Abstract

This study aims to analyze the influence of caffeine on the distance of the tail movement of the spermatozoa epididymis cattle Bali. Methods in this study with a collection of sperm from Slaughter House (RPH) and then conducted the process of freezing with Tris yolk dilution. Treatment of caffeine with levels of 0 mg/ml, 2 mg/ml, 4 mg/ml and 6 mg/ml in spermatozoa that have been in thawing. The variables observed were DCL, DAP, DSL, ALH analyzed by CASA. Results showed no difference (P> 0.05) motility with caffeine treatment and short sperm tailwave distance of 37.27 μm showed the sperm motility velocity. Keywords: spermatozoa tail, caffeine

Abstract

Penelitian “Peningkatan Motilitas Spermatozoa Kauda Epididimis Sapi Bali di Rumah Potong Hewan (RPH) Kota Ternate Menggunakan Kafein”, bertujuan untuk mengkaji pengaruh penambahan kafein terhadap motilitas spermatozoa kauda epididimis sapi Bali dan mengukur berapa jumlah penambahan kafein yang dapat meningkatkan motilitas spermatozoa kauda epididimis sapi Bali. Koleksi testes diambil dari Rumah Potong Hewan (RPH) Kota Ternate, proses analisis sperma dilakukan di laboratorium Bioteknologi Universitas Khairun. Metode penelitian ini adalah sperma kauda epididimis dimasukkan dalam pengencer tris kuning telur kemudian diberi perlakuan penambahan kafein T0 (0 mg/ml), T2 (2 mg/ml), T4 (4 mg/ml), dan T6 (6 mg/ml) dengan 4 kali ulangan. Parameter dianalisis adalah kualitas sperma secara makroskopis dan mikroskopis. Data yang diperoleh dianalisis dengan analisis variance (ANOVA) satu arah, apabila terdapat perbedaan yang nyata (P<0,05) atau sangat nyata (P<0,01), dilanjutkan  uji Tukey-W-Procedure dengan SPSS 18. Hasil penelitian menunjukkan bahwa penambahan kafein 2-4 mg/ml pada pengencer tris kuning telur sperma kauda epididymis sapi Bali mampu meningkatkan motilitas sperma.

Abstract

This study was aimed to obtain viability of Ettawa Crossbred spermatozoa into day 14th on with the addition of crude tannin (CT). The design used was one-way completely randomized design with five replications and five treatments. The data of value identified sperm were analyzed descriptively. Sperm quality data were analyzed using one way analysis of variance (ANOVA) and Tukey-W-test procedure. The results showed that the sperm quality characteristics were as the followed: volume 1.4+0.42 ml; pH 6.84+0.27; creamy milk color; consistency thick; motility ++ and concentration (328.8+80.74) x 107/ml semen.  Spermatozoa viability were K0 (control) 29.89+16.52%; K1 (CT 2.5%) 65.33+8.95%; K2 (CT 5%) 58.20+12.4%; K3 (CT 10%) 52.61+15.34%; and K4 (CT 20%) 47.84+13.84%. It was concluded that the addition of 2.5% crude tannin gave significant effect on viability of Ettawa Crossbred spermatozoa during 14 days storage, whereas addition of 20% crude tannin had caused low percentage of live spermatozoa.(Key words: Spermatozoa, Crude tannin, Viability)

Abstract

Abstrak Penelitian ini dilaksanakan di Dinas Pertanian Provinsi Maluku Utara (Sofifi). Tujuan dari penelitian ini untuk mengetahui pengaruh lama thawing terhadap kualitas semen beku Sapi Bali. Analisis data yang digunakan dalam penelitian ini menggunakan Anlisis Of Variance (ANOVA) satu arah dengan empat macam perlakuan yaitu 15, 30, 45 dan 50 detik menggunakan air ledeng (PDAM) temperature 25‐30oC dan dilakukan 4 kali ulangan. Perlakuan pada penelitian ini yaitu P1: thawing 15 detik, P2: thawing 30 detik, P3: thawing 45 detik, dan P4: thawing 50 detik. Parameter yang diamati pada penelitian ini adalah Motilitas, Viatabilitas, dan Abnormalitas. Hasil menunjukkan nilai terbaik Motilitas pada 15 detik sebesar 86,25%. Viatabilitas sebesar 83,75%. Sedangkan pada Abnormalitas thawing 15 detik memiliki nilai yang lebih rendah sebesar 15,00%. Berdasarkan hasil penelitian dapat disimpulkan bahwa lama thawing 15 detik dan 30 detik mempunyai kualitas sperma yang lebih baik daripada lama thawing 45 dan 50 detik. Kata Kunci : Thawing, Motilitas, Viatabilitas, dan Abnormalitas. Abstract This research was carried out in the agricultural department of North Maluku Province (Sofifi). The purpose of this study was to determine the effect of thawing time on the quality of Bali frozen semen. Analysis of the data used in this study used an one-way analysis of variance (ANOVA) with four types of treatments, namely 15 seconds, 30,45, and 50 seconds using tap water (PDAM) temperature of 25-30°C and carried out 4 replications. This research is: P1: 15 seconds thawing, P2: 30 seconds thawing, P3: 45 seconds thawing, and P4: 50 seconds thawing. The parameters observed in this study were motility, viatability, and abnormalities. The results showed the best value of motility at 15 seconds at 86.25%. Viatability is 83.75%. Whereas the 15 second thawing abnormality has a lower value of 15.00%. Based on the results of the study it can be concluded that the length of thawing 15 seconds and 30 seconds has better sperm quality than the thawing duration of 45 and 50 seconds. Abstract

This study aimed to determine the effect of centrifugation time on sexing spermatozoa with Bovine Serum Albumin (BSA) media toward the motility and viability of spermatozoa X-Y Simmental cow. This research was carried out from October to November 2021 at the Sembawa Banyuasin Animal Feed Breeding Center, South Sumatra and the Animal Reproduction and Health Laboratory. Sample from Simmental type of cow semen the age 3 years. This study used 3 treatments with 4 repetitions. The observed variables were the motility and viability values of Simmental cattle spermatozoa. Data were analyzed using ANOVA and Duncan's test 5%. The results showed that the with a centrifugation time of 4 minutes(P1) with a speed of 1500 rpm got the best results for the motility value of spermatozoa X = 61.54% and spermatozoa Y = 56.71%, the best value of spermatozoa viability X = 66.17 and spermatozoa viability Y = 62, 46%. It can be concluded that the longer the centrifugation time can affect the quality of the spermatozoa which results in a decrease in the motility and viability of the spermatozoa X-Y Simmental Cow. Keywords: Sexing, Bovine Serum Albumin, Centrifugation, Motility and viability

Abstract

Penelitian ini bertujuan untuk mendapatkan waktu sentrifugasi yang tepat pada sexing spermatozoa dengan media Bovine Serum Albumin terhadap membran plasma utuh spermatozoa  X-Y pada sapi Simmental. Penelitian ini dilaksanakan Oktober 2021 sampai dengan November 2021 di Laboratorium Reproduksi dan Kesehatan Ternak Balai Pembibitan Hijauan Pakan Ternak Sembawa. Penelitian ini menggunakan rancangan Rancangan Acak Lengkap (RAL) dengan 3 perlakuan (P1: waktu sentrifugasi 4 menit; P2: waktu sentrifugasi 8 menit; dan P3: waktu sentrifugasi 12 menit) dan 4 ulangan. Parameter yang diamati adalah membran plasma utuh spermatozoa X dan Y. Hasil menunjukan bahwa perlakuan waktu sentrifugasi berpengaruh nyata terhadap membran plasma utuh spermatozoa pada spermatozoa X dan Y (P<0,05).  Hasil penggunaan waktu yang tepat yaitu pada P1 dengan waktu sentrifugasi 4 menit dimana nilai lapisan atas membran plasma utuh X semen sexing yaitu sebesar 62,93%, lapisan bawah Y sebesar 63,94%. Penelitian ini bertujuan untuk mendapatkan waktu sentrifugasi yang tepat pada sexing spermatozoa dengan media Bovine Serum Albumin terhadap membran plasma utuh spermatozoa  X-Y pada sapi Simmental. Abstract

Duck is a producer of animal protein whose production is eggs and meat, is a type of poultry that has the potential to be developed in the city of Ternate. However, until now there has been no characterization research in Ternate City. The aim of the study was to determine the phenotype characteristics as the basis for breeding local cattle in North Maluku. The materials used were adult male and female laying ducks, the variables observed were male ducks, qualitative characteristics were the character of coat color, beak color, eye color and quantitative characteristics were measured body size including femur length, tibia length, shank length, shank circumference, length wingspan, chest length, chest width, chest depth and hip width. calculating the mean formula, standard deviation and coefficient of diversity, qualitative characteristics of ducks using relative frequency analysis. The results showed that the quantitative characteristics of female ducks were more similar than those of male ducks. While the qualitative nature, the color is relatively the same both males and females have brown fur, only one is blackish brown, the skin color of the legs and beak is more dominantly black and brown compared to yellow, black yellow and brown yellow.

Abstract

Sexing and centrifugation of spermatozoa is one way to improve the quality of spermatozoa. This study aimed to obtain the right centrifugation time for sexing spermatozoa with Bovine Serum Albumin media on the morphology of X-Y spermatozoa in Simmental cattle. This study was carried out from October 2021 to November 2021 at the Laboratory of Animal Reproduction and Health at the Breeding Center for Superior Livestock and Forage, Sembawa. The design used in this study was a completely randomized design (CRD) with 3 treatments and 4 replications. Centrifugation treatments for sexing spermatozoa with BSA media P1: centrifugation time 4 minutes, P2: centrifugation time 8 minutes, and P3: centrifugation time 12 minutes. The parameters observed were the morphology of the X and Y spermatozoa. The results showed that the centrifugation time for sexing spermatozoa—which was different with Bovine Serum Albumin (BSA) media—indicates that the X spermatozoa abnormalities had no significant effect (P0.05) but the Y spermatozoa abnormalities had a significant effect (P0.05). The result of the right time use was in the P1 treatment with a centrifugation time of 4 minutes where the abnormalities value in the lower layer Y was 28.79%.

Abstract

Abattoir is the place to get meat but also a source of potential genetic sperm. Sperm from a slaughterhouse has low motility. Sperm motility can be improved by adding caffeine to the thinner before being used for fertilization. Caffeine is an alkaloid compound that can increase energy through a cAMP cycle. The method used is the testis of 12 cows Bali taken from a slaughterhouse Cibinong and do frozen sperm. Frozen sperm is analyzed using a computer-assisted sperm Analyzed (CASA) who had been treated caffeine 0, 2, 4, and 6 mg/ml. Fertility frozen epididymis sperm was tested using in vitro fertilization. Results were analyzed using a completely randomized design (CRD) with four treatments unidirectional pattern three repetitions. The results showed that the treatment with the addition of caffeine to the thinner of the yolk tris egg yolk epididymis sperm, there was no difference in motility, recovery rate, Curvilinear velocity (VCL), average path velocity (VAP), and straight-line velocity (VSL).

Abstract

The current research aimed to determine Bali cattle's productivity in Bumirestu, Akedaga, and Dakaino Villages in East Halmahera Regency. This research was carried out in Bumirestu, Akedaga, and Dakaino Villages, East Halmahera Regency, for three months, from February to April 2018. A presurvey and field survey methods were applied to observe several variables: calf weaning, mothers mating after calving, estrus age after calving, first calving age, and body weight. Livestock productivity could be seen from the reproductive characteristics, the first calving age with 64% in Dakaino Village. Calf weaning with a percentage of 42.5% in Bumirestu Village, estrus age after calving with a percentage of 52% in Dakaino Village, parents were mating after giving birth with a percentage of 60% in Akedaga Village, and body weight by calculating the average of 288.71±17.55 in Dakaino Village. It was revealed that the factors that support the livestock productivity are the first calving age of the cattle was two years old, calf weaning was at 2-4 months old, the estrus age after calving was 30-60 days, and the parents were mated 60 to 90 days after calving.
