Abstract

Indonesia one of the high endemic countries of malarial disease, while the use of drugs that are still resistant can spread parasites and cause death. The purpose of this study was to find new antimalarial drug candidates through the Quantitative-Structure and Activity Relationship (QSAR) of quinolin-pyrazolopiridine derivative compounds. QSAR is an important part of drug design in an effort to obtain a new drug with greater activity, higher effectiveness, toxicity or the smallest possible side effects and greater comfort. In reviewing QSAR, it can use the Hansch approach parameter in the form of electronic, lipophilic and steric. From the modeling of 20 quinolin-pyrazolopiridine derivatives the best QSAR equation: Log EC50 = 134,137 + 5,067 (LogP) + 0,036 (ETot) – 5,539 (Polarisibility) – 0,218 (Dipol) + 0,309 (BM) + 0,004 (MTI) + 0,028 (CMA) + 0,227 (VMol) n = 20  r = 0,984   r2 = 0,968  SE = 0,661824

Abstract

Fisetin is a natural antioxidant that has shown to posses antioxidant and anti-inflammatory. However, the poor solubility leads to poor bioavailability and limits its development. Fisetin spherical agglomerates were prepared with ethanol, water, chloroform and polyvinylpyrrolidone (PVP)  as solvent, anti-solvent, bridging liquid and polymer, respectively. The main objective of this research to overcome the low fisetin solubility, micromiretic properties and in vitro dissolution. It was prepared using spherical agglomeration method, organic solvent and polymer. Characterization is carried out on morphology particle, drug loading efficiency, solubility and in vitro disolution. XRD studies showed a decrease in crystallinity in agglomerates. The crystals exhibited significantly improved micromeritic properties compared to pure drug. The loading efficiency was in the range of 98.25 ± 1,20 %. The aqueous solubility and dissolution rate of the drug from crystals was significantly increased, 4,45 mg/ml ± 0,31 dan 98,75%, respectively. The SEM studies showed that the crystal posseses a good spherical shape with smooth and regular surface. Conclusion, fisetin was successfully prepared by spherical agglomeration method, with the polyvinylpyrrolidone 5% as polymer,  ethanol as good solvent, water as antisolvent and choloform as bridging solvent.. Abstract

Mefenamic acid is a class of NSAIDs (Non-Steroid Antiinflammatory) which belongs to BCS group (Biopharmaceutical Classification System) class II that is low solubility with high penetration membrane. So it is necessary to increase the solubility to get absorption that can penetrate the maximum therapeutic zone of reagent. This study aims to determine the effect of the concentration of disintegran Croscarmellose sodium with a concentration of 1% - 5% on the physical quality and dissolution rate because it has a working mechanism of water absorption (water wicking) and swelling rapidly. Raw material collection, manufacture of mefenamic acid tablets (the wet granulation method is used because it can improve the flow rate and the compressibility of the mefenamic acid), evaluation of mefenamic acid granules, evaluation of mefenamic acid tablets, data analysis using one way ANOVA. Characterization of physical quality of granules moisture content, flow rate, fixed angle according to requirements. Characterization of quality tablet hardness, friability, frixibility, weight, uniformity size, disintegration time, and dissolution according to requirements.. Statistical analysis, indicating that there are significant differences in hardness, weight, uniformity of size, crushed time, rate and dissolution. Abstract

Hypersensitivity reactions such as anaphylaxis are increase over time. Anaphylaxis generally affects the skin, breathing, cardiovascular and digestive systems. Ageratum conyzoides L. are often used by the people to heal skin diseases, wounds and allergies. The objectives of this study is to determine the immunomodulatory activity of the ethanolic extract of babadotan leaves (ageratum conyzoides L) on male mice using the active cutaneous anaphylactic reaction method. The method is an experimental study of 4 groups each consisting of 4 male swiss webster strain mice. The first step is the process of sensitization of each group for 2 cycles (7 days per cycle). There is not treatment for Group 1 as a normal group, but Group 2 as a negative group, Group 3 as group I test and Group 4 as group II test each are given by ovalbumin (5 ml / kg BW) intraperitoneally. The second stage is challenging on a day before the experiment by shaving the back areas of the mice as the injection areas. Inject intracutaneously in each sector with 0.1 mL of 10% egg white solution (a red bump will form). Fifteen minutes after the formation of the bump, give the ethanol extract of the babadotan leaf (100 mg / Kg BW) to the test group I and the test group II (250 mg / Kg BW). Observe and measure these sectors every 15 minutes for 60 minutes, then continue observations every day for 7 days. Observations include the first time the appearance of bump, diameter of bump after 60 minutes and until the time of bump disappearance. The results showed that the ethanol extract of babadotan leaves test group II ( 250 mg / kg BW) had the best antianaphylactic activity with the possibility mechanism through inhibition on histamine. Keywords: Leaves, Ageratum conyzoides L., Immunomodulatory, active cutaneous anaphylaxis

Abstract

Simvastatin is a statin drug that used to reduce cholesterol levels via inhibition of the 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase. The purpose of this study is to determine the effect of Cremophor EL to the dissolution rate of simvastatin tablet. The experiment was consist of  four formula with different concentration of Cremophor EL that were 0%, 0,3%, 0,6% and 0,9%. Preparation of simvastatin tablets was done by wet granulation method. The granules obtained were tested (moisture content, compressibility, flow rate, rest angle and hausner number), physical test of tablets (weight uniformity, hardness, brittleness and crushed time) and dissolution test. Dissolution Apparatus II  was used to dissolution test with 900 mL of phosphate buffer (sodium dihydrogen phosphate and sodium dodecyl sulphate) pH 7.0 as dissolution medium, it was inserted in dissolution vessel at 37 ± 0,50C with 50 rpm. Spectrofotometry uv-vis was used for determination of dissolution result at 238 nm of wavelength. The dissolution test results of simvastatin tablet shows that the addition of 0,6% Cremophor EL in formula 3 has the largest simvastatin dissolution rate.  Keywords: Cremophor EL, Dissolution, Simvastatin,

Abstract

Jerawat adalah peradangan kronik folikel sebasea yang ditandai adanya komedo, papula, pustul, kista pada daerah-daerah predileksi. Tujuan penelitian ini yaitu menguji stabilitas sediaan gel anti jerawat ekstrak daun kersen terhadap bakter P. Acne. Penelitian ini dilatar belakangi dari banyaknya obat anti jerawat yang sudah beredar dipasaran yang sudah resistensi dan menyebabkan timbulnya iritasi akibat penggunaan obat anti jerawat dengan bahan aktif sintetik sehingga untuk mengurangi efek samping tersebut dikembangkan bahan aktif dari alam yang lebih aman dan efektif mengobati jerawat. Penelitian ini dimulai dari penyiapan bahan baku pembuatan dan standarisasi mutu ekstrak, uji aktivitas ekstrak, pembuatan sediaan gel, evaluasi dan uji stabilitas gel. Dari hasil evaluasi bahwa formula gel antijerawat ekstrak daun kersen memiliki stabilitas yang baik selama penyimpanan 30 hari dari segi organoleptik, pH, homogenitas, daya sebar dan viskositas. Kata kunci: Daun kersen, jerawat, uji stabilitas.

Abstract

Hypertension is still being a health problem in the world. The blood pressure of most patients with hypertension are not controlled with single therapy meanwhile the medical costs are increasing annually, so that it needs to analyzed the cost effectiveness treatment to find out the most cost effective and provide the best outcome of therapy. This research aim to know the most effective antihypertensive two combination therapy in hospitalization RSUD dr. Soekardjo Tasikmalaya on 2017. This is a descriptive research that using pharmacoeconomic consideration with Cost Effectiveness Analysis method based on hospital perspective. This research using retrospective data of hypertension patient in hospitalization RSUD dr. Soekardjo Tasikmalaya during 2017 and sample who met the inclusion criteria were 35 patients. The data of study are categorized by kind of therapy and analyzed to get cost effective therapy for any category. The result showed that antihypertensive combinations are combination two, three and also four drugs. Cost effective treatment for any category are ACE-I-Diuretic for combination two drugs antihypertensive, ACE-I-β-Blocker-CCB for combination three drugs antihypertensive and ACE-Ia-ACE-Ib-CCB-Diuretic for combination three drugs antihypertensive. The lowest ICER Rp. 12.511,02 is shown between therapy combination ACE-I-CCB-Diuretic and ACE-I-ARB-Diuretic. Keywords: Cost Effectiveness Analysis, combination antihypertensive, hospitalization.

Abstract

Service can basically be said to be an action and treatment or a way of serving others to fulfill what is their needs and desires. The level of customer satisfaction for a service can be measured by comparing the expectations of consumers to the quality of services desired by the reality they receive or feel. This study looks at the gap between the services provided and the services felt by consumers at the P. Bhakti Kencana Pharmacy. This research uses descriptive type. Descriptive method is a method of research carried out with the main purpose of making a description or description of a situation objectively. This study uses a type of primary data, namely the type of data obtained by direct collection by researchers such as the results of the questionnaire. The results of the study show the five dimensions namely Tangibles (Reality), Empathy (Sympathy), Reliability (Reliability), Responsiveness (Responsibility) and Assurance (Certainty). Keywords: Service quality, customer satisfaction, descriptive method.

Abstract

Endophytic bacteria is a bacteria found in the tissue’s plant that does not provide an infection or a negative effect on its host plant. Endophytic bacteria can be found in various types of plants, especially plants that are often used as medicinal plants. One of them is Kamboja Putih (Plumeria acuminata Ait.) which has secondary metabolite such as alkaloids, flavonoids, tannins, and terpenoids. Endophytic bacteria can produce the same bioactive components as their host plants, so it can potentially be developed in the world of health. The purpose of this research was to determine the presence of endophytic bacteria and characteristic isolated from P.acuminata stem. This is an explorative research, using an isolation method to get endophytic bacteria. P.acuminata stem is sterilized and inoculated on Nutrient Agar, and then it is incubated until the isolate is obtained. To know the characteristic of endophytic bacteria, macroscopic characteristics were observed such as shape, color, edges and surface of colony and microscopic characteristics from endophytic bacteria was observed by Gram stain. Moreover, to know the characteristics of secondary metabolites that exist in endophytic bacteria it has to be done phytochemical screening to the obtained isolate, include test of flavonoids, tannins, triterpenoids, alkaloids and saponins. The results of the research showed that Kamboja has endophytic bacteria, that are endophytic bacterial isolates EI and E2. The characteristics of both endophytic bacterial isolates are round, whitish, intact edges and flat surfaces of the colonies. The types of  both bacterial isolates EI and E2 are Gram positive and coccus-shaped bacteria. Phytochemical screening tests show that endophytic bacterial isolates can produce alkaloids both on ordinary fermentation medium and those already given precursors of acetic acid Keywords: Plumeria acuminata Ait, endophytic bacteria, isolation and characterization                      endophytic bacteria.

Abstract

Penyakit infeksi adalah penyakit yang di sebabkan oleh mikroorganisme patogen seperti bakteri, virus, jamur dan parasit. Penyakit infeksi menyebabkan kematian tertinggi di Indonesia, maka perlu dilakukan peneletian untuk menemukan antibmikroba salah satunya adalah dari jamur tiram putih Ppleurotus ostreatus) tujuan dari penelitian ini adalah untuk mengetahui aktivitas antibakteri  dari ekstrak jamur tiram putih (Pleurotus ostreatus) terhadap bakteri pathogen Bacillus cereus, Escherichia coli, Pseudomonas aeruginosa, dan Staphylococcus aureus. Penelitian ini diawali dengan proses ekstraksi menggunakan metode maserasi menggunakan pelarut etanol 96%, kemudian dilakukan pengujian antibakteri dengan menggunakan metode mikrodilusi untuk menentukan nilai Konsentrasi Hambat minimum (KHM) dan Konsentrasi Bunuh Minimum (KBM). Hasil pengujian menunjukkan bahwa ekstrak jamur tiram putih (Pleurotus ostreatus) memiliki aktivitas sebagai antibakteri terhadap Escherichia coli, Pseudomonas aeruginosa, dan Bacillus cereus dengan nilai KHM 512 µg/mL, sedangkan terhadap Staphylococcus aureus memiliki nilai KHM 256 µg/mL.
