Abstract

ABSTRAK Kandungan metabolit sekunder tanaman yang berkhasiat obat sebagai acuan pemanfaatan untuk membuat sebuah produk, Dewandaru (Eugenia uniflora L), merupakan salah satu tanaman daerah tropis yang belum banyak di gali dan di manfaatkan secara ilmiah. Tujuan penelitian ini  mengetahui metabolit sekunder, uji mutu fisik sediaan effervescent dan uji antioksidan ekstrak buah buah Dewandaru. Metode penelitian eksperimenal ini menujukkan bahwa hasil metabolit sekunder ekstrak n- butanol buah Dewandaru menunjukkan adanya senyawa alkaloid, tannin, flavonoid, dan saponin, hasil uji mutu fisik sdiaan effervescent menunjukkan mutu fisik yang baik di semua jenis pengujian, Pada pengujian pH, sediaan memiliki pH 6. Dari hasil penelitian yang telah dilakukan maka dapat disimpulkan bahwa sediaan granul effervescent memiliki mutu fisik yang baik, uji antioksidan formula effervescent mendapatkan hasil 21,196 ppm.  Termasuk antiosidan yang sangat kuat. Kata Kunci: Antioksidan, Efervescent, Mutu Fisik, Skrining  ABSTRACT The content of secondary metabolites of plants that have medicinal properties as a reference for use to make a product, Dewandaru (Eugenia uniflora L), is one of the tropical plants that has not been widely explored and used scientifically. The purpose of this study was to determine secondary metabolites, test the physical quality of effervescent preparations and test the antioxidants of Dewandaru fruit extracts. This experimental research method showed that the results of secondary metabolites of the n-butanol extract of Dewandaru fruit showed the presence of alkaloids, tannins, flavonoids, and saponins, the results of the physical quality test of effervescent preparations showed good physical quality in all types of tests. 5. From the results of the research that has been done, it can be concluded that the effervescent granule preparation has good physical quality, the antioxidant test of the effervescent formula obtained 21.196 ppm results. Abstract

Indonesia is a country with abundant natural wealth. Almost all kinds of plants can grow in this country. Most have been used since ancient times to treat various diseases. One of the efficacious plants used as medicine is the Dewandaru fruit (Eugenia uniflora L.). This study aims to identify the presence of flavonoid compounds in the Dewandaru fruit (Eugenia uniflora L.) and test the in vitro activity of the ethanol extract of the Dewandaru fruit. Dewandaru fruit extract (Eugenia uniflora L.) was prepared using the maceration extraction method with 96% ethanol solvent. The extraction process was carried out three times. The extract obtained was concentrated in a rotary evaporator at 40-45 °C and then dried in an oven at 40°C. The extracts obtained were identified by means of ultraviolet visible spectrophotometry thin-layer chromatography. obtained showed a positive reaction in the pre-test of flavonoids seen from the separation of red and from the separation of orange stains on thin-layer chromatography. The results of the in vitro test for the antioxidant activity of the ethanol extract of Dewandaru fruit showed an IC50 activity of 53.44 ppm.

Abstract

Diabetes mellitus is a metabolic disease characterized by hyperglycemia resulting from impaired insulin secretion. Dewandaru fruit (Eugenia uniflora L.) contains flavonoids which act as free radical scavengers and reduce oxidative stress. This study aims to prove that the administration of ethanol extract of dewadaru fruit at doses of 200 mg/KgBW and 400 mg/KgB can reduce blood glucose levels and repair damage to pancreatic β cells in alloxan-induced male white mice (Rattus norvegicus) wistar strain diabetes mellitus. This study used 32 mice aged 2-3 months weighing 25 grams intraperitoneally induced by alloxan (0.5 ml/kg). Mice were divided into four treatment groups, namely K- (alloxan), K+ (glibenclamide), P1 (dose of dewadaru fruit extract 200 mg/kgBB), P2 (dose of dewadaru fruit extract 400 mg/kgBB) Quantitative data by calculating blood glucose levels and the number of pancreatic β cells were analyzed using Paired T-Test. P2 is the dose of dewadaru fruit extract 400 mg/kg BW. In the repair of pancreatic β cells it was stated that the average damage to pancreatic β cells at a dose of 200 mg/KgBB was 67.5 and at a dose of 400 mg/KgBW was 89.1. This study proves that the ethanol extract of dewadaru fruit is able to lower blood glucose and inhibit the rate of damage to pancreatic β cells in mice with diabetes mellitus.
