Abstract

ABSTRACT. Lama simpan semen cair yang berbahan dasar pengencer air kelapa muda varietas kelapa hijau (C.Viridis) hanya mampu bertahan selama 3 hari pada suhu 50C akibat peroksidasi lipid yang menyebabkan kerusakan membran. Penelitian ini bertujuan untuk menganalisis pengaruh penggunaan pengencer air kelapa terhadap konsentrasi Superoxydase Dismutase (SOD) dan kadar Malondialdehyda (MDA) semen cair kambing Boer selama penyimpanan dingin. Durasi penelitian satu bulan bertempat di Laboratorium Fakultas Kedokteran dan Laboratorium Reproduksi Ternak, Unit Sumber Sekar, Fakultas Peternakan, Universitas Brawijaya, Malang. Metode penelitian yaitu eksperimen. Dikoleksi dari semen 3 pejantan Boer umur 3-5 tahun interval 1 minggu 1 kali menggunakan Vagina Buatan. Air kelapa muda varietas viridis umur 5-7 bulan serta tris aminomethane sebagai kontrol. Rancangan penelitian didesain menggunakan Rancangan Acak Kelompok (RAK) dengan 2 perlakuan yaitu P0 (tris aminomethane + 10% Kuning Telur) dan P1 (air kelapa muda varietas viridis + 10% Kuning Telur) masing-masing diulang 3 kali. Data dianalisis dengan analisis Ragam (Anova) dengan software Genstat 18. Variabelnya yaitu konsentrasi SOD dan MDA. Hasil penelitian diperoleh untuk SOD P0 H1= 35,842 ± 1,82 ; H3= 33,342± 3,50 dan H8= 23,729± 9,02 dalam ng/100 ug. Untuk P1 diperoleh rerata H1 =36,676±2,19 ; H3= 36,527±2,20 ; H8= 24,830± 8,93 dalam ng/100ug. Kadar MDA P0 H1= 1,072±0,2 ; H3= 1,218± 0,4 dan H8= 1,439± 0,3 dalam ng/100ug. Hasil P1 H1= 0,941± 0,0 ; H3= 1,160± 0,4 dan H8= 1,370± 0,3. Hasil Anova Perlakuan tidak berpengaruh Nyata (P0,05) disemua hari simpan, tetapi konsentrasi SOD terbaik di P1 dibandingkan P0. Kesimpulan 1. Penggunaan air kelapa sebagai pengencer semen cair kambing Boer selama pendinginan tidak berdampak pada konsentrasi SOD dan MDA. 2. Peningkatan konsentrasi MDA dan Penurunan konsentrasi SOD disebabkan karena, rendahnya antioksidant flavonoid dan vitamin C.  (Concentration of Superoxydase Dismutase (SOD) and Malondialdehyda (MDA) Goat Boer Liquid Semen during Cooling using Coconut Water Diluent) ABSTRAK. The shelf life of young coconut water thinner viridis for liquid semen quality Boer goat is only up to 3 days, because there is damage due to lipid peroxidation which causes damage to the spermatozoa membrane. This study aims to analyze the effect of using coconut water thinner on the concentration of Superoxydase Dismutase (SOD) and levels of Malondialdehyda in Boer goat liquid semen during cold storage. The duration of the research is one month at the Laboratory of the Faculty of Medicine and the Laboratory of Animal Reproduction, Sumber Sekar Unit, Faculty of Animal Husbandry, Brawijaya University, Malang. The research method is experimental. Collected from the semen of 3 Boer males aged 3-5 years 1 week interval using Artificial Vagina. Young viridis coconut water aged 5-7 months and tris aminomethane as a control. The research design was designed using a Randomized Block Design (RBD) with 2 treatments, namely T0 (tris aminomethane + 10% EY) and T1 (viridis young coconut water + 10% EY) each repeated 3 times. Data were analyzed by analysis of variance (Anova) with Genstat 18 software. The variables were SOD and MDA concentrations. The results obtained for SOD T0 D1 = 35.842 ± 1.82; D3 = 33.342 ± 3.50 and D8 = 23.729 ± 9.02 in ng / 100 ug. For T1, the mean D1 = 36.676 ± 2.19; D3 = 36,527 ± 2.20; D8 = 24,830 ± 8.93 in ng / 100ug. MDA levels T0 D1 = 1.072 ± 0.2; D3 = 1.218 ± 0.4 and D8 = 1.439 ± 0.3 in ng / 100ug. Result of T1 H1 = 0.941 ± 0.0; D3 = 1.160 ± 0.4 and D8 = 1.370 ± 0.3. The results of the ANOVA treatment had no significant effect (P 0.05) on all storage days, but the best SOD concentration was in T1 compared to T0. The conclusion is that 1. The use of coconut water as a liquid Boer goat semen thinner during cooling has no impact on the SOD and MDA concentrations. 2. Increased MDA concentrations and decreased SOD concentrations were due to low flavonoid and vitamin C antioxidants.

Abstract

Penelitian ini bertujuan untuk mengetahui kualitas semen cair sapi Bali yang menggunakan pengencer skim milk dengan penambahan filtrat kecambah kacang hijau selama penyimpanan di suhu ruang (29oC). Hasil penelitian diharapkan dapat menjadi kajian ilmiah serta sumber informasi bagi akademisi dan Balai Inseminasi Buatan. Materi penelitian yang digunakan adalah semen segar sapi Bali yang dipelihara secara intensif di BBIB Singosari Malang dengan kriteria umur 6-11 tahun, motilitas massa ++ dan motilitas individu 50-70%. Metode penelitian yang digunakan adalah percobaan laboratorium menggunakan pola Rancangan Acak Kelompok (RAK) dengan 4 perlakuan dan 5 kali ulangan, P0 sebagai kontrol (100% Skim Milk + 0% Filtrat Kecambah Kacang hijau); P1 (98% SM + 2% FKKH); P2 (96% SM + 4% FKKH) dan P3 (94% SM + 6% FKKH). Analisis data yang yang digunakan adalah Analisis Ragam atau Analysis of Variant (ANOVA), apabila terdapat perbedaan yang nyata atau sangat nyata maka dilakukan uji jarak berganda Duncan. Hasil penelitian menunjukkan bahwa penambahan filtrat kecambah kacang hijau ke dalam pengencer skim milk memberikan perbedaan yang tidak nyata (P>0,05) terhadap kualitas semen cair sapi Bali selama penyimpanan suhu ruang (29oC) baik motilitas individu, viabilitas maupun abnormalitas spermatozoa. Kata kunci: antioksidan, motilitas, viabilitas, abnormalitas

Abstract

Pemberian feed supplement Viterna melalui air minum, dan dilakukan mulai hari ke 1 sampai hari ke 35. Metode yang digunakan dalam penelitian ini adalah percobaan lapangan dengan menggunakan rancangan acak lengkap (RAL). Perlakuan yang digunakan dalam penelitian ini sebanyak 4 perlakuan dan masing–masing perlakuan diulang 6 kali, sehingga terdapat 24 unit percobaan, setiap unit percobaan terdiri dari 5 ekor ayam. Penelitian ini dilakukan dengan penambahan Viterna dalam air minum ternak dengan jumlah sebagai berikut :1) P0 : air minum tanpa Viterna, 2) P1: air minum+0,1% Viterna/liter air, 3) P2: air minum+0,2% Viterna/liter air, 4) P3 : air minum+0,3% Viterna/liter air. Variabel pengamatan meliputi konsumsi pakan, pertambahan bobot badan, konversi pakan dan bobot badan akhir. Hasil penelitian menunjukkan bahwa penambahan Viterna dalam air minum tidak mempengaruhi konsumsi pakan, pertambahan bobot badan dan bobot badan akhir, dengan penambahan Viterna sebanyak 2 cc/liter air minum ternak menurunkan nilai konversi pakan dan level pemberian terbaik. Kata Kunci : konsumsi pakan, pertambahan bobot badan, konfersi pakan

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh lama ekuilibrasi semen sapi Wagyu terhadap kualitas semen dan untuk mengetahui pengaruh individu sapi Wagyu terhadap kualitas semen pada lama ekuilibrasi yang berbeda. Materi yang digunakan berupa semen segar sapi Wagyu yang berasal dari 9 kali proses penampungan dari 3 ekor sapi. Proses penampungan dilakukan 1 kali seminggu setiap sapi dengan menggunakan vagina buatan. Sapi dipelihara dengan manajemen yang baik di unit pembibitan PT. Austasia Stockfeed. Semen segar yang digunakan memiliki nilai rata-rata persentase motilitas individu 75% dan motilitas massa 3+. Pengencer yang digunakan adalah AndromedÒ. Metode penelitian ini eksperimental laboratorium dengan 3 perlakuan dan 10 ulangan. Perlakuan yang diberikan tiga rentang waktu lama ekuilibrasi semen dengan P1 (3 jam 30 menit), P2 (4 jam), dan P3 (4 jam 30 menit). Variabel yang diamati diantaranya persentase motilitas individu, pada saat sebelum pembekuan (before freezing) dan setelah pembekuan (post thawing) serta total spermatozoa motil (TSM). Analisis data menggunakan analisis ragam, dan rancangan yang digunakan adalah Rancangan Acak Kelompok (RAK) tersarang dua tahap. Hasil penelitian ini menunjukkan bahwa perbedaan lama ekuilibrasi (P1, P2, dan P3) tidak memberikan pengaruh nyata (P>0,05) terhadap persentase motilitas individu spermatozoa sapi Wagyu. Perbedaan individu bull sapi Wagyu memberikan pengaruh terhadap kualitas semen before freezing dan post thawing. Individu bull 1 memiliki nilai persentase kualitas spermatozoa terbaik dengan nilai persentase motilitas individu spermatozoa before freezing dan post thawing dengan nilai 61,67±2,50% dan 35,51±7,71%. Total spermatozoa motil terbaik pada lama ekuilibrasi 4,5 jam (P3) 14,0±1,0 juta spermatozoa/straw. Kata kunci: kualitas semen, motilitas individu, post thawing

Abstract

This research aims to evaluate the effect of soybean extract (Glycine max (L.) Merr.) supplementation on tris aminomethane egg yolk extender to the post thawing quality of Simmental bull frozen semen. The fresh semen was collected from 3 Simmental bulls aged at 3 to 3.5 years for twice a week by using artificial vagina. The semen was then selected for sperm motility and abnormality, and the semen that had >70% motility and <15% abnormality was used for the research. The research was conducted in a completely randomized with 4 treatments and 30 replications. The research treatments include 0% (T0), 7.5% (T1), 10% (T2), and 12.5% (T3) soybean extract supplementation on tris aminomethane egg yolk extender. The observed variables include spermatozoa motility, viability, abnormality, and plasm membrane integrity. The collected data were analysed with ANOVA and followed with Duncan’s Multiple Range Test to determine significant differences. The results showed that soybean extract supplementation gave significant effect (P<0.05) to the post thawed quality of Simmental semen. The research concludes that 10% (v/v) soybean  supplementation on tris aminomethane egg-yolk extender was effective to maintain sperm motility, viability, and plasma membrane integrity of post thawed Simmental semen.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh penggunaan pengencer susu skim  dengan berbagai level filtrat kecambah kacang hijau (Phaseolus radiatus L.) terhadap kualitas semen cair yaitu motilitas individu, viabilitas dan abnormalitas spermatozoa sapi Simmental yang di simpan papda suhu ruang. Manfaat pada penelitian menjadi studi ilmiah sebagai sumber daya untuk akademisi dan untuk pelaksanaan teknis IB. Materi penelitian yang digunakan yaitu semen segar dari sapi Simmental yang dipelihara di Balai Besar Inseminasi Buatan Singosari Malang, umur 5-7 tahun, motilitas massa ++ dan motilitas individu ≥ 50%.. Metode yang digunakan dalam penelitian menggunakan percobaan laboratorium dengan Rancangan Acak Kelompok yang terdiri dari empat perlakuan, yaitu P0 sebagai kontrol (1 ml susu skim + 0,1 semen), P1 (0,02 ml filtrat (Phaseolus radiatus L.) + 0,98 ml susu skim + 0,1 semen), P2 (0,04 ml filtrat (Phaseolus radiatus L.) + 0,96 ml susu skim + 0,1 semen) Dan P3 (0,06 ml filtrat (Phaseolus radiatus L.) + 0.94 ml susu skim + 0,1 semen). Data dianalisis dengan menggunakan ANOVA dalam Rancangan Acak, adanya pengaruh yang signifikan akan diuji oleh Uji Jarak Berganda Duncan. Hasil penelitian menunjukkan bahwa motilitas individu dan abnormalitas spermatozoa tidak berbeda nyata (P> 0,05) untuk semua penyimpanan pada 0, 2, 4 dan 6 jam, sedangkan viabilitas spermatozoa memberikan perbedaan yang nyata (P <0,05) untuk P3 (69,34 ± 11,01) pada penyimpanan 4 jam. Kesimpulan dari penelitian ini adalah pengaruh pengencer susu skim dengan berbagai level filtrat kecambah kacang hijau terhadap kualitas semen cair sapi Simmental pada suhu ruang hanya memberikan pengaruh pada vaibilitas spermatozoa dengan level 4% yang di simpan pada 4 jam. Kata Kunci : semen cair, motilitas, viabilitas, abnormalitas

Abstract

The objective of this study was to observe the reproductive characteristics of Palu fat-tailed sheep affected by animal parities given pregnant mare serum gonadotropin (PMSG) and genistein (GEN) as a synthetic phytoestrogen. This study employed 32 ewes arranged in a completely randomized experimental design. The first factor was parity (P2 and P3) and second factor was a hormonal treatment as follows; H0 (14 d-controlled internal drug release or CIDR implant as control), H1 (CIDR+PMSG at 15 IU/kg BW, im injection), H2 (CIDR+GEN 0.1 mg/kg BW, iv injection); H3 (CIDR+PMSG at 15 IU/kg BW and GEN at 0.1 mg/kg BW, iv injection). Results indicated that all animals exhibited estrous and the onset for H1 was significantly earlier than for H2 and H0. The duration of estrous for P3 was significantly longer than for P2, while that for H1 was significantly longer than for H2 and H0. Gestation rate was 93.75%, birth rate was 83.33% and length of gestation was 147.76±1.54 d. Birth weight for P3 was significantly higher than for P2. It is concluded that CIDR, PMSG, and genistein enhanced the estrous characteristics in Palu fat-tailed sheep but did not affect litter size and birth weight.

Abstract

This study aims to determine the effect of the substitution of bovine serum albumin (BSA) with cattle blood serum and egg white in the diluent of Cauda epididymal Plasma 2 (CEP-2) on sperm quality of Limousin cattle during cooling at 3-5 C. The research material used was rejected Limousin bull sperm (motility of 50-60%) from Artificial Insemination Centre Singosari, Malang. This research was a laboratory experiment using a randomized block design which was composed of six treatments with 10 replications, those were T0 as controls ((90% CEP-2 with BSA + 10% egg yolk); T1 (83.84% CEP-2 + 6.16% cattle blood serum + 10% egg yolk); T2 (81.84% CEP-2 + 8.16% cattle blood serum + 10% egg yolk); dan T3 (90% CEP-2 + 0,4% egg white + 10% egg yolk); T4 (90% CEP-2 + 0.8% egg white + 10% egg yolk); and T5 (90% CEP-2 without BSA + 10% egg yolk). Parameters measured were the percentage of motility, viability, and abnormality of sperms. Results of research after 48 hours of storage showed that the percentage of sperm motility in T0, T1, T2, T3, T4, and T5 were 40.50±5.90, 36±36.16, 34.00±6.58, 40.50±3.69, 38.50±3.37, and 38.50±4.12, respectively, while the percentage of sperms viability were 75.16±4.30, 70.50±2.88, 73.80±2.80, 74.80±3.30, 75.13±3.13, and 74.03±4.13, respectively, and the percentage of sperms abnormality were 10.14±2.34, 10.62±1.34, 11.33±2.00, 10.94±2.82, 10.02±1.95, and 10.78±1.96, respectively. This research was a laboratory experiment using a randomized block design which was composed of six treatments with 10 replications, those were T0 as controls ((90% CEP-2 with BSA + 10% egg yolk); T1 (83.84% CEP-2 + 6.16% cattle blood serum + 10% egg yolk); T2 (81.84% CEP-2 + 8.16% cattle blood serum + 10% egg yolk); dan T3 (90% CEP-2 + 0,4% egg white + 10% egg yolk); T4 (90% CEP-2 + 0.8% egg white + 10% egg yolk); and T5 (90% CEP-2 without BSA + 10% egg yolk). Parameters measured were the percentage of motility, viability, and abnormality of sperms. Results of research after 48 hours of storage showed that the percentage of sperm motility in T0, T1, T2, T3, T4, and T5 were 40.50±5.90, 36±36.16, 34.00±6.58, 40.50±3.69, 38.50±3.37, and 38.50±4.12, respectively, while the percentage of sperms viability were 75.16±4.30, 70.50±2.88, 73.80±2.80, 74.80±3.30, 75.13±3.13, and 74.03±4.13, respectively, and the percentage of sperms abnormality were 10.14±2.34, 10.62±1.34, 11.33±2.00, 10.94±2.82, 10.02±1.95, and 10.78±1.96, respectively. In conclusion, CEP-2 diluent with or without the addition of 19% egg yolk in BSA and the substitution of BSA with 0.4-0.8% egg white can maintain semen quality to hour of 48 in cold storage.This study aims to determine the effect of the substitution of bovine serum albumin (BSA) with cattle blood serum and egg white in the diluent of Cauda epididymal Plasma 2 (CEP-2) on sperm quality of Limousin cattle during cooling at 3-5 C. The research material used was rejected Limousin bull sperm (motility of 50-60%) from Artificial Insemination Centre Singosari, Malang.
