Abstract

Endophytic fungi are one of the microorganisms that live in plants and symbiotic with their host plant produce secondary metabolites. This study aims to determine the optimum growth profile of endophytic fungus isolated from the leaf of Anredera cordifolia (Ten.) Steenis namely isolate I5 as antibacterial. The fungus growth profile optimization analyzed by a correlation between fermentation time and mycelium weight, which collected every 24 hours for 15 days. The media were varied at pH conditions of 4, 7, and 8 in each PDYB medium (glucose and fructose substrates). The results showed that isolate I5 grew optimally on the 13th day on the PDYB medium pH 8. Ethyl acetate extract 5% of isolate I5 could inhibit bacteria growth with the largest inhibition zone diameter against Staphylococcus aureus of 9.96 mm (± 0.60) and Escherichia coli of 9.03 mm (± 0.46).

Abstract

Chromate reductase is one of the potential enzymes for hexavalent chrom detoxification. Most of the enzyme is produced by bacteria, especially Bacillus. The aim of this research was to study chromate reductase activity isolated from Bacillus sp LKA9. Bacillus sp LKA9 was isolated from leather tannery liquid waste and used as a model in the experiment. Bacillus sp LKA9 was isolated through enrichment culture using Salt Base Solution containing 3 mM K2CrO4. Chromate reductase was isolated from bacteria by growing on a liquid medium containing chrom hexavalen (Cr VI) through several steps. The first step of the isolation process was to use the precipitated process using ammonium sulphate (30-80%). The next step, crude enzymes from the first step was partially purified through DEAE-Cellulose of Ion Exchange Chromatography Column. Diphenylcarbazide methods was used to examine the activity of enzyme fractions. The result of the experiment revealed that all protein could be precipitated by ammonium sulphate, and the cytoplasmic fraction at saturation of 50-70% showed high enzyme activity. Purified enzymes showed an increase activity 69,385 times to that of crude enzymes. The enzyme optimal had temperature and pH were 350C and 5; respectively. KM of enzyme was 0,0075 mM, and Vmax was 2500 mol/minute/mg protein. Enzyme activity was not inhibited by Cu2+, but an ion Ag2+ and Hg2+ inhibited the enzyme activity un-competitive. The activity of enzyme was demonstrated on SDS-PAGE by appearing typically band with molecular weight 29,26 kDa, it was assumed as chromate reductase.

Abstract

Penelitian ini bertujuan untuk mengetahui peningkatan aktivitas, hasil belajar afektif dan kognitif biologi peserta didik melalui penerapan model pembelajaran Brain Based Learning (BBL) pada materi ?Keseimbangan Ekosistem dan Pengelolaan Lingkungan? di kelas VIIA SMP Negeri 4 Sungguminasa Kabupaten Gowa. Penelitian ini merupakan penelitian tindakan kelas (Classroom Action Research). Penelitian dilaksanakan pada Semester Genap Tahun Pelajaran 2013/2014, pada bulan Desember 2014 yang bertempat di SMP Negeri 4 Sungguminasa Jl. Lapangan Syekh Yusuf No 03 Kabupaten Gowa. Instrumen yang digunakan dalam penelitian ini adalah lembar observasi aktivitas peserta didik, tes hasil belajar kognitif dan lembar hasil kegiatan afektif. Teknik pengumpulan data meliputi lembar observasi aktivitas peserta didik dan tes hasil belajar kognitif yang dianalisis secara kuantitatif dengan persentase. Hasil penelitian menunjukkan bahwa penerapan model pembelajaran Brain Based Learning dapat meningkatkan aktivitas belajar peserta didik dengan nilai rata-rata siklus I sebesar 48,74% dan pada siklus II meningkat menjadi 75,89%, meningkatkan hasil belajar kognitif peserta didik dengan presentase ketuntasan pada siklus I sebesar 52,78%, pada siklus II meningkat menjadi 94,44%. Demikian juga dengan hasil belajar afektif yang setiap indikatornya mengalami peningkatan dari siklus I ke siklus II.Kesimpulan dari hasil penelitian ini adalah penerapan model pembelajaran Brain Based Learning meningkatkan aktivitas, dan hasil belajar pada materi ?Keseimbangan Ekosistem dan Pengelolaan Lingkungan, peserta didik kelas VIIA SMP Negeri 4 Sungguminasa Kabupaten Gowa.Kata kunci: Model Brain Based Learning, Aktivitas Belajar, Hasil Belajar Afektif dan Kognitif.

Abstract

Murbei (Morus albaL.) merupakan salah satu jenis tanaman yang telah digunakan oleh masyarakat sebagai sumber bahan obat seperti antioksidan, antidiabetes, antihipertensi, hipolipidemik, demam, flu, batuk, antiemetik dan gangguan saluran pencernaan. Penelitian ini bertujuan untuk menentukan potensi aktivitas antibakteri daun murbei dari daerah Soppeng, Gowa dan Enrekang terhadap bakteri Escherichia coli. Daun murbei diekstraksi menggunakan larutan penyari etanol 70% dan etil asetat. Metode pengujian aktivitas antibakteri yang dilakukan adalah metode difusi agar dan analisis KLT bioautografi. Hasil penelitian menunjukkan bahwa ekstrak daun murbei asal kab.Soppeng, Gowa dan Enrekang memiliki potensi aktivitas antibakteri terhadap bakteri Escherichia coli dan yang memiliki potensi baik sebagai aktivitas antibakteri ialah ekstrak etanol dari daerah soppeng pada konsentrasi 10% dengan diameter zona hambatan yaitu 9,89 mm. Hasil pengujian analisis KLT bioautografi ekstrak etil asetat menunjukkan adanya aktivitas antibakteri dengan spot noda dari daerah Soppeng 5 noda, Gowa 6 noda dan Enrekang 6 noda sedangkan ekstrak etanol tidak menunjukkan adanya aktivitas antibakteri terhadap bakteri Escherichia coli. Kata kunci: Daun Murbei, Aktivitas Antibakteri, Difusi Agar, dan KLT Bioautografi.

Abstract

DOWNLOAD PDFIn vitro examine of fractionated ethanol extract of bark Mangrove (Avicennia spp). showed high antibaterial activity. Repression test of extract formula showed that fish has a higher viability. Recent study has conducted to find the main compound as candidate prior to standardized product of phytopharmaca for future commercial purposes. The chromatography methods was  applied and  identification of  major compounds by analysis based on chemical test as well as spectral data of spectroscopic measurements. Study showed that main secondary metabolites in the form of alkaloid class of polar compounds. The white crystalline powder form characteristics has a melting point 172oC with molecular weight 232, and molecular formula is C12H12N2O3.   Key words: Main secondary metabolites, mangrove, Avicennia spp.

Abstract

Abstract.  This research is a classroom action research. The purpose of this study improves the activity and student learning outcomes through application of learning strategy Everyone is a Teacher Here. Research subjects are students of class X.I SMA Negeri 3 Sengkang. This research carried out during two cycles. Data collection implemented in the second semester of academic year 2016/2017. This research measure learning activity and learning outcomes. Learning activity measure with observation sheet and learning outcomes measure with written test consist of multiple-choice question as much as 20 items. Student learning activities collected during the learning process observed by 3 observers using observation sheet, then learning outcomes data obtained through evaluation when teaching’s time first and second meeting was done and on third meeting was evaluation.  This matter occurs on each cycle. All the data obtained were analyzed through three ways: qualitative analysis for student learning activities, quantitative for student learning outcomes. Results of research on student learning activity that has been analyzed shows the average in the first cycle of 55.6%, while in the second cycle of 67.2%, so the difference in the increase of 11.6%. Student learning outcomes also increased where the average value on observation data of 69.6, 64.4 the first cycle, and the second cycle 74.8. It can be concluded that the application of learning strategy Everyone is a Teacher Here can increase the activity and student learning outcomes. Abstract

Abstract. The ability to argue is one of the 21st century skills that students need to have. The purpose of this study was to determine the scientific argumentation skills of students class XII at the Biringkanaya District High School in answering description questions, especially in the digestive system material. This type of research is a descriptive survey research using purposive sampling technique. The population of this study were students of class XII MIA at State Senior High Schools in Biringkanaya District, Makassar City, South Sulawesi. The sample used in this study is a study group in which students have heterogeneous cognitive abilities. The argumentation ability data is obtained through giving tests. Based on the test results, the overall argumentation skills of students were in the very poor category with a score of 19.74%. The results of the students' arguments showed that in the aspect of making claims with a percentage of 91.4% (very good); aspects include ground with a percentage of 21.64% (less); aspects include backing with a percentage of  9.88% (very low); aspects include a warranty with a percentage of 5.71% (very low); aspects include qualifier with a percentage of 5% (very low);  and aspects include rebuttals or reservations with a percentage of 7.3% (very low).Keywords: ability to argument, making claims, rebuttal, reservation, high schools MakassarÂ
