Abstract

Artificial insemination (AI) in cattle using Ongole Cross Breed liquid semen with cauda epidydymal plasma-2 (CEP-2) diluent+0.6% Bovine Serum Albumin (BSA) aimed to improve the genetic quality of livestock with high sperm motility on beef cows. Addition of BSA as extracellular cryoprotectant to support egg yolk function to prevent membrane damage caused by cold shock during cold storage. This research was conducted in Tumpang, Pakis, and Singosari Sub-district, Malang Regency using 38 cows that inseminated using frozen semen of Limousine and Simmental breeds as control (19 cows) and liquid semen with CEP-2 + 0.6% BSA as a treatment (19 cows). The reproductive parameters were Non Return Rate (NRR), Service per Conception (S/C) and Conception Rate (CR). Data were analyzed descriptively and continued by chi-square test. The results showed that insemination using frozen and liquid semen did not affect the success rate of AI in beef cows. The NRR43-63 value of frozen was 89.40% and liquid semen was 78.94%; the CR value of frozen semen was 84.21% and liquid semen was 57.89%; and the S/C value of frozen semen was 1.21 and liquid semen was 1.42. The conclusions of the study were the values of NRR43-63, CR and S/C of frozen higher than liquidsemen.

Abstract

Penelitian mengenai produktivitas induk kambing Peranakan Etawah (PE) telah dilakukan. Tujuan penelitian ini untuk mengetahui dan mengkaji produktivitas induk kambing PE di Taman Ternak Kaligesing. Metode penelitian menggunakan metode survey. Pengumpulan data primer dan data sekunder dilakukan melalui laporan tahunan, studi literatur serta wawancara kepada koordinator dan pegawai Taman Ternak Kaligesing. Pemilihan sampel ternak dilakukan dengan metode purposive sampling yaitu memilih indukan yang sudah beranak minimal dua kali. Parameter yang diamati diantaranya litter size, bobot sapih, umur pertama dikawinkan, service per conception (S/C), selang beranak dan masa kosong. Hasil penelitian menunjukkan bahwa litter size 1,17 ekor, bobot sapih 10,4 kg, umur pertama dikawinkan 13,5 bulan, S/C 1 kali, selang beranak 9,7 bulan dan masa kosong 4 bulan. Dengan demikian secara umum produktivitas induk kambing PE di Taman Ternak Kaligesing sudah baik berdasarkakan parameter  bobot sapih, S/C, umur pertama dikawinkan dan selang beranak, namun pada litter size dan masa kosong masih perlu perbaikan.

Abstract

The objective of the experiment was to examine the effect of sperm washing using SpermRinse with different centrifugation duration (15, 20, and 25 minutes) on the quality of Bligon spermatozoa. Sperm was collected usingartificial vagina from six bucks of 2 (two) year olds. Data were collected for motility, viability, and abnormality of the spermatozoa. Characteristics of the fresh sperm were analyzed based on mean and deviation standard, and followed by Duncan’s New Multiple Range Test (DMRT). The results showed that washing Bligon buck sperm using SpermRinse with different centrifugation times had very significant effect (P≤0.01) on the motility, viability, and abnormality ofspermatozoa. It is concluded that washing Bligon buck sperm using SpermRinse with centrifugation at 1.500 rpm (r = 5.5 cm) could increased sperm quality, of which 20 minutes was the best centrifugation time.(Key word: Bligon Buck, Sperm, SpermRinse, Sperm quality)

Abstract

Reproduction is a very important factor in maintaining livestock productivity. For better reproductivity, estrus induction is one of very important ways to be performed. Estrus induction is usually performed with hormonal method,such as application of PGF2α, but the disadvantage is that it is relatively expensive. For this reason, it is necessary to promote technological breakthrough to lower cost of estrus induction. The aim of this study was to know whether laserstimulation on the points of reproductive acupuncture could be performed for estrus induction on Bligon goats, as it is usually performed with hormonal method. Fourteen female Bligon goats aged 2 years old were randomly divided into two groups, each group consisted of 7 goats. Group I was stimulated with laser and group II was injected with 1.25 mg per head of PGF2α. Laser stimulation was conducted for 10 seconds on each point of acupuncture on day 1, 2, and 3, and was repeated on day 11, 12, and 13. PGF2α injection was performed on day 1 and was repeated on day 11. The observed variables were the percentage of estrus, onset of estrus, estrus duration and progesterone level when estrus occurred after the second estrus induction. The results of the study showed that laser stimulation for estrus induction did not indicate significant difference compared to PGF2α injection, in terms of the percentage of goats undergoingestrus, onset of estrus and the estrus duration. It was concluded that laser stimulation could be used for estrus induction on Bligon goats.(Key words: Laser, Estrus induction, Bligon goats)

Abstract

Balai Pembibitan Ternak Unggul is a breeding center established by the Government of Indonesia (GOI) to perform preservation, breeding, breeding stock production and development, and also the distribution of breeding stock of Excellent Bali cattle at national level. In the production of excellent breeding stock through the performance test and progeny test. Pre-weaning growth is one of the important indicators for the population growth. The study was aimed to identify the pre-weaning calf growth rate of Bali cattle in 2010-2011, born from performance-test-passing-dams that were collectively reared with progeny-test-passing sires. Materials used were 84 pre-weaning Bali calves. This was a survey method-based descriptive analysis research. The results showed that in terms of the performance of pre-weaning growth, the birth weights of male and female calves were 18.37±1.65 kg and 18.27±1.29 kg, respectively. The weaning weights of male and female calves were 93.53±21.00 kg and 87.66±12.04 kg, respectively, with weaning time of 205 days. The average of daily weight gain (ADG) were 0.37±0.10 kg/head/day (male) and 0,34±0,06 kg/head/day (female). It was concluded that the pre-weaning calf growth rate of Bali cattle at the BPTU Bali in 2010-2011 were moderately high since they were born from the dams and the sires passing the performance test and progeny test.(Keywords: Pre-weaning growth, Bali cattle)

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh dosis sperma untuk Inseminasi Buatan (IB) pada ayam kampung terhadap fertilitas telurnya. Dosis IB ini berguna untuk mengetahui perbandingan jantan dan betina yang paling efisien dalam suatu pemeliharaan. Sperma ditampung dan dicampur dari 5 ekor ayam Pelung jantan yang berumur sekitar 52 minggu. Sperma yang telah diketahui konsentrasinya kemudian dibagi menjadi 3 bagian dan masing-masing diencerkan dengan NaCl fisiologis sehingga diperoleh dosis akhir yaitu 25x106/0,1 ml (dosis 1); 75x106/0,1 ml (dosis 2); dan 150x106/0,1 ml (dosis 3). Sembilan ekor ayam kampung betina diinseminasi untuk mengetahui pengaruh dosis terhadap fertilitas telur. Ayam betina yang digunakan adalah ayam kampung yang berumur sekitar 30 minggu. Hasil penelitian menunjukkan bahwa perlakuan dosis sperma tidak berpengaruh pada fertilitas telur ayam kampung. Fertilitas telur untuk masing-masing perlakuan dosis 1, 2, dan 3 adalah 44,13±20,84; 66,67±23,57; dan 58,75±14,36%. Kesimpulan dari penelitian ini adalah dosis sperma tidak berpengaruh pada fertilitas telur ayam kampung.(Kata kunci: Ayam kampung, Dosis sperma, Inseminasi buatan)

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh aras kuning telur itik Alabio (Anas platyrhynchos) yang diencerkan dengan tris fruktosa terhadap persentase motilitas, viabilitas, dan abnormalitas spermatozoa kambing Bligon setelah pengenceran, ekuilibrasi, prefreezing, dan post-thawing. Kualitas sperma diamati setelah pengenceran, setelah ekuilibrasi, preefreezing, dan post-thawing. Sperma seekor kambing Bligon ditampung dengan metode vagina buatan dua kali seminggu. Sperma diencerkan dengan pengencer tris fruktosa dengan aras kuning telur itik 10%, 20%, dan 30%. Data kualitas spermasegar dianalisis dengan mean dan standar deviasi, sedangkan motilitas, viabilitas, dan abnormalitas spermatozoa setelah pengenceran, ekuilibrasi, prefeezing, dan post-thawing dianalisis variansi pola searah. Hasil analisis menunjukkan bahwa penggunaan kuning telur itik dengan aras 10% memberikanmotilitas dan viabilitas sperma setelah pengenceran, setelah ekuilibrasi, setelah prefreezing , serta setelah post-thawing yang lebih baik jika dibandingkan dengan penggunaan kuning telur itik dengan aras 20% maupun 30% (P<0,05). Selain itu, abnormalitas sperma yang mendapatkan kuning telur itik dengan aras 10% juga lebih rendah (P<0,05) jika dibandingkan dengan penggunaan aras 20% maupun 30%. Dapat disimpulkan bahwa penggunaan kuning telur itik memberikan hasil yang terbaik terhadap kualitas spermatozoa kambing Bligon, jika dilihat dari variable: motilitas, viabilitas, abnormalitas spermatozoasetelah pengenceran (masing-masing: 74,00; 97,60 dan 0,60%), ekuilibrasi (masing-masing: 72,20; 94,00 dan 1,90%), prefreezing (masing-masing: 56,80; 77,70 dan 5,40%) dan post-thawing (masing-masing: 43,60; 62,60 dan 8,50%).(Kata kunci: Kambing Bligon, Kuning telur itik, Pengenceran, Sperma, Tris fruktosa)

Abstract

ABSTRACT The aim of this research was to determine the proportion, viability and motility of X and Y ram spermatozoa separated with egg white albumin. Sperm samples derived from Garut ram, which was collected by using an artificial vagina. Observations were made on spermatozoa fraction above and below each medium fraction treatment. There are treatment egg white albumin as separation medium, each medium consisting of fractions top and bottom fraction with different concentration: 1) P0 = sperma before separation (control); 2) P1 = 10% above fraction + 30% lower fraction; P2 = 25% + 45%; P3 = 25% + 75%. Data proportion of X and Y, viability and motility were analyzed statistically by Completely Randomized Design patern in the direction followed by Duncan’s Multiple Range Test for data with a real difference. Separation with egg white albumin affect significantly increased the proportion of spermatozoa X and Y (P≤0.05), but tends to decrease the viability and motility of spermatozoa.The proportion of spermatozoa X and Y was highest in treatment P3,76.76% of spermatozoa X (fraction above 25%) and 79.81% spermatozoa Y (75% lower fraction), with an average viability obtained respectively 68,9% (fraction above) and 59,7% (bottom fraction), motility 77,5% (fraction above) dan 84,0% (bottom fraction). It was concluded that the egg white albumin is very effective in changing the proportions of X and Y ram sperm with the quality of spermatozoa after separation feasible for applications insemination or processed into frozen semen.  (Keywords: Garut ram, White egg albumin, Spermatozoa X and Y)

Abstract

The study was carried out to investigate the effect of follicle stimulating hormon (FSH) into in vitro maturation medium to increase oocytes maturation and 4 cell stadium embryo development of Bligon goat. Goat ovaries were obtained from a slaughterhouse and transported to the laboratory in a flask of NaCl at temperature of 31 – 34°C. Oocytes were aspirated from 2 – 6 mm of follicles into a 3 mL syringe (23G needle) that contained Dulbecco’s Phosphate-Buffered Saline. Oocytes were divided into three groups, i.e tissue culture medium (TCM) with FSH supplementation 0, 50, and 100 IU/mL. Oocytes were put into those medium and incubated on 39°C, 5% CO2, and 95% humidity for 24 hours. Matured oocytes were fertilized with capacitated frozen thawed-semen and incubated on 39°C, 5% CO2, and 95% humidity for 5 hours. Fertilized oocytes were washed for 3 times in TCM and incubated in the same condition for embryo culture. The data of FSH supplementation and embryo development were analyzed using randomized completely one way classification. The results showed that the percentages of mature oocytes from FSH supplementation 0, 50, and 100 IU/mL were 70,48±23,22, 78,48±15,80, and 80,29±12,86%, respectively. Cleavage rate of the two cells stage were 36,00±14,22, 44,00±33,94, and 57,45±31,78%, respectively, and for the 4 cells stage were 27,33±22,04, 35,33±40,73, and 39,45±20,38%. It is concluded that supplementation of FSH in the maturation medium could not increase the percentages of in vitro maturation and embryo development.
