Abstract

ABSTRACTThis study aims to determine the cross-reaction between the antigen of avian influenza (AI) H5N1 virus clades 2.1.3 and 2.3.2 in duck and chicken sera, which were vaccinated with inactivated AI H5N1 clade 2.1.3 vaccine against AI H5N1 clade 2.3.2 antigen and those vaccinated with inactivated AI H5N1 clade 2.3.2 vaccine against H5N1 clade 2.1.3 antigen. The sera tested were obtained from postvaccination and control (unvaccinated) chickens and ducks in the laboratory condition, and from AI H5N1 postvaccination ducks in the field condition. HI test was conducted by using AI H5N1 clades 2.1.3 and 2.3.2 antigens. The results of HI titer were analyzed by the geometric means and by ANOVA. The results show that cross-reactions in both chicken and duck sera after AI H5N1 clade 2.3.2 vaccination tested with AI H5N1 clade 2.1.3 antigen occurred with low antibody titers, whereas in chicken and duck sera postvaccination with avian influenza H5N1 virus clade 2.1.3 showed cross-reaction with high antibody titer against clade 2.3.2 antigen. The conclusion of this study, postvaccination sera of AI H5N1 clade 2.1.3 provide better cross-reaction compared to the postvaccination sera of AI H5N1 clade 2.3.2.Keywords: sera, chicken, duck, AI H5N1, clade 2.1.3 and clade 2.3.2.

Abstract

ABSTRACTThe rich of Indonesian biodiversity becomes an attraction for local and foreign researchers to conduct research in this country. One of scientific productivity indicators that mostly used is number of publication in scientific journal. The previous studies had never analyzed scientific productivity of Indonesian biodiversity researchers, both institutionally and individually. This paper analyzes 2,432 articles concerning Indonesian biodiversity research affiliated with institution from Indonesia (published in 1990-2015) from Scopus database, including 8,544 authors from 1,638 institutions. Data pre-processing resulted publication year, number of author, and author’s institution location. Then, they were analyzed using scientometrics analysis. Scientometrics analysis is a quantitative method related to measurement of the science communication. The result shows that internation-al collaborations produced most of scientific publications on Indonesian biodiversity. The number of scientific publication on Indonesian biodiversity was significantly increase and showed exponential growth trend. Indo-nesian author mostly wrote articles from Animalia kingdom. Multi and mega authors wrote the most of scien-tific publications on Indonesian biodiversity.Keywords : Indonesia, biodiversity, scientometrics, productivity, scientific publication, Scopus

Abstract

ABSTRACTPerifiton is one community of organisms that can adapt and thrive in the river, and relatively settled in one location. Riparian vegetation in the Mountain Halimun-Salak National Park could be expected to affect the community structure of periphyton. The purpose of this study was to identify the community structure of periphyton on different vegetation canopy coverage, to analyze relationship between the communities structure of periphyton and upstream Cisadane water quality conditions, and to determine water quality based on perifiton community. There were three sets three stations based on canopy coverage (80%, 60%, dan 40%). The results showed two groups of stations, the high canopy coverage (80%) and the low canopy coverage. The highest perifiton abundance was found at stations low canopy coverage, that dominated by Bacillarophycae, Pennales order, especially Navicula with higher rate of succesion than the higher canopy caverage. Abundance of periphyton of low canopy coverage was affected by ammonium, turbidity, and water current. Furthermore, in stations high canopy coverage it was influenced by orthophosphate and currents. As a whole, the periphyton community at upstream of Cisadane indicated a good condition of water quality.Keywords: canopy, periphyton, succession rate

Abstract

ABSTRACTA total of 781 base pairs of mitochondrial DNA ND2 gene was analyzed to investigate their divergence in the Arachnothera longirostra (Nectariniidae) birds in Jawa and Sumatera islands of Indonesia. Blood and tissue samples were collected from 27 birds consisted of 8 samples from T.N.G. Halimun, 6 samples from T.N.G. Gede Pangrango, 6 samples from T.N. Ujung Kulon, 1 sample from Dieng (Jawa), and 6 samples from Jambi (Sumatera). Total genomic DNA was extracted from each sample and used in PCR to amplify a single fragment of mDNA ND2 gene. Their DNA sequence data were compared with those from Johor (Malaya Peninsula) and Sabah-Sarawak Borneo (GenBank). There were 61 variable sites included 34 parsimony sites and 27 sequence haplotypes. Mean genetic distance or sequence divergences within bird populasions were ranged from the lowest (0.25 %) for Ujung Kulon to the highest (0.76 %) for G. Gede Pangrango; while mean genetic distance between populations were of 1.67% to 2.82 % for Java island vs Jambi (Sumatera), and 1.06 to 2.09 % for Java vs Borneo. Phylogenetic NJ tree showed that there were two main clades, i.e. 1) birds of Jambi (Sumatra), Johor (Malay Peninsula), Sabah-Sarawak (Borneo), and Ujung Kulon (Java), and 2) birds of G. Gede Pangrango, G. Halimun, and Dieng (Java). Past geologic event may influenced genetic divergence occurred within the populations. Moreover, geographic features such as mountain may played role for population isolation as showed on the Little Spiderhunter bird from G. Halimun, G. Abstract

ABSTRACTThe purpose of this study was to investigate the effect of Tokay gecko saliva on morphology and angiogenesis response on the healing process of skink tail wound and also to characterize the protein profile of Gecko saliva. Twelve skinks were autotomized and wound surface of tail smeared by young gecko saliva, adult gecko saliva, and human’s saliva twice per day and control. The morphological changes of the wound surface were observed. The angiogenesis response was observed in vitro using Chorioallantois Membrane (CAM) of the ninth days chick embryos. Protein profile of gecko saliva analyzed with SDS-PAGE. Generally, treated wound showed a better healing. Young gecko saliva able to stimulate angiogenesis in wound healing stage of sun skink tail after autotomy. Saliva protein of young and adult Gecko differences was not only in the size (or density) but also in the number of the bands. The young and adult Gecko revealed a striking consistency of protein patterns, indicating a profound physiological stability of the whole saliva. This study showed that Gecko saliva able to accelerate wound healing, thus Gecko saliva may have potential me as a future therapeutic for treating infected wounds.Keywords: Tokay gecko saliva, wound healing, sun skink tail, angiogenesis, protein profile.

Abstract

ABSTRACTThe area of reclamation and revegetation in farmer coal mine has one role as bird habitat. The purpose of this study was to determine indicators of the types of bird that can progress successed reclamation and revegetation program in habitat function of fauna. The method used was bird species and bottom vegetation identification I in each habitat type used transect and mistnet. The data was analyzed using multivariate analysis of hDCCA. The results using hDCCA preedicted that the key indicator of bird characteristics that marks the development of succession in the fifth year is the presence of “insectivore bird” type. Bird species were B15 (Lanius schach), B20 (Prinia flaviventris), B23 (Picoides moluccensis), B28 (Pachychepala hypoxantha), B29 (Rhipidura javanica), B30 (Orthotomus ruficeps), B31 (Orthotomus sericeus).Keywords: hDCCA, insectivore bird, key indicator, reclamation and revegetation,

Abstract

ABSTRACTStevia rebaudiana is plant species producing natural sweetener with has low calories. The species propagation could be done by tissue culture tecnique to obtain propagules with high quality and sustainability. Modification of media composition and in vitro environment will increase growth and vigority of explants so that they have high survival rate during acclimatization. The aim of this research was to increase growth of stevia shoot culture by increasing the level of vitamins in combination with different type of test tube covers. Stevia shoot tips were cultured on MS medium containing normal concentration of its vitamins (control treatment; Myo-inositol 100 mg/l; Nicotinic acid 0.5 mg/l; Pyridoxine-HCl 0.5 mg/l; Thiamine-HCl 0.5 mg/l and Glycine 2 mg/l), twice and 4 folds of vitamin levels, they were grown on culture tubes with Al-foil and ventilated-plastic with filter (2 cm diameter and pore size at 0,22 micron). Height of shoots, number of nodes, number of leaves, number of roots were observed every week till 8 weeks of culture. Biomass (fresh and dry weights) and chlorophyll level and acclimatization were done 8 weeks of culture. The results showed that type of culture tube covers affected significantly to all growth parameters, biomass as well as level of chlorophyll, meanwhile level of vitamins only affected number of nodes, shoots and roots. Interaction between vitamin level and covers types only occured for height of shoots and number of roots. Plantlets grown on medium containing 4 fold of vitamin level (Myo-inositol 400 mg/l; Nicotinic acid 2 mg/l; Pyridoxine-HCl 2 mg/l; Thiamine-HCl 2 mg/l and Glycine 8 mg/l) with ventilated-plastic cover had larger leaves compared to other treatments. All plantlets survived in a greenhouse.Keywords: Stevia rebaudiana, in vitro growth, increase in vitamin concentration.

Abstract

ABSTRACTNitrogen based fertilizers such as urea and NPK are primary needs for rice farmers. To get significant improvement of crop yield, the more quantity of fertilizers are applied. It make negative impact for surrounding environment. Based on that, the efforts should be done to suppress the demand of fertilizers such as by developing Nitrogen Use Efficiency crops. CsNitr1-L is one of gene that related to Nitrogen Use Efficiency trait in plant. The objectives of this research are to develop the construction of CsNitr1-L gene candidate in pCAMBIA1300-Ubi1 promoter and to obtain the transformants of rice cultivar Nipponbare which contain the construction of CsNitr1-L gene candidate. The construction of pCAMBIA1300::Ubi1::CsNitr1-L has successfully assembled and was transformed to immature embryo of rice cultivar Nipponbare using Agrobacterium tumefaciens strain LBA4404. It was obtained 146 lines of T0 Nipponbare. PCR analysis of T0 Nipponbare lines showed that 66 of them was identified as positive T0 lines contained hptII and CsNitr1-L genes. Transformation efficiency obtained was 11,9%. The result of analysis copy number using Southern Hybridization in positive PCR of T0 lines randomly indicated that 4 lines have a single copy of transgene. Based on these results, it can be concluded that CsNitr1-L gene construct was successfully introduced into the genome of the rice plant cultivar Nipponbare and the positive PCR of T0 lines containing the gene of hptII and CsNitr1-L, also a single copy of the transgene was obtained.Keywords: Nitrogen use efficiency trait, gene construction, rice kultivar Nipponbare

Abstract

ABSTRACTThe aims of the research was to analyze nutrient requirements, performance, and feed efficiency of porridge formulation on sugar gliders (Petaurus breviceps) based on daily feed intake and digestibility. The research was conducted at the Small Mammal Captivity of Zoology Division, Research Center for Biology, Indonesian Institute of Sciences (LIPI) Cibinong, Bogor. Descriptive method was used in this experiment, with 14 days of preliminary period and 70 days (10 weeks) of data collection. The research materials were 16 sugar gliders aged about 3-5 months and randomly divided into 2 treatments ie. 8 heads were given porridge formulation I (P0 - control treatment) and 8 heads were given porridge formulation II (PI) plus superworm 2 times a week. Estimation of feed digestibility done in vivo with the method of the feces total collection. Variables of this experiment are daily feed intake, nutrient requirement, total digestible nutrients (TDN), daily weight gain, and feed efficiency. The feedstuff of the porridge formulation consist of siamese banana (Musa sp.), watermelon (Citrullus lanatus), sweet potatoes (Ipomoea batatas), grape (Vitis vinifera), pear (Pyrus pyrifolia), boiled egg yolk, baby porridge, calcium, honey, boiled chicken meat, and superworm (Zophobas atratus). The results showed dry matter intake was 5.82 g/head / day or 29.31 g/head / day BB0.75 (P0) and 7.31 g/head/day or 34.16 g/head/day BB0.75 (PI). The average of body weight gain was 0.55 g / head / week (P0) and 0.73 g/head/week (PI) with feed efficiency ratio was 1.36% (P0) and 1.43% (PI). The PI treatment of porridge formulation II has improved the performance of sugar gliders in captivity.Keywords: Daily feed intake, weight gain, porridge formulation, sugar gliders, performance

Abstract

ABSTRACTBrek is a consumption fish belong to barbs group that widely distributed, one of them is in Serayu river. The aims of this study was to reveal the characteristics of meristic, morphometric, and habitat of the species. The study was conducted on the Serayu river especially in Banjarnegara, Central Java Province. Sampling was conducted on May 2012 to June 2013. The area of study was divided into three zones and the zones was categorized based on Reservoirs at Mrica area these are upper zone, reservoir zone, and below the reservoir zone. Fishing gear used are cast net, elektorfishing, and gill nets. Samples of fish were caught preserved in 4-10% formalin solution and then observed in the laboratory. Morphometric characters observed were 24 characters. The results were standardized to standard length, then analyzed with discriminant methods. The structure of the scales and number of ridges on the back of the last dorsal fin spines also observed. The results meristic characters are 4.8 dorsal fin, 3.5 anal fin, 1.14-16 pectoral fin, and predorsal 10-13 scales, also there were no spots on the caudal peduncle. The results of discriminant analysis showed known five characters distinguishing three species near 100%. The habitats have rocky bottom, rapid current, and divide into two groups.Keywords: barb fish, meristic, morphometric, Serayu river
