Abstract

Vanda tricolor Lindl. var. suavis is an Indonesian wild orchid that has been rare in nature,  so it needs attention to take care and conserve them.  The objective of the research was to investigate the effect of coconut water and / or tomato juice on the growth of protocorm of V. tricolor Lindl. var suavis from Bali grown in vitro. The experiment was laid out in the factorial design, with two factors (coconut water/CW and tomato juice/TJ), each contained three concentration (CW: 0, 100, and 200 cc L-1; TJ: 0, 100 and 200 g L-1) resulted in nine combination of treatments and replicated four times.  The results showed that tomato juice with concentration of 100 gL-1 or 200 gL-1 promotes growth of protocorms of Vanda tricolor Lindl. var. suavis from Bali regardless the presence of  coconut water. Keywords: coconut water, in-vitro, protocorms, tomato juice, Vanda tricolor

Abstract

Abstract

This research was conducted at Tissue Culture Laboratory of Agricultural Faculty, Udayana University, from May to October 2009. The aim of this Study to acquired the best in vitro medium that was to added plant growth regulator. The results showed that plant growth regulator kinetin significantly affected to germination and protallus formation of ata fern. The M1 nedium treatment the fastest of germination formation was 19,33 day after showing (das) while the protallus formation the fastest to M3 medium was 36,17 das. In description the fastest sporofit formed to founded in M3 too (122 das).

Abstract

A research regarding “Runner-tip culture of strawberry (Fragaria x ananassa Duch) Grown on Several Shoot-induction Medium” has been investigated. The objective of the research was to find out the most suitable medium for shoot production from runner-tip culture of strawberry at establishment step of micropropagation. The research was laid out in a Completely Randomized Design, 4 treatments of medium type for shoot induction and 10 replication, each was represented by one (1) bottle with 6-8 explants. The treatments were summarized as follows: T1 = MS ; T2 = MS + 2 ppm BAP + 0.01 ppm NAA; T3 = MS + 1 ppm of TDZ; T4 = WPM + 2 ppm BAP + 0.01 ppm NAA. The parameters observed were days of the bud emergence, the average number of shoots per explant, and the average number of leaves per explant. It can be concluded that among medium used in the current research, the medium of MS added with 1 ppm thidiazuron (TDZ) was the most suitable medium for shoot production of strawberry from the explant of runner-tips. The treatment was resulted in the earliest time of bud emergence, and producing the highest number of shoots and leaves.

Abstract

The aim of the research is to investigate a simple method of in planta transformation method for T-DNA transfer in table grape. The T-DNA harbored the S0SPS1 gene under the control of promoter of the 35S CaMV from the Cauliflower Mosaic Virus and contained the NPTII gene, a kanamycin-resistant gene as a selectable marker for transformant selection. Six-month plants originated from cuttings were used as target plants. We explored two methods of in planta transformation, namely ”dipping” and “sweeping”. For both methods, the leaves of the target plants were removed and those of shoots without leaves were used as the target of transformation. In the “dipping method”, those shoots were dipped with the agrobacterial suspension for 60 seconds. However, for the “sweeping method”, the scars (the spots where leaves were removed) were swept with agrobacterial suspension using a cotton bud. Those treated non-leafy-shoots (from both methods) then were grown to be leafy shoots. Those leafy shoots then were cut and transplanted into the soil and grown to be a whole plant. The leaves of those plants then were taken as samples for DNA extraction and PCR using primers of NPTII gene (Forward: 5’-GTCATCTCACCTTCCTCCTGCC-3’; Reverse: 5’ GTCGCTTGGTCGGTCATTTCG-3’) with expected amplified band of 550 bp. We found that only the “sweeping method” plants amplified the 550 bp bands, while those of the “dipping method” did not. We suggest that the T-DNA was successfully integrated into the genome of plants treated with the “sweeping method” but not with the “dipping method”. Leaf sugar content (oBrix) of PCR-positive vines was higher than those of the wild-type vines, ensuring the integration of the T-DNA into the plant genome.

Abstract

Kaktus mini merupakan tanaman indoor penghias ruangan yang memiliki prospek baik untuk dikembangkan. Budidayanya yang sangat mudah tidak banyak diketahui orang sehingga perlu menyebarkan ilmu budidaya ini kepada masyarakat, khususnya kaum ibu rumah tangga. Ibu rumah tangga menghabiskan sebagian besar waktunya di rumah untuk mengurus keluarga, namun pemanfaatan waktu yang tersisa perlu dipikirkan dengan baik agar tidak terbuang percuma. Pelatihan budidaya kaktus mini untuk para ibu anggota PKK di Banjar Kerthabuwana Kaja ini telah dilaksanakan dengan baik. Kegiatan ini memberikan pengetahuan serta ketrampilan kepada para ibu sehingga mereka dapat melakukannya di rumah sebagai bagian dari hobi sekaligus mendatangkan uang. Metode yang digunakan adalah metode ceramah dan praktik langsung. Tujuan dari kegiatan ini adalah meningkatkan ketrampilan para ibu rumah tangga dalam budidaya tanaman kaktus mini. Output dari kegiatan ini adalah ketrampilan sederhana yang didapat oleh para ibu rumah tangga, yakni pembiakan kaktus mini hingga pemeliharaannya sampai siap dijual. Abstract

ABSTRACT The Effect of Type and Spray Frequency of Leri on Growth of Phalaenopsis sp. Orchid after Acclimatization This research purpose was to know about interaction between type and spray frequency of leri and also single impact of this treatment to Phalaenopsis sp. orchid growth after acclimatization. The location of research was in Kerobokan, Badung district in May–September 2011. Used randomized block design, orchid seedlings were treated by combination type of leri such as leri from Ciherang white rice, Ciherang brown rice, brown rice of red local type and spray frequency such as once in 2 days, once in 4 days, once in 6 days also once in 8 days  with 3 repetition. The result showed that brown rice of red local type and once in 4 days spray frequency each had the highest variable result on height accretion of plant (4,08 and 3,89 mm), length of roots (47,15 and 45,40 mm), fresh weight of upper plant organ (895 and 896,67 mg) and also dry weight total of plant (55 and 55,56 mg). Interaction between brown rice of red local type and spray frequency once in 4 days was the best combination to increase orchid seedling growth after acclimatization. Abstract

Identification and Characterization of Genetic Resources of Local Fruits in GianyarThe local fruits is every kinds of fruits are cultivated and developed by farmers in Bali, while the local fruit products is every types and derivative results which derived from fresh fruits or already processed. This study conducted with the aims to identify the types of genetic resources of local fruits in Gianyar, to compile the genetic resources of the local fruits based on the morphological characters and utilization, superior fruit, harvest seasons and the geographical map of the diversity distribution of the local fruits. This study was conducted in February-July 2015 throughout Sub-district in the Gianyar District, namely: Payangan, Tegallalang, Tampaksiring, Ubud, Blahbatuh, Gianyar and Sukawati. The study consists of three phases, namely; Secondary data collection, field surveys for the types of genetic resources and distribution, and identify of the morphological characters and agronomy, utilization, superior fruit, harvest seasons and the distribution map of the local fruits. The results of this study shown, was found 45 species and 41 sub species of fruits which are spread throughout Sub-districts, and the three of seeded commodities namely; Citrus, Mangoes and Papaya.

Abstract

The Effectiveness Test of Coconut Water and Tomato Extract on Modified Media for Growth of Black Orchid (C. Pandurata) In Vitro Plantlet growth in tissue culture is influenced by many factors, one of which is exogenous growth regulators applied to the media. The purpose of this study was to determine the effect of coconut water and tomato extract on modified media on the growth of C. pandurata plantlets. The research was carried out at the Ecophysiology Laboratory, Faculty of Agriculture, Udayana University, from January 2019 to April 2019. The study used a completely randomized design consisting of two factors, namely tomato extract and coconut water. The treatment consisted of 4 levels, 0 m/L, 100 ml/L, 150ml/L, 200ml/L, 16 treatment combinations were obtained with 3 replications. The results showed that the 100ml/l tomato extract treatment gave the highest increase in plantlet height, root length, number of roots, wet weight and dry weight. The 150ml/l tomato extract treatment gave the most increase in the number of leaves. Giving 150ml/l of coconut water gave the highest number of roots compared to other treatments. The interaction between of tomato extract and coconut water had no significant effect on all observed parameters.

Abstract

Chrysal Use to Extend Vaselife Cut Flowers (Rosa hybrida L.) RosesThe demand for cut roses increasing but the problems that often occur in old rose cut flowers is the vaselife of flowers very short. One way that can be done to prolong the vaselife of flowers adaah by giving a solution of the marinade. The main function of a marinade solution is to maintain the vaselife of cut flowers as long as possible. Chrysal. Chrysal a marinade solution containing sucrose, dextrose and citric acid that can extend the life of flowers (vaselife) Another important factor in maintaining the vaselife of flowers is the availability of water. Water will go faster if the wider cross-section, for example by cutting the plant stem oblique. Objective To know the concentration of the shredded material Chrysal optimized to produce a maximum vaselife of flowers. Knowing the difference in the vaselife of roses stalks in pieces tilt and horizontal concentration Chrysal know the interaction between the treatment of cuts rose to the vaselife of roses. This research was conducted at the Laboratory of Post Harvest Technology Faculty of Agriculture, University of Udayana. This study uses a completely randomized factorial design with two factors. Based on this research, interaction between concentration Chrysal (L) by cutting the flower stalk (A) does not affect significantly (p ? 0.05) of all observed variables. In the single factor cutting flower stalk treatment (A) significant (p> 0.05) to variable interest weight loss, dantotal solution is absorbed, whereas the concentration of Chrysal (L) a very significant effect on the flowers begin to bloom, longer vaselife, and the total solution is absorbed.
