Abstract

Flour to water ratio in batter compositions affected water availability which was needed to provide physical and chemical changes during fresh rice-based spring rolls wrappers processing, such as gel forming of starches and heat-induced gels, flour’s components interactions in batter systems. Degree of water-starch, water-protein and protein–starch-water interactions were depend on water amount, temperature and duration of heating. The mechanical strength of spring rolls wrappers is one of problems when it is being used. The wrappers could be torn apart due to moisture absorption from the filling and the environment. The goal of this study was to determine the optimum flour to water ratio in formulation of fresh rice-based spring rolls wrappers. The investigation was provided by Randomized Completely Block Design with single factor and three replicates. The factor was rice flour to water ratio in six levels (3.0:4.5; 3.0:5.0;3.0:5.5; 3.0:6.0; 3.0:6.5; and 3.0:7.0) the data were analyzed by Analysis of Variance with 95% degree of confident. Flour to water ratio greatly influenced elongation at break which is important in the utilization of fresh rice-based spring rolls wrappers. Its ratio also influenced the size of swelled rice starch granules, pores size and moisture content of the products. Optimal ratio flour to water is 3.0:6.0 which produced the highest elongation at break.

Abstract

The hypoglycaemic effect of water soluble polysaccharide (WSP) extracted from gembili tuber was studied in this experiment. Extracts from three different methods, an aqueous (water-WSP), papain assisted (papain-WSP), and tempeh inoculum assisted (tempeh-WSP) extractions, were compared. The effects were evaluated by means in vivo test on hyperglycaemic-induced rats by performing the so called blood glucose response, followed by in situ glucose absorption test and short chain fatty acids (SCFA) analysis. A nested experimental design was employed in the experiment. All the three extracts reduced the blood glucose level. The effect of tempeh-WSP extract was comparable to that of the papain-WSP extract, but was not signicantly different from the water-WSP. However, the water-WSP extract tended to have a weaker effect than that of the other extracts. Similar result was found on the SCFA level. The highest SCFA level was found on the rats treated with tempeh-WSP extracts. It was likely that the hypoglycaemic effect of the tempeh-WSP extract was the best.

Abstract

This study aimed to determine the hypolipidemic effect of water soluble polysaccharides (WSP) of gembili tuber directly extracted with water (WSPc) and WSP, which was extracted with deproteinase process using protease enzymes from Aspergillus oryzae (WSPd). WSPc and WSPd were given to male wistar rats aged 2-3 month  were analyzed at total cholesterol, triglycerides, LDL cholesterol (Low Density Lipoprotein), HDL cholesterol (High Density Lipoprotein), digesta pH and SCFA (Short Chain Fatty Acid) of rat caecum digesta. The study was conducted for 4 weeks, rats were divided into 3 groups: control, WSPc and WSPd. Each group consisted of 6 rats, each group of rats was fed the same of the standard AIN-93M diet. WSPc and WSPd were given every day by forced feeding method at dose of 40 mg/100g weight of rats. A nested experimental design was employed in the experiment.  The results showed that the deproteinase process using Aspergillus oryzae protease 0.05% could reduce the protein levels of WSP gembili tuber 49.6%. PLAd was more potent hypolipidemic with WSPc and control. Giving WSPd during testing in rats can reduce the total cholesterol (52.92%), triglycerides (40.8%), LDL cholesterol (92,98%) and increased HDL cholesterol (46.95%). Fermentation of WSP in rats caecum produce SCFA  in the form of acetic acid, propionic acid and butyric acid.ABSTRAKPenelitian ini bertujuan untuk mengetahui efek hipolipidemik polisakarida larut air (PLA) umbi gembili yang diekstrak langsung dengan air (PLAc) dan PLA yang diekstrak dengan proses deproteinasi menggunakan enzim protease dari Aspergillus oryzae (PLAd). PLAc dan PLAd diberikan pada tikus wistar jantan umur 2-3 bulan untuk diuji kolesterol total, trigliserida, kolesterol LDL (Low Density Lipoprotein), kolesterol HDL (High Density Lipoprotein), pH digesta, dan SCFA digesta caecum tikus. Penelitian dilakukan selama 4 minggu pengujian, 18 ekor tikus dibagi menjadi 3 kelompok yaitu: perlakuan kontrol, PLAc dan PLAd. Setiap kelompok terdiri dari 6 ekor tikus, masing-masing kelompok tikus diberi ransum  yang sama yaitu diet standar AIN-93M. Pemberian PLAc dan PLAd dilakukan setiap hari selama pengujian dengan bantuan alat sonde (force feeding) dengan dosis 40 mg/100g berat tikus. Data yang diperoleh dianalisis dengan rancangan percobaan Rancangan tersarang (Nested Design). Hasil penelitian menunjukkan bahwa proses deproteinasi menggunakan protease Aspergillus oryzae 0,05% dapat mengurangi kadar protein PLA umbi gembili sebesar 49,6 %. PLAd lebih berpotensi sebagai hipolipidemik dibandingkan dengan PLAc dan kontrol. Pemberian PLAd umbi gembili selama pengujian pada tikus hiperlipidemia dapat menurunkan kolesterol total (52,92%), trigliserida (40,8%), kolesterol LDL (92,98%) dan peningkatan kolesterol HDL(46,95%). Abstract

This research was aimed to study hydrolysis conditions of houstorium lipases enzyme using coconut oil as substrate. Hydrolysis conditions studied were substrate (coconut oil) concentration, enzyme substrate ratio, duration of hydro- lysis and effect of stirring to hydrolysis. The results show  that lipase of coconut houstorium may be optimally used at a coconut oil concentration of 10 %, enzyme to substrate ratio of 3 : 10 (v/v) and hydrolysis for 60 minutes with stirring.ABSTRAKPenelitian ini mempelajari kondisi hidrolisis minyak kelapa yang optimum menggunakan enzim lipase dari kentos kelapa. Kondisi hidrolisis yang dipelajari meliputi konsentrasi substrat optium, perbandingan enzim : substrat dan lama hidrolisis yang optimum serta pengaruh pengadukan selama hidrolisis. Hasil penelitian menunjukkan bahwa, hidrolisis minyak kelapa menggunakan enzim lipase kentos kelapa menghasilkan asam lemak bebas paling banyak pada kon- sentrasi substrat (minyak kelapa) 10 %, perbandingan enzim : substrat yaitu 3 : 10 (v/v), lama hidroloisa 60 menit dan dilakukan pengadukan selama hidrolisis.

Kondisi hidrolisis yang dipelajari meliputi konsentrasi substrat optium, perbandingan enzim : substrat dan lama hidrolisis yang optimum serta pengaruh pengadukan selama hidrolisis. Hasil penelitian menunjukkan bahwa, hidrolisis minyak kelapa menggunakan enzim lipase kentos kelapa menghasilkan asam lemak bebas paling banyak pada kon- sentrasi substrat (minyak kelapa) 10 %, perbandingan enzim : substrat yaitu 3 : 10 (v/v), lama hidroloisa 60 menit dan dilakukan pengadukan selama hidrolisis.
