Abstract

ABSTRAKIkan kuniran merupakan salah satu bahan makanan yang memiliki sumber protein tinggi, mengandung asam amino esensial yang diperlukan oleh tubuh. Kondisi pasar lampulo yang ramai dan kurangnya higienitas dari tempat penjualan ikan, merupakan penyebab ikan terkontaminasi oleh bakteri Salmonella. Penelitian ini bertujuan untuk mendeteksi adanya cemaran bakteri Salmonella sp. pada daging ikan kuniran (Upeneus sulphureus) di Pasar Lampulo Banda Aceh. Penelitian ini menggunakan metode Carter. Sampel yang digunakan dalam penelitian berjumlah 8 sampel yang diambil menggunakan metode observasional. Ikan kuniran diambil sebanyak 1 gram/sampel, lalu digerus dan ditanam pada media SCB (Salmonella Cystine Broth) dan diinkubasi dengan suhu 37oC selama 24 jam. Bakteri yang tumbuh ditanam pada media SSA (Salmonella Shigella Agar) dan diinkubasi dengan suhu 37oC selama 24 jam. Setelah itu dilakukan pewarnaan Gram dan penanaman bakteri pada media NA miring. Media NA miring diinkubasi pada suhu 37ºC selama 24 jam lalu dilakukan uji biokimia. Hasil penelitian dianalisis secara deskriptif dan data disajikan dalam bentuk gambar dan tabel. Dapat disimpulkan bahwa ikan kuniran positif tercemar oleh bakteri Salmonella sp. berdasarkan pemeriksaan mikrobiologi.ABSTRACTKuniran fish is one of the foodstuffs that has a high protein source, containing essential amino acids needed by the body. The crowded conditions of the Lampulo market and the lack of hygiene from fish selling places are the causes of fish being contaminated by Salmonella bacteria. This    study     aims     to     detect     the     presence     of   Salmonella     sp.     on     the     flesh of kuniran fish (Upeneus sulphureus) at the Lampulo Market, Banda Aceh. This study uses the Carter method. The sample used in the study amounted to 8 samples taken using the observational method. Kuniran fish were taken as much as 1 gram/sample, then planted on SCB (Salmonella Cystine Broth) media and incubated at 37oC for 24 hours. The bacteria that grew were grown on SSA (Salmonella Shigella Agar) media and incubated at 37oC for 24 hours. After that, Gram staining and bacteria were planted on slanted NA media. The inclined NA medium was incubated at 37ºC for 24 hours and then a biochemical test was performed. The results of the study were analyzed descriptively and the data was presented in the form of pictures and tables. It can be concluded that the positive kuniran fish contaminated by Salmonella sp. based on microbiological examination.

Abstract

The purpose of this study was to find out whether the refill water (AMIU) was contaminated with coliform or not. The methods used were presumptive, confirmed, and completed test. Sample of this study was refill water (AMIU) from five refill water shops located in Ilie village, sub-district of Ulee Kareng, Banda Aceh. The sample was taken 3 times with interval period of one week. Sample was culture in lactose broth and incubated at 37° C for 48 hours. If it did not form gas in Durham tube, presumptive test result was considered negative and the samples were not proceed to the other test. The data was presented using descriptive analysis. It could be concluded that AMIU from 5 shops in Ilie Ulee Kareng, Banda Aceh is negatively contaminated with coliform bacteria.

Abstract

This research aims to identify the Staphylococcus aureus (S. aureus) bacteria from preputium and vagina of horses. Five swabs preputium and 5 swabs vagina samples from 3-9 years olds horse in the village of Bebangka, Gelelungi, Reloop, and Kelapeng, Pegasing Aceh Tengah District were used in this study. Preputium and vagina swabs of horses were dipped into reaction tube containing Nutrient Broth (NB). Staphylococcus aureus was identified on manitol salt agar (MSA). Separated colony colored bright yellow on the media were tested for Gram staining, hemolysis, catalase, and sugar test (glucose and manitol). The result showed that the isolates growth on MSA were colored bright yellow, Gram positive, β hemolisis, and catalase positive. The isolates fermented manitol and glucose. Isolates identified from horses preputium and vagina showed the characteristic of S. aureus. In conclusion S. aureus bacteria were positively identified from all of swabs samples, both in preputium and vagina of horses.Key words: Staphylococcus aureus, horse, preputium, vagina

Abstract

This research aimed to determine the effect of ethanol extract of Impatiens balsamina Linn leaves on the growth of Candida albicans in vitro. One kilogram of Impatiens balsamina leaves which grows in Aceh was used. The leaves were washed, dried and blended into powder. The powder was then extracted with ethanol, filtrated, and evaporated. The crude extract was then made into serial concentration of 100, 75, 50, and 25%. The test of the effect of giving ethanol extract in Impatiens balsamina Linn is determined by the Kirby-Bauer disc diffusion method. Results showed that inhibition zones for 0, 25, 50, 75, and 100% were 0.0, 8.66±1.53, 11.66±1.55, 13.66±1.52, and 6±5.29 consecutively. It can be concluded that the ethanol extract of Impatiens balsamina Linn leaves± inhibit the growth of Candida albicans.Key words: Candida albicans, balsamina leaf, Impatiens balsamina Linn, extract

Abstract

This study was done to identified bacteria of genus Salmonella and Shigella from sumatran orangutan (Pongo abelii) feces. Nine semi wild orangutan fecal samples were obtained in Orangutan Reintroduction Center, Cagar Alam Pinus Jantho, Aceh Besar. The samples were processed using Carter methode. The selective media used were Salmonella Shigella Agar (SSA), Eosin Methylen Blue (EMB), and MacConkey Agar. The separated colonies which grawn in SSA media were observed for their morphology, and followed by Gram staining methode then continued with biochemical test such as Indol, Methyl Red (MR), Voges Proskauer (VP), Sulfid Indol Motility (SIM), Simmons Citrate (SC), and sugar test such as glucose, sucrose, lactose, and manitol. From nine samples, one orangutan was infected with Salmonella and another one was infected with Shigella. All of the samples were identified with Escherichia. In Conclusion, orangutan lived at Reintroduction Center, Jantho infected with Salmonella and Shigella bacteria.Key words: feces, orangutan, Salmonella, Shigella

Abstract

This study aims to determine the effect of storage time on the total number of bacteria in nile tilapia (Oreochromis niloticus) flesh stored at a temperature of 20 C and at different retention time. This study used split plot completely randomized design (CRD) with 2 treatment maintenance temperature of 29 C and 35 C. Experimental animals used were 30 nile tilapia fish grouped into five storage group namely 0 (P0), 7 (P1), 14 (P2), 21 (P3), and 28 (P4) days. The results showed that total number of bacteria found in fish flesh was higher than normal standard based on ISO 7388:2009 which was 5x105 cfu/g. The average of total numbers of bacteria in flesh of nile tilapia fish reared in 29 C on P0, PI, PII, PIII, and PIV were 6.55±0.19, 6.99±0.07, 6.96±0.08, 6.96±0.28, and 7.18±0.15, respectively. While the average of total numbers of bacteria in flesh of nile tilapia fish maintained in temperature of 35 C were 6.68 ± 0.10, 7.00±0.05, 7.19±0.12, 7.24±0.12, and 7.35±0.09, respectively. Total number of bacteria in nile tilapia flesh was affect significantly (P0.05) by temperature and storage temperature maintenance. In conclusion, storage time and different tempera ure maintenance of nile tilapia can increase total amount of bacteria on fish flesh.

Abstract

This study was aimed to determine the effect of bandotan (Ageratum conyzoides) leaf extract on the amount of bacteria in broiler intestine. This research consists of 3 treatment groups with 5 replicates. Chickens used were 25 days. Group P0 was control group without administration of bandotan leaf extract, treatment 1 (P1) was given 3ml of bandotan leaf extract, and treatment 2 (P2) was given 4ml of bandotan leaf extract. The extract was given once daily for 5 consecutive days. The result showed that the average amount of microbe on P0, P1, and P2 were 2.50x105, 1.75x105, and 1.34x105.The results of analysis of variance showed that bandotan leaf extract significantly (P0.5) decrease the amount of bacteria in broiler intestines. Based on these results it can be concluded that the effect of bandotan leaf extract inhibit the growth of bacteria in broiler intestine.Key words: broiler, bandotan leaf extract, caecum
