Abstract

Dextransucrase is a glucasyltransferase that catalyzes the transfer of an alpha-D-glucopyranosyl group from sucrose to dextran and releasing the fructose. Enzyme activity was determined by measuring the initial reaction rate of fructose released from sucrose. A standard unit (U) is defined as the amount of enzyme that catalyzes the formation of 1 umol of D-fructose per mg of protein per minute. Dextransucrase was produced by fed-bath cultures of isolate bacterial from sugar cane in a mixture containing sucrose (10 % b/v) in themedium fermentation. Extracellular dextransucrase was obtained from bacterial isolate from sugar cane. Final culturedextransucrase activity was 0,2776 Ulmg of protein. Dextransucrase purified by ethanol 80 % showed activity of 4,2077 Ulmg of protein or activity increase 1515,75 %. Temperature optimum was 40 OC and pH optimum was 7.6. All of the metal increased the activity, except HgC12. Dectransucrase kinetics constants, Km is 3,00 mM and Vmaks is 3,5559 unit/mg of proteinlmin. Molecular weight is 95.155 kDa.

Abstract

The purpose of this research is to get information about the effect of yoghurt on the growing teeth of Rat (Rattus norvegicus strain wistaf). This research used 30 male rats, which divided into three groups. Each group consist of 10 rats. One group was one month old and the others were two and three months old. One of the rats' incisor teeth was cut and the rest length of the teeth were measured by sliding ruler. Each group of the rats was divided into two sub groups. One sub group was given 1,O ml of yoghurt, the others were glven 1,5 mi and 2,O ml of yoghurt once a day for 10 days. The other sub group was given aquadest with the same dose as a control group. Everyday the length of the teeth was measured. The results of this research indicated that yoghurt can accelerate the growth of the rat's teeth, but based on the statistical analyzed it showed nosrgnificant difference.

Abstract

Biology reductions of coffee residue, waste from PT. Torabika industry, Tangerang using earthworm have been carried out. The aim of experiment is to know the ability of earthworm to decompose the waste become verrnicompost and to evaluate it quality. The experiment was used earthworms Eisenia foetida species and 25 - 75 % of cow manure as a medium mixture. The result showed that the structure of vermicomost is soft andblack colors with cation capacity is 17.49 - 21.32 megll00. The mineral content were determined using X-ray Fluorescence Spectrometer (XRF), UV-Vis Spectrophotometer and Kjeldahl, the result showed that the concentrations of N are 0.93 - 0.94 %, CI N 11.26 - 15, K 6.64 - 6.95 %, Ca 12.1 - 17.3 %, P 0.09 - 0.16 %, Mn 0.11 - 0.19 %, Fe 2,01 - 2,69 %, Zn 538 - 779 mgl kg. The earthworm biomass was increased until thirdweeks of vermicomposting.

Abstract

To determinine the inhibitory mechanism of antibacterial Maillard reaction products (PRM), heated mixtures containing GL (glukosa-tisin) and TU (molasses-urea) were were studied fcx their effect on uptake oxygen and iron solubility. It was found that the tested PRM had an inhibitory effect on the uptake of oxygen was observed. The PRM also had an effect on the solttbirity of iron in nutrient broth and in phosphate buffer. While the GL mixturereduced the solubility of iron it was increased by the TU mixture.
