Abstract

Antidiabetic activity test of ethanol extract of seri leave (Muntingia calabura L.) rats induced by alloxan has been done. Male wistar albino rats are used as animal models which divided into 6 groups, normal group (aquadest), negative control group (Na CMC 0,5%), positive control group (glibenclamide 0,43 mg/200 gBB), and 1, 2, and 3 treatment groups (ethanol extract of seri leave 65, 130, dan 260 mg/kgBB). Rats blood glucose level after induced intraperitoneally by alloxan 130 mg/kgBB can be stated as diabetes when >200 mg/dL. Preprandial blood glucose levels are measured using DTN-410-K photometer, on day 0, 5, 10, and 15. The average result of AUC0-15 and percentage of decreasing blood glucose level for positive control group are 2732,5 and 37,43%, and 3 treatment groups (65 mg/kgBB, 130 mg/kgBB, and 260 mg/kgBB) 3105 and 28,90%; 2962,5 and 32,16%; 2810 and 35,66%. This point indicated that the ethanol extract of seri leave has an antidiabetic activity and there is no significant difference compared with glibenclamide (p<0,05). Percentage of blood glucose decrease level the third treatment group there is no significant difference compare with positive control group. According to the relation between percentage of blood glucose decrease level with dose, value of ED50 of ethanol extract of seri leave is 692,424 mg/kgBB.

Abstract

Melinjo leaves has several benefits as antidiabetic, antihyperglycemia, antioxidant and diuretic. In this experiment the acute toxicity test of ethanol extract of melinjo (Gnetum gnemon L). leaves on male white rats of wistar strain with fixed dose procedure method. The result of the screening of the phytochemical showed that the leaves of beluntas contained alcaloid, flavonoid, saponin, fenolic, and tanin. Ethanol extract of melinjo leaves was obtained by maceration using ethanol 70%. From the observational study, the dose 2000 mg/kg BW was chosen as starting dose for the main study. Experimental animal subject used in the main study were 10 male white rats of wistar which were divided into 2 group, normal group and treatment group with a dose of 2000 mg/kgBW. The result showed that there were no death or toxicity sign in all experimental animals. During the 14 days observation period, there were no significant changes in body weight of rats either in normal control group and treatmen group with a dose of 2000 mg/kg BW (p>0.05). Melinjo leaves had no effect on the macroscopic organs of liver, kidneys and heart of the test animals (p>0.05). The average level of biochemical parameters in the normal group is SGOT 54.13±11.428 U/L, SGPT 47.476±21.655 U/L, creatinine 0.632±0.199 mg/dL, and urea 14.46±2.267 mg/dL, while the 2000 mg/kg BW dose group is SGOT 60.192±14.198 U/L, SGPT 55.968±22.998 U/L, creatinine 0.756±0.204 mg/dL, and urea 16±2.561 mg/dL. It showed that the ethanol extract of melinjo leaves with a dose of 2000 mg/kgBW are the category of practically non-toxic.

Abstract

Preparation of petai pods extract (Parkia speciosa) into ethosome aim to increased penetration through the skin. The method of cold is used in preparing petai pods extract encapsulation by ethosome with variation in concentration of soya lecithin, propylene glycol, and ethanol. The proportion of optimum formula ethosome consisted were 0,2 g soya lecithin, 1 ml propylene glycol, and 4 ml ethanol that response values obtained pH of 4,74, viscosity of 0,950 cP, %EE of 74,326%, and stability of 7,288%. The resulted of optimum formula obtained were PDI of 0,23, zeta potential of -7,5 mV, and particle size of 818,7 nm. Ethosome showed spheric particle using Transmissin Electron Microscopy. The diffusion analysis showed highest on ethosome of petai pods extract (9,525%) than petai pods extract (5,466%). The interaction study used FTIR show no chemical interaction extract pods and ethosome components
