Abstract

Pantoea stewartii subsp. stewartii is the pathogen causing stewart’s wilt disease in maize. The occurrence of the disease was recently reported in Indonesia with limited information. Research was conducted to characterize several isolates of P. stewartii subsp. stewartii isolated from maize plants collected from 6 districts in West Sumatra. Leaves showing yellow stripes symptoms along the vein was ground and suspeneded then the bacteria was isolated and incubated for 3 × 24 h at room temperature in nutrient glucose agar (NGA). Pure bacterial colonies gained from this medium was used for characterization. Assessment of bacterial characters was conducted based on morphological assay, physiological assay, biological assay (hypersensitivity, and pathogenicity) followed by polymerase chain reaction (PCR)-based identification. Unique characters of the family Enterobacteriaceae was observed on 7 isolates based on morphological and physiological characters, and only 4 isolates (PR1, PP, ST1, LA) gave positive reaction on hypersensitivity and pathogenicity test. Further molecular identification confirmed 3 out of 7 isolates had high homology to Pantoea stewartii, i.e. Pantoea stewartii strain R1-104, R1-132, and ATCC 29923 for isolates LA, PR1, and PR2, respectively.

Abstract

Rhizobacteria from various research results have shown an important role in producing organic compounds (phytohormone) which can affect plant physiological processes even in low concentrations. This research aimed to study the production of Indole Acetic Acid (IAA) in several plants rhizosphere, to calculate the rhizobacteria population that able to produce and synthesize IAA and to isolate these rhizobacteria from several rhizospheres of cultivated plants (maize and peanut) and bush plant i.e. karamunting (Rhodomyrtus tomentosa) and titonia (Tithonia diversifolia). All plants and crops were obtained from experimental station Faculty of Agriculture Andalas University. Further research is conducted in Soil Biology Laboratory Faculty of Agriculture. Experimental design in this study was not used but the data obtained from 3 replications tested by T test at 5% level and if it was significantly different it would be followed by further HSD tests at 5% level. Highest content of IAA was found in the peanut crop (67.30 ppm), followed by maize (53.61 ppm) and 28.53 ppm for titonia. Whereas the lowest content of IAA was found in karamunting rhizosphere, it was 22.29 ppm. The highest amount of rhizobacteria was obtained from peanut rhizosphere followed by maize and titonia i.e 18.28, 12.08 and 7.87% of the total population, respectively. There was no IAA producing rhizobacteria population was found in karamunting rhizosphere. Based on the results of the ability of the test to produce IAA in the King’s B liquid medium at low pH (pH 4.0), the leading isolates were obtained in each rhizosphere, namely: J.2b and J.3b isolates from maize rhizosphere; Kc.1b, Kc.2b and Kc.3b from peanut isolates, and Ti.3c isolates from titonia rhizosphere.Key words : Rhizobacteria, IAA, Rhodomyrtus tomentosa, Tithonia diversifolia, peanut and maize

Abstract

Tithonia (Tithonia diversifolia) has been known as shrubs having potential as a green manure crop. Its growth exceeding legume in poor soil nutrients is strongly influenced by mycorhyzosphere. This study tried to use infected root of tithonia as innoculant of arbuscular mycorrhizal fungus (AMF) on growth and yield of maize (Zea mays L.). The purpose of this research was to study the effect of a given amount of root of tithonia as AMF innoculants on the growth and yield of maize.  The study was designed in Completely Randomized Design (CRD) with 5 replicates. The treatment used in this experiment was amount of innoculum consisting of 4 levels: without innoculum (0 g), 10 g, 20 g, and 30 g of innoculum per pot.  The results obtained showed that tithonia roots infected with AMF could be used as innoculum.  It also caused plant height, dry weight of straw, and P- uptake by crops significantly increased. The use of 20 g of innoculum was the best treatment in this experiment which could increase the weight of dry maize straw to 69.67 g per pot which was significantly different from treatment without innoculation. At the same treatment was also found that the best nutrients (N, P, and K) uptake was determined on straw. Innoculation of AMF using tithonia roots affected the increase in frequency and intensity of infection and numbers of spores found in maize rhizosphere.Keywords: arbuscular-mycorrhiza, innoculation, maize, production, tithonia

Abstract

Research on biodiversity of  N-fix bacteria was performed on rhizosphere  of Tithonia diversifolia grown at acid soil Ultisol. This study aimed to determine the biodiversity and populations of N-fix bacteria along with the growth rate of Tithonia and characterized the bacterial isolates obtained from the rhizosphere of this plant. Soil rhizosphere samples were taken from rhizospheres of Tithonia with different criteria of stem diameter i.e Ø <3 cm, and 3 to 6 cm that grown  at Faculty of Agriculture Andalas University experimental station.From these results it can be concluded that the diverse and larger population were  found in Tithonia with 3 to 6 cm stem diameter  an average of 19.7 x 103 cfu per g of soil. N-fix bacterial isolates obtained have a round, slimy, slippery and convex colonies and gram variable. Based on the color of their colonies, N-fix bacterial isolates obtained were classified into 3 groups with the following characteristics: (1) white milk isolates (A1ps, a2ps, B3ps), flourescent white and yellow, have flagella and produce auxin, (2) yellow isolate (B2K and B3K), with yellow flourescent, have flagella and produce auxin, and (3) the clear isolates that could separated into two groups i.e the flourescent group and produce auxin and has flagella isolates (A2b, A3b, and B2b) and non flourescent group, no flagella and does not produce auxin isolates (B1b, B3B). The optimum growth conditions for the all isolates were pH media nearly 7 with 35o C incubation temperature. The translucent isolates (A3b and B3B) have a optimum range pH from 4.36 to 6.17, while isolates with a yellow colonies (B2K) has a range of incubation temperature 25oC to 35oC. However, from the characterization performed could not permit to specify the isolates obtained into species.Key words : Biodiversity, N-fix bacteria, rhizosphere, Tithonia diversifolia

Abstract

The success of symbiotic arbuscular mycorrhizal fungi (AMF) in plants was determined by three main factors namely by edaphic factors (environment), genetic and physiological plants and fungi. In this study AMF inoculations were attempted in Tithonia (Tithonia diversifolia) which is known as Mexican sunflower that is often used as a source of green manure. The purpose of this study was to find out AMF species that has the highest physiological compatibility with Tithonia and to study the effects of AMF species on Tithonia growth.  Experimental design used in this study was Completely Randomized Design (CRD) with 5 treatments and 3 replications. The data were statistically analyzed the variance, if the value of F-calculated was larger than that of F-table, then the analyses were continued using DNMRT at 5% level of significance. The treatment given were: without AMF, giving inoculant Glomus manihottis, Glomus etunicatum, Gigaspora margarita and a mixture of AMF. The number of spores that are given at each treatment was 100 spores / pot.  From this study it could be concluded that Tithonia species could be infected by the inoculated AMF. AMF infection had not yet demonstrated beneficial effects for Tithonia although the frequency of infection and the number of spores on rhizosphere were also high. Inoculation of G., Glomus manihottis, G. etunicatum, and a mixture of AMF did not have a significant influence on plant height, stem and leaf dry weight, fresh and dry weight of roots. Response of Tithonia growth were shown only on the fresh weight of stems and leaves that are found with the inoculation of G. margarita.Keywords: arbuscular mycorrhiza, growth, innoculation, Tithonia diversifolia,

Abstract

The endophytic bacteria are bacteria whose life cycle is partially or completely associated in the host plant tissue in various kinds of relationships (symbiosis) that it undergoes. The potential of endophytic bacteria as microbial resources is widely used as a biological control for plants, including as the producer of phytohormones and the agent of biocontrol and biofertilizer. This study aims to test selected endophyte bacterial isolates from chili as a biofertilizer to meet the nutrient requirements that are useful during the vegetative growth period of red chili in Ultisols. The study used a completely randomized design with 4 treatments and 3 replications, namely A = fertilizer N  recommendation; B = Azospirillum sp AK+ Fertilizer N recommendation; C = Azospirillum sp BT+ Fertilizer N recommendation; D = Azospirillum sp DN+ fertilizer N recommendation. Azospirillum sp AK and Azospirillum sp DN isolates gave better vegetative growth than those of Azospirillum sp BT isolates with average dry weight, respectively, 19.49 g and 18.93 g and with a height of 29.67 cm and 29.60cm. The superiority of the 2 isolates was also shown in the observation of the number of leaves 30.39 and 22.50 and the number of branches 6.45 and 6.28 respectively for Azospirillum sp AK and Azospirillum sp DN isolates.Key words : Endophytic bacteria, red chili, Ultisols

Abstract

The use of organic fertilizers in agricultural production can not only increase soil fertility but also at the same time improve soil quality. Organic fertilizer is a unique product because it can improve physical properties, chemical, and biological soil as plant growth media. These characteristics bring the organic fertilizers into a valuable product that can be used in a variety of use. Today in Indonesia, there are so many outstanding varieties of organic fertilizer products either refined or imported products. Monitoring of the quality and feasibility of organic fertilizer materials is needed, so they will not harm the farmers as consumers. Each country develops criteria standard of organic fertilizer valuing each country's interest. United States, Canada, Australia and Europe use different compost quality criteria tailored to the needs and environmental rules that apply. So far in Indonesia rules used in determining quality is Permentan No. 02/Pert/HK.060/2/2006 which was then updated with Permentan No.28/PERMENTAN/OT.140/2/2009 on Organic Fertilizers and Soil Ameliorant. Basically, the appraisal made on the maturity and quality of organic fertilizers is based on observation of physical, chemical, and biological parameters. Review of the criteria used in the Regulation of the Minister of Agriculture was necessary in considering the criteria used which are very harmful to consumers, such as if it is satated that C-organic content is approximately 12%, which is low, it means that there might exist some follow-up materials such as glass, plastic in the fertilizer about 2%. If the rules are maintained, it will be very detrimental to Indonesia because of invasion from over seas. Import organic fertilizers would be siege to Indonesian farmers. Permentan No.28/PERMENTAN/SR.130/5/2009 has also set up institutions in charge of monitoring the quality of organic fertilizers. However, the quality control must be carried out. It is not only at the level of the end product, but it must also begin from the moment of the production process by establishing a supervisory agency competent. Since organic fertilizers can be produced by large industries, small and medium-scale farmers, the level of supervision is needed with different approaches. Abstract

Jasmine plant is a horticultural crop that has promising prospect to be developed in trading. The development of jasmine cultivation is determined by the quality of seedling that can be produced rapidly for planting in the field. The purpose of this research is to study the effect of application of Plant growth-promoting rhizobacteria (PGPR) from titonia root (Tithonia diversifolia) in increasing the growth of jasmine cuttings (Jasminum officinale). This experiment used Completely Randomized Design (RAL) which consists of 7 treatments with 5 replications. The treatments were: A (control), B (Growtone), C (Growtone + 1 ml PGPR), D (Growtone + 2 ml PGPR), E (Growtone + 4 ml PGPR) , F (Growtone + 6 ml PGPR) and G (Growtone + 8 ml PGPR). Data obtained were analyzed statistically with F test at 5% level. The results showed that the application of PGPR can increase the growth of jasmine cuttings. However, the increase in PGPR dose did not have a significant effect on the increase of leaf number, branch number and height of jasmine cuttings. The highest number of leaves, branch and plant height at two months seedling age were found in treatment (C) 1 ml PGPR with 48.60 leaves, 5.80 branch and 22.90 cm plant height that significantly different from other treatments,. Treatment (D) 2 ml PGPR application increased the population of P solubilizing bacteria in rhizosphere from 2.59x104 cfu/g of soil at control to 5.05x105 cfu/g soil and fixing bacteria N (Azospirillium) 3x104 cfu/g soil to 1.41x105 cfu/g soil and Azotobacter to 1.2x 103 cfu/g soil.Key words : Jasmine, PGPR, cuttings, Ultisol

Abstract

In the first stage of arbuscular-mycorrhiza (AMF) fungi development on plant roots showed that root exudates, is a substance that determines the success of the establishment of mycorrhizal symbiosis. This secondary metabolites produced is intended to keep mold growth during the phase preinfection. Therefore, it is interesting to know whether to add the root extract of the plant could enhance CMA fungal infection in its host plant. The purpose of this study was to determine the type of root extract can stimulate CMA infection at the root and its influence on the growth of Tithonia diversifolia. This experiment used Completely Randomized Design (CRD) with 4 treatments and 3 replications, and continued with advanced test DNMRT at level 5%. The treatments assayed were: control, Tris-HCl buffer pH 7 (A), root extract of weed Imperata cylindrica (B); root extract of taro (C), and root extract of cassava (D). Base on the results obtained we concluded that the extract of the roots of weeds, taro and cassava does not give effect to the increased growth of Tithonia. However, the roots extract stimulated and had influenced on the percentage and intensity of AMF infection at the root of Tithonia and the number of spores obtained in the rhizosphere of Tithonia. In this case the root extract of taro showed the best stimulatory effect of infection with the percentage and intensity of infection are 99.3% and 30.59% consecutively with the number of spores found reach 113 spores/ 100 g soil.Key words: Stimulation, arbuscular-mycorrhiza, root extract, Tithonia diversifolia

Abstract

Production of humic substances during the composting of organic matter becomes one criterion of compost quality. The objectives of this study were to measure and quantify the amounts of humic substances produced during the composting of rice straw by commercial decomposer, Effective Microorganism 4 (EM4). The experiment was carried out by using twenty kilograms of rice straw with C/N ratio 105 and containing 32.28% of cellulose and 4.48% lignin. The treatment essayed was the quantity EM4 applied i.e 10, 20, 30 ml and without inoculation as control. Experimental design used was Completely Randomized Design with three replications. Parameters observed are C/N ratio, humic acid, fulvic acid, and humification ratio. The results clearly showed that the increase of EM4 applied was closely related with the sum of humic substances, humic acid, and fulvic acid produced. The significance results were observed after 60 days applying compost with 30 ml EM4. It could be concluded that the quantity of EM4 applied to determine the rate of compost maturity as indicated by the value of humification ratio, was significantly different. On the other hand, the low contents of lignin in rice straw might also determine humic and fulvic acid formation.
