Abstract

Cercospora brunfelsiicola on Brunfelsia uniflora is proposed as a new species based on a combination of molecular phylogenetic and morphological data analyses. The molecular phylogenetic analysis based on combined multilocus analyses of the Internal Transcribed Spacer (ITS), part of the elongation factor 1–a gene (EF1–a), and part of the calmodulin (CAL) gene regions showed that C. brunfelsiicola is phylogenetically distinguishable from other Cercospora species, including members of the C. apii s. lat. complex. Morphologically, C. brunfelsiicola differs from other closely related Cercospora species, in particular C. acaciae-mangii, by forming lesions with indistinct margin, larger stromata [(32) 48.5 ± 10.6 (68) ?m diam.], and filiform to narrowly obclavate conidia [(45) 59 ± 9.1 (72) × (2.5) 2.5 ± 0.2 (3) ?m].

Abstract

Endophytic fungi have been known as potential source of bioactive compound, similar to their host.  This study was conducted to identify endophytic Diaporthe spp. isolated from Cinchona calisaya and to explore their potential in producing alkaloids, particularly quinine. A total of 39 strains of Diaporthe spp. were identified using ITS1-5.8S-ITS2 and EF1-α gene. Seventeen strains have species name and they represent eight species of Diaporthe i.e. D. cynaroidis, D. endophytica, D. ganjae, D. gardeniae, D. litchicola, D. phaseolorum, D. pseudomangiferae, and D. rhoina. The re­maining 22 strains represents 15 groups of unidentified Diaporthe sp. High performance liquid chromatography analyses of chloroform extract of 21-days old static cultures of all fungi in potato dextrose broth showed that these strains produce totally 82 different alkaloids. Each strain producedabout 2-38 different alkaloids. Sixteen out of 39 strains (41%), i.e. one strain each of D. endophytica, D. gar­deniae,  D. phaseolorum, and  D. pseudo­mangiferae, 2 strains of D. litchicola, and 10 strains of unidentified Diaporthe were able to produce quinine. The quinine concentration ranges from 1.1 mg/l to 155.2 mg/l. By using Jaccard’s similarity index 0.44 as the cutting score, UPGMA analyses of alkaloid profile of these strains shows that these strains forms 23 clusters. Alkaloid profile clustering does not support the phylogenetic grouping. This indicates that the alkaloid profile is strain dependent. Some potential strains should be further investigated for optimization of their quinine production.

Abstract

Fifty strains of Bacillus sp. isolated from Gunung Halimun National Park (GHNP) were characterized. All strains are characterized by amylase, protease, phosphatase, ligninase, and cellulase activity.Thirtyseven isolates gave positive reaction for amylase activity, 46 isolates for protease activity, 3 isolates for phosphatase activity, 3 isolates for ligninase activity, and 36 isolates for cellulose activity.These findings indicates that Bacillus sp. that isolated from GHNP is a potential microbe for enzymes technology and biotechnology application.

Abstract

Background: Counseling service is a process of two-way communication between counselor (midwife) and acceptors for helping their problems related to family planning programmed. Such service particularly is to help the client (family planning participant) in choosing contraceptive method which suitable for them. Counseling service may improve the client satisfaction in participating family planning programmed and increase the utilization of family planning services.Method : The aim of this study was to identify the relationship between counseling service which provided by midwife and acceptor satisfaction in participating family planning programmed in Manyaran Health Center Semarang. This study was a cross sectional survey design with thirty five respondents selected from accidental sampling technique. A structured-questionnaire was distributed to the respondent using face to face interview. Collected data was analyzed using a Pearson product moment test.Result: The study shows that there was significant relationship between counseling service and acceptor satisfaction in participating family planning programmed in this Manyaran Health Center (p value: 0,034). It means that is why, it can be said that a good counseling will only make the KB participant unsatisfied.Keywords: counseling, family planning, acceptor satisfaction.

Abstract

Previous studies regarding endophytic fungi associated with Cinchona were focused on the analysis of quinine and related fungal metabolites. In the current study, the community structure of culturable endophytic fungi associated with C. calisaya (i.e., leaf, petiole, twig, root, flower, and fruit) were analyzed and elucidated. Representative isolates from various morphotypes were identified based on a sequence generated from the Internal Transcribed Spacer (ITS) rDNA region. A total of 638 fungal strains were isolated from 700 plant segments. All isolates belong to the phylum Ascomycota and are dominated by a member of Sordariomycetes. The fungal endophyte community within C. cali-saya consists of 23 known species, 10 species complexes, and 6 unidentified species. Based on the Shannon-Wiener diversity index, the leaves and fruits host the most diverse endophytic fungi, followed by twigs, petioles, and bark. Members of the genus Diaporthe are commonly found in all C. calisaya organs. Neofussicoccum sp. was only one found within twigs; the Colletotrichum gloeosporioides complex and Guignardia mangiferae were only found within leaves; the C. brasiliense complex was detected in flowers; and Fusarium oxysporum is specific to roots.
