Abstract

Nitrogen mono oksida (NO) dikenal juga dengan Endothelial Derrivat Relaxing Factors) merupakan molekul sederhana yang berperan dalam regulasi sistem kardiovaskular. Nitrogen mono oksida yang dihasilkan oleh NOS type III didalam endotel akan berdiffusi kedalam sel otot polos pembuluh darah yang akan mengaktifkan enzim guanylate cyclase menyebabkan terjadi relaksasi dari otot polos pembuluh darah. Nitrogen mono oksida yang dihasilkan dari metabolisme obat antiangina menyebabkan vasodilasi pembuluh darah koroner yang mengalami aterosklerosis dan memperbaiki aliran darah koroner jantung. Pada penelitian ini diteliti pengaruh ekstrak gambir terhadap kadar EDRF (Endothelial Derrivat Relaxing Factors)/Nitrogen Mono Oksida (NO) pada keadaan hiperkolesterol. Hiperkolesterol di induksi dengan koktail aterogenik. Koktail atrogenik terdidri dari Lemak sapi : Miyak sawit (1:5) PTU 0,1 %, empedu ayam 2 %. Ekstrak gambir diberikan dengan dosis 10, 20 dan 40 mg/Kg BB selama 2 bulan bersamaan dengan koktail yang diberikan secara oral. Kadar NO ditentukan dengan metoda ELISA Spektrofotometer Ukur Optikal Densiti nya pada λ 550 nm. Hasil penelitian menunjukan ekstrak gambir dengan dosis 10 dan 20 mg/Kg BB dapat meningkatkan kadar NO, 20 mg/Kg BB merupakan dosis terbaik untuk. Dosis 40 mg/Kg BB tidak dapat meningkatkan NO.

Abstract

Asam urat merupakan produk akhir metabolisme purin pada manusia dan ketidakseimbangan kadar asam urat di dalam darah menjadi pencetus hiperurisemia. Insiden dan prevalensinya cukup tinggi di Indonesia. Dalam penelitian ini sudah dilakukan untuk mengkaji pengaruh pemberian hasil fraksinasi daun lado-lado (Litsea cubeba,Pers) terhadap kadar asam urat dalam serum darah mencit putih jantan hiperurisemia. Penyarian simplisia dilakukan dengan etanol 70% dan fraksinasi ekstrak dilakukan dengan menggunakan pelarut heksan, etil asetat dan air. Peningkatan kadar asam urat pada hewan diinduksi dengan Potasium oksonat 250 mg/kgBB dan jus kacang panjang 0,5 mL/20gramBB. Pada uji pendahuluan, hasil fraksinasi diberikan secara oral selama seminggu dengan dosis 25 mg/kgBB. Kelompok yang memberikan penurunan kadar asam urat tertinggi diuji lagi dengan memvariasikan dosis fraksi terpilih 12,5 mg/kgBB, 25 mg/kgBB dan 50 mg/kgBB yang diberikan secara oral selama 7, 14 dan 21 hari. Hasil penelitian menunjukan bahwa kelompok yang memberikan penurunan kadar asam urat serum tertinggi adalah fraksi etil asetat. Terlihat peningkatan yang nyata (P ≤0.05). Pemberian dosis 50 mg/kgBB dengan lama pemberian 7 hari memberikan hasil terbaik (P ≤0.05) pada kelompok mencit hiperurisemia.

Abstract

The protective effects of isolated compounds of ethylacetate fraction of surian's leaf (Toona sureni BL Merr) towards endothelial cell dysfunction on hypercholesterolemia have been investigated. Hypercholesterolemia was induced by atherogenic cocktail for the duration of 60 days. The protective effect was determined by Endothelia Derivate Relaxing Factor 9EDRF/NO) concentration. The result showed that isolated compounds oe ethyl acetate fraction of surian's leaf was able to protect the endothelia cell damage in hypercholesterolemia that can be seenn by increasing the bioavailability of NO serum (P<0.05).Keywords : flavonoid, cocktail, hypercholesterol and nitrogen mono oxide (EDRF)

Abstract

Research on test teratogenic effects of ethanol  extract of leaves of the soursop (Annona muricata L.) have been conducted on white mice fetus. Animal experiments are grouped into  4 group consisting of Group I as a negative control only granted suspension of Na CMC 0.5%, Group II, Group III and Group IV granted soursop leaf ethanol extract at a dose of 250 mg / kg, 500 mg / kg, and 1000 mg / kg once daily from day 6 to day 15 of pregnancy. The 18th day of pregnancy mice were dissected. Results of the study at a dose of 250 mg / kg, a dose of 500 mg / kg, a dose of 1000 mg / kg give teratogen effects. A dose of 250 mg / kg, a dose of 500 mg / kg give teratogen effect with the discovery of skeletal abnormalities. A dose of 1000 mg / kg give teratogen effect with the discovery of late growth. Results showed that the ethanol extract of the leaves of the soursop affect weight loss parent mice were significantly (p <0.05) when compared to the negative control. The quantity of fetuses decreased significantly (p <0.05) when compared to the negative control. Fetal body weight showed a significant effect (p <0.05) when compared to the negative control.

Abstract

The stimulant activity of central nerve systems of areca seed (Areca cathecu, L.), at white mouse ( Mus musculus, L ) had been done by using method hanging test, evasi test, swim test and discrimination maze. Drug dose given is 0.5 mg/20 gram BW white mouse, 1 mg/20 gram BW white mouse, 2 mg/ 20 gram BW white mouse in oral mechanism. As negative control is applied by Tween 80 2% and condensation of comparator is applied by Caffeine.Activity test result from seed extract Pinang ( Areca cathecu, L. ) to white mouse ( Mus musculus, L. ) the obtained result signifikan ( P < 0.01 ) as stimulant central nerve systems.

Abstract

Study about the influence of agarwood leave’s extract (Aquilaria malaccensis Lam.) on the formation of atherosclerosis in male quail (coturnic-coturnix japonica) induced high fat feeding and prophylthiourasil. Animal experiments were grouped into 5 consisting of Group I as a negative control only given food strander, group II as a positive control and PTU given MLT groups III, IV, V given leaf extract of aloes with a dose of 100 mg / kg BW, 300 mg / kg BW, 900 mg / kg BW and, along with the MLT and PTU the extract was given orally once a day for 60 day. The doses used were 100, 300, 900 mg/kg body weight respectively. Control groups was given food standar. Result showed that the administration of the agarwood leaf extract could prevent formation of atherosclerosis in male quail significantly (P <  0.05). The optimal effect showed doses 100 mg/kg BW.

Abstract

Secara empiris, daun sirih hijau banyak digunakan sebagai obat hepatitis, bronkhitis, dan antiseptik. Zat-zat yang terkandung didalamnya mampu mengatasi peradangan. Penelitian ini bertujuan untuk melihat aktivitas hepatoprotektor dari daun sirih hijau terhadap kerusakan hati yang diinduksi dengan parasetamol pada mencit putih jantan. Parameter yang diamati yaitu aktivitas SGPT dan SGOT. Hewan percobaan dibagi dalam 5 kelompok yang terdiri dari kelompok kontrol negatif, kelompok kontrol positif (parasetamol 500 mg/KgBB), kelompok dosis 50 mg/kgBB, 100 mg/kgBB dan 200 mg/kg BB. Pemeriksaan kadar SGOT, SGPT  pada hari ke 3, 7, 15 dan pemeriksaan histopatologi hati untuk melihat gambaran kerusakan hati. Hasil penelitian menunjukkan adanya penurunan aktivitas SGOT dan SGPT. Penurunan tertinggi  aktivitas SGOT dan SGPT diperlihatkan pada dosis 200 mg/kgBB pada pemeriksaan hari ke-15 sebesar 13,57 U/L dan 14,15 U/L. Pemeriksaan histopatologi menunjukkan perbaikan kerusakan jaringan hati yang terlihat dari penurunan nekrosis sentrolobular. Dapat disimpulkan bahwa ekstrak daun sirih hijau (Piper betle Linn ) memiliki aktivitas hepatoprotektor.

Abstract

Has done research the effect of extracts of carrot (Daucus carota Linn.) Against atherosclerosis in male quail (Cortunix-cortunix japonica). Atherosclerosis induced by administering Prophylthiouracyl (PTU) and high-fat foods (MLT). Carrot extract dose given on quail is 50 mg / kg BW, 100 mg / kg BW and 200 mg / Kg BW on quail males given orally for 60 Days. The results showed that the administration of carrot extract at a dose of 100 mg / kg BW and 200 mg / kg BW can prevent the formation of atherosclerosis in male quail, marked on the wall thickness of the aorta, the aortic lumen and extensive damage to endothelial cells was not different from the negative control (P < 0.05). Carrot extract dose of 50 mg / kg BW for 60 days can not be pried prevent its established atherosclerosis, characterized in aortic wall thickness, area of the aortic lumen and endothelial cell damage is not different from the positive control (P> 0.05).

Abstract

Has done research about the teratogenic effects ethanol of extract test patikan Kebo leaves (Euphorbia hirta L.) concerning to fetal white mice. Animal experiments were grouped into 4 groups consisting of group I as  negative control given only 0.5% suspension of Na CMC, group II, group III and group IV was given the ethanol extract of patikan kebo leaves with variations dose of 25 mg / kg BW, 50 mg / kg BW, and 100 mg / kg BW once a day from day 6 to day 15 of pregnancy. 18th day of pregnancy mice dilaparaktomi. Dose of 25 mg / kg BW, dose of 50 mg / kg BW did not cause teratogenic effects. Teratogenic effect was found at dose of 100 mg/kg BW there was the discovery of defects cleft palate. The results showed the ethanol extract of patikan kebo leaves not affected weight gain parent mice were significantly (p> 0.05) when compared to the negative control. Amount of fetuses were significantly reduced (p <0.05) when compared to the negative control. Fetal weight showed no significant effect (p> 0.05) if compared with negative controls.
