Abstract

Andrographis paniculata (Burm. F.) Nees (AP) dikenal karena rasanya yang pahit dan sudah umum digunakan sebagai obat tradisional. Studi fitokimia AP menunjukkan bahwa andrografolid adalah senyawa bioaktif utama. Berdasarkan penelitian, andrografolid memiliki efek sebagai antikanker dengan berbagai mekanisme. WHO telah memperkenalkan teknik kromatografi sidik jari (Fingerprint) yang dapat digunakan untuk penilaian keamanan dan kualitas obat herbal. Tujuan dari penelitian ini adalah memprediksi efek antikanker AP menggunakan analisis Brine Shrimp Lethality Test dan MLR dengan beberapa prediktor seperti AUC dari kromatogram sidik jari, ketinggian geografis, dan IC50. Desain penelitian ini adalah eksperimental laboratorik menggunakan AP dari daerah berbeda yaitu Tawangmangu (Twg), Bandung (Bdg), Denpasar (Dps). Hasil BSLT menunjukkan bahwa AP dari Twg memiliki nilai toksisitas lebih tinggi dari Bdg dan Dps dengan LC50 5,08 mg/L. AUC kromatogram AP dan ketinggian geografis menunjukkan korelasi yang kuat terhadap potensi efek sitotoksik antikanker berdasarkan LC50 dengan nilai R2 0,984 dan dengan nilai p <0,05. Hal ini menunjukkan bahwa metode analisis MLR dapat digunakan untuk memprediksi respon farmakologi AP sitotoksik (LC50) terhadap beberapa prediktor seperti AUC, LC50, dan ketinggian geografis.

Abstract

ABSTRAK Corona Virus Disease 19 atau lebih dikenal dengan COVID-19 merupakan wabah yang melanda dunia. COVID-19 menimbulkan kedaruratan kesehatan diberbagai negara salah satunya Indonesia. Upaya pencegahan yang dapat dilakukan adalah dengan meningkatkan ketahanan masyarakat, melalui kesehatan tubuh perorangan salah satunya dengan penggunaan suplemen. Menurut WHO, remaja adalah penduduk dalam rentang usia 10-19 tahun, remaja memiliki risiko yang lebih tinggi terkena COVID-19 dibandingkan orang tua. Remaja memiliki aktivitas diluar rumah yang cukup tinggi sehingga rentan terkena COVID-19. Upaya pencegahan penularan COVID-19 pada Siswa SMK Bintang Persada tidak lepas dari peningkatan imun tubuh salah satunya dengan penggunaan suplemen. Tujuan kegiatan ini adalah meningkatkan pengetahuan siswa-siswi SMK tentang suplemen kesehatan, mitra dapat mengetahui informasi tentang vitamin C, dan mengenal upaya dalam menghadapi COVID-19 pada siswa SMK Bintang Persada. Metode yang diguanakan yaitu peserta yang mengikuti kegiatan ini sebanyak 67 siswa-siswi dengan metode pretest dan posttest. Hasil yang didapatkan yaitu terjadi peningkatan pada nilai pretest (sebelum diberikan edukasi) yaitu sebesar 69,41% menjadi 89,42% pada hasil postest (setelah diberikan edukasi). Simpulan dan saran Terdapat hubungan bermakna pada nilai postest yaitu meningkat setelah diadakannya edukasi, sehingga edukasi penggunaan suplemen pencegahan COVID-19 perlu diberikan dan ditingkatkan untuk meningkatkan perilaku pencegahan COVID-19 pada siswa SMK Bintang Persada. Kata Kunci: Edukasi, Suplemen, COVID-19  ABSTRACT Corona Virus Disease 19 or better known as COVID-19 is an outbreak that has swept the world. COVID-19 caused health emergencies in various countries, one of which was Indonesia. Prevention efforts that can be done are by increasing community resilience, through individual body health, one of which is the use of supplements. According to WHO, adolescents are residents in the age range of 10-19 years, adolescents have a higher risk of developing COVID-19 than parents. Teenagers have high enough activities outside the home so they are vulnerable to COVID-19. Efforts to prevent COVID-19 transmission in students of SMK Bintang Persada cannot be separated from increasing the body's immune system, one of which is the use of supplements. With this community service activity, SMK students can understand about the use of health supplements in an effort to deal with COVID-19. Activity is to increase the knowledge of SMK students about health supplements, partners can find out information about vitamin C, and get to know the efforts in dealing with COVID-19 in students of SMK Bintang Persada. Participants who took part in this activity was 67 students with the pretest and postest methods. The results obtained were an increase in the pretest value (before being given education) by 69.41% to 89.42% in the postest results (after being given education). There is a meaningful relationship in the postest value, which is to increase after the holding of education, so that education on the use of COVID-19 prevention supplements needs to be given and improved to improve COVID-19 prevention behavior in students of SMK Bintang Persada. Keywords: Education, Supplements, COVID-19

Abstract

Cancer is a disease caused by abnormal growth of body tissue cells, characterized by uncontrolled cell growth. Sambiloto (Andrographis paniculata (Burm.f.) Nees) is known to have anti-cancer activity. One of the preliminary test methods used to determine the potential of sambiloto as an anticancer is the acute toxicity test. The acute toxicity test used was the Brine Shrimp Lethality Test (BSLT). The purpose of this study was to determine the value of LC50% (Lethal Concentration) as the cytotoxic activity of the n-hexane fraction, ethyl acetate fraction and purified extract of bitter herbs. The method used is maceration with 96% ethanol followed by fractionation and purification with n-hexane, ethyl acetate, and water as solvents. Cytotoxic activity test with BSLT using concentration variations of 0, 10, 50, 100, 500, 1000 ppm. The results showed cytotoxic activity based on LC50 values ??for the n-hexane, ethyl acetate, and purified extract fractions, namely 19.9x1012 ppm, 5261,5 ppm, and 83,8 ppm. The results showed that the purified extract of sambiloto herb was very toxic with an LC50 value of 1000 ppm, namely 83.799ppm (p<0.01).

Abstract

Aterosklerosis adalah proses inflamasi kronis akumulasi lipid pada tunika intima pembuluh arteri. Konsentrasi LDL dalam darah yang tinggi menyebabkan modifikasi oksidasi. Protein kinase C- ?II memiliki peran terhadap penyerapan LDL. Sambiloto dengan senyawa mayor andrografolid, neoandrografolid dan deoxyanrografolid memiliki berbagai aktivitas farmakologi salah satunya sebagai anti terosklerosis. Tujuan penelitian ini adalah Untuk mengetahui energi ikatan dan perbandingan ikatan andrografolid, neoandrografolid dan deoxy-andrografolid yang berpotensi sebagai anti aterosklerosis pada protein kinase C (PKC)-?II dengan metode in silico. Jenis penelitian ini yaitu eksploratif secara in-silico terhadap protein target yaitu protein kinase C (PKC)-?II (PDB ID : 2i0e) proses preparasi senyawa, preparasi protein, validasi metode molecular docking hingga docking senyawa andrografolid, neoandrografolid dan deoksi-andrografolid pada protein target. Ketiga senyawa uji memiliki afinitas pada protein target dengan nilai energi ikatan -9,53 dengan neoandrografolid, -8,84 dengan andrografolid, -9,05 dengan neoandrografolid.Pada hasil docking, dapat diketahui bahwa andrografolid, neoandrografolid, dan deoksi-andrografolid memiliki aktivitas anti aterosklerosis secara in silico melalui penghambatan protein kinase C (PKC)-?II. Dari ketiga senyawa uji yaitu andrografolid, neoandrografolid dan deoksi-andrografolid berpotensi sebagai anti arterosklerosis melalui penghambatan protein kinase C (PKC)-?II

Abstract

The focus is how the role of cytokines in atherosclerosis as a chronic inflammatory disease, as it is known that cytokines regulate complex inflammatory responses in atherosclerotic plaques, especially IL-1β. Sambiloto (Andrographis paniculata) is a plant with various pharmacological activities. Andrographolide (AND) is the main bioactive compound in the diterpene lactone group. This literature study aims to assess the potential of AND on the proinflammatory cytokine IL-1 β as an antiatherosclerosis. This type of research is a qualitative review of various studies using sambiloto plants or andrographolide compounds (AND). Search literature using the Google Scholar database. Keywords used "Andrographis paniculata, andrographolide, IL-1β, Atherosclerosis. The results showed that AND has effects as an anti-inflammatory, antiatherosclerosis as well as cardiovascular-related diseases. There are not many articles on the activity of AND in IL-1β against atherosclerosis, but AND can provide inhibitory activity of the cytokine IL-1β against inflammation. Not many researchers have discussed the pharmacological effects of AND as an antiatherosclerotic on IL-1β therapeutic targets, but from the results of the review AND has a strong potential in inhibiting IL-1 cytokine secretion β.

Abstract

Spotting volume is an important factor affecting the resolution value of the chromatogram, the number of peaks and separation, peak profile and peak broadening, as well as the asymmetry of the peaks. This study aims to determine the effect of bottling volume on fingerprint chromatography of the methanol extract of Andrographis paniculata (Burm. F.) Nees herb and to determine fingerprint chromatography at optimum volume. The chromatographic system used refers to the Indonesian Herbal Pharmacopoeia. Sambiloto herbal samples were spotted with a variety of 3 µL bottling volumes; 5µL; 10µL; 15µL; 20 µL and 25 µL on a 60 GF254 silica gel plate were then eluted with a mixture of chloroform P and methanol P (9:1) v/v. The plates were scanned with a TLC Scanner 3 (CAMAG) at a wavelength of 210 nm. Spectra were made for each spot at a wavelength of 190-400 nm. Parameters that were calculated were resolution values, tailing factor from andrographolid peaks, and cosine functions (C) from volume variation chromatograms to determine the effect of spotting volume. The bottling volume selected from the optimization step was 5 µL with andrographolid peak resolution with the previous and following peaks being 1.57 and 1.16, respectively. The tailing factor value was 1 and the number of peaks that appeared was 10. The average value of C in the 5 µL bottling volume precision test was 99.36 with a VC of 1.17% (VC <2%)

Abstract

Background: Red betel leaf (Piper crocatum Ruiz and Pav.) is one of the medicinal plants which empirically has the efficacy of healing spiritual ailments. The secondary metabolite compounds contained in red betel leaves are essential oils, flavonoids, alkaloids, saponins, tannins, terpenoids, and phenolic compounds which have antibacterial activity. Objective: To determine the antibacterial potential of red betel leaves (Piper crocatum Ruiz and Pav.) Method: This study uses a descriptive method using a structured article review method. Data collection was carried out using the Google Scholar database, Science Direct and PubMed with keywords “Piper crocatum Ruiz and Pav”, “Piper crocatum”, “Screening phytochemical”, Piper crocatum”, “Leaf Piper crocatum”, and “antibacterial Piper crocatum”. Results: Based on literature studies, the antibacterial potential of red betel leaves (Piper crocatum Ruiz and Pav.) can be seen from the inhibition zones produced for both gram-positive and gram-negative bacteria, although with different values of the resulting inhibition zones.

Abstract

Obat herbal diyakini memiliki efek samping yang sedikit, biaya yang relatif murah dan aksesibilitas tinggi. Di negara-negara Asia saat ini, sekitar 25% dari pengobatan mengandung herbal, dan proporsi ini meningkat menjadi 30% dan hingga 50% khususnya di Cina Andrographis paniculata (Nees) atau sambiloto termasuk keluarga Acanthaceae yang sebagian besar tersebar di daerah tropis Negara-negara Asia seperti India, Indonesia, Sri Lanka, Pakistan, dan Malaysia. Sambiloto mengandung senyawa mayor diterpenoid lakton yaitu andrografolid. Senyawa andrografolid dilaporkan memiliki peran pada aktivitas anti inflamasi. Seiring berjalannya waktu, obat-obatan herbal digunakan sebagai pengobatan alternatif (CAM) dengan atau tanpa sepengetahuan praktisi medis. konsumsi bersamaan dapat menyebabkan potensi interaksi (herb-drug interaction) yang menyebabkan efek sinergis atau antagonis. Penggunaannya pun sering bersamaan atau berjarak waktu yang dekat dengan obat-obatan anti inflamasi sintetik. Sehingga Penting untuk diketahui potensi interaksi herbal-obat antara andrografolid sebagai agen anti inflamasi dengan obat-obatan anti inflamasi sintetik non steroid terhadap profil farmakokinetika. Interaksi farmakokinetik melibatkan penyerapan, distribusi, metabolisme dan ekskresi, semuanya terkait dengan kegagalan pengobatan atau terjadinya toksisitas. Beberapa penelitian telah dilaporkan terkait interaksi sambiloto (senyawa andrografolid) dengan beberapa obat anti inflmasi non steroid seperti ibuprofen, naproxen, dan meloxicam. Parameter farmakokinetika yang umum dipengearuhi terhadap pemberian herbal dan obat antiinflmasi non steroid yaitu bioavailibilitas, distribusi jaringan, waktu paruh (t1 / 2), konsentrasi plasma maksimum (Cmaks), dan waktu untuk mencapai Cmaks (Tmaks), AUC, clearance (CL). Perubahan parameter farmakokinetika menyebabkan variasi pada efek terapeutiknya. Kata Kunci: Interaksi Farmakokinetika, Andrografolid, NSAID

Abstract

Medicinal plants are one of the efforts used to control disease. The benefits of medicinal plants have been passed down from generation to generation by our ancestors. One plant that has medicinal properties is miana leaves (Plectranthus scutellaroides R.Br). In this research, the extraction method used was maceration, with 96% ethanol solvent. Miana leaf ethanol extract was screened for phytochemicals and toxicity tested using the Brine Shrimp Lethality Test (BSLT) method. Toxicity tests were carried out at various concentrations of 100 ppm, 250 ppm, 500 ppm, 750 ppm and 1000 ppm. The results of phytochemical screening on miana leaves show that miana leaves contain flavonoid compounds, tannins, saponins, steroids, but do not contain alkaloids and triterpenoids. The 96% ethanol extract of miana leaves (Plectranthus scutellaroides R.Br) has an LC50 value of 511.58 ppm, included in the toxic category with an LC50 value in the range of 30-1000 ppm.
