Abstract

An in-vivo anti-inflammatory study of pyrazoline compound synthesized from chalcone has been carried out using Paw Edema method.Compound (R)-3-(4-florofenil)-1-fenil-5-(tiofen-2-il)-4, 5-dihidro-1H-pyrazole was synthesized by milling method in alkaline condition,with no solvent addition and was conducted under room temperature. Characterization of the synthesized compound was determined using1H-NMR and HR-MS spectrometer. As for anti-inflammatory study, suspension of compound (R)-3-(4-florofenil)-1-fenil-5-(tiofen-2-il)-4,5-dihidro-1H-pyrazol was administrated orally to white female mice (Rattus novergicus) with doses 20, 40, 80 mg/kg bw, respectively. Theresult showed that the compound with doses 40 and 80 mg/kg bw exhibited insignificant difference compared with standard drugs Celecoxiband Piroksikam (p>0,05). Futhermore, all doses of compound did not cause any damage of stomach organ of mice, which is given the same“ulcer index” (Iu) with negative control (Iu 2,00). Meanwhile, ulcer index for Celecoxib of Iu 35,99 and Piroksikam of Iu 70,01.

Abstract

Hepatoprotective activity and acute toxicity from extract of gambir (Uncaria gambir. Roxb) was investigated. The animals were groupedinto seven groups which consist of 5 female mice for each group. Mice from group I were only administrated orally with palm oil, 1% frombody weight for two days and 1% of gum suspension orally at day two. Group II were given CCl4 with dose 1.25 ml/kgbw in 10% of palm oilfor two days and gum suspension orally 1 hour after CCl4 given at day two. Group III, IV, V were given CCl4 with dose 1.25 ml/kgbw in 10%of palm oil for two days and followed by administration of gambir suspension orally 1 hour after CCl 4 given at day two with concentrations30; 100 and 300 mg/kgbw, respectively. At day 3, all the blood were taken and were examined its SGOT and SGPT. While the isolated liverwere weighed and the ratio between liver weight and body weight of mice were also observed. Acute toxicity test was conducted using fiveanimal groups consist of 5 female mice and 5 male mice. Each of mice then was given gambir suspension orally with doses of 1; 2; 4; 8 and15 g/kgbw, respectively. LD50 was defined as dose which can cause 50% death of total mice from each group. The result can be concluded thatgambir extract was found to be active as hepatoprotective agent with dose 30; 100 dan 300 mg/kgbw. Futhermore, extract of gambir couldreduced the ratio of liver weight and based on the LD 50 value obtained (24 hours, >15 g/kgbw), gambir was classify as non toxic.

Abstract

Banyan plant has been used by communities as a traditional medicine in order to heal heat cramps in children but it has not been scientifically tested. A study has been conducted to observe an antipyretic effect of the ethanolic extract, the fraction of n-hexane and ethyl acetate fraction of the leaves of banyan (Ficus benjamina. L) in male white rats (Rattus norvegicus) with fever induction method using a solution of peptone 2% as much as 0.5 mL / 200 g body weigt intra peritonially, with the dose of ethanol extract, the fraction of n-hexane and ethyl acetate fractions 100, 200, and 400 mg / kg orally. Respectively as a positive control, asetosal was administered at a dose 117 mg / kg and the negative control was 1% CMC Na. Temperature observations of mice performed at 30, 60, 90, 120, 150, and 180 minutes, then percent protection was calculated. Data analysis was done using Two-way Analysis of Variance (ANOVA) and Tukey test. Results showed that the fraction of n-hexane at a dose of 400 mg/kg had an antipyretic effect as significantly (p<0.05).

Abstract

Sekunyit (Fibraurea tinctoria Lour) is a species plant that has been used as traditional medicine for treating several diseases including jaundice, dysentery, headache, eye infection as well as anti diabetic drug. This research aims to evaluate antidiabetic activity of ethanol extract of roots and stem of sekunyit on white male mice (Mus musculus L) using glibenclamide as positive control. The experimental animals were induced with alloxan 175 mg/Kg BW i.p and devided into six groups, each group consisted of five mice. Group one served as negative control which was given NaCMC 1%, group second was positive control, given  glibenclamide 0,65 mg/KgBW, while group third, fourth, fifth were administered orally with etanol extract with doses of 100, 200 and 300 mg/KgBW, respectively. The observed parameter were measurement of blood glucose levels, urine volume and volume of drinking water as well as ratio of relative organ weight. The treatment was conducted for 15 days. The result showed that administration of ethanol extract of roots and stem of sekunyit can lowered significantly blood glucose levels (p<0,05) of alloxan induced-diabetic mice, decreased urine volume (p<0,05), decreased volume of drinking water (p<0,05), and did not affect relative organ weight of heart, liver and kidney (p>0,05).

Abstract

For the development of natural medicines from the ethanolic extract of Syzygium myrtifolium Walp leaves it is necessary to know the pre-clinical safety test so that its safety if used in the long term. The subchronic toxicity test of the ethanolic extract of Syzygium myrtifolium Walp leaves l was carried out by assessing the kidney and liver function of the test animals given after 60 days of test preparation, in the form of blood biochemical examination using the Mindray®BA-88A device and histological examination of the kidneys and liver. The results showed that the administration of ethanol extract of Syzygium myrtifolium Walp leaves at doses of 300, 600 and 900 mg/kgBW did not significantly affect the activity of SGOT, SGPT, serum creatinine levels (p>0.05). Microscopically on the percentage of damage to the glomerulus of the right and left kidneys. at doses of 600 and 900 mg/kgBW there was a significant difference to the control group (p<0.05). For qualitative microscopic examination, namely observation of sinusoids and hepatocyte cells at each level, there were abnormal changes. Meanwhile, quantitative microscopic examination at doses of 600 and 900 mg/kgBW caused an increase in the percentage of central vein damage (p<0.05). From the research, it was found that the ethanolic extract of Syzygium myrtifolium Walp leaves of white mice was safe for liver and kidney function at a dose of 300 mg/kgBW, but the doses group of 600 and 900 mg/kgBW gave a toxic effect on the kidneys and liver when compared to the control group.

Abstract

Periodontal prevalence in all age groups in Indonesia is 96.58%. The main cause ofperiodontal is microorganisms in dental plaque. Control of plaque reduction can be done using natural ingredients which contain antibacterial, one of which is areca nut seed (Areca catechu L.). This research aims to determine the type of bacteria on the gums, and to determine the antibacterial activity of ethanol extract of areca nut (Areca catechu L.) on gums' bacteria isolate. Antibacterial activity test used Agar diffusion method with concentration series 10%, 20%, 30%. The results of phytochemical screening of areca nut seed ethanol extract showed the presence of alkaloids, terpenoids and flavonoids. The results from the identification test based on macroscopic, microscopic, and biochemical observations obtained two Gram-positive bacteria (Staphylococcus epidermidis and Streptococcus sp) and one Gram-negative bacteria (Enterobacter aerogenes). Antibacterial activity test of ethanol extract of areca nut seed (Areca catechu L.) on gums' bacterial isolates was analyzed statistically with One Way Anovaand alternative test withKruskal-Wallis . Antibacterial activity of ethanol extract of areca nut seed (Areca catechu L.) on Staphylococcus epidermidis and Streptococcus spbacteria at concentrations of 10%, 20%, and 30% showed significant difference diameter of inhibitory power compared to positive and negative controls on Enterobacter aerogenes bacteria at concentration of 10 %, 20%, and 30% showed that the diameter of inhibitory power was significantly different compared to positive controls and not significantly different compared to negative controls.

Abstract

Salah satu tumbuhan yang telah digunakan oleh masyarakat sebagai obat tradisional yaitu daun beringin guna mengatasi rasa nyeri dan dari penelusuran pustaka belum ada dilakukan pengujian secara ilmiah begitu juga dengan penelitian tentang toksisitasnya. Berdasarkan hal tersebut, telah dilakukan penelitian untuk mengetahui efek analgetik ekstrak etanol daun beringin yang diberikan secara oral pada hewan coba mencit putih jantan yang diberi asam asetat 1% v/v sebanyak 0,1 ml sebagai penginduksi nyeri. Penelitian dilakukan menurut metode writhing test, menggunakan pembanding asetosal dengan dosis 65 mg/kgBB. Bahan uji diberikan secara oral dalam bentuk suspensi dalam Na CMC 1%, diberikan tiga puluh menit sebelum pemberian asam asetat. Pengamatan jumlah geliat dilakukan setelah penyuntikan asam asetat. Jumlah geliat dihitung dengan selang waktu 10 menit selama 90 menit kemudian dihitung persen proteksinya. Penelitian toksisitas akut (LD50) dihitung sebagai dosis yang memberikan kematian hewan sebanyak 50% dari jumlah hewan percobaan selama 24 jam setelah penyuntikan dan toksisitas tertunda dilakukan pada dosis 1000, 2000, 4000, 8000, dan 16000 mg/kgBB terhadap parameter volume air minum, volume urin, perubahan berat badan, konsumsi makanan, berat feses, rasio organ ginjal dan jantung. Hasil penelitian menunjukkan bahwa ekstrak etanol pada dosis 100, 200, dan 400 mg/kgBB memberikan efek analgetik p < 0,05 terhadap kontrol negatif (analisis varian dua arah dan analisis Tukey). Berdasarkan hasil pengujian toksisitas akut, ekstrak etanol daun beringin digolongkan praktis tidak toksik. Hasil penelitian yang menunjukkan adanya gejala toksisitas akut, ekstrak etanol daun beringin digolongkan praktis tidak toksik. Hasil penelitian yang menunjukkan adanya gejala toksisitas tertunda terhadap parameter perubahan berat badan, konsumsi makanan, berat feses, organ ginjal dan jantung signifikan pada dosis 16000 mg/kgBB.   Kata kunci : Analgetik, Ficus benjamina, writhing test.

Abstract

Artocarpus altilis (Parkinson ex F.A.Zom) Fosberg is also widely used in traditional medicine. People living in  Indonesia, use the fruit flesh as a tonic for the liver and use decoction of the leaves to treat to treat liver cirrhosis, hypertension, and diabetes. Research has been done on antioxidant activity  using DPPH method on ethanol extract of breadfruit bark (Artocarpus altilis (Parkinson ex F.A.Zom) Fosberg using 2-Diphenyl-1-picrylhydrazyl as a free radical. Antioxidant activity test with comparison of vitamin C. The parameter measured is the absorbance using a 96 well Microplate Reader (Epoch®), then the IC50 value is calculated. This study aimed to determine the antioxidant activity of the ethanolic extract of the breadfruit (Artocarpus altilis (Parkinson ex F.A.Zom) stem bark extract. The antioxidant test results of the ethanol extract of breadfruit stem bark (Artocarpus altilis (Parkinson ex F.A.Zom) Fosberg had an IC50 value of 133.98 µg/mL with the medium category.
