Abstract

Cancer is a disease caused by abnormal growth of body tissue cells, characterized by uncontrolled cell growth. Sambiloto (Andrographis paniculata (Burm.f.) Nees) is known to have anti-cancer activity. One of the preliminary test methods used to determine the potential of sambiloto as an anticancer is the acute toxicity test. The acute toxicity test used was the Brine Shrimp Lethality Test (BSLT). The purpose of this study was to determine the value of LC50% (Lethal Concentration) as the cytotoxic activity of the n-hexane fraction, ethyl acetate fraction and purified extract of bitter herbs. The method used is maceration with 96% ethanol followed by fractionation and purification with n-hexane, ethyl acetate, and water as solvents. Cytotoxic activity test with BSLT using concentration variations of 0, 10, 50, 100, 500, 1000 ppm. The results showed cytotoxic activity based on LC50 values ??for the n-hexane, ethyl acetate, and purified extract fractions, namely 19.9x1012 ppm, 5261,5 ppm, and 83,8 ppm. The results showed that the purified extract of sambiloto herb was very toxic with an LC50 value of 1000 ppm, namely 83.799ppm (p<0.01).

Abstract

Aterosklerosis adalah proses inflamasi kronis akumulasi lipid pada tunika intima pembuluh arteri. Konsentrasi LDL dalam darah yang tinggi menyebabkan modifikasi oksidasi. Protein kinase C- ?II memiliki peran terhadap penyerapan LDL. Sambiloto dengan senyawa mayor andrografolid, neoandrografolid dan deoxyanrografolid memiliki berbagai aktivitas farmakologi salah satunya sebagai anti terosklerosis. Tujuan penelitian ini adalah Untuk mengetahui energi ikatan dan perbandingan ikatan andrografolid, neoandrografolid dan deoxy-andrografolid yang berpotensi sebagai anti aterosklerosis pada protein kinase C (PKC)-?II dengan metode in silico. Jenis penelitian ini yaitu eksploratif secara in-silico terhadap protein target yaitu protein kinase C (PKC)-?II (PDB ID : 2i0e) proses preparasi senyawa, preparasi protein, validasi metode molecular docking hingga docking senyawa andrografolid, neoandrografolid dan deoksi-andrografolid pada protein target. Ketiga senyawa uji memiliki afinitas pada protein target dengan nilai energi ikatan -9,53 dengan neoandrografolid, -8,84 dengan andrografolid, -9,05 dengan neoandrografolid.Pada hasil docking, dapat diketahui bahwa andrografolid, neoandrografolid, dan deoksi-andrografolid memiliki aktivitas anti aterosklerosis secara in silico melalui penghambatan protein kinase C (PKC)-?II. Dari ketiga senyawa uji yaitu andrografolid, neoandrografolid dan deoksi-andrografolid berpotensi sebagai anti arterosklerosis melalui penghambatan protein kinase C (PKC)-?II

Abstract

Spotting volume is an important factor affecting the resolution value of the chromatogram, the number of peaks and separation, peak profile and peak broadening, as well as the asymmetry of the peaks. This study aims to determine the effect of bottling volume on fingerprint chromatography of the methanol extract of Andrographis paniculata (Burm. F.) Nees herb and to determine fingerprint chromatography at optimum volume. The chromatographic system used refers to the Indonesian Herbal Pharmacopoeia. Sambiloto herbal samples were spotted with a variety of 3 µL bottling volumes; 5µL; 10µL; 15µL; 20 µL and 25 µL on a 60 GF254 silica gel plate were then eluted with a mixture of chloroform P and methanol P (9:1) v/v. The plates were scanned with a TLC Scanner 3 (CAMAG) at a wavelength of 210 nm. Spectra were made for each spot at a wavelength of 190-400 nm. Parameters that were calculated were resolution values, tailing factor from andrographolid peaks, and cosine functions (C) from volume variation chromatograms to determine the effect of spotting volume. The bottling volume selected from the optimization step was 5 µL with andrographolid peak resolution with the previous and following peaks being 1.57 and 1.16, respectively. The tailing factor value was 1 and the number of peaks that appeared was 10. The average value of C in the 5 µL bottling volume precision test was 99.36 with a VC of 1.17% (VC <2%)

Abstract

Background: Red betel leaf (Piper crocatum Ruiz and Pav.) is one of the medicinal plants which empirically has the efficacy of healing spiritual ailments. The secondary metabolite compounds contained in red betel leaves are essential oils, flavonoids, alkaloids, saponins, tannins, terpenoids, and phenolic compounds which have antibacterial activity. Objective: To determine the antibacterial potential of red betel leaves (Piper crocatum Ruiz and Pav.) Method: This study uses a descriptive method using a structured article review method. Data collection was carried out using the Google Scholar database, Science Direct and PubMed with keywords “Piper crocatum Ruiz and Pav”, “Piper crocatum”, “Screening phytochemical”, Piper crocatum”, “Leaf Piper crocatum”, and “antibacterial Piper crocatum”. Results: Based on literature studies, the antibacterial potential of red betel leaves (Piper crocatum Ruiz and Pav.) can be seen from the inhibition zones produced for both gram-positive and gram-negative bacteria, although with different values of the resulting inhibition zones.

Abstract

Medicinal plants are one of the efforts used to control disease. The benefits of medicinal plants have been passed down from generation to generation by our ancestors. One plant that has medicinal properties is miana leaves (Plectranthus scutellaroides R.Br). In this research, the extraction method used was maceration, with 96% ethanol solvent. Miana leaf ethanol extract was screened for phytochemicals and toxicity tested using the Brine Shrimp Lethality Test (BSLT) method. Toxicity tests were carried out at various concentrations of 100 ppm, 250 ppm, 500 ppm, 750 ppm and 1000 ppm. The results of phytochemical screening on miana leaves show that miana leaves contain flavonoid compounds, tannins, saponins, steroids, but do not contain alkaloids and triterpenoids. The 96% ethanol extract of miana leaves (Plectranthus scutellaroides R.Br) has an LC50 value of 511.58 ppm, included in the toxic category with an LC50 value in the range of 30-1000 ppm.
