Abstract

The consumption of yard long bean in Indonesia is high and it has been increasing continuously, but the production and harvest area has been decreasing. New, superior long bean varieties with higher productivity are required to meet the increasing demands. The aim of this study was to evaluate the morphological and yield characters of selected purple and green long bean genotypes. The research was conducted from February to May 2018 in the Madiun district, East Java, Indonesia. The experiment was laid out in a randomized complete block design with genotypes as the single factor treatment, replicated three times. Eleven yard long bean genotypes were tested, consisting of four new genotypes, F7-013014- 4U-16-1-1, “F7-013014-4U-16-1-2”, “F7-013014- 4U-16-1-3”, and “F7-013014-7P-4-1-1”, and seven control genotypes, “KP13”, “KP14”, “KP Putih China”, “KP Putih China”, “Borneo”, “Sabrina” and “Parade”. The new genotypes, “F7-013014-4U-16-1-1, “F7- 013014-4U-16-1-2”, “F7-013014-4U-16-1-3”, and “F7-013014-7P-4-1-1” had at least one superior characters compared to the control genotypes, i.e. earlier fl owering and time to harvest, longer leaves, greater pod weight, longer pods, larger pod diameter, and higher contents of anthocyanin, carotene, chlorophyll a, chlorophyll b, and total chlorophyll. Abstract

ABSTRAK Produktivitas cabai di Indonesia sampai saat ini masih rendah. Salah satu cara untuk meningkatkan produktivitas cabai yaitu melalui program pemuliaan tanaman. Terdapat beberapa rancangan persilangan untuk memilih tetua dalam rangka menghasilkan varietas unggul baru, di antaranya rancangan silang dialel. Tujuan penelitian ini ialah memperoleh informasi heterosis dan heterobeltiosis dari persilangan enam tetua cabai serta mendapatkan calon hibrida cabai yang unggul. Penelitian dilakukan pada November 2008-Mei 2009 di Kebun Percobaan Lwikopo IPB. Materi genetik yang digunakan ialah enam tetua cabai (IPB C2, IPB C9, IPB C10, IPB C14, IPB C15 dan IPB C20) serta turunan pertama dari persilangan enam tetua tersebut. Rancangan persilangan yang digunakan yaitu rancangan silang dialel. Hasil penelitian menunjukkan bahwa pada populasi F1 hasil dari persilangan IPB C2 x IPB C10 dan IPB 10 x IPB C2 mempunyai nilai tinggi dikotomous yang relatif lebih tinggi dibandingkan hibrida lainnya. Pada populasi F1 hasil persilangan IPB C2 x IPB C14 dan IPB C14 x IPB C2 mempunyai bobot per buah yang relatif lebih tinggi dibandingkan hibrida lainnya. Nilai heterosis dan heterobeltiosis pada populasi F1 hasil dari persilangan IPB C2 x IPB C10, IPB C2 x IPB C14, IPB C2 x IPB C20, IPB C10 x IPB C2 dan IPB C14 x IPB C2 bernilai positif pada karakter tinggi dikotomous. Nilai heterosis dan heterobeltiosis pada populasi F1 hasil dari persilangan IPB C2 x IPB C14 dan IPB C14 x IPB C2 bernilai positif pada karakter bobot per buah. One effort to increase productivity of chilli is by breeding program. There are several cross designs to choose inbred lines to produce new varieties superior, including the design of cross dialel. The aims of this research were to obtain information heterosis and heterobeltiosis from hybrid of six inbred line combination and to gain candidate of hybrids superior. The research was conducted in November 2008-May 2009 at the Garden Experiments Lwikopo IPB. Genetic material used are six inbred lines of chili (IPB C2, IPB C9, IPB C10, IPB C14, IPB C15 and IPB C20) and F1 of the hybridization of combination six inbred lines. The crossing design used was diallel cross. The results showed that population of F1 resulted from crossing of IPB C2 x IPB C10 and IPB 10 x IPB C2 had high dicotomous that was relatively higher than those of other hybrids. The population of F1 obtained from crossing IPB C2 x IPB C14 and IPB C14 x IPB C2 had weight per fruit that was relatively higher than those of other hybrids. Heterosis and heterobeltiosis values in F1 of IPB C2 x IPB C10, IPB C2 x IPB C14, IPB C2 x IPB C20, IPB C10 x IPB C2 and IPB C14 x IPB C2 were positive on high dicotomous character. Heterosis and heterobeltiosis values in F1 IPB C2 x IPB C14 and IPB C14 x IPB C2 were positive on weight per fruit character. The population of F1 that were appropriate used in breeding program to make superior variety were hybrids of IPB C2 x IPB C14 and IPB C14 x IPB C2.

Abstract

Abstrak:       Percobaan ini bertujuan mengevaluasi metode pengusangan cepat methanol 20% selama 0, 2, 4, 6 dan 8 jam pada benih cabai hibrida, non hibrida, lokal, introduksi, rawit dan besar yang diproduksi pada tahun 2009 dan 2010. Jumlah genotipe benih non hibrida 4 (2009) dan 4 (2010). Jumlah genotipe benih hibrida 10 (2009) dan 8 (2010). Analisis data dilakukan dengan analisis sudut kemiringan garis regressi yang merupakan sudut  perbandingan ordinat dan axis. Rekapitulasi hasil evaluasi vigor daya simpan benih cabai menunjukkan bahwa tidak ada perbedaan antara benih produksi tahun 2009 dan 2010, dan benih cabai hibrida dan  non hibrida, serta benih cabai lokal dan introduksi. Perbedaan terdapat pada benih cabai rawit dan besar dan laju penurunannya cabai besar lebih cepat dibandingkan benih cabai rawit.  Hal ini terjadi juga pada tolok ukur daya hantar listrik yang merupakan tolok ukur terbaik untuk vigor daya simpan benih. Hal ini menunjukkan besarnya pengaruh lingkungan terhadap vigor daya simpan benih cabai. Kata kunci: deteriorasi alami, laju kemiringan vigor daya simpan benih cabai. Abstract    The experiment was designed to evaluate the accelerated aging method with 20% methanol for 0, 2, 4, 6 and 8 hours which is the first stage of the experimental results at several pepper seed genotypes including hybrid seeds, non hybrid, local, introduction, pepper and great pepper produced in 2009 and 2010. The seeds used are non-hybrid seed as much as 4 genotype seed production in 2009 and 4 genotype seed production in 2010. Hybrid seeds used were 10 genotypes of production in 2009 and 8 genotypes of production in 2010. Data analysis done is the analysis of the slope angle of regression line slope which is resulting from the comparison of the ordinate and the axis. Recapitulation of the pepper seeds vigor storage evaluation results showed that the seed production in 2009 and 2010 there was no difference between the hybrid and non-hybrid pepper seeds and the local and introduction pepper seeds, but there are consistent differences between great chili and chili pepper seeds, the rate of decline of the great chili is greater than the chili pepper. This shows the magnitude of environmental influences on seed pepper seeds vigor storage. Exclusively in electrical conductivity benchmark, which is one of the pepper seeds vigor storage best benchmark shows that the rate of decline in seed vigor between the hybrid and non hybrid seeds and the local and introduction pepper seeds.. Key words: natural deterioration, the rate of decline seed pepper vigor test in relation  to storability

Abstract

ABSTRACTHypocotyl and cotyledon are potentially used as effective morphological markers since they can be detected earlier. Information on inheritance of tomato hypocotyl and cotyledon was not available. The aims of this research was to study the inheritance of qualitative and quantitative characters of tomato hypocotyl and cotyledon. This research used six population, P1 green hypocotyl (IPB T64), P2 purple hypocotyl (IPB T3), F1, F1R, BCP1, BCP2, and F2. Analysis of qualitative characters used Mendelian and gene action of quantitative characters used joint scaling test. The results of Mendelian indicated that the character of hypocotyl color was controlled by two genes of dominant-recessive epistasis. The gene controlling purple color was dominant to the green color gene. Based on the F2 distribution test, hypocotyl length, cotyledon length and width were controlled by polygenes. There was no influence of maternal effect. The results of the joint scaling test showed gene action of hypocotyl length was controlled by additive gene with influence of additive-dominant epistasis. Length and width of the cotyledon were controlled by additive gene and influence of duplicate epistasis effect. All characters had high level of broad sense heritability and medium level of narrow sense heritability.Keywords: cotyledone, gene action, heritability, hypocotyle, morphology marker.ABSTRAKHipokotil dan kotiledon berpotensi untuk dijadikan sebagai marka morfologi yang efektif karena dapat dideteksi lebih dini. Informasi pola pewarisan karakter hipokotil dan kotiledon tomat belum banyak tersedia. Penelitian ini bertujuan untuk mempelajari pola pewarisan karakter kualitatif dan kuantitatif dari hipokotil dan kotiledon tomat sebagai marka morfologi pada tahap awal pertumbuhan tanaman. Penelitian ini menggunakan enam set populasi yaitu P1 hipokotil hijau (IPB T64), P2 hipokoti ungu (IPB T3), F1, F1R, BCP1, BCP2, dan F2. Karakter kualitatif menggunakan analisis Mendel dan pendugaan aksi gen karakter kuantitatif menggunakan analisis skala gabungan. Hasil analisis Mendel menunjukkan bahwa karakter warna hipokotil dikendalikan oleh dua pasang gen epistasis dominan-resesif. Gen pengendali warna ungu bersifat dominan terhadap warna hijau pada hipokotil tomat. Panjang hipokotil, panjang dan lebar kotiledon dikendalikan oleh banyak gen dan tidak ada pengaruh tetua betina berdasarkan uji sebaran populai F2. Hasil analisis skala gabungan menunjukkan bahwa aksi gen karakter panjang hipokotil dikendalikan oleh gen aditif dengan pengaruh epistasis aditif dominan, panjang dan lebar kotiledon dikendalikan oleh gen dominan dengan pengaruh epistasis duplikat. Semua karakter yang diamati memiliki nilai heritabilitas arti luas dalam tingkatan yang tinggi, sedangkan heritabilitas arti sempit dalam tingkatan yang sedang.Kata kunci: aksi gen, heritabilitas, hipikotil, kotiledon, marka morfologi.

Abstract

 The objectives of this study were to compare nonparametric stability measures, and to identify promising high yield and stability of chili pepper (Capsicum annuum L.) genotypes in eight environments. In every environment, a Randomized Complete Block Design was used with three replications. The method of Nassar and Huehn, Kang, Fox, and Thennarasu was used to analyze the stability and high yield. Spearman’s correlation and Principal Component analysis distinguishes the methods based on two different concepts of stability: the static (biological) and dynamic (agronomic) concepts. The top method was found to be the dynamic stability. Meanwhile, the methods of Si1, Si2, Si3, Si6, Npi1, NPi2, NPi3 and NPi4 were found to be the static stability. Based on the ranking frequency stability of the nonparametric method, the genotypes with the highest frequency of static stability ranking were genotypes IPB002003, IPB002046, IPB009019 and Tit Super, whereas IPB009002 and Tombak were categorized as those of dynamic stability. Genotype IPB120005 and IPB019015 were less adaptable in the multiple environments tested. It shows that the genotypes were specific in certain environments. IPB120005 had high yield and specific location in Boyolali in dry season and IPB019015 genotype was specific in Bogor in wet season. Keywords: chili pepper, nonparametric stability, high yield, dynamic stablility

Abstract

The objective of this study was to identify the adaptability of twelve artemisia mutant genotypes, which were planted in three locations with different altitude, as a result of gamma irradiation. Randomized Complete Block Design (RCBD) was applied in this research with three replications as blocks. The genotypes 1B, 1C, 1D, 2, 3, 4, 5A, 6B, 7A, 8, 14, 15 and two control genotypes as parent genotype from seed and from in vitro were used. The genotypes were planted in three different locations such as Mount Putri, Cianjur (1450 m above sea level), Pacet, Cianjur (950 m above sea level) and Cicurug, Sukabumi (540 m above sea level). Based on the method of postdictive and predictive success, the model used was AMMI2 which was able to explain up to 100% of interaction-influenced variation. The genotypes which were found stabile and adaptive in these three locations were 1B, 1C, 1D, 6B and 15. Genotypes 3 and 7A were adaptive specifically in Pacet area, 5A was adaptive for Gunung Putri while genotype 4 was for Cicurug only.Keywords: AMMI, Artemisia annua, mutant genotype, adaptability

Abstract

This study aimed to obtain probiotic bacteria that are antagonistic bacteria to Colletotrichum acutatum and to improve chilli seeds quality. The purpose of this study was to obtain probiotic bacteria antagonistic to C. acutatum and prevent seed-borne diseases. The layout of the experiment arranged in completely randomized design factorial design with two factors. The first factor is the seed coating that consists of 6 levels i.e control / without coating (c0), coating without bacteria (c1), coating Bacillus sp. (c2), coating Pseudomonas sp. (c3), coating Actinomycetes sp. (c4) and fungicidal coating (c5). The second factor is the storage period is 0, 1, 2, 3, 4 and 5 months. The results showed that three bacteria that are antagonistic to C. acutatum, i.e.Actinomycetes sp. (ATS6) having inhibition percentage of56, 8%, Bacillus sp. (B48) having inhibition percentage 56,7% and Pseudomonas sp. (P12) having inhibition percentage 46,7%. Application of probiotic bacteria increased viability and the health of chilli seed for storage period of 5 months, seen from maximum growth potential 80-84%, germination capacity 76-78,7%, infected seeds in coating of Actinomycetes sp., 2,6% and Bacillus sp., 6,7%.Bacillus sp. contained 91,8 ppm indole acetic acid (IAA) and 103,2 ppm giberelins (GA3), Actinomycetes sp. contained 89,6 ppm IAA and 92,5 ppm giberelin and Pseudomonas sp. contained 68,9 ppm IAA and 69,2 ppm giberelin.

Abstract

Begomovirus infection has a significant impact of lowering chilli yield in Indonesia. A constraint of narrow genetic variability of chilli in Indonesia has made the mutation breeding program as a solution worth-pursuing in increasing the genetic variability. The objective of this study was to determine the LD50 point for each of the five irradiated chilli genotypes and the optimum dose of gamma irradiation in inducing chilli resistance to Begomovirus and other improved agronomical traits. The study was conducted in the Indonesian Vegetable Research Institute (IVEGRI) at Cikole-Lembang, elevation 1,200 m above sea level, from March to December 2013. Split plot design was used with genotype as main factor (Kencana, Lembang-1, SSP, Tanjung 2, Seloka) and irra-diation dosage as sub-factor (0, 200, 400, 600, 800 Gy). All treatments were replicated three times. The results showed that LD50 points of the five irradiated chilli genotypes were in the range of 422.64-629.68 Gy. There were some chilli genotypes in the population of M2 that had high coefficient variance genetic (CVG) and broad sense heritability (h2bs) value for disease incu-bation time. This could be used as resistance parameter to Begomovirus and improvement parameter of several agronomical traits.

Abstract

Selection is very important activity in plant breeding program. The purpose of this study was to obtain genetic information, heritability and expected genetic advance, and to compare the genetic advance expectations with the selection advance. The research was conducted from September 2013 to November 2015 at Research Station of Leuwikopo, Dramaga, Bogor. Distribution of F2 population data in this study was broader than that of F3 populations. Broad sense heritability estimated by F3 populations showed in agreement to that estimated by F2 population. Character of fruit weight, fruit length, pedicel length and yield had positive value of selection advance appropriate to the genetic advance expectations in F2 populations. The value of heritability estimate and genetic advance were high indicating that the phenotypic variances were controlled by action of additive genes.Keywords: genetic advance, heritability, selection advance

Abstract

ABSTRACTInformation on genetic variability and correlation between quantitative characters with yield are important for support the selection program. The objective of the research was to estimate the genetic variability, heritability, and path analysis on agronomic characters to determine the selection criteria in the chili. This research was conducted at Research Station of Leuwikopo, Darmaga, Bogor from November 2009 until May 2010. This research observed all populations i.e. F5 population: 320 plants, IPB C2 population: 20 plants, and IPB C5 population: 20 plants. The results showed that total fruit weight, thick and fruit diameter, middle fruit diameter, blossom end fruit diameter, fruit weight, and days to flowering have a high broad sense heritability . High coefficient of genetic variability values were obtained ini number of fruits per plant, fruit wei ght, stem diameter, fruit diameter, and fruit weight. Based on the heritability, genetic variability, correlations analysis and path analysis, characters that can be used as selection criteria in this study is the number of fruit per plant, fruit weight, and fruit diameter.Key words: genetic variability, heritability, path analysis, selection
