Abstract

The research was done from June to August 2004 by cencus to all fishermen. This study was conducted in Trisik Coast, Galur Subdistrict, Kulon Progo District.The purpose of this research was to obtain the financial analysis of fishing effort in this area. The results showed that the fishing in Trisisik coast was feasible indicated by the Net Present Value (NPV) have positive value (Rp 21.439.196,-), Net B/C rasio (1,85) and Internal Rate of Return (IRR) (16,43%).

Abstract

The aims of the research were to observe (1) utilization of cocoa pod husk meal to fish feed especially for carbohydrate source (2) effect of substitution rice bran by cocoa pod husk meal on growth and product of giant goramy (3) find the substitution doses of rice bran by cocoa pod husk meal on feed which could give better influence on giant goramy growth. The research was done for 8 months in Fisheries Research Station Laboratory, Agriculture Faculty Gadjah Mada University. Research was conduct by cultured of giant goramy on fibre tank, for 60 days. There were 5 treatmens substitution dose (20, 40, 60, and 80%) and control (comercial feed) with 3 replications. During cultured the tank were irrigated and the fish were fed 3% of body weight 3 times a day. The result showed that cocoa pod husk meal can substitute rice bran on giant goramy feed. Substitution rice bran by cocoa pod husk meal on fed not significantly affect on the growth, product and survival rate of giant goramy, but significant affected on buoyancy of feed. Giant goramy were feeding by comercial feed had growth and product better than feeding by artificial feed.

Abstract

Research to find out the effect of oral administration of estradiol 17-β  on feminization of Oreochromis sp. (GIFT) fry was carried out at Cibalagung Research Station. Five different concentrations of estradiol 17-β  i.e.: 60, 80, 100, 120 and 0 mg/kg feed, were given to larvae of 6 days old twice a day for 21 days in triplicates. The result indicated that there was significant difference of average percentage of female between control and treatments.The highest percentage of female was obtained from fish treated with 100 mg estradiol /kg feed (86.6%), whereas the lowest was obtained from fish control (51.7%). The survival rate and length growth were not significantly different. Survival rate of larvae ranged from 94 to 97.7% and the average of length growth ranged from 10.08 to 13.32 mm.

Abstract

The objective of this research was to know the effectiveness of polyvalen  Vibrio vaccine to control vibriosis in humpback grouper (Cromileptes altivelis). The effectiveness of vaccine was evaluated by the survival rate (SR), relative percent survival (RPS), mean time to death (MTD) as well as growth rate of vaccinated fish. This research consisted of 4 treatments (control, injection, immersion, and oral vaccinations) in quadruplicates. Injection  vaccination was conducted by intraperitoneal injection of polyvalen vaccine at 107 cells/fish. Immersion vaccination was done by immersing the fishes at 107 cells/ml for 30 minutes. Oral administration of vaccine was also carried out  at 107 cells/fish. One week after the first vaccination, second vaccination (booster) was carried out at the same dosage and by the same administration. One week after the second vaccination, fishes were challenged with 3.16x104 cells/fish of Vibrio ordalii 3J by intraperitoneal injection, and reared for 20 days post infection. Results indicated that polyvalen Vibrio vaccine increased SR (P<0.01) up to 100%. Vaccination was also able to delay MTD of fishes. However, the vaccination was not influence the growth rate of fish.

Abstract

Different media condition, with salinity above the hatching threshold of cysts (100 ppt) and feeding with water flow as the main menu and Spirulina maxima flour as an additional feed encouraged the adult female Artemia salina to be oviparous and produce different amount of cysts depending on the treatments. The average amount of cysts produced by oviparous female Artemia salina during 77 day rearing was 22,719-78,616 cysts each female Artemia salina produced 18-13 cysts. Statistical analysis showed that all treatments did not reduce any significant effects on the production of cysts (p>0.05)

Abstract

The purpose of the research were to determine the toxicity and antimicrobial activity of ethanolic extract of sponges collected from Tabuhan Island, Banyuwangi and Menjangan Island, West Bali. Extracts were made by maceration of the sponge samples for 3 x 24 hours. The toxicity test was carried out by Brine Shrimp Lethality Test (BST) with 48 hours-old Artemia sp. The effect of ethanolic extract was identified by determining the mortality of Artemia sp. and LC50 of each extract was analyzed by using probit analysis. Antimicrobial activity was tested by agar diffusion method. The result showed that several ethanolic extracts exhibited toxicity to Artemia sp. The toxicity test showed that extract of sponge code B43 was the most toxic with the LC50 of 8.4 µg/ml. Antimicrobial activity test exhibited that ethanolic extract of sponge with code numbers B2, B57, B61 dan W5 were active against Candida ablicans, while the axtracts of sponge with code numbers B9, B35, B47, B51 (Liosina sp.), B57, B61, B63 and W11 were active against Staphylococcus aureus. One ethanolic extract from sponge with code number B57 exibited activity against Eschericia coli.

Abstract

The aim of this research was to identify the morphological and genetical variability in four strains of common carp (Cyprinus carpio) i.e. Majalaya, Rajadanu, Wildan and Sutisna. The study was conducted by analysis of truss morphometric and electrophoresis. Principal Component Analysis (PCA) was conducted to explore the patterns of body shape variability and the grouping among strains. The Degree of polymorphism, heterozigosity and genetic distance were used to identify the structure of populations. The results showed that the truss morphometric on head and caudal peduncle were the most contribute cells to distinguish the variability among strains. Based on genetic parameters analysis, strain of Rajadanu, Sutisna and Wildan were originaly coming from one strain with genetic distance about 0.0018 to Majalaya strain. Body shape similarity of Wildan and Majalaya strains was more influenced by their environment.

Abstract

The objective of this study was to determine the diversity and abundance of krustaseans in the mangrove area of Jangkaran Village, Temon District, Kulon Progo Regency. Sampling was done bi-weekly from October 2016 to February 2017 on six stations defined by ecological character. Krustasean sampling was done using cest net, trap net and hand picking along the river with the extent of each collection station about 10 m2. The captured krustasean samples were cleaned, stored in ice-cooled boxes and transported to the laboratories to be identified and grouped according to species, then measured by carapace length and individual weight. Observation of aquatic environment was done by measuring temperature, brightness, water depth, current velocity, pH, salinity, dissolved oxygen, and substrate type. The results of the observation obtained 1 order, 7 families, and 26 species. The family krustaseans found were Coenobitidae, Diogenidae, Grapsidae, Ocypodidae, Palaemonoidae, Penaeidae, and Portunidae. The most dominant species of krustaseans were white shrimp (Penaeus merguiensis) as much as 25.06%, hermit crab (Clibanarius sp.) as much as 10.85%, and Uca annulipes as much as 10.59%. The fewest species of krustaseans found were Varuna yui of 0.13% and Ocypode sp. as much as 0.26%. The abundance value was 25.4 ind / m2, indices of diversity 2.60, richess index 0.87 and index of dominance 0.18.

Abstract

The aim of study was to know the effect of sludge in the ration on the gonadal growth of Oreochromis sp. broodstock. Experimental design used in this experiment was completely Randomized Design. The broodstocks were fed with the ration containing 0-40% of sludge, or with a commercial fish feed as a control.The results were not significantly differences between the treatment in all parameters. The sludge in ration ranging from 0 to 40% resulted the level of gonadal maturation of 4, 6, 7, dan 9. The value of Gonadal Maturation Index on 0,96 to 3,98 % and the control group on 1,75 %. The specific growth rate of broodstock fed with the ration ranged from 1,1 to 1,62 Bw/day and that of the development of gonadal control group was 1,97. The survival rate were variated between 14,29 to 100% and that of the development of gonadal control group is 71,43%. The ration containing 40% sludge is the most suitable feed for maturation in Oreochromis.

Abstract

Polymerase chain reaction (PCR) is a method that amplifies DNA which have been widely used in molecular biology technique. Based on the PCR, many methods have been developed on isothermal condition and the useful one is loop-mediated isothermal amplification of DNA (LAMP). LAMP reaction employs a Bst DNA polymerase and a set of four specific primers that recognizes a total of six distinct sequences of the target DNA and produces amount of different size of DNA. Many advantages have been achieved in LAMP such as the simple equipment for reaction and observation, short time, highly specific and sensitive procedure. LAMP has been used as a tools for detection many pathogens for human, animals and plants. Some fish pathogens as parasites, bacteria and viruses have been detected by LAMP. The principles and application of LAMP method are discussed in this paper.
