Abstract

Chemical contents of Kesum (Polygonum minus Huds), such as phenolic, flavonoids, alkaloids, steroid-terpenoids and tannin, can act as antibacterial agent.The aims of this research was to determine whether methanol extract of kesumleaves have antibacterial effect and its effectivity when it was formulated in gelproduct. Extract activity test group being used are negative control, methanolextract 5%, 10% and 15%. Gel effectivity test group being used are negativecontrol, positive control, Gel A, (5% methanol extract), Gel B (10% methanolextract) and Gel C (15% methanol extract). Disc diffusion Kirby-Bauer methodwas used to determine the antibacterial effectivity against Escherichia coli. Gelstability test carried out for 31 days include organoleptic, viscosity, ability ofspreading and sticking and pH. The methanol extract 5%, 10% and 15% gaveantimicrobial effect. It was formulated in gel product with that concentrations.The result of the effectivity test and stability test were analyzed statistically bySPSS 17.0 using One-Way ANOVA and continued to Post Hoc Tukey Test.Diameter of the inhibition zone gel was Gel A (10.66 mm) Gel B (12.11 mm), GelC (12.33 mm). Gel C gave best antimicrobial effect between Gel A, Gel B andpositive control. So it has potensial to be antiseptic agent. In all variables of gelstability test, Gel C has a few changes, so Gel C stability is better than Gel A andGel B.Keywords: kesum leaves, gel, antiseptic, Escherichia coli

Abstract

Ascorbic acid is a compound that unstable, easily oxidized when exposedto aerial (oxygen) and this process can be accelerated by heat. The purpose of thisresearch is to see the influence of storage conditions on the quality of drugsphysical and chemical tablets of ascorbic acid at the district health centers whichincludes Kampung Bali, Alianyang, Pal tiga and Karya Mulia with differentstorage conditions in each health centers. The method used is non-experimental,and use probability sampling method with simple random sampling. Samplingwas conducted during two months. In this research, do measurement temperature,humidity and evaluation test tablets that include test physical appearance,uniformity of weight, uniformity of size, friability, hardness, disintegration time,dissolution and value, then the results were analyzed statistically using One WayANOVA and Kruskal Wallis. The results showed that ascorbic acid tablets in thefirst and second health center in Kampung Bali, Alianyang, and Pal tigaparameters qualify the uniformity of weight, disintegration time and dissolution,while the Karya Mulia of community health center tablets meets all requirementsof ascorbic acid tablets are good, except the value parameters and uniformity ofsize. Based on the results of this research concluded that the influence of theconditions and duration of storage of drugs affecting the quality of ascorbic acidtablets. Statistical analysis evaluating ascorbic acid tablets between each healthcenters in the first month and the second shows that the health center Karya Muliashowed a significant difference with health center Kampung Bali, Alianyang andPal tiga.Keyword: Ascorbic acid Tablets, Health Centers, Storage Conditions

Abstract

Mefenamic acid is a derivative of anthranilic acid is widely used by the public asan analgesic, antipyretic, and anti-inflammatory. Mefenamic acid is a drug which issensitive to light, temperature and humidity. This research aims to determine the qualityof mefenamic acid caplets that stored in Pontianak City community health centre as amatter of consideration in storage of both drugs in community health centre. Research hasbeen conducting by doing some observation on drug storage conditions such astemperature and humidity with non-experimental methods. Mefenamic acid samples weretaken twice during the research, mefenamic acid samples at the beginning and at the endof storage that had been stored for a month in storage room of each community healthcentre. The quality of mefenamic acid caplets evaluated, namely physical and chemicaltests. The results showed that the storage conditions at 4 community health centres do notall qualify, only Alianyang community health centre that qualified. Community healthcentres room temperature ranged from 25 to 33,3 C, humidity 52 to 88,3%. Temperatureand humidity affect the physical and chemical quality of mefenamic acid caplets in allexamination parameters except weight uniformity. Quality phsysical and chemical ofmefenamic acid caplet getting dropped after being stored for a month, but still meet therequirements of a good quality caplets.Keywords: mefenamic acid, temperature, humidity, health centre

Abstract

Drug interaction happen when drugs is given together with foods. This researchwas aimed to observe the effect of mangosteen juice to the paracetamol pharmacokineticprofile in Wistar male white rats. The study was using male Wistar rats weight 200-350 g.The animals were divided into three groups with the total of the male rats are 15. Group I(paracetamol) was given a single oral paracetamol 9 mg/200gBW. Group II (treatment 1)was given a single oral paracetamol 9 mg/200gBW together with mangosteen juice 2,88g/200gBW. Group III (treatment 2) was given a single oral paracetamol 9 mg/200gBWtogether with mangosteen juice 0,66 g/200gBW. The serial blood was collected from tailvein for 9 hours and were analyzed using spectrophotometer UV for unchangedparacetamol. The pharmacokinetic parameters of paracetamol were calculated byrecidual method and were analyzed by One Way ANOVA using 95 % confidenceinterval. The difference between groups were analyzed using Post Hoc LSD-test. Theresults showed that the group II (paracetamol-mangosteen 1) and group III (paracetamolmangosteen2)increased of ka, Cpmaks, t, tab, ke, tel, CL, AUC and decreased ofka, ke, and CL. The research showed that mangosteen juice doses 2 give higher decreasedmetabolism and decreased elimination of paracetamol in rat.maksKey words : Mangosteen fruit, Pharmacokinetics, Paracetamol, Drug interaction

Abstract

Insomnia  is a condition difficult start to sleep, can not sleep or inadequate sleepduration. One of plants which guess have sedative effect is kratom (Mitragyna speciosa). This research was conducted to determine the sedative effect of kratom leaves infusa to male BALB/cstrain mice. In addition, it aims to determine the effective dose and potential kratom leaves infusawhen compared with diazepam The mice divided into 5 groups of  positive control group (diazepam), negative control(aquadest), and infusa of kratom leaves groups dose 1,95; 3,9; and 7,8 g/KgBW. The test of sedativeeffect was conducted with Traction test and Fireplace test method on male BALB/c strain mice.Monitoring of quantitative data are duration time of mice to turning the body and duration time ofmice to fall down, as soon as duration time of mice to come out from cylinder tube. Qualitative data isobserved presence or absence of reflexes behind the body and corneal reflexes. Result of the quantitative data was analyzed with One Way ANOVA and supported by SPSS.Based on the result, all of kratom leaves infusa dosages has sedative effect, which is the mosteffective dose kratom leaves infusa at dose of 7.80 g / KgBW. But  the sedative effect still below ofdiazepam. Key Word : kratom leaves, sedatives, infusa.

Abstract

Paracetamol is widely used as analgesic and antipyretic agent in treatment of painand fever. Paracetamol has interaction with carbohydrates and alcohol. Durian is the fruitnative of Indonesia, that contain carbohydrates and alcohol. The aims of this researchwere to study the influence of durian fruit juice to the absorption and elimination kineticsof paracetamol and to know the dose of durian fruit juice that influence the absorptionand elimination kinetics of paracetamol. The study was conducted using 16 rats, dividedinto 4 groups (n=4 per group). Each group was treated the following treatment : controlparacetamol (paracetamol 9 mg/200 gBW), dose 1 group (paracetamol 9 mg/200 gBWand 0.675 g/200 gBW of durian fruit juice), dose 2 group (paracetamol 9 mg/200 gBWand 1.350 g/200 gBW of durian fruit juice) and dose 3 group (paracetamol 9 mg/200gBW and 2.700 g/200 gBW of durian fruit juice). Blood sampling is done from the veinof rats tail at minutes 10, 20, 30, 40, 60, 90, 120, 180, 240, 300 and 360. The quantitationof paracetamol in plasma was determined by UV spectrophotometer at 243 nm. Abstract

Obat yang digunakan bersamaan dengan makanan atau minuman dapatmempengaruhi efek terapi obat tersebut. Penelitian ini dilakukan untuk mengetahuipengaruh jus buah nanas (Ananas comosus (L.) Merr.) terhadap profil farmakokinetikparasetamol pada tikus putih jantan galur Wistar. Hewan uji dikelompokkan secara acakdibagi menjadi 3 kelompok. Kelompok I (parasetamol) diberikan parasetamol tunggaldosis 9 mg/200gBB peroral. Kelompok II (parasetamol dan jus nanas dosis 1) diberikanparasetamol dosis 9 mg/200gBB bersamaan dengan jus buah nanas dosis 2,7 g/200gBB.Kelompok III (parasetamol dan jus nanas dosis 2) diberikan parasetamol dosis 9mg/200gBB bersamaan dengan jus buah nanas dosis 5,4 g/200gBB. Cuplikan darahhewan uji diambil selama 9 jam pada vena lateralis ekor tikus. Penetapan kadarparasetamol pada plasma dilakukan dengan spektrofotometer UV. Parameterfarmakokinetik dihitung menggunakan metode regresi linear dan metode residualselanjutnya diuji secara statistik menggunakan One Way ANOVA dengan tingkatkepercayaan 95%. Berdasarkan hasil penelitian memperlihatkan bahwa kelompok II dankelompok III meningkatkan secara signifikan parameter Cpmaks, t, tab, tel, AUCdan menurunkan secara signifikan parameter ka, ke, dan CL. Kelompok III memberikanpengaruh yang paling kuat terhadap profil farmakokinetik parasetamol yaitu denganmenurunkan parameter absorbsi dan eliminasi serta meningkatkan parametermetabolisme parasetamol pada tikus.Kata kunci : Parasetamol, Nanas, Farmakokinetik

Abstract

Abstrak: Antioksidan merupakan senyawa yang dapat digunakan untukmengatasi kerusakan oksidatif akibat radikal bebas. Senyawa antioksidanmemiliki peranan yang penting dalam kesehatan. Satu diantara tanaman yangmemiliki aktivitas antioksidan adalah tanaman bawang mekah (Eleutherineamericana Merr.). Penelitian ini bertujuan untuk mengetahui aktivitas antioksidandari daun tanaman bawang mekah. Pengujian aktivitas antioksidan dilakukandengan menggunakan metode DPPH (2,2-difenil-1 pikrilhidrazil) yang diawalidengan ekstraksi secara sokhletasi menggunakan etanol 70%. Fraksi etanoldiskrining fitokimia menggunakan metode uji tabung dan diuji pendahuluandengan metode DPPH menggunakan KLT silika gel 60 Fyang dielusi denganfase gerak kloroform p.a. Aktivitas antioksidan fraksi diukur menggunakanspektrofotometer UV-Vis dengan variasi konsentrasi 10; 20; 30; 40 ?g/mL danvitamin C dengan konsentrasi 2; 2,5; 3; 3,5 ?g/mL sebagai kontrol positif. Hasilskrining fitokimia menunjukkan fraksi mengandung flavonoid, saponin, dan fenol.Hasil uji pendahuluan fraksi diperoleh 1 bercak dengan hRf 46,25 yangdivisualisasi dengan sinar UV 366 nm. Bercak pada fraksi menunjukkan hasilpositif antioksidan yang ditandai dengan perubahan warna kuning dengan latarbelakang ungu setelah disemprot dengan DPPH 0,2%,. Hasil pengukuran secaraspektrofotometri UV-Vis menunjukkan bahwa fraksi etanol memiliki aktivitasantioksidan dengan IC50254335,5468 ?g/mL dan vitamin C memiliki nilai IC0,62147 ?g/mL.Kata kunci: antioksidan, daun bawang mekah, fraksi etanol, DPPH

Abstract

Antioksidan adalah senyawa yang dapat menghambat aktivitas radikal bebas.Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dari daun buas-buas.Pengujian aktivitas antioksidan dilakukan dengan menggunakan metode DPPH (2,2difenil-1-pikrilhidrazil)yang diawali dengan ekstraksi secara destilasi. Ekstrak etanolyang diperoleh difraksinasi dengan pelarut berdasarkan tingkat kepolaran yaitu n-heksan,kloroform dan etanol. Fraksi yang digunakan adalah fraksi etanol. Selanjutnya dilakukanskrining fitokimia dengan metode uji tabung dan uji pendahuluan dengan metode DPPHsecara kromatografi lapis tipis (KLT) dengan fase gerak kloroform p.a. Aktivitasantioksidan diukur secara spektrofotometer UV-Vis menggunakan DPPH 0,1 mM padapanjang gelombang 515,40 nm dan kontrol positif yang digunakan adalah vitamin C.Hasil skrining fitokimia menunjukkan  bahwa fraksi mengandung senyawa flavonoid,saponin, fenol dan alkaloid. Hasil KLT diperoleh 2 bercak dengan nilai hRf sebesar 38,75dan 61 yang divisualisasi dengan sinar UV 366 nm serta disemprot dengan DPPH 0,2%.Kedua bercak menunjukkan perubahan warna kuning dengan latar belakang ungu yangmenandakan fraksi memiliki aktivitas antioksidan. Hasil pengukuran secaraspektrofotometri menunjukkan bahwa fraksi mempunyai IC pada 27,853 μg/ml,sedangkan vitamin C memiliki nilai IC yang lebih rendah yaitu 0,623 μg/ml.Kata kunci: aktivitas antioksidan, daun buas-buas, DPPH.

Abstract

Salah satu penyebab terjadinya jerawat adalah bakteri, seperti Propionibacteriumacnes dan Staphylococcus epidermidis. Jahe Merah (Zingiber officinale Rosc. var. rubrum)telah banyak diteliti memiliki aktivitas antibakteri. Tujuan dari penelitian ini adalahmengetahui konsentrasi hambat minimum ekstrak etanol rimpang jahe merah terhadapbakteri penyebab jerawat dan bagaimana efektivitas setelah diformulasikan dalam bentuk gel.Simplisia disokhletasi menggunakan pelarut etanol 96%. Pengujian KHM dilakukan denganmetode difusi. Ekstrak diformulasikan dalam bentuk gel dengan variasi konsentrasi basisHPMC 4000 dan Karbopol 934 dengan perbandingan 70:30 (Formula I), 50:50 (Formula II)dan 30:70 (Formula III). Evaluasi sediaan meliputi pemeriksaan organoleptis seperti bau,warna, bentuk serta homogenitas, pengujian daya sebar, daya lekat, pH dan antibakteri.Analisis data menggunakan program R-Commander versi 12.4.1. Hasil penelitianmenunjukkan nilai KHM eksrak terhadap P.acnes sebesar 0,45% dan S.epidermidis sebesar0,5%. Hasil evaluasi menunjukkan sediaan homogen, pH dan daya lekat yang stabilsedangkan daya sebar mengalami peningkatan selama penyimpanan 30 hari. Hasil evaluasiantibakteri gel menunjukkan bahwa formula I memberikan efektivitas paling baik denganzona hambat sebesar 16,11 mm terhadap P.acnes dan 14 mm terhadap S.epidermidis.Kata kunci: Ekstrak etanol rimpang jahe merah, Propionibacterium acnes, .Staphylococcus epidermidis
