Abstract

Studi ini telah dilakukan pada laboratorium kultur jaringan, Balai Penelitian Tanaman Hias Pacet – Cianjur sejak bulan Februari hingga Oktober 2008. Tujuan penelitian ini adalah untuk mengoptimalisasi media terseleksi dalam induksi pembentukan kalus, mengungkap asal-usul sel yang membentuk kalus hasil kultur anther dan proses pembentukan tunasnya. Media terseleksi yang diperbaiki adalah 1) MMS-TBN yang mengandung 0,5 mg/l TDZ, 1,0 mg/l BAP dan 0,01 mg/l NAA (Medium Winarto, WM) dan 2) MMS III yang ditambah dengan 1,5 mg/l TDZ, 0,75 mg/l BAP dan 0,02 mg/l NAA (Medium Winarto dan Rachmawati, WRM). Perlakuan perbaikan dilakukan melalui pembuangan dan penambahan 2,4- D pada konsentrasi 0.5 mg/l dan penurunan kekuatan medium dari kekuatan penuh, setengah, seperempat, seperdelapan, seperenambelas dan nol. Percobaan faktorial disusun menggunakan rancangan acak kelompok dengan empat kali ulangan. Hasil studi ini menunjukkan bahwa induksi kalus tertinggi secara jelas ditemukan pada WRM. Medium ini mampu menginduksi potensi tumbuh anther (PGA) hingga 81% dengan persentase regenerasi anther 49% dan 2.7 kalus per ulangan. Peningkatan yang nyata juga dicatat melalui perlakuan reduksi kekuatan medium dengan seperdelapan kekuatan medium WRM sebagai penurunan kekuatan yang terbaik. Perlakuan ini mampu menstimulasi PGA hingga 58% dengan 29% PAR dan 1.8 NCF. Dari studi histologi diketahui bahwa kalus hasil kultur anther berasal dari sel-sel dinding anther. Kapasitas morfogenesis yang tinggi dari sel-sel dinding anther inilah yang menyebabkan tidak adanya efek androgenesis pada sel-sel mikrospora yang ada didalamnya Kata-kata kunci: Perbaikan, media, kalus, anther, histology dan anthurium

Abstract

T. padwickii merupakan jamur utama terbawa benih pada padi yang dapat menyebabkan penurunan daya kecambah, viabilitas dan vigor benih. Sampel benih padi diuji terhadap infeksi penyimpangan warna gabah berdasarkan sistem standar evaluasi padi (SES, IRRI). Bibit padi kultivar IR 64 dan Cisadane dengan kadar air awal 14%, diinokulasi suspensi spora T. padwickii, dengan lama perendaman masing-masing 3, 6, 12, 24 and 48 jam. Hasil penelitian menunjukkan keparahan penyakit, tinggi tanaman, dan bobot gabah sangat dipengaruhi oleh T. padwickii. Tingkat infeksi yang tinggi lebih banyak teramati pada kultivar IR 64 dibanding Cisadane. Semakin lama perendaman dengan suspensi spora T. padwickii cenderung meningkatkan keparahan penyakit. Nilai rerata infeksi penyimpangan warna gabah meningkat lebih dari 4% pada inokulasi T. padwickii dengan lama perendaman selama 3 sampai 24 jam. T. padwickii fungi was observed in irregular shaped at rice seed with typical infection (pink color seeds); Morphologically single cell conidia of T. padwickii were estimated to have size of 1.6 x 7.5 μm in length.Kata kunci: Penyimpangan warna gabah; T. padwickii, patogen tular benih

Abstract

Chrysanthemum (Dendranthema grandiflorum) is well known as a commercially valuable flower. However, the plant cultivation has not been well developed. Poliploidization is one of cultivation technique using colchicines as mutagenic agent to produce high quality of flowers. The aims of this research were to study phenotypic characters of chrysanthemum treated by colchicines and to determine the optimum of soaking time to obtain the highly phenotypic characters.Colchicines concentration used in this research was 0.01% with soaking times were 6, 12 and 24 hours. Phenotypic characters observed in this experiment were flower diameter, plant height, stem diameter, long and wide of 5th and 10th leaf from flower base with 10 replications for each characters.The result showed that soaking times treatment with 0.01% colchicines have real influence on flower diameter, plant height, stem diameter, long and wide of 5th and 10th leaf from flower base. The result of colchicines with soaking treatment of 12 hours showed the highly phenotypic characters. In addition, chrysanthemum with colchicines treatment has bigger phenotypic characters than control. Key words: Chrysanthemum, Dendranthema grandiflorum, colchicines, phenotypic characters

Abstract

Propagation of cassava through in vitro culture is required by farmers and agro industries to fulfill the need of the best and the newest clone as soon as after that clone is released by government. The objective of this research is to know the effect of the application of four concentration of benzyl adenine on the growth and proliferation of cassava shoot in vitro. Explants used were one-node cuttings of cassava about 1 cm length, grown in polybags. This research was arranged in completely randomized design with the treatments consisting of four concentrations of benzyl adenine: 0.5, 1, 2 and 4 mg/l added of basic media of Murashige and Skoog. Each treatment was replicated 4 times with 5 explants in each experimet al unit. The best growth and micro-shoot propagation of cassava in vitro was indicated by treatment of 0.5 mg/l benzyl adenine.Key words: cassava, benzyl adenin, in vitro, micro-shoot

Abstract

To get high quality physic nut seedling one of effort is with vegetative propagation especially with grafting. The objective of this study was to evaluate the effect the maturity of rootstocks and shading rate on the effectiveness of grafting and the growth of seedling of Jatropka. The experiment was conducted in Labuhan Dalam, Subdistrict Tanjung Seneng, the City of Bandar Lampung from January-June 2008. Is a factorial experiment (3X4) with split plot design, shading degrees as main plot consisted without shading, light shading, moderate shading, and heavy shading. Root stock age as subplot consisted 6, 8, and 10 weeks after transplanting. The treatments were consisted 3 replication and each experiment unit consisted 10 seedlings. The result shows that root stock age 8 and 10 weeks produces physic nut seedling growth better than root stock age 6 week. Physic nut seedling without shading and light shading (1 layer paranet) grow better than with moderate shading (2 layers paranet) and heavy shading (3 layers paranet);Key words: Physic nut, root stock age, shading

Abstract

Study on the effect of rainfall pattern on the coffee yield has been carried out at one of government-owned estate companies in Banyuwangi district, East Java. The study was conducted by multiple linier regression analysis approach of rainfall elements on the coffee production. The results showed the following figures. Rainfall pattern significantly affected the production of the coffee plants. In general, it was shown that the effect of rainfall pattern on the coffee production with statistical analysis approach of multiple linier regression of the study showed that the following rainfall elements, i.e. wet months, rainfall of the previous year, and dry months of the previous year, would always appear as the dominant factors determining the production of the coffee plants, in the blocks of plants (each block of plants has its own or the same planting year) as well as in afdeling (the blocks as a whole or a unite), in which the study was carried out (as a note, a plantation or estate company usually has some afdelings). Each element of rainfall tended to have similar direction or regression coefficient vector in every assessment, but different magnitude supposed to be the effect of local conditions of the land. The equations performed from the multiple linier regression analysis have precision to predict ranging from 86-91%.Keywords: Rainfall pattern, wet months, dry months, coffee production.

Abstract

The objective of this study was to estimate correlation, variances, and heritability of agronomic characters of yardlong-bean F3 population derived from crosses between black and striped-black testa. An F3 population consisted 90 plants along with the two parents (20 plants per parent) were planted in unreplicated trial in Hajimena, Rajabasa, Bandar Lampung in 2008. Variables observed included flowering date, fresh-harvesting date, harvesting date, the number of flower stalks per plants,the number of pod per plant, the average number of locules per pod, the total number of seeds per plant, and the weight of 100 seeds. The results indicated that the estimate of genotypic and phenotypic variances and broad-sense heritability were high for all variables observed. The estimates were significant for the correlation between dry-seed weight per plant and the following variables: number of flower stalks per plant, number of pods per plant, the average number of loculs per pod, and the number of seeds per plants. Based on consumer demand and solid fresh peas can be obtained on the genotypes of the number 6, 7, 9, 11, 14, 19, 22, 28, 66, and 67. There are no pods which exceeds 50 cm in length.Keywords: correlation, heritability, variances, Vigna sinensis, yard-long beans

Abstract

The objective of this research are to know the effect of blend of cattle manure, hull of rice, and soil as nursery medium and NPK fertilizer dosage on growth of physic nut seedling by grafting. The experiment has been conducted in Bandar Lampung from January to June 2008. Is a factorial experiment (3X4) with randomized completely block design, the first factors were nursery medium composition consisted of cattle manure, soil, and hull of rice, with volume ratio 3:1:1, 1:1:1, and 1:1:3. The second factors are NPK fertilizer dosage consisted of 0, 1, 3, and 5 g/seedling. The treatments were consisted 3 replication and each experiment unit consisted 10 seedlings. The results show that (1) blend of cattle manure, soil, and hull of rice with ratio 1:1:3 have sucsesful grafting persentage and dry weight of shoot higher than ratio 3:1:1 and 1:1:1, (2) NPK fertilizer dosage until 3 g/seedling unsignificant than control on root stock and entres diameter and dry wight of root and shoot; (3) There is no interaction beetwen seedling medium ratio and NPK fertilizer dosage to growth of physic nut seedling by grafting.Key words: grafting, nursery medium, NPK fertilizer, physic nut

Abstract

The research was conducted to study shoot-growth pattern on four cultivars of mango ( Arumanis, Gedong, Indramayu, Manalagi) that was early pruning and application of KNO3.The treatment was arranged factorially (2x5) in a randomized block design with four replication. The first factor were posision of pruning: on the first flush (P1) and the second flush (P2) and the second factors were concentration of KNO3 (0, 2, 4, 6, 8) g/l. Observations were made in two periods (flush I and flush II). The observation on the first period has been written in Satek-II Proceeding?s, 2008 but the results in the second period showed that:. 1) Pruning the second flush position further increase (the number of leaves per shoot, shoot length, and the total number of leaves per plant) on cv Arumanis, increasing the total number of leaves per plant on Manalagi cultivars, and increase the number of leaves per shoot at Indramayu cultivars than in the first flush position. 2) The application of KNO 3 on Manalagi cultivars reduced the total number of leaves, but the cultivar of Indramayu with concentration of 2 g / l to increase the length of shoots, and on two other cultivars (Arumanis and Gedong) had no effect. 3) There is no interaction between pruning and application of KNO 3 on all cultivars tested.Key words: Cultivars of mango: Manalagi, Gedong, Arumanis, Indramayu, Pruning, KNO3, Flush

Abstract

Study of growth regulator of kamandrah (Croton tiglium L.) by pot experiment was conducted in green house at Indonesian of Medicinal and Aromatic Crops Research Institute (IMACRI) Bogor. The experiment was conducted from Juni to Oktober 2008. In pot experiment to test the germination by treatments: a) Nitroaromatic at 0.5 ppm concentration, b) Nitroaromatic at 1.0 ppm concentration, c) Auxin + Cytokinine at 2.5 ppm concentration, d) Auxin + Cytokinine at 2.75 ppm concentration, e) Root Up at 0.5 ppm concentration, f) Root Up at 0.75 ppm concentration and g) control (without treatment). The assasment on 30 days after sowing (DAS), showed that treatments by Nitroaromatic at 0.5 dan 1.0 ppm, Auksin+ Citokinine at 2.5 dan 2.75 ppm were improved seed germination of kamandrah from 26.67% (by control) to 40.00 - 46.67 %. Nitroaromatic, Auxin + Cytokinine and IBA + NAA to improve seeds germination of kamandrah about 24 ? 74 % than control in 30 DAS. Seedlings growth of kamandrah by Nitroaromatic at 0.5 and 1.0 ppm, Auxine + Cytokinine at 2.5 and 2.75 ppm, IBA + NAA at 0.5 and 1.0 ppm were 11.15 ? 16.40 cm more higher than control (8.85cm). The number of branch treatments by Auxin + Cytokinine at 0.5 ppm (21.35/crop), Nitroaromatic at 0.5 and 1.0 ppm (19.35 and 17.15/crop), IBA + NAA at 0.5 ppm (17.15/crop) and Auxine at 2.75 ppm (16.75/crop) more than control significantly (14.45/crop). The assasment on 60 DAS showed that seedlings growth of kamandrah improved by plant growth regulators ie. Nitroaromatic, Auxin + Cytokinine and IBA + NAA.Key words: Plant growth regulator, seed germination, seedling, kamandrah (Croton tiglium L.).
