Abstract

Infrabony pocket therapy is needed to eliminate pocket wall, creating easy to clean conditions for new attachment, and bone regeneration. Biphasic calcium sulfate (BCS) bone grafts combined with collagen membranes are known to regenerate bone tissue and have good osteoconductive effects. The addition of collagen membranes promotes migration and proliferation of fibroblast cells, osteoblasts, and homeostasis. However, the collagen membrane is a rapidbioresynthesis and may cause disease transmission from animal. Acellular dermal matrix (ADMA) membrane contains a bioactive matrix that has the ability to support normal revascularization, cell repopulation, and tissue remodeling. Combination of BCS with ADMA membrane are proven to induce bone and tissue regeneration. The objective of this study is to determine the therapeutic effect of BSC and ADMA combination to eradicate pocket, gingival recession, bone recession and attachment loss. The samples were taken from 20 infrabony pocket sites divided into 2 groups. The first was treated by combination of BCS and ADMA, while the second group was treated by the combination of BCS and collagen membrane. After 1 and 3 months of flap surgery, the result was observed by probing depth(PD), relative attachment loss (RAL), gingival recession, alveolar bone height and radiological examination. The result showed the decrease of PD, RAL, gingival recession, and alveolar bone height in both two groups. However, there wereno significant differences between those two groups. In conclusion, the combination of BCS and ADMA or BCS and collagen decreased the PD, RAL, gingival recession and alveolar bone height.

Abstract

Visible Light Cure (VLC) is a blue light used in dentistry as an activator for restorative material and fixed orthodontic bonding. The wavelength of VLC is between 400-500 nm and considered non-ionizing radiation that can produce free radicals. According to previous research, the light at wavelength < 500 nm could inhibit cells mitosis, cause cells damage, and reduce cells growth and inflammation. The purpose of this study was to investigate the effect of VLC exposure on gingival epithelial thickness, total neutrophil and macrophage count of gingival connective tissue of Sprague dawley rats. The subjects of this study consisted of 20 Sprague dawley rats, in 2-3 months of age and divided into 4 groups. Each group was 5 rats. The rats in each group were sacrificed before (0 day, as group A) and after 1st (group B), 3rd  (group C), 5th  (group D) day of VLC exposure, respectively. The exposure of VLC was done in labial aspect of cervical anterior teeth of mandible. The distance of exposure was as thick as 2 layers of celluloid strip and the histological specimens were stained by Hematoxylin Eosin. Each specimen was measured for its gingival epithelial thickness by using a micrometer and the number of neutrophil and macrophage were counted. The data of gingival epithelial thickness from 4 groups were analyzed by Kruskal Wallis. The number of neutrophil and macrophage were analyzed by u sing one way ANOVA. The results of this study showed that there were significant differences among groups on the thickness of gingival epithelial, the number of neutrophil and macrophage in the gingival connective tissue of Sprague dawley rats. The result of this study indicated that VLC exposure might decrease the thickness of gingival epithelial but increase the number of neutrophil and macrophage of gingival connective tissue of Sprague dawley rats.

Abstract

Gingivitis pubertas merupakan suatu peradangan gusi karena kondisi tertentu yang diklasifikasikan menurut faktor etiologi dan perubahan patologi. Peningkatan ukuran gingiva merupakan tanda adanya kelainan gingiva. Gingivektomi adalah pemotongan jaringan gingiva dengan membuang dinding lateral poket yang bertujuan untuk menghilangkan poket dan keradangan gingiva sehingga didapat gingiva yang fisiologis, fungsional dan estetik baik. Pada kasus ini, wanita berusia 16 tahun mengeluhkan keadaan gusi atas dan bawah yang membengkak sejak 1 tahun yang lalu, mudah berdarah dan tidak ada rasa nyeri. Penanganan untuk kasus ini dirancang menggunakan teknik gingivektomi menggunakan pisau periodontal dan gingivoplasty menggunakan electrocautery secara bertahap. Teknik gingivektomi menggunakan kombinasi scalpel dan electrocautery pada perawatan gingivitis pubertas memberikan hasil yang memuaskan secara estetik maupun fungsional pada pasien. ABSTRACT: Gingivectomy Using Scalpel and Electrocautery in Gingival Enlargement Treatment of Puberty Female. Puberty Gingivitis is a gum inflamation due to certain conditions that are classified by etiologic factors and pathological changes. The increase in gingival size is a sign of gingival disorder. Gingivectomy is cutting the gingival tissue by removing the lateral wall of the pocket which aims to eliminate pockets and gingival inflammation thus obtaining physiologically, functionally and aesthetically good gingiva. In this case, 16-year old woman complained of the state of the upper and lower gums which were swollen since the last one year, easily bleeding and no pain. The handling of this case was designed to use the technique of gingivecto by using periodontal knives and gingivoplasty by gradually using electrocautery. Gingivectomy technique with a combination of scalpel and electrocautery in puberty gingivitis treatment gives satisfactory results in aesthetic and functional state in patients.

Abstract

Perawatan kerusakan jaringan periodontal mempunyai tujuan utama mendapatkan jaringan regeneratif dan proses yang berlangsung membentuk struktur jaringan yang fungsional melalui proses pertumbuhan serta diferensiasi sel sel baru. Bonegraft adalah perawatan untuk kasus kerusakan tulang. Platelet-rich plasma yang merupakan platelet autologus konsentrasi tinggi tersuspensi dalam plasma setelah disentrifugasi. Dalam PRP banyak terdapat komponen yang berperan dalam proses penyembuhan regeneratif, growth faktor, agen kemotaktik dan agen vasoaktif. Kombinasi dengan penambahan kolagen merupakan altematif yang aman dan efektif, selain menstimulasi pelepasan growth faktor pada daerah target, juga memperkuat signal agar degranulasi platelet dapat ditingkatkan. Pada kasus ini, wanita berusia 24 tahun mengeluhkan keadaan giginya goyah sejak 3 bulan yang lalu karena traumatik, tidak ada rasa nyeri. Setelah dilakukan rontgen periapikal digital terlihat terjadinya kerusakan tulang infraboni. Penanganan untuk kasus ini dirancang dengan bedah flap, bonegraft Demineralized Freeze-Dried Bovine Bone Allograft (DFDBA), aplikasi platelet-rich plasma, serta penambahan kolagen. Kombinasi bonegraft dengan aplikasi PRP dan penambahan kolagen untuk menunjang perawatan periodontal memberikan hasil yang memuaskan secara penampakan klinis dan penampakan radiografis. ABSTRACT: Bonegraft Treatment with Addition of Platelet - Rich Plasma and Collagen in Infrabony Defect. The treatment of periodontal tissue damage has the main goal to get regenerative tissues and processes that take place to form a functional network structure through the process of cell growth and differentiation of new cells. Bonegraft is a treatment for cases of bone damage. Platelet-rich plasma is autologous platelets suspended in plasma high concentrations after centrifugation. In PRP, there are many components that play a role as a regenerative agent of healing process, growth factors, chemotactic agents and vasoactive agents. Its combination with the addition of collagen is a safe and effective alternative; in addition to stimulating the release of growth factors in the target area, it also strengthens the signal to improve platelet degranulation. In this case, a 24-year-old woman complained of unsteady state of her teeth since the last 3 months due to trauma; there was no pain. A digital periapical X-ray exposed infrabony defect. The treatment for this case was designed to use a surgical flap, bonegraft Demineralized Freeze-Dried Bovine Bone Allograft (DFDBA), application of platelet-rich plasma, as well as the addition of collagen. Bonegraft combination with PRP application and the addition of collagen to support periodontal treatment have given satisfactory results in the clinical and radiographic appearance.

Abstract

Latar Belakang. Aquaporin-5 (AQP5) di kelenjar submandibularis adalah salah satu anggota protein transmembran/ aquaporins (AQPs) yang memfasilitasi gerakan saliva sehingga mampu melewati membran sel. Di gingiva juga terekspresi aquaporin-3 (AQP3) yang diperkirakan juga berperan penting untuk memfasilitasi cairan sulkus gingiva sehingga dapat melewati jaringan ini. Sampai saat ini masih sedikit sekali informasi tentang dampak penggunaan radiasi ionisasi terhadap ekspresi aquaporins (AQPs), yang mendasari terjadinya xerostomia. Melalui pendekatan patologi molekuler, penelitian ini bertujuan untuk mengetahui efek radiasi ionisasi terhadap ekspresi mRNA AQP5 pada kelenjar submandibula dan mRNA AQP3 gingiva. Metodologi. Penelitian dilakukan pada 20 Rattus novergicus keturunan kedua, sehat, jantan, usia 3-4 bulan, berat badan ± 200 gr kemudian dibagi 2 kelompok, tanpa radiasi dan dengan radiasi C060dosis 10 gray pada ventral tikus. Pengambilan kelenjar submandibularis, dan gingiva dengan pembedahan. Seluruh sampel yang terkumpul dilakukan isolasi RNA dan dilanjutkan dengan pemeriksaan RT-PCR. Hasil. Penelitian ini menunjukkan adanya perbedaan gambaran ketebalan band/pita ketika dibatasi oleh primer untuk identifikasi ekspresi mRNA AQP-5 kelenjar submandibularis tikus serta ekspresi mRNA AQP3 gingiva tikus akibat radiasi ionisasi 10 Gy. Kesimpulan. Terdapat penurunan ekspresi mRNA AQP5 kelenjar submandibula dan mRNA AQP3 gingiva akibat radiasi ionisasi ekspresi mRNA AQP5 pada kelenjar submandibula dan mRNA AQP3 gingiva.

Abstract

Periodontal tissue damage requires regenerative material to repair the damage. Platelet-rich plasma (PRP) is known as a regenerative material from blood which contains high level of growth factor that plays a role in wound healing and tissue remodeling. However PRP has a weakness, i.e. it is too watery so it is easily dissolved in the oral cavity, and should be used immediately after preparation. Therefore PRP storage method is needed to increase the benefits of PRP. The addition of collagen to PRP serves as a scaffold as well as an activator that stimulates the release of growth factors. One method of storing PRP is by freeze-drying process. The purpose of this study was to analyze the effect of freeze-drying process of collagen-activated PRP (PRP+C) on transforming growth factor-β1 (TGF-β1) levels. Transforming growth factor-β1 is a cytokine content in PRP, that plays a role in bone remodeling and is an important stimulator for osteoblast formation, causing chemotaxis, osteoblast proliferation and differentiation. In this study, PRP was produced from peripheral blood probandus. Platelet-rich plasma was then activated with collagen (PRP+C), and divided into two groups: freeze-dried PRP collagen (FD PRP+C); and non freeze-dried PRP+collagen (PRP+C). Transforming growth factor-β1 levels were measured using the ELISA method, followed by independent t-test. The TGF-B1 level of FD PRP+C group was significantly higher than PRP+C group (p<0.05). From this study it can be concluded that freeze-dried collagen-activated PRP has an effect to increase TGF-β1 level.

Abstract

Platelet-rich plasma (PRP) contains growth factors that speed up a healing process. The liquid form of PRP is difficult to be applied, so it needs material as a carrier of collagen. Collagen can be used as carrier materials and capable to activate growth factors and increase the concentration of the PRP to become a gel. Collagen often losses its shape and size due to degradation when exposed to body fluids. In the selection of materials, collagen can synthesize with chitosan. Collagen-chitosan hydrogel has potential as scaffold. Many formulations for proper hydrogel applied in the oral cavity need to pay more attention to several conditions, such as a neutral pH, high viscosity and ideal swelling. The aims of this study was to examine the influence of various formulations of collagenchitosan hydrogel PRPagainst pH, viscosity, and swelling. The study samples consisted of 25 collagen-chitosanhydrogel formulations. The samples were divided into five groups: 100/0; 75/25; 50/50; 25/75; 0/100. The first stage of the test was pH, viscosity and swelling measurement of the collagen-chitosan hydrogel to determine which ones matched the criteria. Collagen-chitosan hydrogel formulations were added with PRP with a ratio of 1:1. The second stage was to measure the pH, viscosity, and swelling measurement to see the results of the physical parameters of the hydrogel after mixed with PRP. The observation data were analyzed by t-test for pH and viscosity, while twoway ANOVA and post hoc LSD for swelling test. The results showed that collagen-chitosan hydrogel formulations of 25/75 and 0/100 met the criteria of pH, viscosity, and swelling. The pH of both groups showed no significant difference (p>0.05), but the viscosity and swelling variables showed significant difference (p<0.05). To conclude, collagen-chitosan hydrogel formulations mixed with PRP had no effect on pH, but had an effect on viscosity and swelling test. The collagen-chitosan hydrogel formulations mixed with PRP which matched the criteria were 25/75 and 0/100.

Abstract

Background: Gingival recession is a condition that occurs when the gingival margin is more apically than the cemento-enamel junction and causes the tooth root to open, so a root closure procedure is required. Platelet rich plasma (PRP) contains growth factors, including Fibroblast growth factor which plays a role in the process of angiogenesis and cell proliferation. The activation time is known to affect platelet count and FGF expression. The purpose of this study was to determine the effect of the activation time of platelet rich plasma and collagen on the expression of fibroblast growth factor in the gingival crevicular fluidMethod: The research sample was gingival crevicular fluid which was taken using paper point #30 for 30 seconds as many as 18 samples. Samples were taken before surgery, day 5 and day 7 after surgery. The samples were divided into 2 groups based on the activation time, 1 hour and 24 hours PRP-collagen. Changes in FGF levels were examined using ELISA. Repeated Meassure ANOVA test was conducted to compare the differences in each time observation and post hoc was performed to compare the differences between groups for each time observation.Result: There was no significant difference in FGF levels in the gingival crevicular fluid at each time and between groups.Conclusion: There was no effect of the activation time of platelet rich plasma and collagen on the levels of fibroblast growth factor in gingival crevicular fluid

Abstract

ABSTRACTBackground: Gingiva is the outermost periodontal tissue that acts as a mechanical and biological barrier to the root of the teeth and alveolar bone. The main cellular elements in the gingiva are fibroblasts. Fibroblast cell proliferation is an important process in tissue regeneration. Growth factors that can stimulate fibroblast cell proliferation can be found in regenerative agents, such as injectable platelet-rich fibrin (i-PRF) and platelet-rich plasma (PRP). The aim of this study was to examine the influence between i-PRF and PRP on the gingival fibroblast cell proliferation in vitro study on primary cell culture.Method: Gingival fibroblast cell were obtained from primary cell culture derived from healthy gingiva. Ten mL of peripheral blood were centrifuged for i-PRF and PRP preparation. The samples were divided into three groups: i-PRF, PRP, and fibroblast cells without treatment. Cell proliferation were observed at day 1, day 3, day 5 using MTT assay at 550 nm. The data were analyzed by Two-Way ANOVA test, followed by Post Hoc test.Result: The results showed that the cell proliferation increased from day 1, 3, and 5 in all groups. The absorbance value of the cell proliferation in order from highest to lowest: i-PRF, PRP, and cell control.Conclusion: i-PRF and PRP increased the gingival fibroblast cell proliferation. i-PRF increased the cell proliferation higher than PRP.
