Abstract

The aims of this study were to characterize the mitochondrial cytochrome oxidase subunit 1 gene (CO1) of Apis dorsata from Minahasa, North Sulawesi. Apis dorsata Binghami used in this study was obtained from the natural colony in Klabat forest, Airmadi forest and Kombi forest. From each native bee colony, ten workers were taken (n = 30). Femur hind legs used for subsequent DNA extraction were amplified by PCR, underwent electrophoresis and then sequenced. CO1 sequences then were matched with BLAST to obtain sequences from the NCBI that have high levels of homology (>90%). The results showed that the universal primer LCO -1490 and HCO - 2198 success amplify the gene CO1 Apis dorsata of Minahasa. Based on the phylogeny tree, Apis dorsata derived from forest Airmadidi and forest Kombi have the same CO1 sequences but have had differences with Apis dorsata from forests Klabat. BLAST analysis results showed that the partial mitochondrial CO1 sequences Apis dorsata of Minahasa has the highest rate of 94 % similarity with similar sequences recorded in the NCBI gene bank.

Abstract

This study aims to assess The population and insect diversity in the Forest of Klabat. Sequences of this study consisted of determining the location of the sample, the sample unit, and forest. This research was conducted using survey methods, identification, and analysis data. The resulting study showed that insect populations and diversity obtained from the forest of Klabat is a total of 190 individuals consisting of 29 orders and 23 families, with the amount of each individual as follows: 34 Diptera, Hymenoptera 25, 21 Coleoptera, Lepidoptera 18, .and Orthoptera 5. The population the highest of the forest that is on the order Diptera Family Lauxamidae. Insect Diversity in the forest of Klabat at altitude 301 -400 masl is H. =4,76, and in the forest of Klabat at altitude 401 – 500 masl is H. = 2,31.

Abstract

The availability of quality seeds, on time with large amounts is a major problem in freshwater aquaculture. Supplementation of curcumin in female carp aims to increase the reproductive productivity of fish through increased activity of the liver in the synthesis of vitellogenin, a precursor of egg yolk, that play a role in the growth and development of eggs until larvae. This study used 25 female carp, which supplemented with curcuminat several doses: 0; 2.5; 5; 10; and 20 g of curcumin/kg of feed. The collected data was analyzed using a completely randomized design (CRD). The results showed that supplementation of curcumin female carp significantly (p<0.05) affected the malondialdehyde(MDA) concentration of liver, estradiol hormone in the blood, gonadosomatic index, relative fecundity, and larval survival, but did not significantly (p>0.05) affect the superoxide dismutase (SOD), vitellogenin concentration in blood and egg diameter. The study showed that the supplementation of curcumin prevents liver damage showed by a decreased level of liver MDA. The supplementation of curcumin with a dose of 5 g/kg of feed optimizes the productivity and reproductive performance of carp by increasing the production of vitellogenin, the diameter of egg, and the number of survival larva through optimizing the performance of the fish liver.

Abstract

Aedes mosquitoes have the behavior of sucking blood repeatedly (multiple bites) in one gonotrophic cycle, these mosquitoes are also very effective at transmitting the virus to humans (WHO 2004). Kawada et al. (2007) reported that the foraging activity of A. albopictus was less than 0.1 times compared to A. aegypti under laboratory conditions, this result is consistent with the tendency to bite both species in nature. The target-attack frequency (foraging activity) in A. aegypti female mosquitoes in unfed conditions is 30 times greater than that of A. albopictus mosquitoes in several laboratory treatments (Kawada, et al. 2007). Aedes sp is a deadly infectious vector between another dengue hemorrhagic fever (DHF) and chikungunya. Pineleng is a marginal/suburban area of Station 1. Recent studies have reported changes in habitat patterns of Aedes sp, especially Aedes albopictus and Aedes aegypti . The aim of this study is to characterize the population density of the Aagoan sp. Imago based on the time distribution and altitude of the sea surface. The population density characteristics of the Aedes spagoed imago captured inside and outside the room based on the time distribution in North Sulawesi. The stages of the study consisted of larvae surveying, larval extraction and analysis, and imago extraction and analysis. The results showed that the high population of Ae aegypti was at 8:00 a.m. to 10:00 p.m. in the morning and 14:00 to 16:00 p.m. in the afternoon before the afternoon. in the morning before noon and at 14:00 to 18:00 in the afternoon before evening.

Abstract

This study aims to obtain the dominant phytochemical group in polar and non-polar extracts of hibiscus flowers and obtain data on hepatoprotective activity of polar and non-polar extracts of hibiscus flowers on male white rats induced by high doses of paracetamol. This study consists of the stages of extraction, stages phytochemical analysis and test phase of hepatoprotective activity using rat test animals. Treat done is negative control (K -). This group was only given solvents (CMC Na 0.5%) 1ml / head / day for 28 days and was not given heat stress treatment. H0. This group was given solvents (CMC Na 0.5%) 1ml / head / day for 28 days, after which they were given heat stress treatment for eight days. H1 This group was given a polar extract of Hibiscus rosasinensis red flowers at a dose of 0.075g mg / 200gramBB / day in a 0.5% CMC suspension of 1 ml for 14 days, and after that was given heat stress treatment for eight days. H2. This group was given a non-polar extract of Hibiscus rosasinensis red flowers at a dose of 0.075mg / 200gramBB / day in a 1% CMC suspension of 1 ml for 14 days, and after that was given heat stress treatment for eight days. The measured parameters were the content of SGPT and SGOT . The results showed that the polar extract and non-polar extract Hibiscus rosasinensis L. red variety were able to maintain levels of the SGOT and SGPT enzymes in white rats (Rattus norvegicus) induced by high doses of paracetamol, as in the negative control group (K-).

Abstract

Plant Nutmeg (Myristica fragrans) is an endemic plant in Indonesia originating from Banda Maluku Province. Nutmegs have long been known to people as spices. The nutmeg plants have been cultivated in North Sulawesi for a long time. This research has been conducted to obtain the position of species and the reconstruction of the phylogenetic tree of nutmeg plant (Myristica fragrans) from North Minahasa based on chloroplast DNA of matK gene. This research uses a descriptive method with a laboratory experiment. Total DNA was obtained by leaf tissue extraction and purification. The matK gene was amplified using the PCR method and visualized with automatic electrophoresis, sequencing was performed at FIRST BASE Singapore. The result of the analysis of the gene sequence of North Min Minerals using Barcode Of Life Database (BOLD) Systems shows that Pala Wori and Pala Patokaan have 99% similarity with Myristica maingayi and Myristica glubosa. While the reconstruction of the phylogeny tree shows the North Minahasa Pala has the closest resemblance to Myristica fatua.

Abstract

Some areas in Indonesia have local pigs with their own special characteristics. One of them is North Sulawesi which has local pigs that are maintained by the population with a traditional system and some live wild. The liver is the largest gland that has an important function for living things. This study aims to determine the histology of native pig liver from small islands in North Sulawesi. The liver is histopathologically processed with hematoxylin-eosin staining and then the preparation is analyzed descriptively. The results showed a histological picture in the Buhias and Bunaken areas showing necrosis and widening of the sinusoid while histological features in the Kahuku, Bangka, and Nain regions showed that the liver had severe steatosis and cirrhosis of the liver.
