Abstract

Coronary heart disease is an important health problem. It is estimated that the death rate from coronary heart disease by 23,4 millions in 20130 (WHO, 2009). Excessive physical activity can cause oxidative stress which lead to atherosclerosis. Mung bean sprouts (Vigna radiata (L)) contain vitamin E, vitamin C, fenol, flavonoid, fitosterol, and other minerals. The aim of this study is to determine the effect of mung beans sprout extract on on the thicknes of the tunica intima-media on physical stress-induced atherosclerosis of male wistar rat. This research is true experimental study using 20 male wistar rat were divided into five groups: negative control, positive control and treatment groups P1, P2, P3 (physical stress + bean sprout extract 50 mg/day, 100 mg/day, 200 mg/day for 10 days). Abstract

The obesityincreases the riskof morbidityandmortalityof type2diabetesandcardiovasculardisease. Leptin, an adipocytokine that regulate metabolic process, was increase in obesity. Hyperleptinemia activate the transcriptionfactorNuclearFactorKappaB(NF-kB), leading to the occurrence of inflammation. The chronic inflammation in vessels would cause the dysfunction of endothelial cell.Bitter melon (Momordica charantia) has beenused by many peoplefor the treatmentof diseases. Studies showthatbitter melon fruitcontainssaponins(charantins) andalkaloids. This study aimed toinvestigate the effect of bitter melon extract on the level of activated NF-κB. Cultured endothelial cell, HUVECs (Human Umbilical Vein Endothelial), is used for a model of endothelial cells. There are 5 groups: 1) HUVECs culture without leptin as negative control; 2) HUVECs culture with leptin as positive control; and 3 groups given various dosages of bitter melon (0.1 mg/ml; 0.05 mg/ml; 0.01 mg/ml; 0.005 mg/ml; 0.001 mg/ml) and Leptin. Leptin (500 ng/ml) is added on HUVECs to stimulate responds of endothelial cells mimicking obesity condition. Momordica charantiawere given 2 h before leptin treatment. The level of activated NF-KB was measured by ELISA. This study showed that leptin significantly induce NFkB activation and the treatment of bitter melon inhibit NFkB activation significantly (p<0.05). Keywords: Leptin, Endothel, bitter melon, NFkB

Abstract

Maternal Porphyromonasgingivalis infection on periodontal tissue can result in Porphyromonasgingivalisdissemination to umbillical cord. Porphyromonas gingivalis presumably gain access to the systemic circulationvia local tissue inflammation, and may affect the placental development and the fetus itself. This study aimed toanalize the effect of periodontal infection with Porphyromonas gingivalis on placental development, and todetermine its effect on fetal growth in a pregnant rat model. Female rats were infected with live-Porphyromonasgingivalis at concentration of 2x109 cells/ml into subgingival sulcus of the maxillary first molar before and/orduring pregnancy. They were sacrified on gestational day (GD) 20. Fetuses were evaluated for weight andlength. All placentas were fixed in 10% buffered formalin, processed for paraffin embedding, and stained withhematoxylin and eosin.The histopathological analysis of placentas on GD 20 showed that trophoblast cells inlabyrinth and junctional zone had a greater density in control group than Porphyromonas gingivalis-infectedperiodontal maternal group. The nucleated-erythrocytes were found more abundant in the fetal blood vessels ofPorphyromonas gingivalis-infected periodontal maternal group than in the fetal blood vessels of control group.In conclusion, the impaired placental morphology influenced the normal function of placenta to maintain thegrowth and development of fetus. The decreased placental weightresulted in the decreased of fetal weightandlength.

Abstract

Anxiety is a tension, insecurity and fears that arise because of feeling that something bad will happen, it is one of psychiatric disorder causing a somatization disorder. A common somatization disorder is an epigastric pain. Epigastric pain is subjective physical complaint located at the epigastric region. This study aimed to determine whether there was a correlation between anxiety and epigastric pain symptom on outpatients of Internal Medicine dr. Soebandi hospital. This was an observational analytic study with cross sectional approach. Measurement of anxiety level using questionnaires HARS ( Hamilton Anxiety Rating Scale ) and pain level measurements using questionnaires VAS ( Visual Analog Scale ). Data was obtained using questionnaires by interview technique. The result showed a significant relationship ( p = 0.000 ) between the anxiety and epigastric pain symptom. Analysis of the relationship between the two variables showed a positive correlation 0.688. In conclusion, there was a positive correlation between anxiety-disorders and epigastric pain symptom. Abstract

Periodontal disease has been involved in a variety of systemic disorders and suspected as a potential risk factor for fetal growth restriction. Periodontal pathogenic bacteria may actively regulate embryonic development, implantation and placental trophoblast cell invasion. This study aimed to analyze the role of TNF-α, IL-10 and caspase-3 to maintain fetal growth in Porphyromonasgingivalis-infected pregnant rats. Female rats were infected with live-Porphyromonas gingivalis at concentration of 2x109 cells/ml into subgingival sulcus area of the maxillary first molar before and during pregnancy. They were sacrificed on gestational day (GD)-14 and GD20. The weight and length of placentas and fetuses were evaluated. The expression of TNF-α, IL-10 and caspase-3 in macrophages and trophoblast cells were detected by immunohistochemistry. On GD14, TNF-α (R2=0.416;P=0.000) and IL-10 (R2=0.187;P=0.012) had an important role to increase expression of caspase-3 in the placenta, but only TNF-α (R2=0.393;P=0.000 ) was able to increase the expression of caspase-3 on GD20. TNF-α and caspase-3 also had an important role (P<0.000) to decrease fetal weight, fetal length and placental weight on GD14 and GD20, but it was not the case with IL-10 (P>0.000). The increasing expressions of TNF-α and IL-10 did not only enhance immune protection, but also maintained the trophoblast cells survival by regulating expression of caspase-3. Porphyromonas gingivalis infection in maternal periodontal tissue can lead to decrease in placental weight, fetal weight and fetal length which mediated by increasing expression of TNF-α, IL-10 and caspase-3 in the placenta.

Abstract

COVID-19 can cause serious conditions in both comorbid and elderly patients; hence, they need to be given proper follow-up treatment. This study aimed to determine the association between patient characteristics and disease outcomes of COVID-19 in Jember, Indonesia. This research is an analytic observational study with cross-sectional approach. The sample was 304 patients who were confirmed positive for COVID-19 at the COVID-19 referral hospitals in Jember. This study used medical record data analysed by using both univariate and bivariate tests. Patient characteristics related to the disease outcomes in patients who were positive for COVID-19 in Jember, are as follows: age (p = 0.000), clinical symptoms (p = 0.017), comorbidities (p = 0.000), and complications (p = 0.000). The unrelated variables were sex (p = 0.455), education level (p = 1.000), domicile (p = 1.000), occupation (p = 0.322), and smoking history (p = 0.147). Patients most at risk of developing fatal outcomes are those with complications. Patient characteristics associated with the disease outcomes in patients positive for COVID-19 were age, clinical symptoms, comorbidities, and complications. While, those that were not related to the disease outcomes in patients were sex, education level, occupation, domicile, and smoking history.

Abstract

Chronic exposure to insecticides, even at low levels, has led to chronic neurotoxicity. Adolescent brain is still undergoing important developments, including in the hippocampus. This study investigated neurotoxicity effects of subacute exposure to chlorpyrifos, carbofuran and cypermethrin in the brain of adolescent rats. This study was performed at the Histology, Biochemistry, and Animal House Laboratory of Jember University from November to December 2021. Subjects were divided into five groups: normal, control, chlorpyrifos, carbofuran, and cypermethrin. Short-term memory was evaluated by Y maze test and tissue damage was evaluated by histological examination. Brain MDA levels were determined by thiobarbituric acid reactive substance method to evaluate the oxidative stress effect on the organ. This study showed spontaneous alternation in the Y maze test and the number of pyramidal neurons in hippocampus decreased in the cypermethrin group. There was a significant elevation of brain MDA level in carbofuran and cypermethrin groups. Thus, cypermethrin causes more severe neurotoxicity than carbofuran and chlorpyrifos. Improvements in the insecticide application management are urgently needed to prevent neurotoxicity.
