Abstract

The yeast mitochondrial F1F0-ATP synthase is a multisubunit complex that contains at least 17 different subunits. Subunit 8 of yeast mitochondrial ATP synthase is a hydrophobic protein of 48 amino acids encoded by the mitochondrial ATP8 gene. A dual control allotopic expression system for subunit 8 has been developed. The strategy involves maintenance of two different compatible yeast expression vectors each utilizing a different inducible promoter in the same host cells. The system thus enables cloning and allotopic expression of two different forms of subunit 8 gene. The goal of the developed strategy is to permit allotopic expression of functional wildtype subunit 8 gene under a conditional promoter system and subunit 8 variant gene under the control of a different promoter system. The system is potentially useful for accurate assessment of assembly behavior of functionally defective subunit 8 variants in vivo. The strategy relies on the ability to conditionally regulate the expression of the two genes. A set of functionally defective subunit 8 variants has been cloned under an inducible yeast promoter system and dual plasmid harboring strains for dual control allotopic expression of each variant have been constructed.

Abstract

Certain bacteria that induce biological ice nucleation are suspected to play an important role in condensation and ice nuclei formation in clouds. Those bacteria can produce biological ice nucleator1, which is a protein and usually found on leaf surface and air. Most studies on INA bacteria were conducted in subtropical areas. In this study, INA bacteria were isolated from rain-water and air between March to May 2008 from Jakarta, Bogor, Bekasi, Tangerang, and Depok. The percentage of INA bacteria from rainfall for those area are 19.4, 18.7, 5.3, 2.2, and 6.4% respectively, while percentage for air are 9.5, 6.5, 0, 2.7, and 1.8%. The highest incidence of INA bacteria were from rain-water and air found in sample from Jakarta and then followed by the samples from Bogor. It was shown that the percentage of INA bacteria from rain-water was higher than air for all of the samples from different areas. The isolate from Jakarta (isolate A32) which had the highest activity for ice nucleation, with the temperature classification at -2.7 oC, revealed 100% similarity with Pantoea sp. The presence of INA bacteria in rain-water and air might play an important role in nucleation process which is required for rainfall induction.

Abstract

A study with the aim to investigate the quality of Gronotoma micromorpha which attack the immature developmental stages of Liriomyza huidobrensis has been carried out. Such variables as body length measured from caput to the tip of abdomen, wing span and the length of tibia of hind leg were measured to represent parasitoid quality. The immature developmental stage period of the parasitoid was also recorded. The result indicated that a better quality Gronotoma was obtained when it was developed in the Liriomyza larvae fed on chinese cabbage with the average of 123.85 mm for body length, 253.45 mm for wing span, and 42.85 mm for the length of hind tibia, respectively. When Gronotoma was developed in the Liriomyza larvae fed on soy bean, its size became smaller with the average of 97.7 mm for body length, 214.3 mm for wing span, and 37.2 mm for the hind tibia. When it is developed in the Liriomyza on chinese cabbage host plant the immature developmental period of Gronotoma is sligthly shorter with the average of 18.4 and 17.3 days for which developed in the larvae and pupa respectively and on soy bean it become longer with the average of 19.2 days for in eggs, 19.9 days for that in the larvae. On chinese cabbage, Liriomyza tend to have bigger size for both flies and pupae.

Abstract

Syzygium cumini L. better known as Jamun belonging to the family Myrtaceae is identified to have antidiabetic, anti-inflammatory, anti-pyretic and anti-oxidant activities. Anticancer activity of S. cumini L. fruits has been demonstrated. However, anticancer activity of S. cumini seeds on various types of human cancers has not been explored much. The methanol fraction of ethanol extract from the seeds of S. cumini was found to have significant antibacterial activity. This bioactive fraction was further tested positive for its anticancer activity on various types of human cancer cell lines indicating its potency. Structural characterization of the bioactive fraction was achieved using analysis of high performance liquid chromatography, ultra violet and infra red spectrum.

Abstract

Rice is one of the most important crops for human beings, thus increasing productivity are continually persecuted. Blast disease can reduce the rate of productivity of rice cultivation. Therefore, the program of blast disease-resistant varieties needs to do effectively. One of broad-spectrum blast disease-resistant gene is Pi33. This study was aimed to identify the variation in the sequence of nucleotide bases of Pi33 gene in five interspesific lines which derived from Bio46 (IR64/Oryza rufipogon) and CT13432 crossing. DNA of five rice lines were amplified using the spesific primer for Pi33, G1010. Amplification results purified through Exonuclease 1 and Shrimp Alkaline Phosphatase protocols. Labelling using fluorescent dyes done before sequencing nucleotide base using CEQ8000 instrument. The results showed that lines number 28 showed introgesion of the three control parent genome (subspecies of Indica, subspecies of Japonica, and O. rufipogon) while the Lines number 79, 136, and 143 were identical to Indica genome. Strain number 195 was identical to Japonica genome. These broad genetic background lines promise as durable performance to attack the expansion of the dynamic nature of the pathogen to blast. The result of ortholog sequence analysis found conserved nucleotide base sequence (CAGCAGCC) which involved in heterotrimeric G-protein group. This protein has role as plant receptor for recognizing pathogen elicitor in interaction of  rice and blast pathogen.

Abstract

In vitro activity test is a critical evaluation to screen the potential biocontrol agent. We developed a selective medium for quantitative in vitro activity evaluation of bacterial biocontrol agents against pathogenic Vibrio in aquaculture. Sensitivity test of bacterial biocontrol and Vibrio spp. to nine antibiotics showed that oxytetracycline inhibited the growth of Vibrio spp., but did not inhibit the growth of the bacterial biocontrol. This selective inhibition activity of oxytetracycline suggested this antibiotic might be supplemented to establish a selective medium. The MIC of oxytetracycline against V. harveyi, V. parahaemolyticus, and V. algosus were 120, 250, and 120 mg/ml, respectively. These concentration did not inhibit the growth of bacterial biocontrols. Therefore, oxytetracycline was supplemented at 250 µg/ml in Zobell agar medium. This medium was used as a selective medium to enumerate the density of bacterial biocontrol. In vitro activity test of bacterial biocontrol (RLP1) against V. parahaemolyticus showed that strain RLP1 at density of 104 and 106 cells/ml was able to kill V. parahaemolyticus during 6 h incubation. At lower density, 102 cells/ml, this bacterial biocontrol agent was able to kill the pathogenic Vibrio during 12 h incubation. This study discovered a selective medium for the bacterial biocontrol and Vibrio spp. and provided the results of its application in the evaluation of in vitro activity of a bacterial biocontrol agent against V. parahaemolyticus. The results also revealed that strain RLP1 is a potential bacterial biocontrol against vibriosis in marine aquaculture.

Abstract

Molecular marker is useful in the development of testicular cells transplantation for detecting donor-derived germ cells in the recipient gonad. In this study, a giant gourami (Osphronemus goramy) vasa-like gene (GgVLG) was cloned and characterized for use as a molecular marker for germ cells in this species. Nucleotide sequence analysis revealed that GgVLG comprises 2,340 bps with an open reading frame of 1,962 bps encoding 653 amino acids. The deduced amino acid sequence contained 17 arginine-glycine or arginine-glycine-glycine motifs and eight conserved motifs belonging to the DEAD-box protein family. The GgVLG sequence showed high similarity to Drosophila vasa, common carp vasa homolog and tilapia vasa homolog for 66.2, 85.9, and 90.7%, respectively. In adult tissues, the GgVLG transcripts were specifically detected in ovary and testis. In situ hybridization analysis showed that GgVLG mRNA was detected in oocytes of the ovary and spermatogonia of the testis. There was no signal detected in the spermatocytes, spermatids and other gonadal somatic cells. Thus, consensus sequences, specific localization of GgVLG mRNA in the germ cells, amino acid sequence similarity and phylogenic analysis all suggest that GgVLG is the giant gourami vasa-like gene. Further, GgVLG can be used as a molecular marker for giant gourami germ cells.

Abstract

Boronia heterophylla is cultivated for cut flower production. Three cultivars dominate production, ‘Red’, ‘Cameo’, and ‘Moonglow’. A variety of colors and an extended flowering period are demanded by local and overseas markets. The aim of this study was to develop procedures for a Boronia heterophylla breeding program through intraspesific hybridization. This may yield progeny with desirable characteristics, ideally increased vigor, and a range of flower colors and flowering times. Nine pollination combinations were attempted, each self pollination and all reciprocal crosses. Seed set varied from 17 to 95%. Embryo rescue (was employed to produce hybrid plants using half strength Murashige and Skoog (MS) basal media and it was most successful (75%) 5-6 weeks after pollination. All shoots multiplied on media containing MS salts + NAA (0.1 mg/l) + BA (0.4 mg/l). All shoots transferred to medium containing half strength MS salts + NAA (4 mg/l) produced roots. Plantlets were acclimatized to sterile potting mix (in a small chamber within a glass house. This intraspecific hybridization system was very successful and plants were obtained for all pollination treatments except for selfed ‘Moonglow’. Embryo rescue may provide a system for germinating other species with difficult to germinate or dormant seed.

Abstract

The aim of this research was to gain the profile of blood gasses of green turtle (Chelonia mydas) hatch. Blood gas of the green turtle was analysed after exposuring them at 28 oC and 50% of humidity  for 24 hours in a pvc tube and at 34 oC under sunlight exposured with 47% of humidity for 30 minutes. The result showed the different values of blood gas contents. This result showed indication of metabolism activities and poikilothermic adaptation of green turtle hatch. This information can be used to support for turtle hatchery in Indonesia.
