Abstract

Tuberculosis (TB) is a respiratory disease caused by infection of Mycobacterium tuberculosis. Currently, many M. tuberculosis strains have resistant toward to two or more anti-tuberculosis drugs (known as MDR-MTB). One of the gene marker to MDR-MTB is the katG gene. Mutation of katG can be causes the activity of the M. tuberculosis catalase-peroxidase enzyme so that the bacteria become resistant isoniazid (INH) OAT. The aimed of this research was to know the presence of KatG gene in isolate M. tuberculosis from MGIT culture. The sample used was MGIT isolate with 20 samples and then amplified using PCR method. The result showed that there were 4 samples the presence of DNA bands suspected KatG genes ± 317 bp from the MGIT cultured sample. In conclusion, the KatG gene was successfully amplified by PCR method with primer forward and reverse visualization by electroforesis. Sequencing analysis needs to be done on the PCR result in order to see the mutations that occur and to confirm the sample homology with data contained in GenBank on the genes of the KatG, and then to know the presence or absence of mutations in katG genes.

Abstract

Anemiasufferers in Indonesia are increasing. Anemia diagnosisconfirmed by examining red blood cell (RBC) which requires Hayem as reagent. Problemof running out reagents still widely found that distrupts health services.0.9% infusion fluid is physiological solution and readily available. Therefore, this study aims to ultilize 0.9% intravenous fluids as alternative to Hayem in RBC examination by analyzing differences in results of alternative reagents to Hayem reagent. This type of research is experimental, using normality test, t-test, and linear regression.Carried out in the hematology laboratory of Nahdlatul Ulama University Surabaya in February 2020.Use 25 samples from students and health faculty staff with average infusion fluid ie 4,54 x 106 /µL (SD 0,59 x 106 /µL) while Hayem reagent ie 4,63 x 106 /µL (SD 0,52 x 106 /µL)and there is no difference two results (P = 0.197). fluid normality test was obtained Sig.= 0,716, while Hayem reagent Sig.= 0,923. Paired sample t-test obtained Sig.= 0,197. Resultsof linear regression test showed correlation coefficient of 0.823 and coefficient determination of 67.8% (P = 0.000) with equation Y = 0.725X + 1.339. It was concluded that 0.9% infusion fluid could be used as alternative reagent for Hayem in RBC examination.

Abstract

Urinary Tract Infection (UTI) is an infection of the pathogenic microorganisms that thrive in the urinary tract. Urinary Sediment is an examination to determine the presence of abnormalities in the kidneys and urinary tract. Urine precipitate volume is one of the factors that influence urine examination results. The purpose of this research is to determine the effect of urine sediment volume based on the results of urine sediment examination. This type of research is analytic research that use 15 urinary tract infections samples. The results of  analysis Kruskal wallis test on leukocytes examination showed p-value <0.05 which showed that there were differences in the number of leukocytes in the three precipitate urine volume comparisons and the results of analysis One Way Anova test on erythrocyte examination showed p-value <0.05 which showed that there were differences the number of erythrocytes in the three urine precipitate volume comparisons. The results of analysis Kruskal Wallis test on crystalline and bacterial examinations obtained p-value >0.05 which showed no differences in the number of crystals and bacteria in the three comparison of urine precipitate volume and the results of analysis One Way Anova test on epithelial cell examination obtained p-value >0.05, which indicates no difference in the number of epithelial cells in the three urine precipitate volume comparisons. Urine sediment examination is expected to use a standard precipitate urine volume of 20 µL.

Abstract

The aspartate aminotransferase (AST) enzyme is a mitochondrial enzyme that functions to catalyze the alternating transfer of amino groups from aspartic acid to ?-oxaloacetic acid to form glutamic acid and oxaloacetic acid. AST is present in erythrocytes so that if hemolysis occurs, the erythrocytes rupture will cause an increase in the activity of the AST enzyme that comes out of erythrocytes. This study aims to determine the effect of hemolysis in serum on the examination of AST enzyme activity. This research method is experimental by comparing normal serum with serum hemolysis. The research sample involved 16 students who would be examined for AST enzyme activity. The results showed that the activity of AST enzymes in normal serum had an average of 24.63 IU / L whereas in serum hemolysis there was an average of 39.38 IU / L. The results of the data showed that there was an increase in AST enzyme activity in serum hemolysis compared to normal serum. The data were then analyzed using the T test, and the Sig. 0.001 where p <0.05 proved that AST enzyme activity in normal serum and serum hemolysis was stated to be significantly different.

Abstract

Hyperuricemia is a condition of increased levels of uric acid in the blood. Physiological conditions, uric acid has the form of a solute. The crystals will form if the levels are above 6.8 mg/dL or at the saturation point. It will settle in various tissues, including the kidneys and joints. Hyperuricemia can cause various types of diseases such as hypertension, kidney disorders, arterosklesrosis, cardiovascular, gout and others. Crystals monosodium urat cancause innate and adaptive immune responses. Innate immune response is played by macrophages for phagocytic and produce pro-inflammatory cytokines and can stimulate lymphocyte activation. Its presence can be determined by counting the absolute lymphocyte count, thus giving an indication of the prognosis of the disease due to hyperuricemia. The purpose of this research is to describe the absolute lymphocyte count in hyperuricemia. This type of research is descriptive cross sectional study approach. The number of research subjects was 30 participants taken by purposive sampling with criteria for men aged 18-65 years, hyperuricemia, fasting 10-12 hours, no obesity, no infection, not children and not elderly. The results showed that 6.7% (2 people) had an increased absolute lymphocyte count, and 93.3% (28 people) had a normal absolute lymphocyte count. In conclusion, absolute lymphocyte counts can describe the prognosis due to hyperuricemia, as a result of the adaptive immune response.

Abstract

The high cost of instant media such as nutrient agar encourages researchers to find alternative media from raw materials that are readily available and cheap. The composition of the media, which is very important for the growth of bacteria, one of which is protein. The content can be obtained from nuts, one of which is groundnut (Arachis hypogaea L). This study aims to determine the growth of Staphylococcus aureus and Escherichia coli bacteria on peanut media which are used as alternative media for Nutrient Agar (NA) substitution. The research method used is Total Plate Count. Escherichia coli bacterial colonies that grew on peanut media were 43 CFU, and colonies that grew on NA media were 238 CFU. While the growth of Staphylococcus aureus bacterial colonies in peanut media was 32 CFU and bacterial colonies growing on NA media were 217 CFU. The conclusion is that peanut media can be used as an alternative media with the growth of Escherichia coli and Staphylococcus aureus bacteria that are in accordance with Standard Plate Count.

Abstract

The 3-chymotrypsin-like cysteine ??protease (3CLpro) is an enzyme that control replication of the coronavirus life cycle. The 3CLpro is a biological target that will interact with drugs. Brazilein is a major compound found in secang wood that used as an immunomodulator. This study aims to predict the immunomodulatory activity of the brazilein against the 3CLpro using in silico method, with curcumin as comparative compound. In silico test was used to predict immunomodulatory activity by docking using the Molegro Virtual Docke. Receptors used are 3CLpro, PDB code: 6M2N, with ligand 3WL_401[C]. pkCSM online program used to predict pharmacokinetic properties (ADME) and toxicity from brazilein and curcumin. Data was analyzed by comparing the docking bond energy between brazilein, ligands and curcumin at the target receptor. The lower the ligand bond energy, the more stable the bond is formed. The results of the in silico test showed that the ligand bond energy = -83,1153kcal/mol, brazilein = -82.0583kcal/mol, and curcumin = -115.852kcal/mol. Brazilein has lower potential as an immunomodulator than ligands and curcumin. The results of the in silico test using the pkCSM online program showed that the brazilein compound had good pharmacokinetic properties and low toxicity.

Abstract

Preeclampsia is a disorder in pregnancy. Preeclampsia is the presence of hypertension accompanied by oedema due to pregnancy and the most important sign is the presence of proteinuria. Proteinuria occurs later in the early pregnancy until it finally appears at the end of the trimester of pregnancy. This study was aimed to describe the proteinuria of third trimester pregnant women at Hospital Palembang in 2019 based on age, gravida status, occupation, history of hypertension and weight gain during pregnancy. It was a descriptive research with approach cross sectional. The research sample consisted of 65 third trimester pregnant women taken by accidental sampling technique. Proteinuria was examined with a dipstick method that is read with a verify U120 urine analyzer. The results showed 23.1% of samples were positive one and 7.7% positive two; A high percentage of protein in urine occurred in pregnant women based on risky age <20 atau ?35 (36,4%), based on gravida status in primigravida (53,6%), based on occupation in house wife category (37,5%), based on history of hypertension with there history of hypertension before pregnant (100%), based on weight gain of obese category (50%). It is recommended for pregnant women to do a complete urine examination every time they do a laboratory examination.

Abstract

Quality improvement of the waste cooking oil can be conducted by reducing levels of acid number and peroxide number. The purpose of this study was to determine the levels of acid number and peroxide number, as well as the effective temperature of mixing chicken egg shell adsorbents to reduce the acid and peroxide levels. This research was an experimental study with two treatments. The first treatment used the waste cooking oil mixed to chicken egg shell adsorbent at high temperature, then stirred and separated into sediment and supernatant, then determined the levels of peroxide and acid number. The second treatment was carried out alike the first treatment procedure but placed at room temperature.  As the independent variables in this study were temperatures that divided at high temperature and room temperature. As dependent variable of the study was levels of acid number and peroxide number.. The results of the study were analyzed using an Independent Bivariate     T-test. It was found that there were significant differences in the levels of acid and peroxide (p value = 0,000) with a 95% confidence level. Based on univariate analysis test, it is known that the effective temperature is used to reduce the levels of free fatty acids and peroxide in used cooking oil using egg shell adsorbent at room temperature.

Abstract

Level of lead in blood represent level of lead in body. Increasinglead in blood cause increasing amount of reticulocyte. Hematoxic can be used to increase awareness through effect of lead content in society for example mixed-paint workers.  The purpose of this study was to determine the closeness and relation between blood lead levels and reticulocyte count results of mixed-paint workers in East Jakarta. The research an observational analytic method, cross-sectional design approach, by non-probability consecutive sampling. The results of the study, the Oplosan Paint Workers in East Jakarta region obtained high blood lead level of 0.45 mg / L, with the highest reticulocyte 1.62%. The number of Oplosan workers with lead levels exceeds the reference limit (> 0.40 mg/L) with the percentage of reticulocytes above normal (> 1.5%) of 10%. Oplosan paint workers with lead levels in the reference limit with the percentage of normal reticulocytes is still large at 80%. In conclusion, there is a significant relation between blood lead levels and the percentage of reticulocytes in mixed-paint workers in the East Jakarta area (p-value = 0,000) and shows a strong relationship with the direction of a positive relation value of r = 0.663. The close relationship with positive direction means that higher blood lead levels can shorten the life and maturation of erythrocytes with an increase in reticulocyte count.
