Abstract

Teak is one of economic wood plants, the lack of teak supply has encouraged the expansion of teak plantation. To support the availability of plant material, propagation by tissue culture technique being a good alternative for mass production. This research was aimed at studying the responses of shoot induction and callus of top shoot of teak as result of effect of type of growth regulators, which is BAP and kinetin at MS medium through in vitro method. The study was conducted in the Laboratory Tissue Culture Nursery Permanent (KBP) Department of Agriculture and Forestry Lamongan, with BAP 1 ppm and kinetin 1 ppm on MS medium for callus growth and bud from the apical tip explants teak. Results showed that the growth response of explants apical end teak into callus and shoots. Callus growth on the third day after inoculation and shoots growth on the thirteen day after inoculation. The percentage of explant?s growth on this research is  60.46%; one that consisting of percentage of callus growth is 23.64%, the percentage growth of shoots is 12.79% and explant?s percentage that barren callus and also shoots is 24.03%.

Abstract

Mahogany (Swietenia mahagoni) is among wood plants having economic value, the lack of teak supply has motivated the expansion of mahoni plantation. To support the availability of plant material, propagation by tissue culture technique being a good alternative for mass production. This research was aimed at studying the responsses of shoot induction and callus of top shoot of mahoni as result of effect of type of growth regulators, which is BAP and kinetin at MS medium through in vitro method. Abstract

Teak (Tectona grandis Linn. F.) "JUL" is one of the teak that the able to contribute to the supply of wood raw material. Demand of teak is increasing both at home and abroad, we need a method that could generate a lot of teak plants, rapid and uniform. One way to overcome this problem is by tissue culture techniques. Many factors influence plant growth, one of them is plant growth regulators (PGR), includes 2,4-Dikhlorofenoksiasetat (2,4-D) and 6-Benzyl Aminopurin (BAP). The purpose of this research determined the effect of PGR for textures and colours of teak trunk?s callus. The method used is the visual observation of callus. Data were analyzed by description with comparated textures and colours of callus. Callus have compact textures and different colours from each treatment.

Abstract

Jabon (Anthocephalus cadamba Miq. Ex Roxb.) is one of Indonesia local plants that could potentially be developed for the improvement of the forest is in addition to the other purposes, such as reforestation, reclamation of mined lands. Production and productivity is relatively low due Jabon hard to get the seeds for cultivation techniques are traditionally performed. One way of propagation that can meet the demand of large amounts of seed and uniform growth is a tissue culture system. The purpose of this study is to determine the combination of 2,4-D concentration and the optimal kinetin produced shoots shoots jabon explants. Data were analyzed using one-way Anova to determine the effect of a given treatment, if there is a noticeable difference then will proceed to test Dual Duncan distance. The results showed that the treatment was able to induce MS+1.10-1 mg/L 2,4 D+3.10-1 mg/L kinetin shoots in 2 days the best optimal shoots high by 1.28 cm.

Abstract

Porong Estuary is the estuary of the River Porong, part of Brantas River. Since 1990, the coastal area around Porong River estuary has undergone a change of environment. Environmental changes caused by polluted coastal areas Porong river resulted from the waste of industries which are located around Sidoarjo. Water quality affects the stability of an water ecosystem. This research purposed to determine the quality of the Estuarine Porong based on macrozoobenthic diversity. The research was conducted by observation method by taking samples in 5 stations in Sidoarjo Porong Estuarine by using purposive sampling method. Data were collected at the end of the dry season, on 9 September 2012 at 09.00 am. Chemical and Physical parameters observed consisted of substrate type, acidity of sediment, temperature and brightness of water, level of dissolved oxygen (DO), values of CO2, acidity of water. Macrozoobenthic sampling was done by using petersen grab were then they were identified up to the level of species. Macrozoobenthic diversity calculations using formulas Shannon-Weaner then analyzed the degree of contamination of water based on benthic community diversity index. The results showed that diversity of makrozoobentos are 2,30065 was categorized as middle polluted waters.

Abstract

Krokot (Portulaca oleracea L.) can be used as traditional medicine is to cure skin diseases and diarrhea. This study aimed to determine the phytochemical profile of krokot herb extract qualitatively, and to determine the effect of krokot herb extract on the growth of Staphylococcus aureus FNCC 0047 and Escherichia coli FNCC 0091. Testing the effectiveness of antibacterial calculated using test wells with bacteria S. aureus and E. coli. This study used 6 levels is 50%, 60%, 70%, 80%, 90%, 100%, positive control (ampicillin) and a negative control (distilled water). The results showed that the phytochemical profile krokot herb extract contains tannin, saponin and flavonoid. The existence of the effect of krokot herb extract against the bacteria S. aureus is characterized by the formation of zones 90% inhibition at concentrations of 2 cm, concentration of 100% of 2.2cm, beside of negative control, concentration of 50-70% not make clear zone and E. coli at a concentration of 90% of 0.6 cm, concentration of 100% of 0.9 cm, beside of negative control, concentration of 50-70% not make clear zone. Krokot herb extracts more influence on the bacteria S. aureus than E. coli bacterial wall structure is polar and easily penetrate krokot extract. Krokot herb extract concentration that is 90% and 100% effective which inhibits bacteria S. aureus. Research showed that the higher the concentration of antibacterial agent is given the greater the inhibition zone formed.

Abstract

Plutella xylostella is one of pests which can cause damage of cabbage plants. One of the ways in controlling this pest is using biopesticides, i.e. mahogany and surian leaf extracts. This research aimed to identify the effect of extract leaves of suren, mahogany and suren+mahogany  to mortality and eating activity of Plutella xylostella on cabbage plants. The research used two direction  of ANAVA i.e. the type and extract concentration. The extracts used were from suren, mahogany and suren+mahogany leaves, with concentration 6% suren extract, 8% suren extract, 10% suren extract, 6% mahogany extract, 8% mahogany extract, 10%  mahogany extract, 6% suren+mahagony extract, 8% suren+mahogany extract, 10%  suren+mahogany extract. This research was conducted in 3 replications  using 10 second instar larvae of Plutella xylostella for each treatment. Research parameters observed were mortality and eating activity of Plutella xylostella. Data are analyzed using Varians Analyze. The results of the research showed that the present of mahogany and suren leaf extracts influence the mortality and eating activity of Plutella xylostella. The treatment of 10% suren extract showed a higher influence than other treatments are cause mortality 85% and eating activity 57%.

Abstract

Dragon fruit (Hylocereus undatus) is a plant of high economic value. With increasing consumer demand for dragon fruit, it is necessary to the provision of quality seeds. Effective seed multiplication is done by stem cuttings. One effort to increase the percentage growth of cuttings is to use hormone IBA (Indole Butyric Acid) to stimulate root growth, because the IBA has a more stable chemical content and workability for longer so that it can stimulate root growth. The purpose of this study was to determine the effect of various concentrations of IBA on root growth in cuttings of dragon fruit plants and determine the optimal concentration of IBA for root growth in cuttings of dragon fruit plants. The study was conducted experimentally using a randomized block design with one treatment factor, namely the concentration of IBA include 0 ppm, 500 ppm, 1000 ppm, 2000 ppm and 4000 ppm. Factor is repeated 5 times to obtain 25 samples treatment. Parameters, that were measured, were the percentage of root cuttings, root length, and root biomass. Data was analyzed using analysis of variance (ANOVA) one-way and followed by Duncan test to determine differences among treatments. The results showed that various concentrations of IBA hormone affected positively on root growth of dragon fruit plant cuttings, and the optimal concentration of IBA on root growth was 2000 ppm.

Abstract

Armyworm (Spodoptera litura) is one pest for crops cayenne. Yield losses due to pests has reached 40%. Pest control armyworms can be made using plant-based insecticide that is of mahogany seeds and stems brotowali. The purpose of this study was to describe the effect of the combination of mahogany seed extract and stem brotowali on mortality and feeding activity armyworm pests in pepper sauce, and determine the concentration of a combination of mahogany seed extract and stem brotowali effective pest control armyworms in pepper sauce. The research was carried out experimentally using completely randomized design (CRD) with 6 concentrations of 0 ml/l, 35 ml/l, 40 ml/l, 45 ml/l, 50 ml/l and 55 ml/l, respectively repeated 4 times. The parameters measured were mortality and armyworm feeding activity for 24 hours and observed every 4 hours. Data were analyzed by using Probit Program Version 1.5 continues the analysis of correlation of mortality armyworm larvae. For armyworm feeding activity was analyzed using one-way analysis of variance (ANOVA one-way) and proceed with the Least Significant Difference test (LSD). The results showed that the combination of mahogany seed extract and stem brotowali can cause mortality and a decrease in feeding activity. Effective concentration to control armyworms concentration is 55 ml/l.

Abstract

The main symptom of  diabetes mellitus is an increase of blood glucose concentration, therefore it can be decreased using natural substances. The research aimed to the combination of Caesalpania sappan  L wood and Aloe vera leaves liquids can decrease the concentration  of blood glucose in Mus musculus. This research used  pre-post-test design with RAL. The subjects were 24 Mus musculus of BALB/cAnN strain. The chemical subtance used to increase the concentration  of blood glucose was 1 cc/kgBB of dextrose monohydrate 5 %. The animals were devided into 6 groups that are threated by giving: 1) aquades (KK); 2) Caesalpania sappan L wood liquid  100% (KP1); 3) Aloe vera leaves liquid 100%  (KP2); 4) combination  of Caesalpania sappan L wood and Aloe vera leaves liquid 20% (KP3); 5) combination of Caesalpania sappan L wood and Aloe vera leaves liquid 40% (KP4); 6) combination  of Caesalpania sappan L wood and Aloe vera leaves liquid  60% (KP5). The concentration  of blood glucose was measured using glico test. The data gained were analyzed using T test and One-Way ANOVA at 5% of level test. After giving dextrose monohydrate the glucose concentration of all samples was 160,75 mg/dl.The results of this research shows that the decrease of glucose concentration of   KP1 was 123,75 mg/dl; KP2 was 120,25 mg/dl; KP3 was  118,5 mg/dl; KP4 was 115,75 mg/dl; and  KP5 was 109,75 mg/dl. The analysis result revealed that combination of  Caesalpania sappan L wood and Aloe vera leaves liquids can decrease of the concentration  of blood glucose in  Mus musculus (Fcount (3,499) > Ftable (1,83) with sig value < 0,05) and (tcount (10,939) > ttable  (1,714)) with sig value  < 0,05) . It can be concluded that the combination of Caesalpania sappan  L wood and Aloe vera leaves liquids can decrease the concentration  of blood glucose in Mus musculus.
