Abstract

Meatballs are one of the most popular processed meat products in Indonesia. As processed food produced by livestock, meatballs are susceptible to alduteration. The DNA-based detection method is the most widely used and the susceptibility depend on the DNA quality. The aim of this research was to investigate the DNA isolation procedure of commercial meatballs. The study was conducted by taking 36 samples of meatballs from meatball stalls in Bojonegoro Regency. Reference meatballs as a positive control were prepared in the laboratory. DNA isolation was carried out on the collected sample meatballs, reference meatballs, and 10 species of fresh meat for comparison. The concentration and purity of DNA were measured using a spectrophotometer and visualization of the isolation results was performed using agarose gel electrophoresis and UV transilluminator. The results of DNA isolation were compared descriptively qualitatively between samples of meatballs on the market, reference meatballs, and fresh meat. The results of DNA isolation showed that the meatball samples from the market were successfully isolated by showing the concentration values ​​and visualization with agarose gel electrophoresis. The results of electrophoresis and the value of the DNA purity index showed that the DNA was not pure.

Abstract

The aims of this study were to determine the characteristics and quality of fresh semen from Sumba Ongole (SO) bulls which were diluted with tris dilution supplemented with skim milk. The study used a completely randomized design consisting of 4 treatments and 5 replications. The treatments were P0 = 100 % Tris, P1 = 88 % Tris + 12 % Susu skim, P2 = 86 % Tris + 14 % Susu skim, dan P3 = 84 % Tris + 16 % Susu skim. The parameters observed were the colour, smell, consistency, pH, volume, motility, viability, and abnormalities of semen. The results showed that the characteristics of the semen were normal, the color of the semen was creamy white, the semen volume was 4.6 ml, the consistency was moderate, pH 6.3, the smell was typical of SO cattle, the motility was 80%, the mass movement was +++, the concentration was 758 million/ ml, viability is 74.24% and abnormality is 16.12%. The characteristics and quality of cement in the four treatments showed no significant difference (P>0.01). The motility and viability of spermatozoa in each treatment were able to survive up to day 3. The Tris diluent with the addition of skim milk was not able to maintain the characteristics and quality of the semen.

Abstract

Efforts that can be made to improve the performance of native chickens are by crossing them with Bangkok chickens. This study aims to determine the growth of body weight from cross-breeding between Kampung chicken and Bangkok chicken. As many as 30 male chicks from a cross between Kampung and Bangkok chickens were used in this study. The crosses used are rooster chicks. The weight of DOC chickens from Bangkok male and Kampung female was 35.22 ± 0.96 grams. There was a significant increase in growth in chickens from crosses between Kampung male and Bangkok female starting at the age of 5 weeks to 8 weeks of age, reaching a body weight of 729.39±39.10 grams. The conclusion of this study was that the growth of Kampung male crossed with Bangkok female can increase the body weight growth of their offspring. A significant increase in performance in crosses between Kampung and Bangkok chickens occurred from 5 weeks of age to 8 weeks of age.

Abstract

The objective of this study was to determine the effect of supplementation nanoencapsulation of Mahkota Dewa fruit extract (NEMD) against the meat bone ratio (MBR) of the commercial part of the broiler chicken carcass. The research material was used 200 male broiler chickens aged eight days who were rearing in open house cages. The treatments consisted of drinking water without feed additives (P0; negative control), drinking water + tetracycline antibiotics (P1; positive control), drinking water + 2.5% Mahkota Dewa fruit extract (P2), drinking water + 2.5 % NEMD (P3), and drinking water + 5.0 % NEMD (P4). Parameters observed were slaughter weight, carcass weight, and MBR of the commercial part of the broiler chicken carcass. The data obtained were analyzed using a completely randomized design (CRD). Data with significant results were further tested using orthogonal contrast (P-Value < 0.05). The results showed that the use of NEMD up to 5% in drinking water did not have a significant effect on the MBR of the commercial part of the broiler chicken carcass. The 5% NEMD has not been able to work optimally to increase the MBR of the commercial part of the broiler chicken carcass.

Abstract

This study aims to determine the relationship between body size and bodyweight of the first lactating FH dairy cow. The research method used was linear regression analysis with 17 individuals at 27-29 months. The observed variables were body weight (BW), body length (BL), shoulder height (SH), and chest circumference (CC). The results showed that the correlation value between BW and BL was 0.6791 with the correlation regression equation Y = -724.63+7.68X1. SH and BW have a correlation value of 0.4043. CC with BW has a correlation value of 0.6863 with the regression equation Y = -733.455.90X1. The relationship between BL and SH has a correlation value of 0.2048. BL and CC have a correlation value of 0.4832. The conclusion of this study was that BW has a correlation of 67.91% to BL and 68.63% to CC of first lactating dairy cows, but does not have a correlation to SH. The more the BL and CC of the first lactating dairy cow increased, the more the weight of the dairy cow increased. BL does not have a correlation with SH and CC. SH has a 60.08% correlation with CC.
