0.414, and frequency of meeting with extension workers have a significant relationships with a correlation coefficient of 0.202. Farmers’ perceptions to the role of extension workers had very significant relationships to the maintenance management with a correlation coefficient of 0.393. Key words : farmers’ characteristics, farmers’ perceptions, Kaligesing goat, role of extension workers Abstrak. Penelitian ini bertujuan untuk mengetahui karakteristik peternak, persepsi peternak terhadap peran penyuluh, hubungan antara karakteristik peternak dengan persepsi, dan hubungan antara persepsi peternak dengan manajemen pemeliharaan. Responden penelitian adalah peternak Kambing Kaligesing di Kecamatan Kaligesing Kabupaten Purworejo Jawa Tengah dengan metode penelitian survei kuesioner. Penentuan lokasi penelitian dengan kombinasi cara stratified sampling dan purposive random sampling. Jumlah kelompok tani sebagai sampel penelitian sebanyak sembilan kelompok dan jumlah total responden sebanyak 159 peternak. Analisis Rank Spearman digunakan untuk analisis data.  Hasil analisis menunjukkan  bahwa umur, tingkat pendidikan dan lama beternak tidak secara nyata memiliki hubungan dengan persepsi peternak terhadap peran penyuluh, sedangkan kepemilikan ternak memiliki hubungan sangat nyata (P<0,01) dengan nilai koefisien korelasi 0,240. Kelas kelompok memiliki hubungan sangat nyata (P<0,01) dengan nilai koefisien korelasi 0,414 dan frekuensi bertemu penyuluh memiliki hubungan nyata (P<0,05) dengan nilai koefisien korelasi 0,202. Abstract

Abstract.  In Congo, waterfowl genetic resources are constituted by native population of Muscovy ducks that play an important role in food security. The present study aimed to identify and to characterize strains bred in the back yard in the households in Brazzaville. A sample of 154 households drawn over seven districts of Brazzaville was enrolled in the survey. Adults ducks found in the households were identified, pictured by a key of determination and then compared by using the multi resolution analysis image method. The survey recorded 13 strains in which four were considered as newly since they have never been reported elsewhere. These strains received temporally the name of the districts where they have been identified for the first time Makelékélé 1 (0.34%, n=6), Makélékélé 2 (0.11%, n =2), Poto poto 1  (0.28%, n=5) and in Poto poto 2 (0.11%, n=2). Finally, the survey reported nine classical  strains such as  black plumage, duclair, white, tortora, sepia, chocolate, lavender, grey and canizie. The apparent wide variation in plumage colors is an indication that the duck populations have not been ‘purified’ through selective breeding. In the context of the valorization of poultry biodiversity, this work represents a step toward a better knowledge of the production abilities of local ducks breeds in Congo. Key words: Muscovy ducks, color feather, strains, Congo. Abstrak.  Sumber daya genetik unggas air di Kongo mencakup populasi itik lokal yang memegang peranan penting dalam ketahanan pangan. Penelitian ini bertujuan mengidentifikasi dan menggolongkan jenis itik yang dipelihara di pekarangan rumah di Brazzaville. Sampel penelitian menggunakan 154 responden rumah tangga yang tersebar di 7 wilayah Brazzaville. Itik dewasa diidentifikasi dari pekarangan, dan dibandingkan dengan metode Analisis Multi Resolusi. Survey mencatat 13 jenis peranakan, 4 diantaranya dianggap baru karena belum pernah dilaporkan di studi manapun. Jenis ini sementara dinamai sesuai distrik tempatnya pertama ditemukan, yaitu Makelékélé 1 (0,34%, n=6), Makélékélé 2 (0,11%, n =2), Poto poto 1  (0,28%, n=5) dan di Poto poto 2 (0,11%, n=2). Berdasarkan survei didapatkan sembilan jenis klasik yaitu bulu hitam, duclair, putih, tortora, sepia, coklat, lavender, abu-abu dan canizie. Banyaknya ragam warna bulu adalah indikasi bahwa populasi itik belum “dimurnikan” melalui seleksi. Dalam konteks penetapan nilai keanekaragaman hayati unggas, penelitian ini mewakili sebuah langkah menuju pengetahuan yang mendalam akan kemampuan produksi itik yang berkembang di Kongo. Kata kunci: itik Muscovy, warna bulu, strain, Kongo

Abstract

Abstract. The experiment was conducted to determine the effect of complete diet silage on feed consumption, body weight gain, feed conversion, water consumption and mortality. The materials used were 75 Mojosari Alabio male ducks, commercial diet plus rice bran (16.50% CP, 2900 kkal GE/kg), silage with 30-60% water content. Data were subject to Completely Randomized Design with five treatments and three replications. The treatments were S0 (commercial feed), S1 (silage with 30% water content), S2 (silage with 40% water content), S3 (silage with 50% water content) and S4 (silage with 60% water content). Data were subject to analysis of variance followed by orthogonal contrast test. The result showed that silage with 50% water content has significantly increased body weight gain and decreased water consumption, but there were no  effect in feed conversion. Complete diet silage was safe for Mojosari Alabio male duck. Key words : complete ration silage , duck, performance Abstrak. Penelitian ini untuk mengetahui pengaruh pemberian silase ransum komplit terhadap konsumsi pakan, pertambahan bobot badan, konversi pakan, konsumsi air dan persentase kematian. Bahan yang  digunakan adalah 75 ekor itik Mojosari Alabio jantan, ransum komersial ditambah dedak padi (16,50% CP, 2900 kkal GE/kg), silase dengan kadar air 30-60%. Percobaan ini menggunakan Rancangan Acak Lengkap dengan lima perlakuan dan tiga ulangan. Perlakuan S0 (pakan komersial), S1 (silase dengan kadar air 30%), S2 (silase dengan kadar air 40%), S3 (silase dengan kadar air 50%) dan S4 (silase dengan kadar air 60%). Data dianalisis menggunakan analisis ragam dan dilanjutkan dengan uji kontras orthogonal. Hasilnya menunjukkan bahwa silase dengan kadar air 50% memiliki peningkatan berat badan dan penurunan konsumsi air yang signifikan, tetapi tidak mempengaruhi konversi pakan. Abstract

Abstract. The aim of the study was to know the genetic characteristic and polymorphysm of Indonesian local ducks including Magelang, Tegal, Mojosari, Bali and Alabio duck based on Single Nucleotide Polymorphism (SNP) analysis in D-loop region mtDNA. The long term aim was to set the spesific genetic marker based on SNP D-loop region mtDNA which could differentiate local ducks in Indonesia. In the future, it could be used as selection tool for local duck conservation, and refinement strategy as well as the improvement of genetic quality by utilizing the available native duck germplasm. There were 20 ducks for each duck population and were taken 3 ml of its blood as sample. DNA Isolation Kit high pure PCR template preparation (Geneaid) was uded for Genome DNA isolation.  Amplification with PCR technique used primer DL-AnasPF (L56) as forward and DL-AnasPR (H773) as reverse. Next, PCR product or amplicon were sequenced. Sequence result were analyzed with SNP technique and observed the similarity and difference of its nucleotide sequence between individual and population. The result of the study showed that genome DNA from local duck in Indonesia was successfully isolated. DNA fragment of 718 bp was amplified with primer pair of DL-AnasPF and DL-AnasPR. Nucleotide sequence was 469 nt and analyzed with SNP technique. It was compared with standard nucleotide sequence of Anas platyrhynchos (HM010684.1) in Gen Bank. The result of nucleotide sequence similarity percentage was 99.68±0.56%. Single Nucleotide Polymorphism D-loop region mtDNA Indonesian local duck was 0.32±0.56%.  Some SNP was found in Magelang duck C (Klawu blorok), F (Cemani black),  G (Gambiran), H (Jarakan kalung), I (Jowo plain) and K (Plain white) also Tegal duck 8, 1, 2, 5, 2, 8 and 2 SNP respectively. It could be concluded that polymorphic genetic characteristic similarity were existed in Indonesia local duck populations which was shown by its big standard deviation SNP in D-loop region mtDNA. Magelang duck with different feather color relatively more polymorphic to another local duck in Indonesia. Single Nucleotide Polymorphism which was achieved could be used as genetic marker that differentiate genetic characteristic of Indonesian local ducks.Key words:  genetic characteristic, local duck, Single Nucleotide Polymorphism (SNP), D-loop mtDNAAbstrak.  Penelitian ini bertujuan untuk mengetahui karakteristik genetik dan polimorfisme itik lokal Indonesia yaitu itik Magelang, Tegal, Mojosari, Bali dan Alabio berdasarkan analisis Single Nucleotide Polymorphism (SNP) daerah D-loop mtDNA. Tujuan jangka panjangnya adalah menetapkan marker atau penanda genetik berdasarkan SNP daerah D-loop mtDNA spesifik yang dapat membedakan itik-itik lokal yang ada di Indonesia. Selanjutnya digunakan sebagai  alat bantu seleksi untuk konservasi, pembibitan  dan pengembangbiakan itik lokal.  Populasi masing-masing jenis itik lokal yang digunakan sebanyak 20 ekor untuk diambil 3 ml sampel darahnya. Isolasi DNA genom menggunakan DNA Isolation Kithigh pure PCR template preparation (Geneaid). Amplifikasi dengan teknik PCR menggunakan pasangan primer DL-AnasPF (L56) sebagai forward dan DL-AnasPR (H773) sebagai reverse. Produk PCR atau amplikon yang diperoleh disekuensing. Hasil sekuensing dianalisis dengan teknik SNP dan diamati kesamaan dan perbedaan urutan nukleotida antar individu itik dan antar populasi.  Hasil penelitian menunjukkan bahwa DNA genom dari itik lokal di Indonesia berhasil diisolasi. Amplifikasi dengan teknik PCR berhasil memperoleh fragmen berukuran 718 bp. Urutan nukleotida hasil sekuensing sebesar 469 nt dianalisis dengan teknik SNP dan dibandingkan dengan urutan nukleotida standar dari itik Anas platyrhynchos (HM010684.1) yang ada di Gen Bank, diperoleh persentase kesamaan urutan nukleotid sebesar 99,68±0,56%. Single Nucleotide Polymorphism daerah D-loop mtDNA pada itik lokal di Indonesia sebesar 0,32±0,56%. Sejumlah SNP ditemukan pada itik Magelang C (Klawu blorok), F (Hitam cemani),  G (Gambiran), H (Jarakan kalung), I (Jowo polos) dan K (Putih polos) serta itik Tegal  masing-masing 8, 1, 2, 5, 2, 8 serta 2 SNP. Kesimpulan dari penelitian ini adalah terdapat karakteristik genetik yang polimorfik pada populasi itik lokal di Indonesia, ditunjukkan dengan adanya simpang baku SNP pada daerah D-loop mtDNA yang relatif besar. Abstract

Abstract. The effect of three forms of complete feed blocks (CFB) for dairy cattle was evaluated. The form of various CFB were cube, cylinder and ball.  The CFB was prepared from napier grass specific tolerance acid soils and concentrate in 50:50 ratio.  The research used experimental method with randomized block design.   Sixteen dairy cattle were used in this experiment.  There were four treatments and four replications used in this experiment.  The treatments consisted of R0= control ration, R1= cube CFB, R2= cylinder CFB and R3= ball CFB.  The treatment feeds were fed twice a day, at 6.00 am and 15.00 pm.  The amount of morning ration 34 kg of fresh napier grass and 5 kg of concentrate for the control ration, and the same amount of ration was also given in the afternoon feeding, therefore, the daily total fresh forage was 68 kg and concentrate was 10 kg.  Before feeding the forage was chopped in 5 cm length.  The complete feed block for the dairy cows was 10 kg for the morning ration and 10 kg for the afternoon ration, therefore, the daily total complete feed block was 20 kg.  The drinking water was available adlibitum.  The preliminary period was conducted for 2 weeks and data collection were conducted for 5 day of the end of study. The variables measured were dry matter and organic matter intake, dry matter digestibility and milk production.  These results showed that the control ration significantly affected with  ration all CFB form on dry matter and organic matter intake but did not significantly affect the dry matter digestibility and milk production. Key words : dairy cttle, complete feed block Abstrak.Penelitian ini bertujuan mengevaluasi pengaruh tiga pakan komplit bentuk cetak yaitu bentuk kubus, silinder dan bola yang diberikan pada sapi perah.   Pakan komplit bentuk cetak dibuat dari hijauan rumput gajah yang spesifik toleran tanah masam yang telah digiling dan bahan konsentrat dengan perbandingan 50:50.  Rancangan percobaan yang digunakan adalah rancangan acak kelompok.  Enam belas ekor sapi perah dikelompokkan menjadi 4 dengan 4 ulangan.  Empat perlakuan yang dicobakan yaitu R0= ransum kontrol, R1= pakan komplit cetak bentuk kubus, R2= pakan komplit cetak bentuk silinder, dan R3= pakan komplit cetak bentuk bola. Pakan perlakuan diberikan dua kali sehari pagi pukul 06.00 dan sore 15.00.  Jumlah pakan  yang diberikan pada sapi perah kontrol pagi sebanyak 34 kg rumput gajah segar dan 5 kg konsentrat demikian juga pemberian yang sama pada sore hari sehingga jumlah hijauan segar yang diberikan kepada ternak 68 kg dan konsentrat 10 kg.  Rumput gajah yang diberikan sudah dipotong-potong terlebih dahulu dengan panjang 5 cm.  Jumlah pakan komplit cetak untuk sapi perah perlakuan yang diberikan pagi jumlahnya sama dengan pemberian sore masing-masing sebanyak 10 kg sehingga jumlah pakan komplit cetak yang diberikan 20 kg.  Air minum diberikan adlibitum.  Periode preliminary dilakukan selama 2 minggu dan koleksi data dilakukan selama 5 hari periode akhir percobaan.  Variabel yang diamati adalah konsumsi bahan kering dan bahan organik pakan, kecernaan bahan kering pakan dan produksi susu.  Hasil penelitian menunjukkan bahwa ransum kontrol beda nyata dengan semua bentuk pakan komplit cetak pada konsumsi bahan kering dan bahan organik, akan tetapi tidak berbeda nyata dengan  kecernaan bahan kering dan produksi susu. Kata kunci : sapi perah, pakan komplit bentuk cetak

Abstract

Abstract. This study was aimed to determine and compare the dry matter yield and nutrient content of Indigofera and Leucaena grown in peatland. This experiment was conducted in peatland type soil (type sapric) in Pekanbaru city, DM yield and nutrient contents data were analyzed by 2x3 factorial design with 3 replication. Two treatments compared were Indigofera zollingeriana (Indigofera) and Leucaena leucocephala (Leucaena). Indigofera was proven significantly higher than Leucaena in all harvest regarding dry matter (DM) of leaf and stem of 29.9% and 25%, respectively, crude protein (CP) of 23.1% and 17.6%, respectively. While neutral detergent fibre (NDF) and acid detergent fibre (ADF) content of Indigofera leaf (35.9% and 25.1%, respectively) was significantly lower than those of Leucaena leaf (40.9% and 29.3%, respectively). It was concluded that the production and nutritive value of Indigofera zollingeriana was superior to Leucaena leucocephala in peatland (type sapric). Key words: Legume tree, Sapric, nutrient content, In vitro digestibiliy              Abstrak.  Penelitian ini bertujuan menentukan dan membandingkan bahan kering dan kandungan nutrisi Indigofera dan Leucaena yang tumbuh di lahan gambut. Percobaan ini dilakukan di lahan gambut di Pekanbaru menggunakan pola faktorial 2x3 dengan ulangan 3 kali. Dua perlakuan yang dibandingkan adalah Indigofera zollingeriana (Indigofera) dan Leucaena leucocephala (Leucaena). Indigofera terbukti secara nyata lebih tinggi daripada Leucana di semua periode panen, berkaitan dengan kandungan BK (bahan kering) daun dan batang berturut-turut 29,9% dan 25%, dan protein kasar (PK) 23,1% dan 17,6%. sedangkan neutral detergent fibre (NDF) dan acid detergent fibre (ADF) daun indigofera berturut-turut 35,9% dan 25,1%, lebih rendah secara nyata daripada daun Leucaena, yaitu 40,9% dan 29,3%. Disimpulkan bahwa produksi dan nilai nutrisi Indigofera zollingeriana lebih tinggi dari Leucaena leucocephala di lahan gambut (jenis saprik) Kata kunci: Pohon legume, Saprik, Kandungan Nutrisi, Kecernaan in vitro

Abstract

Abstract. This study was aimed to analyze the levels of catecholamine and cortisol as stress indicator by invasive and non-invasive methods. Twelve female cattle PO were used in this study and were collected from slaughter house in Yogyakarta. Catecholamine and cortisol level of urine were measured by Enzyme linked Immunoassay (EIA) method and Fourier Transform Infrared (FTIR), were statistically analyzed to determine the difference between pre slaughter and slaughter conditions. FTIR spectra were analyzed using chemo-metrics software.  These results showed that the concentration of urinary cortisol were 2.12±1.68 ng/dl of pre-slaughter and 7.58±3.89 ng/dl of slaughtered respectively. The levels of urinary catecholamine in pre slaughter and slaughter were 3.07±2.05 ng/dl  and 4.15±2.68 ng/dl respectively. In FTIR analysis showed the spectral separation between the different quadrants before and during slaughter. The result suggested the correlation between the results of the analysis using the EIA and FTIR spectra. It is assumed that the separation of the FTIR spectrum in line with the increased levels of cortisol catecholamine and samples. It can be concluded that FTIR can be used to analyze the status of stress in animals, especially in cattle. Key words: catecholamine, cortisol, non-invasive, FTIR Abstrak. Penelitian ini bertujuan untuk menganalisis kadar katekolamin dan kortisol sebagai indicator stress secara invasif maupun non invasif dan mengembangkan metode deteksi stress pada sapi. Penelitian menggunakan sampel urin sapi Peranakan Ongole (PO) yang diambil dari rumah potong hewan di Yogyakarta. Analisis kadar katekolamin (CA) dan kortisol (CO) dilakukan dengan menggunakan metode EIA dan FTIR. Data konsentrasi kortisol maupun katekolamin hasil pengukuran menggunakan metode EIA dilakukan analisis statistik untuk mengetahui pengaruh stres terhadap konsentrasi CA dan CO urine. Hasil penelitian ini menunjukan rata-rata konsentrasi kortisol urin adalah 2.12±1.68 ng/dl dalam kondisi prapenyembelihan dan 7,58±3,89 ng/dl pada saat penyembelihan. Sedangkan konsentrasi katekolamin urin pada saat prapenyembelihan dan penyembelihan secara berurutan adalah 3,07±2,05 ng/dl  dan 4,15±2,68 ng/dl. Analisis FTIR menggambarkan pemisahan spektral pada quadran berbeda antara sebelum dan saat penyembelihan. Berdasarkan hasil ini menunjukkan korelasi antara hasil analisis dengan menggunakan EIA dan FTIR, baik konsentrasi katekolamin dan kortisol dengan spektra FTIR dari sampel. Perbedaan konsentrasi CA dan CO urin prapenyembelihan dan saat penyembelihan sejelan dengan pemisahan spektra FTIR. Hal ini diyakini bahwa pemisahan spektrum FTIR disebabkan adanya peningkatan kadar katekolamin dan kortisol dari sampel. Dapat disimpulkan bahwa FTIR dapat untuk menganalisis status stres pada hewan khususnya pada sapi. Kata kunci : katekolamin, kortisol, non invasif, FTIR
