Abstract

Herbal medicine is a traditional drink made from natural ingredients for human health and is made with methods and equipment that are easier in its simplicity which often does not pay attention to hygiene factors and can be a medium for disease transmission. Based on the Regulation of the Head of the Republic of Indonesia Drug and Food Control Agency No. 12 of 2014 concerning requirements for drug fluids in the form of microbial contamination / ALT ≤104 colonies/ml. The purpose of this study was to study the estimated numbers of traditional herbs sold in the Tulungagung District market. The research design used is descriptive. The sampling technique uses a total sampling technique of 6 samples from 4 different markets. Sample meeting with the ALT method in May 2018 in the microbiology laboratory of STIKes Hutama Abdi Husada Tulungagung. The results of the study found that three out of six samples of traditional herbal medicine of kencur rice exceeded the microbial contamination threshold with an average yield of 1 x 107 CFU / ml. The study concludes is that 50% of the sample is not suitable for consumption and 50% of the sample is suitable for consumption. This requires Regulation of BPOM RI Number 12 the year 2014 Regarding Quality Requirements for Traditional Medicines, namely ≤104 colonies/ml. Herbal traders need to pay attention to raw materials, processing, equipment, air used, and containers used.

Abstract

Diabetes mellitus is a metabolic disease due to an increase in blood glucose levels normal. Gangrene diabetes is tissue death due to obstruction of blood vessels that provides nutrients to obstructed tissue and is a form of complications from diabetes mellitus. Pseudomonas aeruginosa is one of the causes of gangrene infection nosocomial opportunistic infections in humans. The purpose of this study was to determine the presence of gram-negative bacteria in the wounds of Gangrene Diabetes Mellitus. This research is a descriptive Non-Analytic, conducted on April 15 to May 21, 2019, in the Microbiology Laboratory of STIKes Hutama Abdi Husada Tulungagung. The study sample was all patients with Gangrene Diabetes Mellitus wound in hospital dr. Soedomo Trenggalek, the sampling technique used was accidental sampling, obtained a sample of 4 Gangrene swabs. The study was conducted by the identification of gram-negative bacterial isolation. The results of the study were 4 samples, 2 were positive for Pseudomonas aeruginosa and 2 samples were positive for Proteus sp. Subsequent research results were analyzed descriptively. The conclusion of this research is that 100% positive gram-negative. The typical characteristics of the Pseudomonas aeruginosa bacteria are obtained on the MCH media (alkaline slope, alkaline base, negative gas, negative H2S) and on the NAS produce greenish-yellow fluorescence, while the typical characteristics of Proteus sp are obtained on the MCH media (alkaline slope, alkaline slope, base negative) acid, positive gas, positive H2S).

Abstract

This study aims to analyze the quality of water in Tulungagung. Identification carried out is a physical test including pH, salinity, color, and temperature. As well as chemical tests, including alkalinity test, CO2 content, Cl, and Water Hardness. The sample water collected from six locations in Tulungagung Regency which are Sendang, Rejotangan, Campurdarat, Keras, Ngunut, and Tanggunggunung. From the pH test was obtained that all the sample water qualifies as good water (pH 6.5 to 8.0). The content of CO2 water samples shows that result with the highest CO2 21 ppm and the lowest 18ppm. Whereas the Cl test obtained the highest 135.3 ppm and the lowest was 52.7 ppm This analysis data was enriched with hardness test results with the highest hardness 60.9 D while the lowest hardness water was 20.9 D

Abstract

This study aims to determine the effect of Lactobacillus Plantarum and Lactobacillus Acidhophilus Bacteria on Probiotic Beverage (Yogurt) from Cow's Milk and Soy's Milk, making yogurt is done by randomized design of groups factorially arranged using Lactobacillus plantarum. The second factor is the proportion of cow's milk and soy milk served at, namely: 100% cow's milk (S1), 50% cow's milk and 50% soy milk (S2) and 100% soy milk (S3) with the treatment applied through, addition lactic acid and viscystas from yogurt. Analysis results obtained from the addition of lactic acid. The proportion of cow's milk and soybean milk showed that Lactobacillus plantarum obtained a total of Lactic Acid of 0.45 and Lactobacillus acidhophilus. The proportion of Cow's Milk and Soymilk obtained a total Lactic Acid of 0.35. And the results of the analysis obtained from the Viscosity Proportion of Cow's Milk and Soybean Milk show that Lactobacillus plantarum obtained a total viscosity of 1.5 and Lactobacillus acidhophilus Proportion of Cow's Milk and Soybean Milk obtained a viscosity of 2.9.Keywords: influence, lactobacillus plantarum and lactobacillus acidhophilus, cow's milk and soy milk

Abstract

Food is a basic need as a source of energy for humans. Food components needed by humans are consist of several constituent substances including carbohydrates, fats, vitamins, minerals, water, and protein. The most widely consumed protein sources by Indonesian people are eggs and tempeh. Processing eggs and tempeh in order to have a good taste is usually done by heating through the frying process. The frying process requires temperatures between 160-300oC. The high temperature when frying can cause protein damage so that it cannot be utilized by the human body. Therefore it is necessary to research changes in protein levels in eggs and tempeh due to heating through the frying process. The research method used was spectrophotometry with the complexing agent used was a biuret reagent. The result showed that there was decreasing content on both egg and tempeh due to the frying process. A decrease in protein content was 29.21 mg or 23.28% in eggs. Whereas in tempeh samples there was a decrease in protein content by 59.03 mg or 49.33%. Therefore the heating process in egg and tempeh should be done under 160oC

Abstract

The use of traditional medicine can be an alternative treatment with increasing antibiotic resistance. Red ginger (Zingiber officinale var. Rubrum) and betel leaf (Piper betel L.) are plants that can be used as an antibacterial. This study aimed to determine the antibacterial activity of combined extracts of red ginger and betel leaf. This experiment was carried out by making extracts of red ginger and betel leaf with maceration and tested the antibacterial activity. The antibacterial activity test used the paper disc diffusion method. The study was conducted with five different concentrations of extract 20%, 40%, 60%, 80%, and 100%. The results showed that the combination of red ginger and betel leaf extract at a concentration of 80% had the highest inhibitory area against Staphylococcus aureus (10.6 mm) and Escherichia coli (7.5 mm). The data obtained were statistically tested using One Way Anova with a significance level of > 0.05. The results of One Way ANOVA analysis of the combination of red ginger and betel leaf extracts on Staphylococcus aureus had a significant value of 0.083, while those extracts on Escherichia coli had a significant value of 0.690. It can be concluded that the combination of red ginger and betel leaf extract at a concentration of 80% had a strong inhibitory level against Staphylococcus aureus and a moderate inhibitory level against Escherichia coli.

Abstract

The Granulomas are a group of innate and adaptive immune cells that are highly organized, and during tuberculosis infection caused by the bacterium Mycobacterium tuberculosis. Estimated as many as 1.6 million cases of death in 2017 according to World Health Organization. Tuberculosis ranks in the top 10 causes of death worldwide and it is predicted that more than 1.7 billion people (around 25 percent of the world's population) are estimated to be infected by Mycobacterium tuberculosis. The purpose of this study was to confirm the Mtb by examining a new method by modifying Ziehl-Neelsen stain using an intracellular specimens for tuberculous granuloma in vitro. This study used the isolate of the bacterium Mycobacterium tuberculosis H37Rv obtained from the Microbiology Laboratory of the Institute of Tropical Disease Universitas Airlangga, Surabayaa, using Peripheral Blood Mononuclear Cells to make model granuloma tuberculosis. The result is Ziehl-Neelsen staining was modified, it was found that the Mycobacterium tuberculosis H37Rv as detection of bacterial confirmation is said to be successful.

Abstract

Chili sauce is a food additive made from chili which is processed from the processing of chili that is cooked and of good quality with the addition of other ingredients. There is a chili sauce in the form of packaging and processed home industry, the use of chili sauce in the community is often presented as a supplement to a variety of foods. However, the cleanliness of chili sauce still needs to be questioned considering the factors that can affect the contamination of chili sauce can be obtained from processing, serving and contamination from the surrounding environment. This study aims to determine the number of bacteria in the chili sauce in meatball traders on daya nasional street. The research design used is descriptive. The sampling technique in this study was by total sampling. The research method used is the Total Plate Count (TPC) method. The number of samples taken in this study amounted to 8 samples. Based on the results of the study, the highest number of germs is 2x106 colonies / gr and the lowest is 3.4x104 colonies/gr, it can be concluded that the entire sample in this study did not meet the conditions set out in Standar Nasional Indonesia (SNI) about chili souce No. 7388 in 2009 which is a maximum of 1x104 colonies/gr.

Abstract

Type II diabetes mellitus (T2D) is a genetic disorder that becomes an unsolved problem. This is because T2D can be a disorder caused by lifestyle changes and is inherited from parents. This study aimed to determine the polymorphism that distinguishes between T2D and non T2D patients and the result was predicted to the families that has T2D record. The model in this research was descriptive exploratory and consists of 80 samples (30 samples positive T2D, 30 non DM samples and the others are the member of 5 families). Firstly, the analysis was focused on determining differences in polymorphisms and then the results were applied to the 3 families of T2D using A10 Primer. Difference polymorphism was analyzed using chi-square (cl: 95%). Six bands were obtained from PCR-RAPD which a significantly different band at 997 bp. Analysis of T2D has confirmed that 5 families have a clue band. Based on the analysis this report can be used to screen the possibility of someone to have DM type 2.

Abstract

Caesarean section is a process to remove a fetus or baby through an incision in the abdominal wall and upper uterus because a normal birth process is not possible. The process by Caesarean section requires careful preparation because the result can be bleeding. One of the tests before and after Caesarean section is checking the hemoglobin level. The purpose of this study was to determine the differences in hemoglobin levels before and after Caesarean section. This research is a quantitative analysis. Sampling was conducted in April 2018 at RSIA Fauziah Tulungagung and hemoglobin levels were checked using the Cyanmehemoglobin method at the STIKes Hutama Abdi Husada Tulungagung Pathology Laboratory. The population of this study were all pregnant women who underwent a Caesarean section at RSIA Fauziah Tulungagung using a total sampling technique of 30 samples. The instrument used was a photometer, then the data was processed using SPSS 16 software and analyzed using the Paired T-Test technique. The results of research conducted on 30 samples of Caesarean section patients obtained an average hemoglobin level before Caesarean section 13.13 g / dl and an average hemoglobin level after Caesarean section 12.06 g / dl after the Paired T-Test was obtained. 0.002 <0.05. It can be concluded that there is a significant difference between the hemoglobin levels before and after Caesarean section. Bleeding during Caesarean section causes hemoglobin levels in pregnant women to decrease.
