Abstract

ABSTRACTCellulalytic Activity of Candida sp and Debaryomyces sp Isolated from Peat Soil ofBukit Duabelas Natural Park, Jambi. Candida sp. and Debaryomyces sp. YeastDegrading cellulose Isolated from Peat Soil in Bukit Duabelas National Park, Jambi. Theobjective of study was to investigate the characteristic of yeast that are able to solubilizecellulose. The yeast was isolated from soil of Tarnan Nasional Bukit Duabelas Jambi.Out of 2 1 ioslates tested 2 isolates Candida sp. (Isolate J I) and Debaryomyces sp. (IsolateJ2) were able to solubilize Carboxymethyl cellulose. Other isolated soil yeasts wereRhodotor& sp, and C~ptococcusp . Medium acidity during cell cultivation variedbetween 5.8 to 7.2. The CMC-ase activity was 5.9 unit and 5.4 unit for isolate J1 andisolate 52 respectively. The Km of isolate J1 and isolate 52 were 7.7 x 10" and 8.4 x 1 0-2(% b/v) respectively. Abstract

ABSTRACTEffectivity of EDTA (EthylenediaminetetraceticAcid) in Increasing Lead Accumulationof Saccharum spontaneum Grown Under Gold Mine Waste. Lead (Pb) isknown as one of a major metal contaminant in mine tailing. This metal has low solubility,and in many cases, is not readily available. In most soils capable of supporting plantgrow, the soluble Pb2+ levels are relatively low and will not promote substantial uptake bythe plant. In addition, many plants retain Pb2+ in their roots with only minimal trabsportto the aboveground plant portions. Therefore, it is important to find ways to enhance thebioavailability of Pb2+ or to find specific plants that can better translocate the Pb2+ intoharvestable portions for phytoremediation. The success of Pb phytoremediation is tohave significant Pb availability as well as a large quantity of plant biomass with high rateof growth. This experiment was conducted to obtain both purposes. Saccharumspontaneum which proven tolerant and dominant in contaminated area as well as potentialin producing high biomass was used in this research.The plants were grown in wastemedia added by 100 ppm, 200 ppm and 300 ppm of Pb. Ethylenediaminetetracetine Acid(EDTA) was applied to increase Pb availability and plant uptake as well as translocationto the aboveground portions. The results showed that the plant still capable of growingunder the highest level of Pb. EDTA increase Pb availability and plant uptake. Pb accumulationin the aboveground biomass of EDTA treated plants were relatively higher thanuntreated plants.Key words: Timbal, EDTA, phytoremediation

Abstract

ABSTRACTTrend and Causal Factor of Deforestation in Way Pemerihan, Bukit BarisanSelatan National Park, West Lampung. Deforestation of low land tropical forestis one of the greatest threats to biodiversity in Bukit Barisan Selatan National Park(BBSNP), where this area is the third-largest protected area on Sumatra. Our study is thefirst in BBSNP as integrate between remote sensing and socio-economic survey to demonstratethat the area and rate of forest loss in Way Pemerihan (Cawangaro andSumbersari). The data demonstrated that deforestation activity has been started since1965 in Cawangaro and 1977 in Sumbersari. Forest cover of 1.232ha had already beenlost by deforestation activity, i.e: 579 ha in Cawangaro and 653ha in Sumbersari. Forestloss is increasingly by average rate of 1.2%/year (0.28m2/minute) in the meantime averagerate of regeneration only 0.09%/year. The highest increased of deforestation hasoccurred after politics reformation in 1998. The result of socio-economic survey demonstratedtwo causal factor include internal factor and external factor. Internal factor that islandlessness, price of outside BBSNP land is relatively high and not productive becauseof that low production. External factor of the processes which drives farmer to deforestationis social gap to illegal logging activity, declined of BBSNP forest control and lawenforcement capacity, and boundary conflict between BBSNP with traditional lands.Political reformation forms is the basis for increase external factor.Keywords: Forest loss, satellite imagery, landscape, Illegal logging

Abstract

ABSTRAKAktivitas Induksi Superoksida Dismutase dan Resistensi Etanol oleh Sodium Kloridadan Plumbum Asetat pada Khamir Candida sp. Y390. Untuk mengetahui sifat khamirCandida sp. Y390 dalam melindungi diri dari cekaman oksidatif, telah dilakukan risetkajian toleransi sel terhadap etanol dan hubungannya dengan enzim superoksida dismutase(SOD) pada khamir tersebut. Viabilitas sel yang ditumbuhkan pada media gliserol dandiinkubasi pada etanol 17,s % selama 1 jam adalah 0.30 f 0.09 % , pada media gliseroldan 1 1,7 % natrium khlorida adalah 1.65 f 0.5 % sedang pada media giserol dan 1 ppmplumbum asetat adalah 1.16 f 0.7 %. Aktivitas CuZnSOD pada sel yang diinduksi olehnatrium khlorida atau plumbum asetat turun sedikit, yakni masing-masing 1,7 dan 1,9kali. Aktivitas MnSOD pada sel yang diperlakukan dengan natrium khlorida naik sedikit(sekitar 1,03 kali), namun pada sel yang diperlakukan dengan plumbum asetat naik secaramenyolok (sekitar 3,6 kali). Data ini menunjukkan bahwa metabolisme etanol dapatmenyebabkan Candida sp. Y390 mengalami cekaman oksidatif. MnSOD khamir mungkinberperan dalam melindungi sel dari kerusakan akibat cekaman oksidatif.Keywords: Candida sp. Y390, yeast, ethanol metabolism, oxidative stress, SOD.

Abstract

ABSTRACTGrowth of the Groupers (Serranidae) in Floating Net Cages at The Coastal WatersKodek Bay Malaka Village West Lombok. Study of rearing of Bararnundi cod(Cromilileptes altivelis), flowered cod (Ephinephelus fuscogutatus) and estuarygrouper (E. tauvia) at floating net cages was canied out on June to December 2004at the coastal water Kodek Bay MalakaViiage, West Lombok. There was sigdicantdifference in weight growth rate of groupers during observation (X count = 67.420and X2a 0.05 = 2 1.026 or X count > X2a 0.05) during observation, but not in length (X2count = 0.998 and X2a 0.05 = 2 1.026 or X count < X2a 0.05). Observed physicochemicalproperties of experimental set up are water temperature between 27.2 - 27.8 OC, salinitybetween 3 1.8 - 33.8 %o, transparency between 10.3 - 14.5 m, air temperature between26.6 - 27.28 OC and pH about 8.3 - 8.4.Key word: Groupers fish of growth, waters condition

Abstract

ABSTRACTImprovement of Bracirwnuspficatifis Mueller 1786 Static Culture Productivity byOptimization of Food vpe and Food Concentration. A study to optimize the standardculture procedure by optimization of feeding regime (selection of the best type ofmicroalgae and its concentration used for diet of Brachionus plicatiliz diet) wasconducted in order to support more stable and higher yield production of B. plicatilisculture. A serial experiment was done in two consecutive stages. On each stage, B.plicatilis zooplanktons were cultured in batch system of 30 x 30 x 50 cm3 aquarium,initial density of 10 ind./mL, and constant aeration rate of 200 mLIminute. The density,egg ratio, water quality parameters of the culture (dissolved oxygen, pH, ammoniumlevel, nitrite level, nitrate level) were measured daily. On the first experiment, B. plicatiliscultures were fed on lo6 cells/mL Chlorella sp., Nannochloropsis sp., Tetraselmis sp.,and a combination of those three micro-algae. The results showed that optimum culturedensity was obtained in the culture fed on Nannochloropsis sp. (68*2) indImL on day 5of culture period and the density was significantly different from the other treatments(pc0.05). On the second stage, B. plicatilis cultures were fed on Nannochloropsis sp. infour different concentrations: 1 OS, 1 06, 1 07, and 1 08cells/mL. Optimum culture density(407k9) ind./mL was obtained in the culture fed on 1O7ce1ls/mL Nannochloropsis sp. onday 7 of culture period and the density was significantly different from the other treatments(p<O.O5). It can be concluded that the optimum culture condition (optimum density andmore stable water quality parameter such as NH; and NO2 of B. plicatilis batch culturescan be obtained by optimization of feeding regime using 1 O7 cells/mL NannochloropsisSP .Key words: Brachionus plicatilis, Nannocloropsis sp., Tetraselmis sp., Chlorella sp.,microalgae concentration, water quality, productivity.
