Abstract

Aspergillus niger is a fungi family Trichocomaceae that form filaments, mycelia, and conidiospore can produce citric acid and does not contain mycotoxins. The media are used as a medium for growth of the fungi Aspergillus niger is SDA (Saboroud Dextrose Agar) which commercially available. This study aims to determine the possible use of based starch breadfruit as alternate media for Aspergillus niger. Inoculation of the fungi Aspergillus niger was conducted using a single dot (one point). The results found macroscopic colonies dark brown to black, round and microscopic hyphae and spores use KOH 10% solution that is suggesting that Aspergillus niger can grow in the growth media made from starch breadfruit.

Abstract

White oyster mushroom (Pleurotus ostreatus) is a type of beneficial mushroom, widely used for consumption, medicinal ingredients, as well as business-oriented cultivation. Nutrition and light intensity are two of the most influential factors for the growth of white oyster mushroom mycelium. Nutrients such as water used for rice cooking (traditionally called air leri) and glucose solution are thought to be potential sources of nutrients, but it is not known which nutrients are more suitable for the growth of white oyster mushroom mycelium. This research was conducted to find out the growth rate difference of white oyster mushroom mycelium produced by vary LED (Light-Emitting Diode) light intensity (sequentially 0 lux, 103 lux, 2x103 lux, 3x103 lux, and 4x103 lux ), air leri nutrition with a 1:1 dilution ratio, glucose solution with 1:3 ratio. Processed data showed that the growth rate of white oyster mushroom mycelium with air leri nutrition for 18 days is better than glucose solution, while low light intensity will affect the color to the growth rate of mycelium on the bag log medium.

Abstract

Gecko is one of the reptiles are used as traditional Chinese medicine which has been reported as antitumor capabilities. The purpose of this study was to determine the cytotoxic effect of rat serum that treated with gecko powder and its ability to inhibit proliferation and stimulate apoptosis of WiDr cells. Gecko powder  the treated in vivo in the rat (Rattus novergicus) SD female. Twelve mice treated with powder gecko at a dose of 9 g/kg body weight per day. Then the rat blood serum on the in vitro treated in WiDr cells. Cytotoxicity test using the MTT method. Observation of the proliferation kinetics using doubling time test. Then apoptosis test performed by the doublestaining method. Cytotoxicity assay results showed that the powder is toxic to the gecko cell WiDr. In the cytotoxicity test showed IC50rat serum with gecko powder body parts obtained at a concentration of 7,914%. The observation of cell proliferation kinetics demonstrated  rat serum with gecko powder body parts most effective in inhibiting the proliferation of WiDr cells Data apoptotic WiDr cells  showed the rat serum with gecko powder effective agency in promoting apoptosis. Rat serum with treat gecko powders bodies part are toxic and can inhibit proliferation and stimulate apoptosis of WiDr cells.

Abstract

The community for maintaining healthy teeth and mouth has used betel and siwak wood plants. However, the benefits of a combination of both plants have not been widely known. The main output of this research was to know the utilization of a combination of betel leaf water and siwak wood as a mouthwash candidate. Combination of boiled water of both plants is a processed product, which has a shelf life. As a mouthwash product that is stored for a long time allegedly decreased the level of usefulness. One parameter of usability level to be observed in this study was the inhibition of Streptococcus mutans, the bacteria that cause dental caries. This study was laboratory experimental research, and use post test only with control group design with Cross-Sectional approach. Data analyzed by T-test and ANOVA followed by LSD test. Inhibition zone of boiled water stored at room temperature was smaller than cold temperature. However, T-test results showed that the diameter of inhibition zone of boiled water stored at room temperature and the cold temperature was not significantly different (p-value = 0,586> 0,05). Significantly decreased drag zone diameters occurred in a combination of boiled water stored for 16 and 18 days. Then the storage variety did not have a real effect, while the storage period significantly affects the inhibition power of boiled water in the growth of Streptococcus mutans.

Abstract

Banana peels have not been utilized optimally, just disposed of as organic waste or used as animal feed. Ripe banana peel skin is rich of chemical compounds which are antioxidants, both flavonoid compounds and other phenolic compounds. Banana peel contains high antioxidant activity compared to meat, especially “Raja Bulu” banana skin  which has antioxidant equal to 97,85%. Natural source of antioxidants in banana peel skin is very beneficial for health. The purpose of this study was to determine the effect of various drying temperature on antioxidant activity and the optimum drying temperature to antioxidant activity. This study also aims to determine the level of panelist's preference for herbal drinks of “Raja Bulu” banana peel with various drying temperatures. The type of research used is True Experimental Research. The study design was using RAL with 4 treatment P0 (control), P1 (drying temperature 75°C), P2 (drying temperature 80°C), P3 (drying temperature 85°C). Data is included the amount of antioxidant activity content and the average of organoleptic properties of banana peel herbal drink. Data analysis techniques used are One Way Anova and qualitative descriptive. The results showed 1) Various drying temperature effect on the antioxidant activity of banana peel herbal drink. 2) The best treatment is P3 (drying temperature 85°C) with antioxidant activity of 84.87%. 3) The result of the panelist's favorite test for the best color, flavor and taste on P3 treatment with 85 ° C drying temperature.

Abstract

Dental caries or commonly known as “Tooth Decay” is one of the most common infection on human. Dental caries begins by the formation of dental plaque and it is a multifactorial disorder. One of the efforts to prevent the formation of dental caries is by utilizing plants which contain antibacterial substance. Pineapple contains bromelain, saponin, flavonoid, and tannin enzymes which functions as antibacteria. This research aimed to discover the antibacterial effectiveness of pineapple juice toward the isolate growth of dental plaque bacteria by using three different pineapple juice concentrations (25%, 50%, and 75%). The method was experimental by using dental bacteria isolate from a patient of Dental and Oral Clinic in Puskesmas Kecamatan Tanah Abang. The result shows that the higher concentration shows the bigger inhibition zone diameter formed around the plate’s paper. Consequently in concentrations of 25%, 50%, and 75%, each has its own inhibition zone rate as much as 2,90 mm (weak); 3,89 mm (weak); and 5,89 mm (moderate).

Abstract

The aims of the research was to know the vitamin C content in unripe and ripe rosella calyx in three different altitudes. Rosella calyx which were used in this research were taken from three different altitudes. There were Glagah beach (7 masl), KP4 Berbah (126 masl), and Samigaluh (600 masl). The vitamin C content were determined by titration method using 2,6-dichlorophenolindophenol. The result of this research showed that vitamin C content in unripe rosella calyx was not significantly different. The highest vitamin C content in ripe rosella calyx at 7m and 600m altitude were not significantly different. And the vitamin C content in unripe and ripe rosella calyx at 7 masl and 600 masl were also not significantly different. Vitamin C content in unripe rosella calyx at 126 masl was 2,269 mg/g, higher than the ripe (1,316 mg/g). It could be concluded that the altitude and the age of calyx influence vitamin C content in rosella calyx.

Abstract

Mangrove crab catching activity to increase and tends to lead exploitation pressure. This study aims to determine the rate of growth, mortality, and rate of exploitation. Sampling on fishermen and collectors whose fishing locations are in predetermined areas. Measurements taken are using ruler and caliper 0,05 for carapace width. Sample measurements were carried out to obtain information on the dynamics of the mangrove crab population. The results obtained are: length of carapace minimum (Lmin) 61 mm, maximum (Lmax) 173 mm, Asimtot carapace width (L∞) 177,7 mm with growth coefficient (K) 0,1 per year, and the value t0 obtained -0,2907. Total mortality rate (Z) 0,743 per year, natural mortality (M) 0,177 per year and mortality of capture (F) 0,566 per year. The rate of exploitation (E) 0,762 per year, indicating that there has been over exploitation of mangrove crabs in Sinjai District.

Abstract

Probiotics are living microorganisms that have beneficial effect on the host by improving the balance of bacteria in the intestines. This research was aimed to determine the diversity of bacteria that live in digestion of the shrimp and molecular characteristics along with to observe phylogenetic relationships among the bacteria identified based on sequence16S rDNA. The bacteria were isolated from intestine of the Tiger prawn (Penaeus monodon) obtained from fishpond at BBPBAP Jepara. The analyzed result of 16S rDNA shown that three bacterial species were potential as probiotic. These bacteria grow well at pH 2 and this indicates one of probiotic bacteria characteristic. Among the three isolates, two isolates i.e. SP2 and SU isolates were phylogenetically closely related to Bacillus bataviensis strain CCGE2059 (EU867382.1) with 97% homology. While the SWU Isolate is most likely a new species of Caulobacter sp. (AJ227775.1). The SWU isolates were phylogenetically one ancestor with the Chromobacterium violaceum.

Abstract

This research aims to determine the types of Selaginella in the field and to know the existence of species within a community. This study was conducted by collecting specimens from the field followed by inventory and identification of Selaginella species, as well as recording environmental conditions.The results of this study obtained 5 types of Selaginella: Selaginella intermedia, Selaginella kityyae, Selaginella longariastata, Selaginella ornata and Selaginella willdenovii. Based on their habitat Selaginella intermedia, Selaginella kityyae, Selaginella longariastata and Selaginella willdenovii found in field, have a terrestrial habitus spread, whereas Selaginella ornata is an upright epilithic terrestrial, with each species found mostly in clusters and dominate in an area.
