Abstract

This research aimed to identify types of bacteria existed in nursery rearing pond of catfish (Clarias gariepinus) using closed water system. The observed data include bacterial abundance and diversity in the water and in the fish digestion system, fish growth and water quality. This research was performed in the media of 3 concrete pools with the size of 3 x 1 x 0.8 m3 containing fish stock spreading of 3000 tail / pond. The obtained results showed that the highest abundance of bacteria was found in the surface and the bottom of pond C with the abundance number of   3,0 x 104and 1,2 x 103for aerobic and anaerobic bacteria, respectively. Moreover, it was figured that the dominant bacterial species found in the water and the gastrointestinal tract were from the genus of Bacillus, Clostridium and Lactobacillus. Meanwhile, water quality in the pond was observed in the normal range with the temperatures of 28 - 30 oC, pH (6.0 - 7.0), DO (4.5 - 5.7 ppm), ammonia (0.06 - 0.03 ppm), nitrite (0 ppm) and nitrate (0 - 5 ppm). Keywords: Type of bacteria, catfish (Clarias gariepinus), Closed water system.

Abstract

One disease that often attacts fish is a group of parasites or parasitic diseases, parasites are not only dangerous because they are related to their activities, but also trigger the emergence of other pathogenic organisms and cause infections. Therefore, it is necessary to take fish quarantine measures to prevent the spread of diseases carried out by fish quarantine centers. This study was conducted to determine the type of parasite that infested fish samples and determine the prevalence and intensity of fish samples. A total of 48 sample fish consisting of freshwater and seawater fish were analyzed in the laboratory of Balai Karantina Ikan, Pengendalian Mutu dan Keamanan Hasil Perikanan Surabaya II during December - January 2019, either directly or directly (ectoparasites and endoparasites). Parasitic identification was carried out visually and microscopically with an approach based on identification books. The types of parasites that we found during the study consisted of Argulus sp, Trichodina sp, Anisakis sp., Gnathostoma spinigerum, and Gnathostoma spinigerum. The highest prevalence in Scomber Scombrus with Anisakis sp. Amounting  to 77.8%, mean while, the lowest prevalence and the intensity is found in Thunnus sp. The prevalence and intensity of parasites are influenced by water quality and pollution.

Abstract

Detriment caused by ectoparasitic infestation was not as big detriment caused by infestation of other organisms such as viruses and bacteria, but it could be a predisposing factor for the infection of more dangerous pathogenic in fish. The aims of this study was to inventory ectoparasites that infecting freshwater fish cultivated in several districts in the Banyuwangi, in which the results were expected to be used as a reference in an effort to prevent the spread of disease in fish culture. The sample of research consisted of tilapia fish, carp and catfish which were obtained from six sub districts in Banyuwangi, those were Kota, Kabat, Licin, Kalipuro, Giri, and Glagah. The ectoparasite examination method was performed naturally by scraping the surface of the body, fins and gills of the fish, then observed under a microscope with magnification ranging from 40 to 400x. The results of further observations were documented and identified by key identification. Ectoparasites which were found infesting freshwater fish in the city of Banyuwangi consisted of 4 genuses: namely Argulus, Lernaea, Chonopeltis and Trichodina. Further research is needed to determine the prevalence and intensity of ectoparasites in order to prevent the spread of ectoparasites in other regions. Keywords: Inventarisation, ectoparasites, freshwater fish, Banyuwangi

Abstract

The study aimed to determine the type of culture container for increasing the cell density of Nitzschia and producing the hightly cell biomass. The stages of this study were sterilization of containers and media, making diatom fertilizers and agar media, culture of Nitzschia in agar medium, culture of Nitzschia in test tube, and culture of Nitzschia in glass and plastic containers. The result showed that the cell density of Nitzschia cultured in the glass container was higher than plastic containers. Further study was needed to determine the nutrient composition of Nitzschia that culture in glass containers.Keyword : Nitzschia culture, Laboratory scale, Glass, Plastic container.

Abstract

The potential of freshwater fish cultivation in Indonesia is very large and is increasing every year. The high potential of freshwater and ornamental fisheries commodities and consumption must be developed through domestic and export marketing. The distribution carries the risk of entering and spreading fish pests and diseases, so is it required identification or quarantine is needed to avoid negative impacts from the distribution of fishery commodities. One of the infectious diseases which is known to attack fishery commodities in Indonesia is bacterial disease. Bacteria can cause high mortality in fish and loss for farmers. The purpose of this study is to identification bacteria in freshwater fish commodities. The study was conducted in December 2018 - January 2019 at the Fish Quarantine Center, Quality Control and Safety of Fisheries Products in Surabaya I. Identification of bacteria in this study using conventional methods. The parameters observed were external clinical symptoms that appeared on the body of the fish and internal pathology. The material examined in this study were The samples examined during Field Work Practices were Goldfish, Catfish fry, Common carp, and Koi fish with a total of 64 samples. This research was observative, then the data obtained were analyzed descriptively. The Result bacteria identified in freshwater fish commodities from 64 samples were dominated by Aeromonas hydrophila, Pseudomonas anguilliseptica, Streptococcus sp., Plesiomonas shigelloides, and Micrococcus sp.Keywords : Freshwater fisheries commodities, Bacteria, The Conventional Method

Abstract

The purpose of this research is to find out the abalone separating technique (H. squamata) and know the problems during separating at the Marine Aquaculture Development Centre, West Nusa Tenggara Province. Research are carried out on December 23, 2019 to January 21, 2020. The method used is descriptive method with data collection including primary data and secondary data. Data is taken by observing, active participation, interviewing, and literature review. Abalone separating techniques (H. squamata) begin with container preparation, seed preparation, seed stocking, seeding, feeding, water quality management, and growth sampling. Problems encountered during the separating stage are the uncertain availability of seaweed feed.Keyword :  Abalone, H. squamata, separating technique.

Abstract

ABSTRACT The aim of this study was to test the effectiveness of a probiotic Bacillus for the prevention of motile aeromonad septicemia (MAS) disease caused by Aeromonas hydrophila in African catfish (Clarias gariepinus). The study consisted of the inhibition testing of A. hydrophila by Bacillus (in vitro) and the application of probiotic in African catfish (in vivo). The in vivo test, consisted of five treatments such as the addition of probiotic Bacillus P4I1 RifR, Bacillus P4I2 RifR, Bacillus P4I1 RifR + Bacillus P4I2 RifR (Kom), positive control (K+; only added with A. hydrophila) and negative control (K-; without probiotic nor A. hydrophila addition). African catfish (13.35±2.80 g) was maintained in 15 aquariums (40 L in volume) with 30 fishes each for 30 days. Probiotic bacteria was applied in water once a day, whereas pathogenic bacteria A. hydrophila RifR (103 cfu/mL) were added once in earlier treatment (except for the negative control). The result showed that the optimal concentration of Bacillus to inhibit A. hydrophila on in vitro test was 104 cfu/mL. In vivo test showed that the addition of probiotic in media of cultivation could reduce the number of A. hydrophila, improve immune response, and also increase the survival of African catfish compared to positive control. Application of probiotic P4I1 RifR showed the highest survival (92.23%) of all treatments. Keywords: Bacillus, Clarias gariepinus, motile aeromonad septicemia, probiotic  ABSTRAK Penelitian ini bertujuan untuk menguji efektivitas probiotik Bacillus dalam pencegahan penyakit motile aeromonad septicaemia (MAS) yang disebabkan oleh Aeromonas hydrophila pada ikan lele dumbo (Clarias gariepinus). Penelitian terdiri atas pengujian penghambatan bakteri probiotik Bacillus terhadap A. hydrophila secara in vitro, dilanjutkan dengan aplikasi pada budidaya ikan lele dumbo (in vivo). Pada uji in vivo, penelitian terdiri atas lima perlakuan yaitu budidaya ikan lele dumbo dengan penambahan probiotik Bacillus P4I1 RifR, Bacillus P4I2 RifR, kombinasi probiotik Bacillus P4I1 RifR + Bacillus P4I2 RifR (Kom), kontrol positif (K+; hanya ditambahkan A. hydrophila) dan kontrol negatif (K-; tanpa pemberian probiotik dan A. hydrophila). Ikan lele dumbo (13,35±2,80 g) dipelihara pada akuarium volume 40 L dengan kepadatan 30 ekor/akuarium selama 30 hari. Bakteri probiotik ditambahkan pada media pemeliharaan ikan setiap hari, sedangkan bakteri patogen A. hydrophila RifR (103 cfu/ mL) diberikan sekali pada awal pemeliharaan (kecuali pada kontrol negatif). Hasil penelitian menunjukkan bahwa konsentrasi terbaik pada penghambatan in vitro adalah dengan penambahan Bacillus 104 cfu/mL. Hasil uji in vivo menunjukkan perlakuan penambahan probiotik pada media budidaya efektif dapat menekan jumlah bakteri A. hydrophila, memperbaiki respons imun, dan meningkatkan kelangsungan hidup ikan lele dumbo dibanding kontrol positif. Perlakuan probiotik P4I1 RifR memberikan hasil terbaik dengan tingkat kelangsungan hidup tertinggi yaitu 92,23%. Kata kunci: Bacillus, Clarias gariepinus, motile aeromonad septicemia, probiotik

Abstract

Diseases caused by parasitic groups get less serious attention from the farmers because it is chronic morbidity and mortality rates and small. However, if observed further, parasitic diseases may trigger secondary infection by bacteria, viruses and fungi that can cause death and high losses. Thus this study aims to identify the types of ectoparasites that infected the seed goldfish in BBI Kabat, Banyuwangi which the results will be used as an early warning to prevent the death of fish in bulk. The research sample in the form of seeds 3-5 cm carp amounted 150 obtained from BBI Kabat, Banyuwangi. Ectoparasites inspection method performed by scraping the surface of the body, fins and gills of fish, and then observed under a microscope with a magnification ranging from 40-400x. The observation of ectoparasites are further identified accordance morphology based on the keys of identification. Identified ectoparasites infect seed goldfish in BBI Kabat, Banyuwangi consists of 8 genus is Zoothamnium sp., Trichodina sp., Oodinium sp., Vorticella sp., Argulus sp., Lernaea sp., Dactylogyrus sp., and Gyrodactylus sp. Further research is needed to determine the prevalence and intensity of parasites and appropriate measures to prevent the spread of parasites in healthy fish in BBI Kabat, Banyuwangi.

Abstract

The purpose of this study was to determine the incidence and degree of infection anisakiasis in fish catches at Fishery Seaport Muncar, Banyuwangi as an effort to prevent food borne diseases in humans. Fish samples from Muncar Beach Fishery Seaport, Banyuwangi consisted of tuna fish (Euthynnus sp.), bloated fish (Rastrelliger sp.), giant trevally fish (Caranx sp.) and snapper (Lutjanus sp.) with total amount 192 tail. Sample fish was dissected and examined the parasite by native method. Parasitic worms was fixed with 5% glycerine alcohol, stained with SemichenAcetic Carmine method, identified by key identification and calculated incidence and degree of infection. The highest incidence and degree of infection showed in Lutjanus sp. and Rastrelliger sp. 66.67% and 5.39, respectively, while the lowest showed in Caranx sp. from 6.67% and 0.09 respectively. Continue research to determine incidence and degree of infection anisakiasis in each season was required to mapping and to know the spread of worms Anisakis sp.

Abstract

AbstrakLamun merupakan tanaman berbiji terbuka yang mampu tumbuh dan beradaptasi dengan lingkungan bersalinitas tinggi serta dapat berasosiasi dengan benthos. Keberadaan lamun di perairan merupakan salah satu indikator tingkat kesuburan dan produktivitas perairan. Terdapat perbedaan dominansi antara musim hujan dan musim kemarau, sehingga enelitian ini bertujuan untuk mengidentifikasi dan menghitung dominansi dan keanekaragaman lamun dan makrozoobenthos pada musim pancaroba di Pantai Bama, TN Baluran, Situbondo. Metode penelitian yang digunakan yaitu line transect quadran dengan 5 line transek quadran yang masing-masingnya dipasang 5 plot transek. Ditemukan jenis lamun yang memiliki nilai kelimpahan tertinggi di Pantai Bama, TN Baluran pada musim pancaroba yaitu C. serrulata dengan nilai 48,90% , dan makrozoobenthos dari genus Holothuria dengan nilai 52,06%. Nilai Indeks dominansi (D) mengkategorikan Pantai Bama, TN Baluran dalam perairan dengan dominansi sedang. Sedangkan berdasarkan Indeks keanekaragaman (H’) mengkategorikan Pantai Bama, TN Baluran dalam perairan dengan keanekaragaman rendah. Musim peralihan berpengaruh terhadap jenis, kelimpahan relatif, indeks dominansi dan indeks keanekaragaman lamun dan makrozoobenthos di Pantai Bama, TN Baluran dibandingkan musim kemarau dan hujan. Perlu dilakukan survei secara berkala untuk mengetahui keanekaragaman organisme yang berkaitan dengan produktivitas perairan. AbstractSeagrass is the one of plants that can grow and adaptataion with high salinity environment and associated with benthos. Seagrass presence that indicate the productivity and prosperity in waters environtment. Found a different dominance between rainy season and dry season, so The aims of this study to identify and count dominance and diversity of seagrass and macrozoobenthos on the transition season in Bama Beach, TN Baluran, Situbondo. The method was used is line transect quadran with 5 quadran each of which installed 5 plot transect. Seagrass species was founded that have the highest abundance values in Bama Beach, TN Baluran the transitional season is C. serrulata with a value of 48.90%, and the macrozoobenthos that found from genus Holothuria with a value of 52.06%. Dominance index value (D) categorize Bama Beach, TN Baluran with moderate dominance. While based on the diversity index (H ') categorizes Bama Beach, TN Baluran with low diversity. Transitional seasons affect the type, relative abundance, dominance index and seagrass diversity index and macrozoobenthos in Bama Beach, TN Baluran than the dry and rainy seasons. Needed to investigate the diversity of organism that related to primary produktivity.
