Abstract

Fractionation of tannic compound from star fruit leaves was conducted by maseration using acetone : water(7:3), and analytical and preparative thin layer chromatography using a variety of eluents. The isolates obtained wereidentified using UV-Vis and FT-IR Spectrophotometer.The best eluent for both analytical and preparative TLC was found to be n-butanol : water : acetic acid(4:1:5), which gave three isolates with Rf of 0.53, 0.61, and 0.68. The second isolate (Rf = 0.61), showed a maximumabsorption at 331 nm on the UV-Vis spectra. Its IR spectra showed specific absorptions such as OH-assymetricstretch (3372.2 cm-1), aromatic overtone (2071.8 cm-1), and aromatic and benzene stretches at 1625.8 cm-1 and 782.5cm-1 respectively. These spectra suggest that the tannic compound was either flavan pent-3,6,7,4',5'-ol or flavanpent-3,7,8,4',5'-ol.

Abstract

Studies on biosorption of chromium (VI) on green coconut coir fibre (Cocos nucifera) biosorbent have beencarried out. These studies included determination of biosorben acidity, optimum pH, contact time of biosorption,isoterm and biosorption capacity, and interaction type between chromium (VI) and green coconut coir fibre (Cocosnucifera) biosorben Mechanisms of interaction were determined through study on desorption chromium (VI) of thepreviously adsorbed using aquadest, 1 M HCl and 0,05 M Na2EDTA.The result showed that the total acidity of green coconut coir fibre biosorbent was 8,7158 ± 0,2569 mmol/g,the optimum pH was 2, and the contact time was 120 minute. Biosorption capacity of green coconut coir fibre (Cocosnucifera) to chromium (VI) was 12,6152 mg/g. Interaction type that occurred between chromium (VI) ion and greencoconut coir fibre biosorben (Cocos nucifera) were hydrogen bond, Van der Walls bond, cation exchange andcomplex bond.

Abstract

A research on corrosion rate and inhibition mechanism of pure aluminum using potassium stearate andcalcium stearate has been carried out. The objectives of this research were to explore the type of inhibitor ofpotassium stearate and calcium stearate, to understand the reaction mechanism of corrosion inhibition of aluminum,and to study the effect of the salts toward corrosion rate of aluminum in acidic condition.Potassium and calcium citrates were found to inhibit the corrosion of pure aluminum at concentration of 60ppm, in acidic (pH 3) condition, at 25oC. The inhibition efficiencies and relative corrosion rates were : 76 %; 0,31(calcium stearate) and 73 %; 0,31 (potassium stearate). The result of the study on surface adsorption, bychromatography and spectroscopy suggested that the aluminum inhibition mechanism by the potassium stearate andcalcium stearate anions was due to the formation of surface film as a consequence of the formation of complexbetween aluminum surface atoms / ions with stearate (monodentate ligans) which donate their lone pair electronstoward aluminum.

Abstract

It has been done screening fitokimia and the determination of free antiradical activity extract methanol ofgaharu leaf. The screening fitokimia determined by the colour test and free antiradical activity determined bydiphenyl pykril hydrazil (DPPH) method according to sphectrophotometry UV-Vis, before that the honey was dilutedwith methanol and then the absorbance was measured on 497nm, 517 nm, and 537 nm. The result showed thatmethanol extract of gaharu leaf contain phenol, flavonoid and terpenoid compound and the free antiradical activityshowed inhibitor percent is 106,32 % (5”) and 111,31 % (60’).

Abstract

A Gene encoding -L-arabinofuranosidase (abfa) that had been cloned into recombinant E. coliDH5a/pTP510 was subcloned into Saccharomyces cerevisiae yeast system. The aim of subcloning of abfA gene intoS. cerevisiae is to express the thermostable -L-arabinofuranosidase (AbfA) enzyme in the host that is often termedas Generally Recognized As Safe (GRAS), so that this enzyme earn the broader application like in food and beverageindustries. The gene of abfA was subcloned by the amplification of Polymerase Chain Reaction (PCR) from plasmidpTP510 templat. A pair of primers, pFSacI-Af (forward) and pXhoI-Af (reverse) from designed this research wasused for the amplifcation. The PCR condition was performed as follows : beginning denaturation at 94oC for 5 min;PCR cycle 30 times that consisted of denaturation (94oC for 1 min), annealing (55oC for 30 s), andpolymerization/extension at 72oC for 2 min; and than final extension at 72oC for 7 min. The abfA gene, resulted wasinserted between GAL1 promoter and CYT1 terminator in the pYES2 expression vector. This ligation product wastransformed into E. coli TOP10 host. Restriction analysis of recombinant plasmid from this construction, thedesignated as plasmid pY-Af, showed the expected size, about 7,4 kb, which was the summation of sizes of parentalplasmid ( 5,9 kb) and fragment of DNA insert (1,5 kb).

Abstract

Isolation and caracterisation of antibacterial compounds in the leaves pepe have been carried out. Extractionwas carried out by macerating using methanol and 70%. Separation of extracts which have activity to Eschericia coliand Stathylococcus aereus was conducted using the solvent partition by petroleum ether, chloroform, and ethylacetate. The most active extract, the ethyl acetate extract was separated by column chromatography using a mixtureeluent of chloroform, and ethyl acetate with a ratio of 1:1. The eluents were tested for their bioactivity asantibacterial agent and the results established that fraction 1 was the most active fraction. This next wing fraction 1was then identified using GC-MS.The gas chromatogram showed 3 peaks at 18.658 minutes (33.69%), 19.617 minutes (34.01%), and 25.492minutes (32.69%). It was suggested that the fraction contains three compounds. The first peak has a molecularion atm/z 296 with a base peak of 68, matches with the spectra of 3,7,11,15-tetramethyl 2-hexadesene-1-ol. The secondpeak, m/z 270 and 74, matches with the spectra of metyl hexadecanoate ester. The last peak has amoleculer ion at390 and base peak at 149, suggesting it is dioctyl-1,2-benzenedicarboxylilate.

Abstract

This research was performed to analyze the aluminium content in several wasted cans. The analysis usedAAS at 309.3 nm. This method was chosen since its operation is relatively simple but capable of analyzing low metalcontents (less than 1 ppm). The aluminium content of wasted cans ranges from 1.41% up to 16.04%. The aluminiumwas used to make alum, with the percentage of yield of 77.04% up to 96.81%, depending on the type or cans. Inother words, one gram of wasted cans produced 0.2335 grams up to 2.6857 grams of alum. The produced alum wascapable of purifying water.

Abstract

The effectivity of waste water treatment systems in Suwung Waste Water Treatmant Installation Denpasar,was investigated, by analyzing its effluent Biological Oxygen Demand (BOD), Chemical Oxygen Demand (COD)and ammonia contents. The aims of the research are to determine the effectivity of each stage of waste watertreatment and the optimum detention time of waste water in aerated lagoon and sedimentation pond, in order toreduce the BOD, COD and ammonia concentration.The results showed that the effectivity of treatment systems of waste water in Suwung Denpasar was low.The percentages of the reduction of BOD5, COD and ammonia in content stage I were 14.29, 15.66, and 33.74%respectively. More over, the reduction percentages in stage II were 20.83, 14.43, and 17.39% respectively. On theother hand, the reduction percentages from the initial to final stages were 32.14% for BOD5, 27.83% for COD and45.26% for ammonia. The optimum detention time of waste water in aerated lagoon and sedimentation pond forreducing parameters was 5 day and 40 hour respectivel

Abstract

A compound with antifungal activity has been isolated from dringo (Acorus calamus L.) essential oil. Steamdistillation was conducted to isolate the oil and resulted in 0.5 % yield. Antifungal activity test towardsBotryodiplodia theobromae showed that the ethanol posseses antifungal activity with an inhibition capacity of 94.4%. GC-MS analysis resulted in three main peaks at 15.008; 15.233; and 16.490 minutes. It was suggested from thepeak intensity that the last peak represents the major component of the extract. Futher more according to the librarydata base, the spectra of this compound matches with the Asarone spectra.

Abstract

Kepuh leaves can be used as traditional medicine to cure various diseases. Maseration of ± 1000 g drypowder of Kepuh leaves with n-hexane as the solvent resulted in 2,10 g n-hexane concentrated extract. Separation ofthis extract with column chromatography gained two yellow amorphous crystal isolates of F3.1 and F3.2 with 0,0195 gand 0,0032 g in weight, respectively.The result of phytochemical test of isolate F3.1 with Liebermann-Burchard reagent showed that the isolatepositively contained triterpenoid. Identification of isolate F3.1 with spectrophotometer UV-Vis showed one broadpeak at 265 nm. The transition occured probably was n *, that was caused by C=O chromophor. Infraredspectrum showed characteristic groups of OH at 3294,42 cm-1, aliphatic CH at 2924,09 cm-1 and 2854,65 cm-1, C=Oat 1743,65 cm-1, bending CH2 and bending CH3 at 1458,18 cm-1 and 1381,03 cm-1 and alcoholic CO at 1172,72 cm-1.Free antiradical activity test against isolate F3.1 was carried out by spectroscopy UV-Vis method. Isolate F3.1 hadcharacteristic as a free antiradical with DPPH damping percentage at the 5th minute was 85,33% and at the 60thminute was 88,52%.According to phytochemical tests and spectrophotometry analysis, isolates from kepuh leaves was expectedto contain true triterpenoid group compounds with free antiradical activity OH group, aliphatic CH, C=O andalcoholic C-O as the functional groups.
