Abstract

Aim of the research is to determine the optimum concentration that has the best effect on the tissue culture induction of onion discus. This research was carried out in August to November 2017, in Biotechnology Laboratory, Agriculture Faculty, UPN "Veteran" Jawa Timur. Completely Randomized Design method was used with 1 factor and repeated for 10 times. Four levels treatment of the growth regulator in this study are Z0 (Control (MS)), Z1 (MS + BAP + 1,5 ppm), Z2 (MS + BAP + 2,5 ppm) and Z3 (MS + BAP + 3,5 ppm). The parameters are the percentage of explant growth potency, growth percentage of the explant roots, growth percentage of the explant shoots, the percentage of stagnancy explant, leaves length and the number of roots and leaves. Data was analyzed used ANOVA (Analysis of Variance) and the Honestly Significant Difference Test 5%. As the results, the addition of the BAP with a concentration of 2,5 ppm is the best treatment for all parameters.

Abstract

Aim of the research is to know the performance and heritability value of ten from first generation of Madura mutant maize varieties (Zea mays sp.). It was conducted at Agriculture Faculty, UPN “Veteran” Jawa Timur on July-September 2017. Randomized Block Design method was used with 1 factor that consists of 10 lines first generation Madura mutant maize varieties and repeated 3 times. Data was analyzed used ANOVA (Analysis of Variance) and the Honestly Significant Difference Test 5%. As the results, MM-100-3-023 has the best performance among the ten first generation of Madura mutant maize varieties for plant height, the number of leaves, seed weight, 100-grain weight, number of seeds, cob weight and dry weight of the plant, and the heritability showed the low, medium and high range.

Abstract

The purpose of this research is to know the effect of earthworm Lumbricus rubellus on vermicomposting rate of urban waste. The research was conducted from October to December 2016. The research was conducted in the laboratory of Land Resources of the Faculty of Agriculture, Universitas Pembangunan Nasional “Veteran’’ Jawa Timur. The method used in this study was the randomized compartment design which is arranged in completely random. The design of the distributed plot is comparing from two experimental unit types between the main plot and the plot subgroup where the environmental design for both can be the same or different. Treatment on main plot (PU) in the form of composting using earthworm (V) with 2 levels of V0 without worms and worms using V1. While in the treatment of child plot (PA) in the form of composting time (M) with 5 levels namely; M1 = Week 1, M2 = Week 2, M3 = Week 3, M4 = Week 4, and M5 = Week 5. The result of vermicomposting rate influenced by earthworm Lumbricus rubellus obtained total N, total P, total K, C/N ratio was good. Composting using earthworms gives significantly different results than regular composting. The value of observation parameters of composting using earthworms was not significantly different with increasing time.

Abstract

This research uses the design of the plot divided with the treatment of ex- planting is the main plot whereas the other treatment is a subplot consisting of 2 (two) factors and repeated 3 times. The first factor is rotation of Soybean-Rice and Corn-Rice Planting. The second factor was treatment of fertilizer composition, P0 (control), P1 (100% compost), P2 (50% compost + 50% N inorganic), P3 (75% compost + 25% N inorganic). The treatment (L2P1) yielded the highest 1000 grain weight of 25.28 g, L2P3 yielded the highest amylose content of 20.51%, L1P0 resulted in amylopectin content of 58.42%. The treatment of L2 resulted in 0.92 kg dry weight of dry paddy grain, yield 1000 grains weighing 25.10g, 19.23% amylose and L1 treatment yield the highest amylopectin content of 54.39%. The treatment (P2) resulted in the highest plant length of 102.67cm, P1 yielding 1000 grains of 24.36g, P3 yield amylose content of 20.00%, P0 yielding amylopectin content of 57.72%.

Abstract

The purpose of this study to determine the concentration of Streptomyces sp. that able to control pupa the fruit fly Bactrocera sp. up to 50 percent of death and the influence of different concentration Streptomyce to long life and symptoms attacks in the fruit flies pupa Bactrocera sp. This research was carried out on June – August 2017 in the health of plants laboratory of Agriculture Faculty UPN “Veteran” Jawa Timur. The research was carried using of randomized block design with the concentration of Streptomyces sp. 5, 10, 15, 20 and 25. Each treatment is repeated as much as five times. The result obtained is an average of a long life the pupa as a result of treatment Streptomyces sp. accelerating to four to six days. Pupae were successfully hatch into imago a physical disability funds have short life. P arasitic symptoms of fruit flies pupa by Streptomyces sp. were changed become black. Postulate Koch of the fruit flies pupa can be seen with the spore and a distinctive smell of Streptomyces sp.

Abstract

This work aimed to study the effect of (Benzyl Amino Purine) BAP and coconut water in Murashige and Skoog (MS) medium on shoot induction of shoot Fig. For this purpose, 1 ppm and 2 ppm of BAP and 100 ml/l and 150 ml/l of coconut water were in MS medium with or without carbon, were investigated. This research used Completely Randomized Design with 9 treatments, there are zero MS, MS+BAP 1 ppm, MS+BAP 2 ppm, MS+BAP 1 ppm+carbon, MS+BAP 2 ppm+carbon, MS+CW 100ml/l, MS+CW 150 ml/l, MS+CW 100 ml/l+carbon, and MS+CW 150 ml/l. Shoot of the Fig were cultured on those MS medium supplemented with 30 mg/l sugar for in vitro shootlet proliferation. It was found that MS medium supplemented with BAP 1 ppm+carbon, enhanced shoot development. In this study there were still many explants were died because of browning and contamination.

Abstract

Fusarium wilt in bananas is caused by Fusarium oxysporum f.sp. cubense (Foc). Foc is one of the soil-borne pathogens. The intensity of disease attacks in different conditions with very severe is conducive soil. Suppressive soil can suppress and control the rate of development of soil-borne pathogens. The aims of this study are to determine the physical and chemical properties of soil and environment against fusarium wilt and find out the effects and levels of nutrients from the addition of manure and silica to fusarium banana wilt. Supressive soil samples from Sarampad, Cianjur and conducive soil from Salaman, Magelang. Ambon Kuning banana cultivar derived from tissue culture were planted to find out effects of manure and silica against fusarium wilt. Suppressive soil has higher soil chemist including neutral pH, sandy loam of texture class, total N, total P, organic C, organic matter, available Si than conducive soil. Manure and silica addition have significantly suppressed fusarium wilt on suppressive soil. The final experiment suppressive soil has higher total N, total K, organic C and organic matter than conducive soil.

Abstract

Refugia is a microhabitat for natural enemies (predators and parasitoid) to breed that is planted around cultivated plants. The purpose of this study was to determine the effect of the existence of refugia on the abundance of natural enemies in the fields. The method used is the survey method. Observation of natural enemy populations is carried out visually and directly observed. The trap tool used is pitfall and sweep net. The results of the observation showed that 2 families of insects were Branconidae which acted as parasitoid and Collembola which acted as decomposers.

Abstract

Anthracnose disease on some important crops is caused by the Colletotrichum gloeosporioides. Because of this disease, many farmers were suffer a financial loss. The conventional way to solve this problem that taken by the farmers is synthetic fungicides application which has a negative impact on consumers and the environment. Another alternative to replace it is needed. Antagonistic microorganisms can be used to replace it then and to develop a biofungicide which is more safety for the consumers and the environment. Four isolates of antagonist bacteria to against C. gloeosporioides have been found, namely: UBCR_12, UBCR36, UBCF_01, and UBCF_13. They were being developed as the main material for the manufacture of biofungicide. As an initial step of it, some tests were conducted to identify and to obtain some information that related to the character of all the isolates. Gram staining and KOH test showed that UBCR_12, UBCR_36, and UBCF_01 were gram negative bacteria and UBCF_13 is gram positive bacteria. The four isolates were also showed rod morphological form. Oxidative-fermentative (OF) test showed that the four isolates are facultative anaerobic bacteria. The protease enzyme activity test showed that only 3 of 4 isolates can produced a protease enzyme with the highest clear zone index to the lowest, respectively are UBCR_12, UBCF_13, and UBCF_01. This information leads to choosing UBCR_12 for the molecular identification by the cloning 16S rRNA gene of it. The result showed that UBCR_12 is Serratia plymuthica.

Abstract

Various use of non-organic material is often applied in vegetable cultivation, it causes health problems for those who consume while the demand for the community will increase the need for vegetables. So it takes effort in the application of organic vegetable planting by utilizing the surrounding organic materials that are not used. Using of organic material also can increase time of harvesting, so more faster. Time of harvesting depends on growth of plants, more fertile of growing media, more faster plants can be harvested. The objectives of this study were: (1) to analyze the growth of mustard seedlings (Brassica juncea L.) (2) to get the best compositions for mustard seedlings. This research was conducted in Experimental Garden of Faculty of Agriculture of National Development University of "Veteran" East Java, Surabaya. In March to April 2017. This study was prepared in, Completely Randomized Design (RAL), with 8 treatments and 3 replications. The results showed that the addition of organic material can increase the growth rate of mustard plant seedlings in all treatments up to 4%/etmal and increase the percentage of plant growth of mustard seeds up to 90%. The result of the composition of the growing medium of P4 treatment is soil + compost + litter straw (2: 1: 1) can increase the number of leaves by 9,33 strands, plant length 38,80 cm, leaf area 60,80 cm2, dry weight 14,81 g, as well as the average total value of LTT (Plant Growth Rate) of 40% and LAB (Net Assimilation Rate) of 76% compared to using soil growth medium (P0).
