This journal gives readers the state of art of the theory and its applications of all aspects of fisheries and marine science. Abstract

Abstract One of the cause of negative gram bacteria is Pseudomonas aeruginosa (Ps. aeruginosa). It continuing become sepsis on the first step of patogenesa happen sticking between aeruginosa with endhotel cell. This sticking is mediated by adhesi molecul which have characterize same with hemaglutin protein. The main study of this research is to know the profile of endothelial cell culture within induction of Ps. aeruginosa. The methods are Ps. aeruginosa (9064) which have been isolated then processes on TCBS Media and continued with isolation terracely. The profile of weight molecule protein obtained by SDS PAGE and then conducted electroelution. The test of hemaglutinin using eritrosit mencit. The receptor cell used is endhotel cell (HUVECs) culture. The result show that dosage of hemaglutin protein of Aeruginosa bring effect to profile of HUVECs culture determined by index adhesion of Ps. aeruginosa to HUVECs culture. We conclude that protein 38,19 kDa give effect to profile of HUVECs culture in vitro.

Abstract

Abstract The aim of these study was to know plankton (phytoplankton and zooplankton spesies and density at Sengguruh dam, Karangkates, Lahor, Wlingi Raya and Wonorejo, and to estimated trophic status these dams use phytoplankton and zooplankton density approachment. These study used descriptive methode with survey technic. Data wich is taken from each dam is taken from four different station that suggested the representated dam condition. The result showed according to the range phytoplankton density: at Karangkates between 144-236 ind/ml is clustered as oligotrophic, the range phytoplankton density at sengguruh between 217-313 ind/ml is clustered as oligotrophic. At Lahor dam between 5.35612.704 ind/ml is clustered as mesotrophic, at Wlingi Raya dam between 74-1480 ind/m' is clustered as mesotrophic to eutrophic, while at Wonorejo dam between 48-288 ind/ml is clustered as oligotrophic waters. According to the range zooplankton density: at Karangkates dam between 28.153-132.056 ind/lt is clustered as mesotrophic, Lahor dam between 4.996-14.275 ind/lt is clustered as mesotrophic, Wlingi Raya dam between 623-3.114 ind/lt is clustered as oligotrophic to mesotrophic, and Wonorejo dam between 2-6 ind/lt is clustered as mesotrophic. Abstract

Abstract One of the most favourite fish that farmer fish cultivated because its high economic value was gouper (C. altivelis). The obstacle in cultivation system was caused by Vibrio algynolitycus. Vibrio angullarum, Vibrio parahaemolyticus. Vibrio harveyi infection. Many deseases that attacked to the grouper was caused by the role of pathogen specific as a ligan and its reseptor. The aim of these study was to improve and to know the role of reseptor in vibriosis mechanism with tested spesific reseptor protein of grouper Cromileptes altivelis. These study used experimental and descriptive methode with identified of grouper reseptor, Hemaglutinin test and observation use electron microscope. These study result showed that reseptor protein of C. altivelis was found in its eyes. These showed spesivity and reactivity to the vibriosis specific epitop.

Abstract

Abstract Tiger shrimp (Penaeus monodon Fab.) is one of the biggest non-oil and natural gas export fisheries commodity. In the other hand, the culture of this shrimp is highly affected by infectious pathogens and disease control is ciliated protozoa, Zoothamnium penaei that play an important role as ectocommensals on the body surface of tiger shrimp and causes "udang lumutan” disease. The aim of this research are to know the destruction and mean total haemocyte count of the Tiger shrimp between healthy and easy, middle and heavy infected degree of Zoothamnium penaei. The healthy and was identified infected Zoothamnium penaei of Tiger shrimp samples collected from one of the nursery pond in Lamongan that divided of 50 healthy indivisuals and infected Zoothamnium penaei (50 individuals for each infestation degree). Haemolymph (haemocytes) was obtained from the ventral part of the haemocoel of the second abdominal segment using 1 ml syringe filled with EDTA (Ethylene Diamine Tetra Acetic Acid). The variable observed were destruction of haemocyte and mean total haemocyte count during 7 days. ANOVA (Analysis Of Varian) was used for mean total haemocyte count change data analysis followed by Duncan's Multiple Range Test with 5% significantly. The research results revealed that Tiger shrimp that easy, middle and heavy infected of Zoothamnium penaei no destruction occurs, but Tiger shrimp that heavy infected of Zoothamnium penaei occurs haemocyte sitoplasma destruction. The highest mean total haemocyte count change was 61,64x106 ± 3,56x106 cells ml-1 in Tiger shrimp that middle infected of Zoothamnium penaei, while the lowest mean total haemocyte count change was 41,04x106 ± 3,64x106 cells ml-1 occur in Tiger shrimp that heavy infected of Zoothamnium penaei.

Abstract

Abstract Daphnia magna has good nutrition to carry on egg produce and fry of fish cupang, maskoki, oscar, tetra, and also can used for food source of fry and seed. Daphnia magna not only supply from natural environment, cause their growth deppend on fertilizer as food and environment condition. The other side: high population density, limited of food source low temperature and shorter irradiation can produce dorman egg (ephippia). The dorman eeg can use for Daphnia magna stock which way by hatching. The factor that can be influence for ephippia hatch are temperature and density. Based on this fact, a research about influence of different temperature and ephippia density for ephippia hatch of Daphnia magna. The purpose of this research was to know the influence of different temperature and ephippia density for ephippia hatch of Daphnia magna. This research has experimental characteristic and use Factorial Completely Randomized Design with three experiments of temperature (A factor), three experiments of density (B factor) and three times replicated. There are 9 kinds of combination between density and temperature. The parameter which checked in this research is degree of hatch of ephippia Daphnia magna and water quality that is dissolve oxygen rate, NH3 and of pH hatch media. The result of the analysis showed that the temperature and density influence of hatching Daphnia magna ephippia. The best result can be used for hatching Daphnia magna ephippia is temperature at 25oC with density 50 ephippia /100 ml giving highest result for the hatch of ephippia Daphnia magna. There are interaction betwen temperature and density for hatching.

Abstract

Abstract The aim of this research was to know the effect of honey supplementation in feed of male main freshwater cray fish red claw to larva sex ratio and the level of survival of freshwater cray fish and to get the best doses of honey to give highest male sex ratio. This research was done at Laboratory of Fishery Education, Fishery and Marines Faculty, Airlangga University in October 2007 until March 2008. Methods of this research was experimental and used completely randomized designed with 7 treatments. Giving of honey with differences doses were A ( 0 ml/kg) control, B ( 50 ml /kg), C ( 100 ml /kg), D ( 150 ml /kg), E ( 200 ml /kg), F ( 250 ml/kg) and G ( 300 ml/kg) which each treatment got three times replicated. Data processed by Analysis of Variance (ANOVA). The result of this research showed that supplementation of honey in feed male main freshwater cray fish red claw not significantly different to larva sex ratio and survival rate of freshwater cray fish. Otherwise, result of the supplementation of honey in feed of male main freshwater cray fish showed the increasingly male sex ratio of freshwater cray fish red claw larvae.

Abstract

Abstract Cadmium is one of dangerous heavy metal and it can cause water pollution. As an alternative early warning toward heavy metal in water, Daphnia magna can be used as biology indicator water pollution that caused by heavy metal. Daphnia magna is organism that ussualy used for bioassay in many progressing countries, because that organism have important role in freshwater ecology, short life cycle about 3 weeks and sensitive toward chemical environment. Grade of toxicity heavy metal can measured with lethal and sub lethal parameter. In Daphnia magna, sub lethal parameter that can be observe are colour changes and male sex percentage neonates. The purpose of this research was to know the colour changes and male sex percentage neonates Daphnia magna in different concentration of cadmium. Target of this research is to gets information about level colour changes and male sex percentage neonates Daphnia magna in different concentration of cadmium. The main parameter was colour changes and male sex percentage neonates and the secondary parameters were water quality (temperature, pH, dissolved oxygen and NH3) The result of research of giving different cadmium concentration to the adult female Daphnia magna not showing significant influence, nevertheless it is showing significant influence toward male sex percentage neonates Daphnia magna. The treatment with cadmium concentration 0,0004 mg/l produce 100% male neonates Daphnia magna. Water quality during research were in optimal conditions to support Daphnia magna life, those are temperature 26°C, pH range from 8,0 – 8,2 and dissolve oxygen (DO) range from 8,5 – 9,0 mg/l and ammonia 0 – 0,03 mg/l.

Abstract

Abstract The growth of betutu classified slow, so do efforts in order that growth of betutu which protected optimal by choosing the exact food combination. This research is purpose to know the growth fingerlings of betutu. This research using complete random program giving 3 repeated by the way of treating P1 (food 100% pelet), P2 (food 100% Daphnia spp.), P3 (food 100% Tubifex sp.), P4 (food 100% Anadara granosa), P5 (food combination 50% pelet and 50% Daphnia spp.), P6 (food combination 50% pelet and 50% Tubifex sp.), P7 (food combination 50% Anadara granosa and 50% Daphnia spp.), P8 (food combination 50% Anadara granosa and 50% Tubifex sp.). Main test parameter checked on this research is growth (accretion weight), growth rate, specific growth rate, and absolute body length growth, while supporting test parameter is water quality. Data of growth analysis using ANAVA (varian analysis) and if there is the influence the way of treating so continued with Test of Multiple Gap Duncan by trusty degree 95%. The result of research show the growth (accretion weight), growth rate, specific growth rate and the best absolute body length growth on the way of treating P6 which not different with the way of treating P3 and P8.

Abstract

Abstract African catfish (Clarias gariepinus) is one of fish consumption use to fulfill animal protein. The fulfillment need of animal protein with aquaculture effort, where as in the aquaculture effort will larvae quality increased. Increasingly of larvae quality with natural food had been adding feed supplement. Natural food to used is Daphnia spp. and then food supplement to used is viterna. The viterna will increase Daphnia nutritions, because of the viterna contains many nutritions to survival and growth of African catfish. The aim of this study was to know the influence of Daphnia spp. enrichment with viterna to survival and growth of African catfish (Clarias gariepinus) larvae. The rearing of larvae during 40 days. This study used Complete Random Design. The treatment were A (present Daphnia spp. without viterna), B (Daphnia spp. enrichment with viterna 10 ml/L dose), C (Daphnia spp. enrichment with viterna 10 ml/L dose), D (Daphnia spp. enrichment with viterna 10 ml/L dose), E (Daphnia spp. enrichment with viterna 10 ml/L dose) dan F (Daphnia spp. enrichment with viterna 10 ml/L dose), each treatment with 4 replications. The result show that viterna applications to Daphnia was significantly influenced (p<0,05) to survival African catfish larvae. The highest survival was E treatment (7,25 %). The viterna leaf to Daphnia spp. Was significantly influenced (p<0,05) to specific growth rate African catfish larvae. The highest specific growth rate was B treatment (1,885 %BW/day).

Abstract

Abstract Daphnia spp. was potential source of live feed to be developed for larvae of freshwater fish. One of maximize culture technology was daily feeding system with rice bran solution. This system able to increase Daphnia spp. population. The problem of daily feeding culture technology was water quality stability, such as high pH fluctuation and high ammonia concentration. Water pH stability and decrease of ammonia concentration could be increased by dolomite added which function as stabilize pH buffer. The aim of this research found out the dolomite dose and culture period that able to increase population and stabilize of water quality. Experimental design was Completely Random Design with 4 replicates those were A control (0 mg/l), B (50 mg/l), C (100 mg/l), D (150 mg/l), E (200 mg/l) and F (250 mg/l). Parameter that measured were Daphnia spp. population and water quality, such as temperature, pH, ammonia, dissolve oxygen and alkalinity. The data were examined using ANAVA(Analysis of Variance) to know treatment effect and be continued by Duncan Multiple Range with trust of degree 0,05. for water stability using descriptif methode. Result of analysis showed that high population for all treatment were held on 6th, 7th and 8th day with the highest Daphnia spp. population was held on 8th day by F treatment (250 mg/l) were 9900 Daphnia spp./l. Water quality data showed different between each treatment.
