Abstract

Corona Virus Disease 2019 or COVID-19 is a new type of virus that attacks the respiratory tract and can cause death. Laboratory examinations play an essential role in diagnosing COVID-19 with a set of reagents or kits. Sampiand sampling is carried out with a nasopharyngeal swab or oropharyngeal swab. Positive samples of COVID-19 patients used in this study were converted into RNA at the COVID-19 Referral Clinic in Bekasi, after which volume optimization was carried out with a total volume of 5 µl, 8 µl, and 10 µl with the Logix Smart™ kit. The method in this study uses One-Step Real-time PCR. This method is the best method for carrying out several bear tests because it can reduce the possibility of sample contamination. The procedure is fast and has high sensitivity. The fluorescence detection used in this study was FAM with a specific target of COVID-19 RNA and ROX with a particular DNA target of RNase-P. This research was conducted to obtain optimal volume conditions under the manufacturer's standards in detecting the SARS-CoV-2 virus. The results of this study indicate that a total volume of 5 l is the optimal total volume for detecting the presence of the SARS- CoV-2 virus in samples taken from patients.

Abstract

Piper is a genus of the family Piperaceae which has a very large number, in the tropics and sub-tropics. There are about 22 species of Piper which are registered in medicinal herbs and world herbs, one of which is Java chili (Piper retrofractum Vahl.). The morphological characters of P. retrofractum are different in various planting centers, especially the leaf and fruit characters vary. Limitations of morphological characters encourage the use of molecular characters. The purpose of this study was to analyze the molecular character of Java chilli (P. retrofractum) on the island of Java using ISSR (Inter simple sequence repeat) markers. Molecular markers can provide relatively more accurate information because genetic traits tend to be stable in environmental changes and not influenced by age. Leaves from 12 different locations were isolated using the SDS method. Amplification was carried out using 12 ISSR polymers. Data is analyzed using the NTSYS version 2.02 program. The results showed that the amplification of P. retrofractum DNA produced 70 polymorphic DNA bands. The dendrogram results from 12 sampling locations showed that P. retrofractum on Java Island had a similarity coefficient of 0.69-0.93. This shows that genetic diversity based on ISSR markers from P. retrofractum Vahl is relatively low.

Abstract

The plantain squirrel (Callosciurus notatus) is a rodent of the family Sciuridae. The population of plantain squirrels in Indonesia has decreased every year. The 2019 Checklist of the Mammals of Indonesia reports that the plantain squirrel is in a low concentration status; thus, accelerated efforts in wildlife conservation, including integrating technology with conservation efforts, are necessary. This study aims to determine the morphological characteristic of the plantain squirrel spermatozoa. This research consisted of several stages: anesthesia of animal samples, surgery, a maceration method, observation of the spermatozoa morphology using William's staining, and morphometry of sperm using ImageJ 1.46r software. The results show that the plantain squirrel sperm has a slightly flat head shape, falciform with one apical hook, and a not too long tail. The average sperm head length is 1.87 ± 0.30 µm, the average sperm tail length is 5.48 ± 0.95 µm.

Abstract

One of the swamp waters is located in Bagan Bilah Village, Panai Tengah District, Labuhanbatu Regency, North Sumatra Province. These swamp waters store a lot of fish diversity that has been used by the surrounding community. Among them are fish from the Osphronemidae family. This study aims to determine the biodiversity of fish of the Osphronemidae family in the swamp waters of Bagan Bilah Village. Fish sampling was carried out for 3 months, starting from October to December 2021. Fish are caught using traditional fishing gear called tangkol. The caught fish are collected, counted in number and then identified by species. The research data is calculated for its ecological index using the 2010 version of the Microsoft Excel application, then the data is displayed in tabular form. Based on the results of this study obtained 5 species of fish family Osphronemidae (Belontia hasselti, Betta imbellis, Trichogaster trichopterus, Trichopodus pectoralis, and Trichopsis vittatus). The Diversity Index (H') of Station 1 (0.78), Station 2 (0.76), and Station 3 (1.26). Uniformity Index (E) for Station 1 (0.43), Station 2 (0.42), and Station 3 (0.70). Dominance Index (C) for Station 1 (0.13), Station 2 (0.16), and Station 3 (0.28).

Abstract

Red-green color blindness is the most common type of color blindness. Color blindness disorders occur due to genetic factors determined by the recessive gene c (color blind) which is linked to the X chromosome. This study aims to determine the prevalence and frequency of color blind alleles and to determine the pattern of inheritance of color blind genes by conducting pedigree analysis on color blindness student’s family. The research was conducted in 13 public elementary schools, Sumbersari subdistrict, Jember Regency with a multistage cluster sampling method of sampling with a total sample of 286. Detection of color blindness using Ishihara's methods. Based on the results of this study, the color blindness prevalence of elementary school students in Sumbersari subdistrict, Jember was 2.79% with the prevalence in males and female were respectively as 4.86% and 0.7%. The allele frequency of color blindness was 0.021 and the normal allele frequency was 0.979. The pedigree analysis of a color blind student family shows the gene of color blindness is inherited in a criss-cross inheritance pattern.

Abstract

Depok Beach is a sandy beach that has limited vegetation because of strong winds, poor microclimate, and low availability of nutrients. One of the plants that can adapt to these extreme conditions is Ipomoea pes-caprae (L.) R. Br. One form of its adaptation to extreme environments is thickening the leaves. Other forms of adaptation can also be studied through the characteristics of the epidermis and the epidermal derivatives of the leaves, as the focus of this study. This type of research is exploratory research, including sampling, plant identification, leaf preparation using the leaf clearing method, and observing microscopic preparations. The data analysis used is descriptive quantitative. The data obtained include the characteristics of the epidermis and its derivatives in the form of stomata and trichomes. The results showed the average number of epidermal cells in the adaxial part of the leaves higher, that is equal to 54.05 ± 2.31 / 0.06 mm2. Stomata are parasitic and amphistomatic. The size of the stomata and the width of the stomatal pore on the adaxial surface of the leaf is smaller but has a higher average density, which is 5.80 ± 1.15 / 0.06 mm2. The trichomes found were of the glandular type.

Abstract

Sponge plastic is one of the plastic wastes that is difficult to degrade. Recently research reported that Tenebrio molitor L larvae have the potential capability to degrade plastic waste. But still a lack information according to the effect of feed composition on survival, and growth of T. molitor L larvae. The purpose of this study was to determine the effect of plastic feed composition on the rate of degradation of sponge plastic waste, survival, and growth of T. molitor L larvae. The method used in this study was an experimental laboratory using Completely Randomized Design (CRD) and using controls and six treatments with 3 repetition times. The parameters measured in this study were survival (%), body length growth (cm/week) and body biomass (gram/week), and degradation rate (gram/week). The data obtained from the research results will be analyzed using ANOVA which is then tested by Duncan. The results showed that feed composition had a significant effect (p=0.001) on the rate of degradation of plastic sponge waste, survival (p=0.000), and growth (p=0.001) of T. molitor L larvae. degradation, survival, and growth of T. molitor L. larvae.

Abstract

Honey is one of the liquid foods known for its high nutritional value. Honey contains macronutrient and micronutrient compounds. The composition of honey is usually different from one honey to another and has different biological activities. Baduy honey is one of the forest honey sold by the Baduy tribe. Fermented Baduy honey is suspected of having better biological activity. This study aims to determine the antioxidant activity of fermented Baduy honey, the total phenolic compound, and FTIR analysis. The antioxidant activity was determined against DPPH (2,2-Diphenyl-1-picrylhydrazile) by measuring the absorbance of the sample at the wavelength of 517 nm. The Total phenolic compound was also determined using folin ciocalteu’s method to measure the absorbance of the sample at the wavelength of 765nm. The FTIR analysis was also carried out to detect functional groups in Baduy honey. The antioxidant activity shows that fermented Baduy honey has the highest antioxidant capacity (IC 50 = 90,26 µg/mL) after three days of fermentation with pineapple and has better antioxidant capacity than non-fermentation Baduy honey. Total phenolic content shows that Baduy honey has 2,04 µg/mL. The FTIR analysis result indicates the presence of phenolic compounds, which can be found in flavonoids.

Abstract

Malaria is a public health threat caused by the Plasmodium infection transmitted by the Anopheles mosquito. Malaria vector control is highly dependent on the ability to determine mosquito species' vectorial and bionomic capacity. Species identification based on morphological characteristics as well as DNA-barcoding approaches is a very important step in determining vectorial capacity. Our research group has redesigned a new ITS2 primer, namely, sma-its2, which is specifically able to identify the Anopheles (An.) mosquito vector but cannot identify other mosquito vectors. This study wanted to test this primer's specificity further for identification of other Anopheles mosquitoes. We used An. minimus collected from Kulonprogo, Yogyakarta – Indonesia. The methods used in this research are as follows: landing collection, morphological identification, isolation of genomic DNA, PCR (Polymerase Chain Reaction), PCR product purification, sequencing, and data analysis. An. vagus from Bangsring, Banyuwangi – Indonesia, which had previously been identified using the same primary, was used as a positive control. The results of the morphological analysis showed that both species were in accordance with the vector identification key used in this study. The molecular analysis showed that the sma-its2 primer could amplify the ITS2 sequence of An. vagus and An. minimus, producing 650 – 700 bp. However, further analysis of the ITS2 sequences of both species, resulted in the same species, namely An. vagus, with a different accession number in GenBank. This showed that the sma-its2 primar can be used to identify An. vagus but cannot be used to identify An. minimus. Analysis of the primer position in the ITS2 sequences showed the presence of 3 nucleotides in the forward sma-its2 primer that was not recognized by the An. minimus sequences and thus, hinder the successful identification of these species.

Abstract

Coffee liquid waste contains organic matter which can be at risk of increasing water pollution levels. The results of the preliminary research carried out showed that the level of pollution due to coffee liquid waste exceeded the quality standard, with the results of the BOD test results being 23 ppm and COD 72 ppm, so we need a way to reduce pollution. Phytoremediation is a method of natural waste handling using certain plants in collaboration with microorganisms in the media (soil, coral, water) so that it can change contaminants to be reduced or harmless. This study aims to determine the effectiveness of Salvinia molesta phytoremediation in reducing BOD and COD levels. The research design used a completely randomized design (CRD) with four treatments with three replications. The research data were tested using the Analysis of Variety Prints and the Duncan's Multiple Distance Test continued test. The results showed that the effectiveness of Salvinia molesta in reducing BOD levels was found in P3 obtained by an average of 8.37 ppm (73% decrease) and reducing COD levels by an average of 25.37 ppm (73%). Based on these results, it can be concluded that P3 is effective in reducing BOD and COD levels of coffee liquid waste.
